The aim of this project was to analyse critically the potential use of prebiotics in aquaculture. Initially, the intestinal microflora of rainbow trout, raised in the aquarium facilities at CEFAS, Weymouth, was investigated using both conventional bacteriological (culture, BiOLOG) and molecular (Restriction Fragment Length Polymorphism (RFLP), 16S rRNA gene sequencing) techniques. Dominant colony types were identified as Aeromonas sobria, Carnobacterium piscicola and Clostridium gasigenes. A series of in vitro growth curve studies was carried out using a selection of intestinal bacteria and known fish pathogens, to determine the ability of intestinal bacteria to utilise the potential prebiotic candidates inulin, lactulose …

There is an urgent need to understand the effects of climatic variability on species' distributions. This thesis uses an integrative approach to identify and explain the changes in geographical distribution of two rocky intertidal, warm water species of trochid gastropod, Osilinus lineatus (da Costa) and Gibbula umbiiicalis (da Costa) that have occurred since the onset of rapid climate warming in the mid-1980s. The potential physiological mechanisms causing the range shifts are examined, and the effects on population dynamics quantified. Free-living nematodes are abundant in all marine habitats, highly diverse and can be important ecological indicators for monitoring anthropogenic impacts on the environment. Despite such attributes, nematode diagnostics has traditionally relied on detailed comparison of morphological characters which is often difficult and laborious, and as a result there is an increasing 'black hole' in faunal inventories where the biodiversity of groups such as nematodes is typically underestimated. The aim of this thesis was to investigate the effects of human disturbance on animal foraging behaviour using oystercatchers foraging on the rocky shore as a model system. The primary focus of this thesis was the balance between vigilance and foraging, and the variation in this balance with changes in environmental factors and the application of experimentally controlled human disturbance. On the structurally complex rocky shore foraging and vigilance are generally mutually exclusive behaviours and so an individual must trade-off energy acquisition with predator avoidance. Over the last 10-15 years there has been increasing concern within Europe as to the effects of ammonia emission and subsequent deposition to sensitive ecosystems, causing eutrophication and soil acidification. Transboundary transport of emissions has led to legislation at EC level with member states being given emission ceiling targets. Research has therefore aimed at quantifying national emissions, modelling emission processes and developing mitigation strategies. Agriculture accounts for >80% of total UK ammonia emission, therefore an accurate and robust model is required to estimate emissions from this sector. Bioluminescent imaging (BLI) is particularly well suited for imaging small animals, and it can be readily used by scientists who are already routinely using the luciferase gene as a reporter in cell-based assays. This strategy relies on ATP and an O_2 dependent photochemical reaction between luciferin and luciferase, resulting in the release of photons from only live cells. In BLI, the intensity of the light detected by the device is proportional to the intensity of light emitted and the relationship is quantitative. Radiotherapy treatment planning utilizing PET and CT is rapidly gaining acceptance in oncology. A limiting factor of the dual modality is the PET/CT alignment. A small error in PET/CT alignment may result in giving large doses of radiation to healthy tissues as a result of poor treatment planning. For this purpose, regular quality assurance testing of PET/CT must be performed. Separate QA procedures and phantoms have been developed for the two different modalities. In particular, many existing phantoms cannot be used for both modalities, which is a requirement for evaluating PET/CT alignment. Our goal is to evaluate several existing phantom …

It was designed to be an alternative to more difficult science classes. Students at our school are required to have three years of science to graduate. This class was added to our curriculum to be an alternative to students who probably will not be going to college and just need the science credits to graduate. Most of the students who take this class do not like science and they believe that they cannot "do" science.

The West Delaware school district is located in northeast Iowa, halfway between Waterloo and Dubuque along highway 20. The district has approximately 650 students in grades 9-12 and employs 48 teachers. Four of these teachers teach science full-time, and two teachers from other departments pick up extra courses. The 150+ 8th grade students represented in this study were taught by two different science teachers, who also teach 7th grade science. 8th grade science is taught in a 45 minute day for the entire year, while 9th and 10th graders are taught on a 90 minute block schedule, with each class …

This project has been completed to improve the quality of instruction of the biology students at Iowa City West High School. A curricular unit covering plants has been designed keeping in mind the National Science Education Standards, the curricular benchmarks established by the Iowa City Community School District and the current research on how people learn. Applicable research that supports the inquiry approach to teaching biology, an outline of the daily events that occur during the course of the curricular unit, and examples of the use of inquiry teaching styles are included throughout this work.

The theoretical base of Wilderness Therapy is discussed to provide conceptual framework and describe how the therapy could be launched into mainstream therapy. The techniques of Wilderness Therapy are explained for better understanding of how the therapy works. Eighty-six primiparous or multiparous sows and their pigs were used to evaluate the effects of changing dietary cation-anion difference (DCAD) in late gestation and in lactation on sow productivity. Twenty sows were used in Exp. 1, and the dietary treatments consisted of a positive control (PC) corn-soybean meal (C-SBM) diet with four levels of DCAD (140, 99, 75, and 45 mEq/kg). These DCAD’s were achieved by four levels of SoyChlor (SC) additive (0, 1.5, 2.5, and 3.5%). The influence of donor animal sex or age on in vitro bovine cell culture was evaluated to provide foundation information for the selection of donor tissue for nuclear transfer. Skin biopsies were taken from each of sixteen individuals including four bulls (B), four cows (C), four male calves (MC), four female calves (FC). At passage 2, cells from in vitro culture of cell lines were influenced not by gender but by age in the mean cell generation time (MGT). A new structure-activity relationship (SAR) approach to modeling was utilized to study mammary gland carcinogens. A set of chemicals tested for mammary tumorigenesis that have been analyzed in the Carcinogenic Potency Database (CPDB) were subjected to several computational analyses in an attempt to predict each chemical’s carcinogenic potential. A total of six learning sets (rat and mouse mammary gland carcinogen, CPDB rat and mouse, and female-specific rodent models) were developed and validated using a SAR modeling algorithm called categorical-SAR (cat-SAR). Development of protocols for storing desiccated cells at ambient temperatures offers tremendous economic and practical advantages over traditional storage procedures such as cryopreservation and freeze-drying. An integrated frame work of experimentation and modeling is adopted in the present work to develop procedures for storage of adult stem cells at ambient temperature. Contextualism is the epistemological thesis that holds context to significantly affect the truth value of claims such as “S knows that p.” A shift in context can lead to a shift in the standards by which we evaluate propositional knowledge claims, and thus a shift in the truth values of these claims: a statement “S knows that p” may be true when evaluated in one context while simultaneously false when evaluated in another context. This study reports on the demography and osteological profile of the Poole-Rose Ossuary. Excavated in 1990 under the direction of Heather McKillop and at the request of the Alderville First Nation, the Poole-Rose Ossuary is a Late Woodland burial site in southern Ontario, Canada. Lack of European artifacts in the burial suggests that this site predates European contact. The Poole-Rose Ossuary is radiocarbon dated to A.D. 1550 ± 50 years. The skeletal remains were commingled. This ossuary likely represents the mass re-burial known as the “Feast of the Dead” or the “Kettle.” For the most part, closely related individuals were …
