Reconstructing Cetacean Brain Evolution Using Computed Tomography, Lori Marino, Mark D. Uhen, Nicholas D. Pyenson, Bruno Frohlich

Until recently, there have been relatively few studies of brain mass and morphology in fossil cetaceans (dolphins, whales, and porpoises) because of difficulty accessing the matrix that fills the endocranial cavity of fossil cetacean skulls. As a result, our knowledge about cetacean brain evolution has been quite limited. By applying the noninvasive technique of computed tomography (CT) to visualize, measure, and reconstruct the endocranial morphology of fossil cetacean skulls, we can gain vastly more information at an unprecedented rate about cetacean brain evolution. Here, we discuss our method and demonstrate it with several examples from our fossil cetacean database. This …

A Pcr Assay For Specific Detection Of The Pandemic Vibrio Parahaemolyticus O3:K6 Clone From Shellfish, P.S. Marie Yeung, A. Depaola, C. A. Kaysner, K. J. Boor

The current standard method for identifying Vibrio parahaemolyticus serotype O3:K6, an emerging pathogen with apparent enhanced virulence characteristics, typically takes 4 to 6 d to complete and requires serotyping. To provide a more rapid strategy, we optimized a polymerase chain reaction (PCR)-based assay for specific detection of V. parahaemolyticus O3:K6. Of 78 V. parahaemolyticus isolates and other related species; only strains classified into the V. parahaemolyticus O3:K6 clonal group (n = 39) showed positive results in the PCR assay. The assay detected 2.3 cells/PCR reaction and 310 cells/g using bacterial cultures and inoculated oyster samples, respectively. Sensitive and specific detection …

The Role Of Pleckstrin Homology Domains In Phosphoinositide Binding, Evan D. Christensen

Phosphoinositides are membrane-bound lipids that exist as eight different isoforms resulting from the phosphorylation of their inositol head group at three distinct positions. Proteins with Pleckstrin Homology (PH) domains bind to phosphoinositides involved in the regulation of cellular processes. The binding of several yeast PH domains to phosphoinositides was examined using dot-blot overlay assays, Surface Plasmon Resonance, and in vivo localization methodologies. While most of these yeast PH domains exhibited low specificity and/or low binding affinity, several proteins showed strong in vivo localization to either the plasma membrane or the Golgi membrane in mammalian cells. Ca2+ entry through voltage-gated Ca2+ channels contributes to the regulation of neuronal membrane excitability, neurosecretion, and gene expression. The nervous system regulates these physiological processes through G protein-coupled receptors (GPCRs), which strongly modulate Ca2+ channel activity. This dissertation provides significant new information concerning modulation of neuronal L-, N,- and R-type Ca2+ channels by muscarinic acetylcholine receptors. The major findings of this work are: 1) CaV2.3 channels are differentially modulated by Gαq-coupled muscarinic receptors; 2) reconstituted slow muscarinic inhibition of CaV1.2c can be triggered by either M1, M3, or M5 muscarinic receptors; 3) …

The plant mitochondrial genome is retained in a multipartite structure that arises by a process of repeat-mediated homologous recombination. Low-frequency ectopic recombination also occurs, often producing sequence chimeras, aberrant ORFs, and novel subgenomic DNA molecules. This genomic plasticity may distinguish the plant mitochondrion from mammalian and fungal types. In plants, relative copy number of recombination-derived subgenomic DNA molecules within mitochondria is controlled by nuclear genes, and a genomic shifting process can result in their differential copy number suppression to nearly undetectable levels. Platelet Endothelial Cell Adhesion Molecule-1 (PECAM-1), an immunoglobulin family adhesion molecule, is involved in endothelial migration and angiogenesis. We found that PECAM-1 deficient endothelial cells exhibit enhanced nondirected single cell motility and extension formation at the expense of wound-healing migration. These in vitro endothelial behaviors model aspects of highly developmentally and pathophysiologically relevant processes. Wound healing migration may model the reendothelialization of denuded vasculature following balloon angioplasty or the remodeling of vascular endothelium under variant flow conditions, while single cell motility is prominently involved in developmental processes such as endocardial-mesenchymal transition within the cardiac cushion. It therefore becomes necessary to raise the consciousness of older people to their changing role in American and global societies to ensure fulfillment in their eldering years. Virulent Ethnocentrism And Conflict Intractability: Puzzles And Challenges For 3rd Party Intervenors, Dennis J. D. Sandole
Peace and Conflict Studies
This article addresses complex identity-based conflicts, such as those associated with the ending of the Cold War (e.g., Bosnia). It suggests that in many identity-based conflicts, historical memories of outrage and victimhood ("chosen traumas") have persevered across centuries, thereby keeping the conflicting parties "in history." The paper examines the role of virulent ethnocentrism in such intractable conflicts. It also examines the role of "nature" and "nurture" in embedding the universal tendency for humans to divide their species into "them" and "us" within a highly charged emotional context. The paper argues that the complexity of these conflicts has at least four …

Assessing Storage Reliability Of Farm Dams, D Farmer, N Coles
Resource management technical reports
This report provides information on a method to estimate the volume of water in a farm dam and to determine how long this water will last. During periods of low rainfall, dams with farmland catchments receive limited run-off. Alternative water sources or management strategies need to be considered before the water runs out. The methods provided in this report are intended to provide to the landholder an indication of water supply over short periods only (i.e. in the order of 4-20 weeks).

This paper reports on the Pacific Northwest National Laboratory (PNNL) DOE Initiative in Image Science and Technology (ISAT) research, which is developing algorithms and software tool sets for remote sensing and biological applications. In particular, the PNNL ISAT work is applying these research results to the automated analysis of real-time cellular biology imagery to assist the biologist in determining the correct data collection region for the current state of a conglomerate of living cells in three-dimensional motion. The real-time computation of the typical 120 MB/sec multi-spectral data sets is executed in a Field Programmable Gate Array (FPGA) technology, which has …

Recent studies on potential mercury (Hg) contamination of fish from East Texas lakes and waterways have caused concern about mercury levels in East Texas waters. Historical records of Hg concentrations in 33 East Texas streams showed that median concentrations for each stream segment were no different than other U.S. streams. All the means and medians for stream segments having at least 20 recorded measurements were less than Texas (2.4 µg/L) water quality standards. Water samples collected in December 1995 and March 1996 from 6 different stream sites in Nacogdoches County had concentrations similar to historical records. Due to biological magnification, …

Less than six months after Ronald Reagan was inaugurated as the 40th president of the United States, the federal Centers for Disease Control in Atlanta reported pneumocystis carinii pneumonia, in five gay men. In the United States, AIDS was first diagnosed among gay men in New York City and San Francisco. There were several media trends for AIDS coverage in the 1980s. In the early 1980s, AIDS was mainly thought of as a gay-related disease. In the mid- l 980s, AIDS cases were found in hemophiliacs, intravenous drug users and "celebrities." Coverage in the late 1980s focused on heterosexuals and …
