Microglia Activation In Alzheimer's Disease, Kirk Phillip Townsend

The work detailed in this dissertation has an overarching theme of modulating microglia activation in both in vivo and in vitro models relevant to AD. The premise is that understanding microglia function in this context may lead to a better understanding of AD pathogenesis and thus to effective therapeutic interventions. In chapter 3, we employ a well-defined model of microglia activation whereby the intraperitoneal delivery of LPS results in CNS microglia activation and TNF-α production. Oocyte development in Xenopus laevis spans six morphologically distinct stages (stage I-VI), and is associated with a decrease in protein O-GlcNAc levels. As a first step in elucidating the role of O-GlcNAc in developing oocytes, initial efforts were focused on isolation and identification of fifteen modified proteins that decrease during oocyte development. Stage I oocytes due to their high amounts of these proteins, were used as starting material for purification. Multiple affinity and specific antibody based purification technique were initially used in an attempt to enrich the O-GlcNAc proteins. Due to the unique properties of the proteins …

Real-Time Registration And Display Of Confocal Microscope Imagery For Multiple-Band Analysis, Scott E. Budge, A.M. Mayampurath, J.C. Solinsky

Multiple images acquired in real-time from a confocal microscope in different illumination wavebands are used to construct a three-dimensional understanding of living cell signal propagation. Registration, fusion, and display of these images are performed in real-time using field programmable gate array (FPGA) technology to allow the biological scientist to observe time critical signaling processes as they occur. The registration and display system operates at a rate of up to 15 frames/s, and performs dark offset correction and noise reduction, followed by non-linear correction for lens warp and focal plane tilt, and finally correction for spatial mismatch between the different wavebands. …

Electrotransformation Of Clostridium Thermocellum, Michael V. Tyurin, Sunil G. Desai, Lee R. Lynd

Electrotransformation of several strains of Clostridium thermocellum was achieved using plasmid pIKm1 with selection based on resistance to erythromycin and lincomycin. A custom-built pulse generator was used to apply a square 10-ms pulse to an electrotransformation cuvette consisting of a modified centrifuge tube. Transformation was verified by recovery of the shuttle plasmid pIKm1 from presumptive transformants of C. thermocellum with subsequent PCR specific to the mls gene on the plasmid, as well as by retransformation of Escherichia coli. Optimization carried out with strain DSM 1313 increased transformation efficiencies from <1 to (2.2 􏰍 0.5) 􏰎 105 transformants per 􏰃g of plasmid DNA. Factors conducive to achieving high transformation efficiencies included optimized periods of incubation both before and after electric pulse application, chilling during cell collection and washing, subculture in the presence of isoniacin prior to electric pulse application, a custom-built cuvette embedded in an ice block during pulse application, use of a high (25-kV/cm) field strength, and induction of the mls gene before plating the cells on selective medium. The protocol and preferred conditions developed for strain DSM 1313 resulted in transformation efficiencies of (5.0 􏰍 1.8) 􏰎 104 transformants per 􏰃g of plasmid DNA for strain ATCC 27405 and 􏰁1 􏰎 103 transformants per 􏰃g of plasmid DNA for strains DSM 4150 and 7072. Cell viability under optimal conditions was 􏰁50% of that of controls not exposed to an electrical pulse. Dam methylation had a beneficial but modest (7-fold for strain ATCC 27405; 40-fold for strain DSM 1313) effect on transformation efficiency. The effect of isoniacin was also strain specific. The results reported here provide for the first time a gene transfer method functional in C. thermocellum that is suitable for molecular manipulations involving either the introduction of genes associated with foreign gene products or knockout of native genes.

2004 Freshwater Mussel Survey Of The Big Piney River And Roubidoux Creek, Pulaski County, Missouri, M. Christopher Barnhart, Jennifer Wigger, Matt Duzan

Native freshwater mussels are an ecologically important group of aquatic mollusks. This group is of particular concern to conservation biologists because many species are classified as species of conservation concern (SCC). The freshwater mussel community of the Big Piney River and Roubidoux Creek, on and in the vicinity of Fort Leonard Wood, was previously surveyed by Sternburg et al. (1998). The present project was a resurvey of the area to determine the current status of the mussel community, so that appropriate measures can be taken to preserve and protect that community.

Using lines of mice having undergone long-term selection for high and low growth, a large-sample (n ≈ 1000 F₂) experiment was conducted to gain further understanding of the genetic architecture of complex polygenic traits. Composite interval mapping on data from 10-week-old F2 females (n = 439) detected 15 quantitative trait loci (QTLs) on 5 chromosomes that influence reproduction traits characterized at day 16 of gestation. Stat 5 Activators Can Replace The Requirement Of Fbs In The Adipogenesis Of 3t3-L1 Cells, William C. Stewart, James E. Baugh, Z. Elizabeth Floyd, Jacqueline M. Stephens

The 3T3-L1 cells differentiate into fat cells that have many properties of native adipocytes including: substantial lipid accumulation, insulin sensitivity, and the ability to secrete endocrine hormones. A substantial expense in using these cells is fetal bovine serum (FBS), a critical component of efficient adipogenesis. Our recent studies on STAT 5 proteins have revealed that these transcription factors are phosphorylated and translocate to the nucleus immediately after the initiation of differentiation. Studies by several other laboratories also suggest that STAT 5 proteins can have pro-adipogenic properties. Growth hormone (GH) and prolactin (PRL) are both potent activators of STAT 5A and …
