The proopiomelanocortin (POMC) gene is expressed in the pituitary and arcuate neurons of the hypothalamus. POMC arcuate neurons play a central role in the control of energy homeostasis, and rare loss of function mutations in POMC cause obesity. Moreover, POMC is the prime candidate gene within a highly significant quantitative trait locus on chromosome 2 associated with obesity traits in several human populations. Here, we identify two phylogenetically conserved neuronal POMC enhancers designated nPE1 (600 bp) and nPE2 (150 bp) located approximately 10 to 12 kb upstream of mammalian POMC transcriptional units. We show that mouse or human genomic regions …

Aerodynamics Of The Hovering Hummingbird, Douglas R. Warrick, Bret W. Tobalske, Donald R. Powers Jan 2005

Aerodynamics Of The Hovering Hummingbird, Douglas R. Warrick, Bret W. Tobalske, Donald R. Powers

Faculty Publications - Department of Biological & Molecular Science

Despite profound musculoskeletal differences, hummingbirds (Trochilidae) are widely thought to employ aerodynamic mechanisms similar to those used by insects. The kinematic symmetry of the hummingbird upstroke and downstroke has led to the assumption that these halves of the wingbeat cycle contribute equally to weight support during hovering, as exhibited by insects of similar size. This assumption has been applied, either explicitly or implicitly, in widely used aerodynamic models, and in a variety of empirical tests. Here we provide measurements of the wake of hovering rufous hummingbirds (Selasphorus rufus) obtained with digital particle image velocimetry that show force asymmetry: hummingbirds produce …

Operational sex ratio (OSR) has been traditionally thought of as a force imposing competition for mates rather than also a cue used to regulate the intrasexual competition individuals encounter. To assess whether eastern red-spotted newts, Notophthalmus viridescens, could appropriately compare OSRs, we quantified field responses to traps containing four males, a sexually receptive female, four males plus a female, or nothing as a control. Early in the breeding season, males from two populations chose competitive mating opportunities over no mating opportunity at all, but generally preferred less competitive mating prospects. Later in the breeding season, as the OSR of …

Probing The Regulatory Mechanisms Of Protein Tyrosine Kinases, Using C-Terminal Src Kinase (Csk) As A Model System, Xiaofeng Lin Jan 2005

Probing The Regulatory Mechanisms Of Protein Tyrosine Kinases, Using C-Terminal Src Kinase (Csk) As A Model System, Xiaofeng Lin

Open Access Dissertations

Protein tyrosine kinases (PTKs) are important signal transducers. Most PTKs have a catalytic domain, which carries out catalysis, and regulatory domains, which regulate their activity. Autophosphorylation, the Tyr phosphorylation in the activation loop, is also a key regulatory mechanism for most PTKs. In this study, I probed the regulatory mechanisms of PTKs using Csk as a model system. Csk has two regulatory domains, SH3 and SH2, and a catalytic domain. In this study, I probed how the regulatory domains modulate Csk activity. First, the regulatory domains were required for Csk full activity, yet not involved in the recognition of Src, …

Sex Change In Crepidula Fornicata: Influence Of Environmental Factors On Reproductive Success And The Timing Of Sex Change, Dina A. Proestou Jan 2005

Sex Change In Crepidula Fornicata: Influence Of Environmental Factors On Reproductive Success And The Timing Of Sex Change, Dina A. Proestou

Open Access Dissertations

Many organisms experience a sex change at some point in their life cycle. In order for this life history strategy to be adaptive, lifetime reproductive success must be maximized. The size advantage hypothesis predicts a universal size at sex change for all individuals; however, for most species, size at sex change varies both across and within populations, indicating that the reproductive success of the first sex is influenced by external factors. The objective of this research was to investigate the influence of environmental variables on the sex, reproductive success, and timing of sex change of the protandrous sequential hermaphrodite, Crepidula …

Iucn As Catalyst For A Law Of The Biosphere: Acting Globally And Locally, Nicholas A. Robinson

Elisabeth Haub School of Law Faculty Publications

Unique among international organizations, the International Union for the Conservation of Nature and Natural Resources (IUCN) operates at the global, regional, and national levels to build governmental capacity to protect the environment. With a membership of over 75 sovereign states and 800 nongovernmental organizations, IUCN functions as an intergovernmental organization at the transnational level while operationally embodying the maxim "think globally, act locally." IUCN acts as a consortium of environmental scientists and professionals, including environmental lawyers who have proposed and secured adoption of significant environmental treaties such as the Convention on the International Trade in Endangered Species (CITES) and their …

The Utility Of Non-Use Values In Natural Resource Damage Assessments, Jason J. Czarnezki Jan 2005

The Utility Of Non-Use Values In Natural Resource Damage Assessments, Jason J. Czarnezki

Elisabeth Haub School of Law Faculty Publications

Non-use values are frequently underestimated or ignored in natural resource damage assessments, despite the fact that there are significant social and economic benefits to assessing costs for lost non-use values. The regulations of the Department of the Interior, which bind some CERCLA trustees, create unusual barriers to the consideration of non-use values and are potentially vulnerable to a reasonableness challenge under Chevron v. NRDC. Trustees who are not bound by the DOI regulations should consider calculating and assessing non-use values because of the economic and social benefits of recognizing non-economic injury caused by the destruction or degradation of natural resources.

Mechanical Properties Of The Intervertebral Disc As An Estimator Of Postmortem Interval, Jennifer Noelle Jackson Jan 2005

Mechanical Properties Of The Intervertebral Disc As An Estimator Of Postmortem Interval, Jennifer Noelle Jackson

Electronic Theses and Dissertations

Currently, forensic scientists are only able to determine time since death (or postmortem interval) up to the first 60 hours. This is based largely on insect activity. Herein, it is proposed to use the degradation of the intervertebral disc (IVD) after death to determine a relationship between the mechanical properties of cadaveric tissue and time since death in order to extend the 60-hour window. To that end, 1 fresh human spine and 6 pig spines were each separated into sections (6 human and 48 pig), with each section having one intact disc. The sections were buried, unearthed, and cleaned, leaving …

Isolation And Enrichment Of Abundant Microsatellites From A Channel Catfish (Ictalurus Punctatus) Brain Cdna Library, Dan Nonneman, Geoffrey C. Waldbieser Jan 2005

Isolation And Enrichment Of Abundant Microsatellites From A Channel Catfish (Ictalurus Punctatus) Brain Cdna Library, Dan Nonneman, Geoffrey C. Waldbieser

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Efforts to construct a genetic linkage map of channel catfish have involved identification of random genomic microsatellite markers, as well as anchored Type I loci (expressed genes) from channel catfish. To identify Type I markers we constructed a directional cDNA library from brain tissue to obtain expressed catfish sequences that could be used for single nucleotide polymorphism (SNP) marker development. These cDNA sequences surprisingly contained a high proportion of microsatellites (about 14%) in noncoding regions of expressed sequence tags (ESTs), many of which were not associated with known sequences. To further identify cDNAs with microsatellites and reduce the number of …

Prevalence Of Pathogenic Yersinia Enterocolitica Strains In Pigs In The United States, Saumya Bhaduri, Irene V. Wesley, Eric J. Bush Jan 2005

Prevalence Of Pathogenic Yersinia Enterocolitica Strains In Pigs In The United States, Saumya Bhaduri, Irene V. Wesley, Eric J. Bush

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Yersinia enterocolitica is considered an important food-borne pathogen impacting the pork production and processing industry in the United States. Since this bacterium is a commensal of swine, the primary goal of this study was to determine the prevalence of pathogenic Y. enterocolitica in pigs in the United Sates using feces as the sample source. A total of 2,793 fecal samples were tested for its presence in swine. Fecal samples were collected from late finisher pigs from 77 production sites in the 15 eastern and midwestern pork-producing states over a period of 27 weeks (6 September 2000 to 20 March 2001). …

Risk And Ethics In Biological Control, Ernest S. Delfosse Jan 2005

Risk And Ethics In Biological Control, Ernest S. Delfosse

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

All introduced natural enemies present a degree of risk to nontarget species. Since most biological control programs use relatively host-specific natural enemies, the risk to nontarget species is generally very low, particularly from biological control of weeds, which uses extensively tested and validated host-specificity testing procedures to predict risk. However, many of the published comments about risks of biological control are superficial or misleading, often inappropriately lumping risk from all taxa of agents as “the risk of biological control,” and ignore the potential benefits, rather than dealing with species-by-species risk and benefits. Particularly confounding accurate predictions is the common mixing …
