The goal of these projects has been to build capacity among Alaska federally recognized tribes to address their concerns about adverse changes in the environment. The University of Alaska’s Institute of Social and Economic Research and the Alaska Native Science Commission collaborated on both projects. Since the projects are complementary, we have combined the two final reports. Over the past fifteen years, Alaska’s salmon industry has experienced dramatic losses in income, market share, permit and boat values, and tax revenues to communities and the state. The economic crisis in the salmon industry—driven by competition from farmed salmon and other factors—has prompted numerous task forces and summits to call for improved quality, new products, better marketing, and other measures to enable Alaska’s salmon industry to compete more effectively in world salmon markets. When the US purchased Alaska from the Russians in1867 it quickly stepped into Russia's role in the colonial relationship. The US exploited salmon as the primary base resource from about 1878. Gold was discovered in lode deposits in the 1880s in the southeast of Alaska (Juneau and Treadwell), about the same time as the Lena goldfields of Irkutsk began to step up operations. Denali National Park, Alaska substantially owes its stature as Alaska's premier wilderness park to Adolph Murie. Forty years after he retired as park biologist, Murie still influences the perception and management of Denali National Park. Murie's development from childhood to esteemed scientist and wilderness advocate followed a linear progression. His rural upbringing under the tutelage of his older brother, Olaus Murie, cultivated his desire to be a biologist and his appreciation for wild places. His academic training in animal ecology solidified his belief that the management of natural areas must consider all species as essential and equally valuable. His pioneering …

Life-History Patterns Of North American Elk: Effects Of Population Density On Resource Partitioning, Reproduction, And Plant Productivity, Kelley Merlet Stewart

I examined density dependence in North American elk (Cervus elaphus ) and effects of density dependent processes on resource partitioning, physical condition, reproduction, and ecosystem processes. Specifically, I examined spatial, temporal, and dietary niche partitioning among elk, mule deer (Odocoileus hemionus) and cattle (Bos taurus ). I tested hypotheses related to density-dependent processes in elk by creating populations at high (20.1 elk/km2) and low (4.1 elk/km2) density. I hypothesized that physical condition and fecundity of females would be lower in an area of high population density than in the low-density area. Quantifying Variability In The Alaskan Black Spruce Ecosystem: Linking Vegetation, Carbon, And Fire History, Teresa Nettleton Hollingsworth

The boreal forest is the largest terrestrial ecosystem in North America, one of the least disturbed by humans, and most disturbed by fire. This combination makes it an ideal system to explore the environmental controls over species composition, the relative importance of abiotic factors and floristic composition in governing ecosystem processes, and the importance of legacy effects at a large regional spatial scale. In the boreal region of interior Alaska, Picea mariana (black spruce) is the predominant tree species and spans a wide range of habitats, including north-facing slopes with permafrost, lowland bogs, and high dry ridge-tops. Screening For Differentially Expressed Genes: Are Multilevel Models Helpful?, Dongmei Liu, Giovanni Parmigiani, Brian Caffo

Screening for changes in gene expression across biological conditions using microarrays is now a common tool in biology. Efficient use of these data for identifying important biological hypotheses is inherently a statistical problem. In this paper we present a broad Bayesian multilevel framework for developing computationally fast shrinkage-based screening tools for this purpose. Our scheme makes it easy to adapt the choice of statistics to the goals of the analysis and to the genomic distributions of signal and noise. We empirically investigate the extent to which these shrinkage-based statistics improve performance, and the conditions under which such improvements takes place. …

Determining The Biosynthetic Sequence In The Early Steps Of The Fumonisin Pathway By Use Of Three Gene-Disruption Mutants Of Fusarium Verticillioides, Ravi S. Bojja, Ronald Cerny, Robert H. Proctor, Liangcheng Du

Fumonisins are polyketide-derived mycotoxins produced by Fusarium verticillioides, a fungal pathogen of corn plants. Although a gene cluster for the biosynthesis of fumonisins has been cloned, the biosynthetic pathway is still not clear. We have used three gene-disrupted mutants, designated ΔFUM1, ΔFUM6, and ΔFUM8, to study the early steps of the pathway. Fumonisins were not produced in single-strain cultures of the ΔFUM1, ΔFUM6, and ΔFUM8 mutants. However, fumonisins were produced by ΔFUM1 or ΔFUM8 mutants when they were cocultured with the ΔFUM6 mutant. No fumonisins were produced when the ΔFUM1 and ΔFUM8 mutants were cocultured. Background The pap smear test is highly effective at detecting pre-cancerous and cancerous cervical cells. However, only 68.3 percent of women in the U.S. received a Pap smear in 2001. Previous research studies have shown that overweight and obese women are less likely to receive routine pap smears. A Systemic Gene Silencing Method Suitable For High Throughput, Reverse Genetic Analyses Of Gene Function In Fern Gametophytes., George Rutherford, Milos Tanurdzic, Mitsuyasu Hasebe, Jo Ann Banks

Background

Ceratopteris richardii is a useful experimental system for studying gametophyte development and sexual reproduction in plants. However, few tools for cloning mutant genes or disrupting gene function exist for this species. The feasibility of systemic gene silencing as a reverse genetics tool was examined in this study.

Results

Several DNA constructs targeting a Ceratopteris protoporphyrin IX magnesium chelatase (CrChlI) gene that is required for chlorophyll biosynthesis were each introduced into young gametophytes by biolistic delivery. Their transient expression in individual cells resulted in a colorless cell phenotype that affected most cells of the mature gametophyte, including the …

Analysis Of Clinically Important Compounds Using Electrophoretic Separation Techniques Coupled To Time-Of-Flight Mass Spectrometry, Zlatuse Durda Peterson

Capillary electrophoretic (CE) separations were successfully coupled to time-of-flight mass spectrometric (TOFMS) detection for the analysis of three families of biological compounds that act as mediators and/or indicators of disease, namely, catecholamines (dopamine, epinephrine, norepinephrine) and their O-methoxylated metabolites (3-methoxytyramine, norepinephrine, and normetanephrine), indolamines (serotonin, tryptophan, and 5-hydroxytryptophan), and angiotensin peptides. While electrophoretic separation techniques provided high separation efficiency, mass spectrometric detection afforded specificity unsurpassed by other types of detectors.

Both catecholamines and indolamines are present in body fluids at concentrations that make it possible for them to be determined by capillary zone electrophoresis coupled to TOFMS without employing any …
