The delineation of dopamine dysfunction in the mentally ill has been a long-standing quest of biological psychiatry. The present study focuses on a recently recognized group of dopamine receptor- interacting proteins as possible novel sites of dysfunction in schizophrenic and bipolar patients. We demonstrate that the dorsolateral prefrontal cortex in schizophrenia and bipolar cases from the Stanley Foundation Neuropathology Consortium display significantly elevated levels of the D2 dopamine receptor desensitization regulatory protein, neuronal calcium sensor-1. These levels of neuronal calcium sensor-1 were not influenced by age, gender, hemisphere, cause of death, postmortem period, alcohol consumption, or antipsychotic and mood stabilizing …

Regulation of ciliary and flagellar motility requires spatial control of dynein-driven microtubule sliding. However, the mechanism for regulating the location and symmetry of dynein activity is not understood. One hypothesis is that the asymmetrically organized central apparatus, through interactions with the radial spokes, transmits a signal to regulate dynein-driven microtubule sliding between subsets of doublet microtubules. Based on this model, we hypothesized that the orientation of the central apparatus defines positions of active microtubule sliding required to control bending in the axoneme. Lake Vida, one of the largest lakes in the McMurdo Dry Valleys of Antarctica, was previously believed to be shallow (<10 >m) and frozen to its bed year-round. New ice-core analysis and temperature data show that beneath 19 m of ice is a water column composed of a NaCl brine with a salinity seven times that of seawater that remains liquid below - 10°C. The ice cover thickens at both its base and surface, sealing concentrated brine beneath. The ice cover is stabilized by a negative feedback between ice growth and the freezing-point depression of the brine. The ice cover …

Punishment Theory, Holism, And The Procedural Conception Of Restorative Justice, Erik Luna

This Article has argued that the ongoing battle among punishment theories and the resulting consequences for real-life practice pose neither an inevitable nor preferable position on the criminal sanction. Instead, various theories can be accommodated using holism and holistic reasoning through a process-based methodology that avoids the prevailing problems of linear reasoning. In particular, the procedural conception of restorative justice offers an important component of a more holistic approach to sanctioning, allowing all interested parties to air their positions on punishment, both in theory and with regard to a particular offense. Recognition of a protein's fold provides valuable information about its function. While many sequence-based homology prediction methods exist, an important challenge remains: two highly dissimilar sequences can have similar folds --- how can we detect this rapidly, in the context of structural genomics? High-throughput NMR experiments, coupled with novel algorithms for data analysis, can address this challenge. We report an automated procedure for detecting 3D-structural homologies from sparse, unassigned protein NMR data. Our method identifies the 3D-structural models in a protein structural database whose geometries best fit the unassigned experimental NMR data. It does not use sequence information and is …

Richard Goldschmidt is remembered today as one of the most controversial biologists of the twentieth century. Although his work on sex determination and physiological genetics earned him accolades from his peers, his rejection of the classical gene and his unpopular theories about evolution significantly damaged his scientific reputation. This article reviews Goldschmidt's life and work, with an emphasis on his controversial views.

Tissue-Specific Structure/Function Differentiation Of The Liver Isoform Of 6-Phosphofructo-2-Kinase/Fructose-2,6-Bisphosphatase, Yong Hwan Lee, Yang Li, Kosaku Uyeda, Charles A. Hasemann

The crystal structures of the human liver 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase in three different liganding states were determined and compared with those of the rat testis isozyme. A set of amino acid sequence heterogeneity from the two distinct genes encoding the two different tissue isozymes leads to both global and local conformational differences that may cause the differences in catalytic properties of the two isozymes. The sequence differences in a β-hairpin loop in the kinase domain causes a translational shift of several hydrophobic interactions in the dimeric contact region, and its propagation to the domains interface results in a 5° twist of the …

Changes in scleral extracellular matrix, which is composed of proteins including glycoproteins, proteoglycans, and collagen, allow axial elongation of the eye resulting in myopia. Theoretically, changes in any scleral component could result in changes leading to increased axial length. The present study was designed to assess the validity of the White Racial Identity Attitude Scale (WRIAS: Helms & Carter, 1990) and Modem Racism Scale (MRS: McConahay, Hardee & Batts, 1981) in the prediction of discriminatory behavior. These measures were hypothesized to predict prejudiced behavior associated with simulated criminal sentencing as reflected in the Criminal Justice Questionnaire (CJQ) developed for purposes of the present study. The CJQ was administered on two occasions separated by a one month interval. While electronic structure methods that are able to describe accurately excited states exist (e.g., MRCI), they are often prohibitively expensive for all but the smallest systems (i.e., up to three atom systems with a moderate-sized basis set). This is because the inclusion of dynamic electron correlation, which is necessary for obtaining accurate energies and often required for obtaining reliable geometries, is computationally demanding. Since reactions typically involve more than three atoms, techniques able to describe excited states and include dynamic correlation in a much more computationally reasonable fashion than MRCI are highly desirable. The COL9A1 gene contains two promoter regions, one driving expression of a long alpha1(IX) chain in cartilage (upstream) and one driving expression of a shorter chain in the cornea and vitreous (downstream). To determine how the chondrocyte-specific expression of the COL9A1 gene is regulated, we have begun to characterize the upstream chondrocyte-specific promoter region of the human COL9A1 gene. Transient-transfection analyses performed in rat chondrosarcoma (RCS) cells, human chondrosarcoma (HTB) cells, and NIH/3T3 cells showed that the COL9A1 promoter was active in RCS cells but not HTB or NIH/3T3 cells. The purpose of this study was to investigate the relationships between student success in college-level introductory physics courses and various educational and background characteristics. The primary variables of this study were gender, high school mathematics and science preparation, preference and perceptions of learning physics, and performance in introductory physics courses. The purpose of this study was to identify those factors that affect the social integration of Hispanic adolescents in a predominately white public school. The sample includes seventh through twelfth grade students (N = 954) in two school districts located in small size Midwest cities. Subjects completed a questionnaire about themselves, their family and their educational experiences. Survey items specifically addressed the student’s sense of belonging to the school environment, their perceptions o f peers and school staff, their social involvement in school-sponsored activities, substance use, and experiences of being bullied. The location and function of the electron transport chain render the mitochondria the primary source for potentially damaging reactive oxygen species in aerobic cells. Normally, the concentration of superoxide anion in mitochondria is controlled, at least in part, by the sequential action of superoxide dismutase (SOD) and catalase. However, when the terminal steps of the electron transport are impaired or inhibited, superoxide anion production may rise to levels that overwhelm the normal protective enzymes. Studies have shown that copper deficiency causes a reduction in the activity of cytochrome-c oxidase, the copper dependent, terminal respiratory complex of the electron transport chain. …

This invention relates to the identification and characterization of the maize HSP22 protein including the corresponding nucleic acid molecules, nucleic acid fragments and protein fragments. Further, this invention relates to both polyclonal and monoclonal antibodies to HSP22 and the production thereof. These antibodies can be used in a method for detecting the presence of HSP22 expression in whole leaf samples. Further, the nucleic acid molecule and fragments thereof can be used in assays to detect the levels of nucleic acid molecules involved in HSP22 expression. Five hundred thirty-four steers were evaluated over a 2-yr period to develop and validate prediction equations for estimating carcass composition from live animal ultrasound measurements and to compare these equations with those developed from carcass measurements. Within 5 d before slaughter, steers were ultrasonically measured for 12th-rib fat thickness (UFAT), longissimus area (ULMA), rump fat thickness (URPFAT), and body wall thickness (UBDWALL). Carcasses were fabricated to determine weight (KGRPRD) and percentage (PRPRD) of boneless, totally trimmed retail product. Data from steers born in Year 1 (n = 282) were used to develop prediction equations using stepwise regression. Final models using …

Data from 534 steers representing six sire breed groups were used to develop live animal ultrasound prediction equations for weight and percentage of retail product. Steers were ultrasonically measured for 12th-rib fat thickness (UFAT), rump fat thickness (URPFAT), longissimus muscle area (ULMA), and body wall thickness (UBDWALL) within 5 d before slaughter. Carcass measurements included in USDA yield grade (YG) and quality grade calculations were obtained. Carcasses were fabricated into boneless, totally trimmed retail products. Five hundred thirty-four steers were evaluated over a 2-yr period to determine the accuracy of ultrasonic estimates of carcass 12th-rib fat thickness (CFAT) and longissimus muscle area (CLMA). Within 5 d before slaughter, steers were ultrasonically measured for 12th-rib fat thickness (UFAT) and longissimus muscle area (ULMA) using an Aloka 500V real-time ultrasound machine equipped with a 17.2-cm, 3.5-MHz linear transducer. Overall, correlation coefficients between ultrasound and carcass fat and longissimus muscle area were 0.89 and 0.86, respectively. Correlations for UFAT with CFAT were similar between years (0.86 and 0.90), whereas the relationship between ULMA and CLMA was stronger in …

Paramecium bursaria chlorella virus (PBCV-1) is the prototype of a family of large, icosahedral, plaque-forming, dsDNA viruses that replicate in certain unicellular, eukaryotic chlorella-like green algae. Its 330-kb genome contains ~373 protein-encoding genes and 11 tRNA genes. The predicted gene products of ~50% of these genes resemble proteins of known function, including many that are unexpected for a virus, e.g., ornithine decarboxylase, hyaluronan synthase, GDP-D-mannose 4,6 dehydratase, and a potassium ion channel protein. Phalangogonia debilidens Ohaus is placed in synonymy with P. sperata. This report analyzes the decision to limit the cleanup of the Hanford 300 Area to “Industrial Use” only. This decision raises legitimate concerns that providing such a minimal cleanup may be insufficient for protecting the Columbia River ecosystem and natural environment surrounding the Hanford site throughout the extraordinarily long life span of the hazardous waste. This report also provides a background on how the cleanup decision was made, the legal drivers, and an analysis of the risk assessment processes used in reaching the decision.
