RNA silencing in plants likely exists as a defense mechanism against molecular parasites such as RNA viruses, retrotransposons, and transgenes. As a result, many plant viruses have adapted mechanisms to evade and suppress gene silencing. Tombusviruses express a 19 kDa protein (p19), which has been shown to suppress RNA silencing in vivo and bind silencing-generated and synthetic small interferingRNAs (siRNAs) in vitro. Most birds are uricotelic. An exception to this rule may be nectar-feeding birds, which excrete significant amounts of ammonia under certain conditions. Although ammonia is toxic, because it is highly water soluble its excretion may be facilitated in animals that ingest and excrete large amounts of water. Birdpollinated plants secrete carbohydrate- and water-rich floral nectars that contain exceedingly little protein. Thus, nectarfeeding birds are faced with the dual challenge of meeting nitrogen requirements while disposing of large amounts of water. Structures of the DNA sequences d(CCGGCGCCGG) and d(CCAGTACbr5UGG) are presented here as four-way Holliday junctions in their compact stacked-X forms, with antiparallel alignment of the DNA strands. Thus, the ACC-trinucleotide motif, previously identified as important for stabilizing the junction, is now extended to PuCPy, where Pu is either an adenine or guanine, and Py is either a cytosine, 5-methylcytosine, or 5-bromouracil but not thymine nucleotide. We see that both sequence and base substituents affect the geometry of the junction in terms of the interduplex angle as well as the previously defined conformational variables, Jroll (the rotation of the stacked duplexes …

It is now established that the hypothalamus is essential in coordinating endocrine, autonomic, and behavioral responses to changes in energy availability. However, the interaction of key peptides, neuropeptides, and neurotransmitters systems within the hypothalamus has yet to be delineated. Recently, we investigated the mechanisms through which serotonergic (5-hydroxytryptamine, 5-HT) systems recruit leptin-responsive hypothalamic pathways, such as the melanocortin systems, to affect energy balance. Through a combination of functional neuroanatomy, feeding, and electrophysiology studies in rodents, we found that 5-HT drugs require functional melanocortin pathways to exert their effects on food intake. Hypothalamic proopiomelanocortin (POMC) neurons are critical for controlling homeostatic functions in the mammal. We used a transgenic mouse model in which the POMC neurons were labeled with enhanced green fluorescent protein to perform visualized, whole-cell patch recordings from prepubertal female hypothalamic slices. The mouse POMC-enhanced green fluorescent protein neurons expressed the same endogenous conductances (a transient outward K current and a hyperpolarization-activated, cation current) that have been described for guinea pig POMC neurons. In addition, the selective -opioid receptor agonist DAMGO induced an outward current (maximum of 12.8 1.2 pA), which reversed at K equilibrium potential (EK), in the majority …

C75, a recently derived compound that potently suppresses feeding and induces weight loss, has been proposed to act mainly by inhibiting fatty acid synthase (FAS) in central neurons that control feeding. For example, normal, fasting- associated, hypothalamic increases in neuropeptide Y (NPY)/Agouti-related protein (AGRP) expression and decreases in proopiomelanocortin (POMC)/cocaine and amphetamine regulated transcript (CART) expression were reported to be blocked by C75. Intracellular Dynamics Of Lead And Calcium In Lead-Exposed Cultured Neurons Of Aplysia Californica (Mollusca: Opisthobranchia), Catherine T. Tamse

The neurotoxin lead (Pb2+) is a continuing health and environmental problem. However, its neurotoxic mechanism remains unclear. Since calcium (Ca2+) is vital in cellular processes and with Pb2+ reported to interfere with Ca2+-mediated processes, the hypothesis that Pb2+ subverts Ca2+ homeostasis by entering cells through Ca2+ transport pathways and displacing intracellular Ca2+ ([Ca2+] i) was investigated. A multi-technique approach tested this hypothesis. Using cultured Aplysia bag cell neurons, changes in transmembrane Ca2+ flux after Pb2+ exposure were directly measured with the Ca2+-selective self-referencing electrode. Cumulative concentrations of extracellular Pb2+ produced a corresponding and significant (P < 0.05) increase in Ca 2+ efflux, independent of time exposure. Ca2+ channel blockers Co2+, La3+, and verapamil inhibited Pb2+-induced flux. The sarco/endoplasmic reticulum Ca 2+-ATPase pump inhibitors, thapsigargin and cyclopiazonic acid, both elicited a Ca2+ efflux reflecting inhibition of ER pumps and [Ca2+]i release. Furthermore, Pb2+ evoked a moderate and sustained elevation in thapsigargin-stimulated Ca 2+ efflux. For morphological evidence that Pb2+ can mobilize [Ca 2+]i, potassium pyroantimonate (KPA) cytochemistry of bag cells were analyzed. 2% KPA resulted in widespread electron-dense precipitates. 10 mM EGTA + KPA or 200 μM Pb2+ + KPA both produced marked reductions of precipitates in neurons. X-ray microanalysis confirmed antimony (Sb) in precipitates but could not separate the overlapping Ca2+-Sb spectral peaks. However, reduced precipitates in EGTA-incubated cells indicate presence of Ca2+ in intracellular precipitates observed. No apparent pathological changes were correlated to Pb2+ exposure. The effects of Pb2+ on [Ca2+]i using the fluorescent Ca2+ indicator, chlortetracycline (CTC), was examined. Control neurons incubated in 10 μM CTC demonstrated extensive yellow-green fluorescence while reduced fluorescence intensities were seen in cells exposed to 200 μM Pb2+. Multi-sized punctate fluorescence were observed along the perimeter and sometimes, throughout the soma. No neuronal autofluorescence nor neuritic fluorescence in control and Pb2+-exposed cells were detected. Average fluorescence intensities between control and Pb2+-exposed neurons were statistically significant (P < 0.0005). Collectively, these results are consistent with the hypothesis that Pb 2+ perturbs Ca2+ homeostasis via mobilization of [Ca 2+]i. Pb2+ operates through Ca2+ transport pathways as shown by changes in transmembrane Ca 2+ flux, in KPA precipitate reactions, and in CTC fluorescence.

Nitrate (NO3−) assimilation in perennial grasses occurs predominantly in leaves. A growing body of evidence indicates that NO3− transport to and assimilation in leaves reduces the partitioning of photosynthetic products to roots. Since roots are dependent on leaf derived carbohydrates to support metabolism and growth, concentrating NO3− assimilation in roots may increase their sink activity and growth. The rate-limiting step in the assimilation of NO3− is its reduction to nitrite (NO2−) catalyzed by nitrate reductase (NR). The lily leaf beetle, Lilioceris lilii (Scopoli), first officially reported in the U.S. in 1992, has become a serious pest of native and cultivated lilies throughout its North American distribution. With no effective North American natural enemies, this exotic insect poses a potential economic threat to horticulturists and an environmental threat to native lilies. This study presents the results of an ongoing classical biological control program initiated against L. lilii in North America. Habitat enhancement and stock enhancement methods were examined as to their effect on abundance of the lobster, Homarus americanus, in Dutch Harbor in Narragansett Bay, Rhode Island. Six experimental artificial reefs for lobster were established in Dutch Harbor in February 1997. The reefs were constructed identically consisting of two portions made up of cobble and boulders. The area was monitored for 6 months before placement and five years after placement using a combination of visual surveys by SCUBA divers, trap sampling, a tag-recapture program and airlift sampling for young of the year. The article first summarizes CWA requirements relevant to TMDLs and outlines elements of an effective trading program. It then examines the program recently established by the State of Connecticut to allow trading of nitrogen credits among sewage treatment plants on Long Island Sound to achieve an established TMDL, and the CWA issues presented. Finally, it gives a brief comparison to the program being designed for the Chesapeake Bay, for which no TMDL has been established. Current brief descriptive summaries of several often cited programs are appended.

This dissertation studied cognitive packet networks (CPN) which build networked learning systems that support adaptive, quality of service-driven routing of packets in wired networks and in wireless, mobile ad hoc networks.

The first part of this study completed the design of the CPN algorithm for wired systems by revisiting the decision making procedure employed by smart packets in the former initiative. To conduct evaluation studies, this work proposed two implementation architectures, both of which were integrated into the Linus 2.4 kernel.

The second part of this dissertation extended CPN to ad hoc networks by introducing mechanisms to handle node mobility …

Seizures cause selective neuronal cell loss in vulnerable regions such as the hippocampus, cortex, thalamus or substantia nigra (SN) in human or experimental animals. While the HF can propagate seizures, the SN pars reticulata (SNR) regulates the spread ol seizures in an age-dependent fashion, and the pars compacta (SNC) affects extrapyramidal motor control. In mature animals, both structures are wlnerable to seizure-induced damage. In prepubescent rats, seizure susceptibility is reduced and CA 1 region rs primarily affected. Computer mediated multimedia learning is playing an increasingly important role in education. This study seeks to create and implement an interactive multimedia tutorial on the process of communication. The tutorial will be based on the traditional basic model of communication in used entry level undergraduate speech communication courses. The study examines the evolutionary development of the basic model of communication as well as models currently in in use popular textbooks. The study also examines educational theory, visual communication and multimedia learning as they pertain to the creation of the communication tutorial.
