Characterization Of Electrospun Dlplga Nanofibers For A Drug Delivery System For Intracranial Tumors And Avms, Kimberly A. Griswold May 2004

Characterization Of Electrospun Dlplga Nanofibers For A Drug Delivery System For Intracranial Tumors And Avms, Kimberly A. Griswold

Theses

Electrospinning processes apply electric fields to a polymer solution in order to produce strands of polymer in the nanoscale range. These polymer fibers are manipulated for their porosity, high surface area, fineness and uniformity. In order to create the ideal drug delivery system for intracranial arteriovenous malformations (AVMs) and tumors, D, L poly-(lactice-poly-glycolide) (DLPLGA) nanofibers were electrospun in a tetrahydrofuran (THF) solvent. Three different concentration ratios of DLPLGA, 85/15, 80/20, and 75/25 were analyzed to obtain the prime base for drug annexation. 1 ,3-bis (2-chloroethyl)- 1 -nitrosourea (BCNU) was then codissolved with the DLPLGA nanofibers to form a homogenous solution, …

Analysis Of Auf1 Targeted Mrna Sequences, Jiebo Lu

Theses

AUF 1, an A+U rich element (ARE) binding protein, plays an important role in mRNA decay. To identify the mRNAs that interact with AUF 1, mRNA derived from a human cardiac cDNA expression library was purified by AUF 1 affinity chromatography and cloned following RT-PCR. 261 sequences were obtained. The sequences were searched against two protein databases and four nucleic acid databases, and the sequence information and database search results were input into a local Microsoft Access database, BLAST-AUF 1, by Java applets for parsing document. Analysis of protein information by querying BLAST-AUF 1 identified 194 function-known proteins, which were …

Design And Implementation Of Heart Rate Variability Measures In The Time Domain, Binoy John Mathews May 2004

Design And Implementation Of Heart Rate Variability Measures In The Time Domain, Binoy John Mathews

Theses

The following study was conducted to design and implement computer programs to derive five time domain and two frequency domain Heart Rate Variability (HRV) measures from 24 hour Holter monitor recordings and to generate activity plots that display the variation of three of the five time domain measures and two frequency domain measures over a period of 24 hours.

Working with known pre and post experimental ECG data taken from a cyclical exercise program, standard parameters in both the time and frequency domains were derived using programs designed in Labview 7.0 engineering software package. Five parameters in the time domain …

Review Of Algorithms For Rna Secondary Structure Prediction With Pseudoknots, Ingrid Helene Nielsen May 2004

Review Of Algorithms For Rna Secondary Structure Prediction With Pseudoknots, Ingrid Helene Nielsen

Theses

Pseudoknots are structures that are formed from the base pairing of an RNA secondary loop structure with a complementary base which lies somewhere outside of the loop. The result is a structure, which plays a vital role in cell structure rigidity, regulation of protein synthesis, and in the structural organization of RNA complexes. Deciphering RNA folding patterns would begin to unravel some of the mysteries surrounding the cell and its functions and open a new world to scientists. Many algorithms have been written in this quest to predict RNA's secondary structure but not many have been very successful.

In this …

Extrusion And Evaluation Of Degradation Rate And Porosity Of Small Diameter Collagen Tubes, Bipinkumar G. Patel May 2004

Extrusion And Evaluation Of Degradation Rate And Porosity Of Small Diameter Collagen Tubes, Bipinkumar G. Patel

Theses

The limited availability of autografts and failure of small diameter synthetic vascular grafts has stimulated continuing efforts to develop small diameter vascular grafts based on natural materials. The small diameter collagen tubes were extruded using bovine collagen type I. The biodegradation rate was determined and compared for small diameter collagen tubes crosslinked with glutaraldehyde and N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide hydrochloride (EDC) and Nhydroxysuccinimide (NHS) to evaluate physio-chemical properties. The samples were also evaluated for their surface area and porosity using mercury porosimeter. The noncrosslinked tubes almost completely degraded after 2 hours. The tubes crosslinked with EDC/NHS degraded slower than the ones crosslinked with …

Particle Size Dependence On Immobilized Molecular Sensors : Assay Of Nucleic Acid Probes Attached To Paramagnetic Particles, Celin Sunny May 2004

Particle Size Dependence On Immobilized Molecular Sensors : Assay Of Nucleic Acid Probes Attached To Paramagnetic Particles, Celin Sunny

Theses

Nanotechnology is concerned with materials and systems whose structure and components exhibit novel and significantly improved physical, chemical and biological properties, phenomena and processes due to their nano scale size.

The transition from micro scale to nano scale leads to a number of changes in physical properties, possibly including new physical principles, some of which may be yet to be discovered. One of the major factors in this is the increase in ratio of surface area to volume.

This thesis research uses the increase in ratio of surface area to volume property of going from macro to micro to nano …

Spinning Of Collagen Fibers And Characterizing Thermal, Mechanical, Tensile And Structural Properties, Grace Evangelin Tatagiri May 2004

Spinning Of Collagen Fibers And Characterizing Thermal, Mechanical, Tensile And Structural Properties, Grace Evangelin Tatagiri

Theses

Collagen is the most important building block in the entire animal world. It plays an important role in regeneration of broken bones and wound healing, also helps grow blood vessels to feed the healing areas. The use of synthetic materials has been extensive, but these materials have there own limits and drawbacks. Poor biocompatibility of these materials is the main issue and often result in inflammations. Hence collagen is being heralded as one of the most appropriate replacement as an implantable material. The reasons being that it is a naturally occurring material, exhibits high biocompatibility low antigenicity and host response. …

Mathematical Modeling Of Transient State Transdermal Drug Delivery, Alison Nickol Weltner May 2004

Mathematical Modeling Of Transient State Transdermal Drug Delivery, Alison Nickol Weltner

Theses

In this work, a two-pathway mathematical model for transdermal drug delivery with iontophoresis is presented. The partial differential equations are described and then solved. An alternative, two-pathway, three-layer model is also presented, and the implications of the coefficients within the equation are discussed. Using Franz cell iontophoretic delivery data from three drug substances (amitriptyline HCl, clomipramine HCl, and nortriptyline HCl), the two-pathway model is regressed to determine the diffusion coefficient and the concentration within the skin at the drug reservoir interface. ANOVA analysis indicates a correlation between iontophoretic current and concentration of drug within the stratum corneum.

Statistical Analyses And Reproducible Research, Robert Gentleman, Duncan Temple Lang May 2004

Statistical Analyses And Reproducible Research, Robert Gentleman, Duncan Temple Lang

Bioconductor Project Working Papers

For various reasons, it is important, if not essential, to integrate the computations and code used in data analyses, methodological descriptions, simulations, etc. with the documents that describe and rely on them. This integration allows readers to both verify and adapt the statements in the documents. Authors can easily reproduce them in the future, and they can present the document's contents in a different medium, e.g. with interactive controls. This paper describes a software framework for authoring and distributing these integrated, dynamic documents that contain text, code, data, and any auxiliary content needed to recreate the computations. The documents are …

A Model Based Background Adjustment For Oligonucleotide Expression Arrays, Zhijin Wu, Rafael A. Irizarry, Robert Gentleman, Francisco Martinez Murillo, Forrest Spencer May 2004

A Model Based Background Adjustment For Oligonucleotide Expression Arrays, Zhijin Wu, Rafael A. Irizarry, Robert Gentleman, Francisco Martinez Murillo, Forrest Spencer

Johns Hopkins University, Dept. of Biostatistics Working Papers

High density oligonucleotide expression arrays are widely used in many areas of biomedical research. Affymetrix GeneChip arrays are the most popular. In the Affymetrix system, a fair amount of further pre-processing and data reduction occurs following the image processing step. Statistical procedures developed by academic groups have been successful at improving the default algorithms provided by the Affymetrix system. In this paper we present a solution to one of the pre-processing steps, background adjustment, based on a formal statistical framework. Our solution greatly improves the performance of the technology in various practical applications.

Affymetrix GeneChip arrays use short oligonucleotides to …

State Coastal Observations And Monitoring Needs: Results Of A Survey To Assess Coastal Management Needs (Draft Report), Jeff Benoit, Chantal Lefebvre, Dan Hellin, Regan Maund, Jack Thigpen

Urban Harbors Institute Publications

The success of the U.S. Coastal Ocean Observing System will be measured, in part, by how well the needs of the coastal management community are being addressed. The results of this survey indicate that the two most important management issues facing coastal programs are land use and habitat change. It is essential that the planning and implementation of the USCOOS take this fact into account and place a priority on addressing these high priority management needs. This can only be accomplished through the direct long-term involvement of the coastal management community with USCOOS efforts at the national and regional levels. …

Potential Of Modis Ocean Bands For Estimating Co2 Flux From Terrestrial Vegetation: A Novel Approach, Abdullah Rahman, Vicente D. Cordova, John A. Gamon, Hans Peter Schmid, Daniel A. Sims May 2004

Potential Of Modis Ocean Bands For Estimating Co2 Flux From Terrestrial Vegetation: A Novel Approach, Abdullah Rahman, Vicente D. Cordova, John A. Gamon, Hans Peter Schmid, Daniel A. Sims

School of Earth, Environmental, & Marine Sciences Faculty Publications

A physiologically-driven spectral index using two ocean-color bands of MODIS satellite sensor showed great potential to track seasonally changing photosynthetic light use efficiency (LUE) and stress-induced reduction in net primary productivity (NPP) of terrestrial vegetation. Based on these findings, we developed a simple ‘‘continuous field’’ model solely based on remotely sensed spectral data that could explain 88% of variability in flux-tower based daily NPP. For the first time, such a procedure is successfully tested at landscape level using satellite imagery. These findings highlight the unexplored potential of narrow-band satellite sensors to improve estimates of spatial and temporal distribution in terrestrial …

Characterization Of Human Udp-Glucose Dehydrogenase: Cys-276 Is Required For The Second Of Two Successive Oxidations, Brandi J. Sommer, Joseph J. Barycki, Melanie A. Simpson May 2004

Characterization Of Human Udp-Glucose Dehydrogenase: Cys-276 Is Required For The Second Of Two Successive Oxidations, Brandi J. Sommer, Joseph J. Barycki, Melanie A. Simpson

Department of Biochemistry: Faculty Publications

UDP-glucose dehydrogenase (UGDH) catalyzes two oxidations of UDP-glucose to yield UDP-glucuronic acid. Pathological overproduction of extracellular matrix components may be linked to the availability of UDP-glucuronic acid; therefore UGDH is an intriguing therapeutic target. Specific inhibition of human UGDH requires detailed knowledge of its catalytic mechanism, which has not been characterized. In this report, we have cloned, expressed, and affinity-purified the human enzyme and determined its steady state kinetic parameters. The human enzyme is active as a hexamer with values for Km and Vmax that agree well with those reported for a bovine homolog. We used crystal coordinates …

Fall case studies of three‐dimensional circulation, plankton, and benthos models explored the consequences of interannual changes in ice cover and water motion on carbon/nitrogen cycling by the end of September within the Chukchi/Beaufort Seas. The coupled model scenarios were those of reduced (greater) northward flow, colder (warmer) temperatures, and more (less) extensive ice cover over the preceding ∼60 days of August and September during the negative (positive), anticyclonic (cyclonic) phase of the Arctic Oscillation in 1980 (1989). On the inner Chukchi shelf, stronger flows in 1989 advected nitrate and silicate stocks of Pacific origin ∼130 km farther northwest toward Wrangel …

Photosynthesis And Respiration In Five Species Of Benthic Foraminifera That Host Algal Symbionts, Robert A. Walker

USF Tampa Graduate Theses and Dissertations

Oxygen production and consumption were measured in five species of benthic foraminifers using a "Clark-type" oxygen electrode. Net photosynthesis and respiration were calculated and normalized to both μg Chl a and mm² upper surface area for the chlorophyte-bearing soritid foraminifers, Archaias angulatus and Cyclorbiculina compressa, and the diatom-bearing amphisteginids, Amphistegina gibbosa, A. lessonii and A. radiata. Photosynthesis/Irradiance curves were generated by fitting data to the hyperbolic tangent equation P = Pmax tanh (α I/ Pmax). Derived photosynthetic parameters, Pmax, α, Ik were found to correspond to the general responses of …

A Comparison Of Weekend And Weekday Travel Behavior Characteristics In Urban Areas, Ashish Agarwal May 2004

A Comparison Of Weekend And Weekday Travel Behavior Characteristics In Urban Areas, Ashish Agarwal

USF Tampa Graduate Theses and Dissertations

Travel demand analysis has traditionally focused on exploring and modeling travel behavior on weekdays. This emphasis on weekday travel behavior analysis was largely motivated by the presence of well-defined peak periods, primarily associated with the journey to and from work. Most travel demand models are based on weekday travel characteristics and purport to estimate traffic volumes for daily or peak weekday conditions. Much of the planning and policy making that occurs in transportation arena in response to weekday travel behavior and forecasts.

More recently, there had been a growing interest in exploring, understanding, and quantifying weekend travel characteristics. The ability …
