Mortality from natural causes has been documented throughout the range of the cougar (Puma concolor), including California (Beier and Barrett 1993), Canada (Ross et al. 1995), Florida (Taylor et al. 2002), Idaho (Hornocker 1970), Nevada (Ashman et al. 1983), New Mexico (Logan and Sweanor 2001), and Utah (Gashwiler and Robinette 1957, Lindzey et al. 1988). Intraspecific killing by adult males is the most common cause of death in unhunted populations; adult males have killed kittens, subadults, and adults of both sexes (Beier and Barrett 1993, Maehr 1997, Logan and Sweanor 2001, Taylor et al. 2002). Copolymer 1 [Cop1, glatiramer acetate, Copaxone, poly(Y,E,A,K)n] is widely used in the treatment of relapsing/remitting multiple sclerosis in which it reduces the frequency of relapses by ≈30%. In the present study, copolymers with modified amino acid compositions (based on the binding motif of myelin basic protein 85–99 to HLA-DR2) have been developed with the aim of suppressing multiple sclerosis more effectively. The enhanced efficacy of these copolymers in experimental autoimmune encephalomyelitis (EAE) induced in SJL/J mice with proteolipid protein 139–151 was demonstrated by using three protocols: (i) simultaneous administration of autoantigen and copolymer (termed prevention), (ii) …

Matrix isolation infrared spectroscopy has been combined with theoretical calculations for the characterization of the 1:1 hydrogen-bonded complex between H2O and dimethyl methylphosphonate (DMMP). The symmetric O-H stretching mode was observed to shift 203 cm-1 to lower energy upon hydrogen bond formation, while a 32 cm-1 blue shift was noted for the H-O-H bending mode of the H2O subunit in the complex. These values compare extremely well with the (unscaled) shifts of -203 and +32 cm-1, respectively, that were calculated theoretically at the MP2/6-31+G** level. The ENergy and WATer BALance (ENWATBAL) model is a mechanistic, numerical model that simulates soil water and temperature profiles, evaporation from soil, and transpiration from crops, but it does not simulate the effects of a mulch layer. Surface vegetative mulches are becoming more common, especially in reduced -tillage systems, limiting the model’s applicability. Our objective was to modify ENWATBAL to enable physically based simulation of the effects of a dense mulch. The impacts that setback distances of 1/4, 3/8, 1/2, 3/4, and 1 mile would have on the land area available to the livestock industry for expansion in Cuming County, Neb. were estimated using a geographic information system and the current county zoning requirements. These setbacks seem to be typical of distances cited in many county zoning regulations. Setback distances greater than 3/8 of a mile appear to be very exclusive to expansion of the livestock industry in Cuming County. Reciprocal setbacks that apply to new housing construction do not appear to be restrictive. It is expected that other Nebraska counties …

This study was conducted to investigate the effects of increasing dietary protein intake on growth performance and serum insulin-like growth factor-I (IGF-I) concentration in growing-finishing gilts. Thirty-nine crossbred gilts with an initial body weight of 74.3 lb were used in a 28-day growth study. The gilts were randomly allocated to one of five dietary treatments. The diets were standard corn soybean meal diets, which were formulated to contain 10, 14, 18, 22, or 26% crude protein by changing the ratio of corn to soybean meal in the diet. Research was conducted to assess the effects of the type of zone heater and floor mat used in a wean-to-finish building on the thermal environment created for newly weaned pigs and resulting pig performance. Gas-fired brooder heaters were compared to electric heat lamps and farm-cut wood sheathing was compared to commercial [unheated] rubber floor mats. No consistent differences in air temperature near the heating zone were found between either of the treatments. However, black-globe temperatures in pens having gas-fired heaters and/or wood mats were consistently warmer than in their comparison pens. In this paper, we develop a finite-deformation model for cell membranes with a view toward characterizing the local mechanical response of membranes in atomic force microscope (AFM) experiments. The membrane is modeled as a 2-D fluid continuum endowed with bending resistance. The general theory is used to obtain equations that describe axisymmetric equilibrium states. The membrane is assumed to enclose a fluid medium, which transmits hydrostatic pressure to the membrane, and a point load is applied at the pole to simulate an AFM probe. Kcv, a 94-aa protein encoded by Paramecium bursaria chlorella virus 1, is the smallest known protein to form a functional potassium ion channel and basically corresponds to the ‘‘pore module’’ of potassium channels. Both viral replication and channel activity are inhibited by the ion channel blockers barium and amantadine but not by cesium. Genes encoding Kcv-like proteins were isolated from 40 additional chlorella viruses. Differences in 16 of the 94 amino acids were detected, producing six Kcv-like proteins with amino acid substitutions occurring in most of the functional domains of the protein (N terminus, transmembrane 1, pore helix, selectivity filter, …

Alien parasitic wasps, including accidental introductions and purposefully released biological control agents, have been implicated in the decline of native Hawaiian Lepidoptera. Understanding the potential impacts of alien wasps requires knowledge of ecological parameters that influence parasitism rates for species in their new environment. Sophora seed-feeding Cydia spp. (Lepidoptera: Tortricidae) were surveyed for larval parasitoids to determine how native and alien wasps are partitioned over an elevation gradient (2200–2800 m) on Hawaii Island, Hawaii. Parasitism rate of native Euderus metallicus (Eulophidae) increased with increased elevation, while parasitism rate by immigrant Calliephialtes grapholithae (Ichneumonidae) decreased. In situ characterization of colloidal particles under hydrous conditions is one of the key requirements for understanding their state of aggregation and impact on the transport of pollutants in aqueous environments. Scanning transmission X-ray microscopy (STXM) is one of the few techniques that can satisfy this need by providing element- and chemical-state-specific 2-D maps at a spatial resolution better than 50nmusing soft X-rays from synchrotron radiation wiggler or undulator sources tuned to the absorption edges of different elements. Quantifying patterns is a key element of landscape analysis. One aspect of this quantification of particular importance to landscape ecologists is the classification of continuous variables to produce categorical variables such as land-cover type or elevation stratum. Although landscape ecologists are fully aware of the importance of spatial resolution in ecological investigations, the potential importance of the resolution of classifications has received little attention. Here we demonstrate the effects of using two different land-cover classifications to predict avian species richness and the occurrences of six individual species across the conterminous United States. Feeding and maturation by the soybean looper, Pseudoplusia includens (Walker) (Lepidoptera: Noctuidae), were investigated in a 2-yr study on 'Davis' soybean, Glycine max (L.) Merr., grown alone and combined with the weed hemp sesbania, Sesbania exaltata (Raf.) Rybd. ex. A. W. Hill, the root-knot nematode, Meloidogyne incognita (Kofoid & White) Chitwood, and the charcoal rot fungus, Macrophomina phaseolina (Tassi) Goid. Of the three pests, hemp sesbania had the greatest effects on plant growth and insect feeding and maturation. When fed foliage from soybean stressed by hemp sesbania, soybean looper larvae remained longer in feeding stages, consumed more foliage, and showed …

The major heat shock protein, Hsp70, can protect against cell death by directly interfering with mitochondrial apoptosis pathways. However, Hsp70 also sensitizes cells to certain apoptotic stimuli like TNF. Little is known about how Hsp70 enhances apoptosis. We demonstrate here that Hsp70 promotes TNF killing by specifically binding the coiled-coil domain of IκB kinase γ (IKKγ) to inhibit IKK activity and consequently inhibit NF-κB-dependent antiapoptotic gene induction. An IKKγ mutant, which interacts with Hsp70, competitively inhibits the Hsp70–IKKγ interaction and relieves heat-mediated NF-κB suppression. Depletion of Hsp70 expression with RNA interference rescues TNF-mediated cell death. Although TNF may or may …
