Engineering For Expression Of The Cold Regulated Barley Protein Hvcr21 In E. Coli, Kathleen Graupman
The Journal of Undergraduate Research
HVCR21 is a barley protein known to be cold regulated at the mRNA level. However, its function as well as the affect of low temperature on its translation are unknown. The purpose of this project was to engineer E. coli to express recombinant HVCR21. PGR primers were designed for the 5' and 3' ends of the HVCR21 coding region. The primers were also designed with a 5' SacI restriction site and a 3' Pst I restriction site. After ligation of the PCR product into the pCR4-TOPO vector, bacteria were transformed and plated and the successful transformant verified by PCR. The …

Influence Of Normal Daytime Fat Deposition On Laboratory Measurements Of Torpor Use In Territorial Versus Nonterritorial Hummingbirds, Donald R. Powers, A R. Brown, J A. Van Hook

Fat deposition and torpor use in hummingbirds exhibiting distinct foraging styles should vary. We predicted that dominant territorial hummingbirds will use torpor less than subordinate nonterritorial species because unrestricted access to energy by territory owners allows for fat storage. Entry into torpor was monitored using open-flow respirometry on hummingbirds allowed to accumulate fat normally during the day. Fat accumulation was measured by solvent fat extraction. Territorial blue-throated hummingbirds (Lampornis clemenciae) had the highest fat accumulation and used torpor only 17% of the time. Infectious bursal disease virus (IBDV) is a pathogen that primarily infects B lymphocytes in domestic avian species. This viral infection has been associated with immunosuppression, clinical disease/mortality, and enteric malabsorption effects. The purpose of this experiment was to compare the effects of a classic (USDA-STC) and a new variant IBDV (RB-4, known to induce primarily the enteric disease) on immune cell populations in lymphoid organs. Seventeen-day-old specific-pathogen-free (SPF) White Leghorn chickens were either not infected (control) or inoculated with either USDA-STC or RB-4 IBD viral isolate. The melanocortin 1-receptor (MC1-R) gene plays a key role in the expression of fur and feather color in mammals and birds by regulating the distribution of two melanin pigments: eumelanin (black/brown) and pheomelanin (red/yellow). MC1-R corresponds to the classical Extension (E) locus in mice, pigs, dogs, horses, and chickens. Three E locus alleles, the wild-type (e+), brown (E), and redhead (erh) have been identified in Japanese quail (Coturnix japonica). The objective of this research was to evaluate a potential time-saving method for surface lipid content (SLC) measurement of milled rice by utilizing new extraction technology. The SLC is often used as the basis for quantifying the degree to which bran has been removed from kernels during the rice milling process; this quality factor is often referred to as degree of milling (DOM). The SLCs of two long-grain cultivars of rice, ‘Cypress’ and ‘Cocodrie’, were determined using an accelerated solvent extraction system (ASE) and compared to the conventional, manual system (Soxtec extraction system) that is typically used for SLC measurement. …

Chitosanase is an enzyme, similar to chitinase, capable of hydrolyzing the β-1,4-linkages between N-acetyl-D-glucosamine and D-glucosamine residues in partially acetylated chitosan polymers found in fungal cell walls. When attacked by pathogenic fungi, many plants exploit this hydrolytic action as a component of a larger post-attack defense response, but these enzymes may also play a role in the initial plant-pathogen interaction via the generation of elicitors resulting from the hydrolysis of fungal cell walls. To gain insight into these mechanisms, a Paenbacillus chitosanase was cloned, sequenced, and modified for plant expression. The modified gene was delivered to tobacco (Nicotiana tabacum L. …

On The Thermohaline Circulation In Flat Bottom Marginal Seas, Michael A. Spall

The thermohaline circulation in an idealized marginal sea is studied using both numerical and analytical models. The circulation is forced by repeating an annual cycle of cooling in the interior of the marginal sea for 2 months and turning off the surface forcing for 10 months. The marginal sea is connected to an open ocean through a narrow strait, thus providing a means to equilibrate the cooling within the marginal sea by exchange with the open ocean. The resulting upper ocean circulation consists of a warm boundary current that flows into the marginal sea through the strait that is gradually …

The Large-Scale Time-Mean Ocean Circulation In The Nordic Seas And Arctic Ocean Estimated From Simplified Dynamics, Ole Anders Nøst, Pål Erik Isachsen

A simplified diagnostic model of the time-mean, large-scale ocean circulation in the Nordic Seas and Arctic Ocean is presented. Divergences in the surface Ekman layer are extracted from observed climatological wind stress fields. Similarly, divergences caused by the meridional thermal wind transport (relative to the bottom) are calculated from an observed climatological density field. These known quantities are then used to "force" the model's bottom geostrophic velocities. Both scaling arguments and direct observations show that for long time scales the bottom currents are closely aligned with contours of f/H, (where f is the Coriolis parameter and H is the …

In situations where the direct physical forcing of biological processes, through nutrient supply or light limitation of phytoplankton, does not show significant spatial variability, it is demonstrated that the effects of the mesoscale circulation and population dynamics on plankton patchiness are in principle, separable. In this case, the physical stirring and mixing control the size and shape of structures seen in plankton distributions, whilst the biological dynamics controls the size of the populations within those structures. Modes Of Internal Thermohaline Variability In A Single-Hemispheric Ocean Basin, Lianke A. Te Raa, Henk A. Dijkstra

In this study, the internal modes of variability of the thermohaline ocean circulation in an idealized single-hemispheric basin are investigated. The modes are determined systematically by performing linear stability analyses of fully three-dimensional thermohaline flows. Relevant for the large-scale low-frequency variability of the flows are two kinds of oscillatory modes. The modes in one of these classes, which have an interdecadal time scale, are truly three-dimensional phenomena and are characterized by westward propagating temperature anomalies, while modes in the other class have a centennial time scale. Cultural Context Matters: Terry's "Seesaw Effect", Frank Rudy Cooper

This Article investigates why the enforcement of a given legal doctrine may vary with changes in the cultural context in which it is applied. It argues that officials apply the law along an "enforcement practices continuum" in accord with changes in the prevailing articulations of the meaning of cultural identity norms associating particular groups with crime.
Terry v. Ohio doctrine allows police officers to make "stops" and "frisks" of limited scope upon reasonable suspicion of crime rather than requiring the higher standard of probable cause. The Article contends the officer discretion resulting from this "scope continuum" approach permits cultural identity …

A Model Of Fluff Layer Erosion And Subsequent Bed Erosion In The Presence Of The Bioturbator, Hydrobia Ulvae, Francis Orvain, Pierre Le Hir, Pierre-Guy Sauriau

Previous studies have shown that the gastropod Hydrobia ulvae destabilizes the top layers of fine-grained sediments. This process is mediated by the formation of a "biogenic" fluff layer that includes tracks, faecal pellets and mucus. This fluff layer has been shown to be easily resuspended before general bed erosion. In order to examine how fluff layer and bed erosion interact, flume experiments were performed with fluid sediments of varying water contents. Ten thousand snails were placed and allowed to crawl for 5 h on the sediment surface, and then the resuspended sediment mass was measured in response to step-wise shear …
