Purple loosestrife is an introduced invasive weed that is overrunning thousands of acres of wetlands and waterways in the Midwest. Once purple loosestrife invades a wetland, natural habitat is lost and the productivity of native plant and animal communities is severely reduced. Growers need to understand how the various pathogens causing plant disease respond to environmental conditions and cultural practices — what keeps the level of disease below an economic threshold and how these conditions can be developed and maintained for more sustainable production.

This NebGuide provides information on two common pathogens that cause seedling disease (damping off) in many crops. Although vegetable production is used in the examples, the principles apply to many cropping systems.

This NebGuide explains how to plant and raise nut trees and shrubs from seed. Seeding methods, planting design, site preparation and post-planting care are discussed.

Introduction

Conservation tree and shrub plantings create wildlife habitat, improve water quality, produce marketable nuts, and enhance landscape diversity. Usually, nursery-grown seedlings are used to establish these plantings. However, a number of problems exist when using seedlings, including increased costs and reduced initial tree vigor and survival. Direct seeding of tree and shrub seeds avoids many of these problems, and is a proven, effective means to establish conservation plantings in Nebraska and elsewhere.

A recombinant heavy chain fragment C of botulinum neurotoxin serotype F (BoNTF(Hc)) has been expressed in Pichia pastoris for use as an antigen in a proposed human vaccine. P. pastoris cells were grown using glycerol batch, glycerol fed-batch, and methanol fedbatch methods to achieve high cell densities. The total cellular protein recovered after homogenization was 72 mg/g of cell paste. BoNTF(Hc) was purified from soluble Pichia cell lysate employing ion-exchange chromatographic (IEC) and hydrophobic interaction chromatographic (HIC) methods developed at the bench scale using 10–100mL columns. Formin-homology-domain-containing proteins interact with Rho-family GTPases and regulate actin cytoskeleton organization and gene transcription. FHOD1 is a member of this family, interacts with Rac1 and induces transcription from the serum response element. In this study, we examined the effects of FHOD1 expression on cytoskeletal organization and function in mammalian cells. FHOD1 proteins were stably expressed in WM35 melanoma cells and NIH-3T3 fibroblasts. Cells expressing full-length FHOD1 demonstrated an elongated phenotype compared with vector-transfected cells and cells expressing a truncated FHOD1 (1-421) that lacks the conserved FH1 and FH2 domains. The effects of single meals of different sucrose concentrations on feeding responses and survival of 8–24-h-old, 1–2-, 10–12- and 31–36-day-old female and male western cherry fruit flies, Rhagoletis indifferens Curran, were determined. Feeding time and food consumption response patterns in both sexes within age groups were curvilinear. Feeding times increased as sucrose concentrations increased, and were longest when the sucrose concentration was 100% (dry). Consumption of dilute wet sucrose was low, whereas consumption of concentrated wet sucrose was high. However, consumption of dry, 100%sucrose was also low. Most research that has attempted to predict verdict preferences on the basis of stable juror characteristics, such as attitudes and personality traits, has found that individual differences among jurors are not very useful predictors, accounting for only a small proportion of the variance in verdict choices. Some commentators have therefore concluded that verdicts are overwhelmingly accounted for by "the weight of the evidence," and that differences among jurors have negligible effects. But there is a paradox here: In most cases the weight of the evidence is insufficient to produce firstballot unanimity in the jury (Hans & Vidmar, 1986; Hastie, Penrod, …

The past two decades have been a period of rapid evolution in the science of biotechnology and therefore in patent strategies, if not in patent law itself. Patent law takes a long time to catch up with science, and commentators take a long time to catch up with the law, but patent lawyers don’t have that luxury. They have to keep ahead of the game, figuring out claiming strategies that allow their clients to capture the value of future discoveries. I want to discuss some of these strategies today.

Flanking Sequences Modulate Diepoxide Cross-Linking Efficiencies At The 5'-Gnc Sste, Gregory A. Sawyer

Diepoxybutane, diepoxyoctane, and mechlorethamine are cytotoxic DNA crosslinking agents that vary in their carcinogenic versus chemotherapeutic potentials. Interstrand cross-linking occurs between the N7 positions of deoxyguanosine residues on opposite strands of the DNA duplex. Each synthetic DNA oligomer used in this study contained four 5'-N\GN2CN3 sites (within a 32 base sequence) and we have systematically varied the bases in the N1 and N2 positions to determine the resulting cross-linking efficiencies of each cytotoxic agent. Each duplex was 5' -end labeled and incubated with cross-linking agent. Interstrand cross-links were purified through denaturing polyacrylamide gel electrophoresis and then subjected to piperidine cleavage. …
