Monitoring Immediate-Early Gene Expression Through Firefly Luciferase Imaging Of Hrs/J Hairless Mice, Michael E. Geusz, Anne M. Collaco

Background: Gene promoters fused to the firefly luciferase gene (luc) are useful for examining gene regulation in live transgenic mice and they provide unique views of functioning organs. The dynamics of gene expression in cells and tissues expressing luciferase can be observed by imaging this enzyme's bioluminescent oxidation of luciferin. Neural pathways involved in specific behaviors have been identified by localizing expression of immediate-early genes such as c-fos. A transgenic mouse line with luc controlled by the human c-fos promoter (fos::luc) has enabled gene expression imaging in brain slice cultures. For too many birds their environment includes airfields and aircraft. Knowing avian sensory abilities, researchers can design experiments and develop new devices and techniques to deter birds from aircraft on and away from airfields. How birds perceive the world about them determines many choices, including foraging, predator avoidance, and flight. Most experiments to investigate the sensory abilities of birds have been developed and analyzed using only human sensory capabilities, which often differ markedly from those of birds. Blackbirds (Icteridae) create human health and safety concerns when roosting in large numbers near airports or other urban areas. Therefore, there is considerable demand for effective nonlethal techniques to deter these birds. We tested the effectiveness of rotating mirrors (Peaceful Pyramid®) to keep blackbirds away from decoy traps. Mirrors were in place on traps for 5 consecutive days with control traps having similar a device in place but without mirrors. The total number of birds captured in traps with clear mirrors and control traps did not (P = 0.62) differ. When red mirrors were used, the total number of birds …

An ongoing challenge for wildlife managers is that target species habituate to or forget about hazing, even though the animals must be somewhat stressed by the hazing. Although stress and efficient memory formation/retrieval are considered to be incompatible, research suggests that the effect of stress on memory is complex. Mild and intense stress can impair memory formation and chronic stress can disrupt memory retrieval. Intermediate stress, however, actually enhances the formation of new memories, particularly long-term memories. Unfortunately, these conclusions are based data from only a very few domesticated species under highly controlled conditions. In nature the variation in baseline …

There are many criteria which need to be assessed when carrying out site selection for new airport developments. One of those which should be assessed is prospective bird hazard and bird strike risk – unfortunately, this is rarely considered as a factor. One reason for this may be the lack of available methodology for such work. This paper presents the methodology used during a recent hazard assessment for a proposed new airport for London. Wildlife observations collected over time at a US Air Force Base (AFB) have been visually displayed through Geographic Information System (GIS) technology. By continuing to expand upon the capabilities of GIS, USDA Wildlife Services has been able to manage the Seymour Johnson AFB airfield and surrounding areas in a practical manner. A Wildlife Hazard Assessment of the AFB used GIS to clearly display the location of birds and mammals observed on the airfield. Baseline themes of natural features (i.e. rivers, woodlands, etc.) were related to wildlife observations to determine areas requiring management. Additionally, GIS has been a useful tool to …

Abundance And Feeding Ecology Of Humpback Whales (Megaptera Novaengliae) In Kodiak, Alaska, Briana Harmony Witteveen

A feeding aggregation of humpback whales (Megaptera novaeangliae) in the Kodiak Island region has received little previous study. A mark-recapture experiment was conducted in 2001 and 2002 to estimate its abundance. Historical abundance was back-calculated from this estimate, whaling records, and suspected survival and productivity values within a population model. The current population was estimated at 157 whales and the pre-whaling population at 343 whales. Prey consumption by humpback whales was modeled using three methods for two hypothetical diets based on prey availability surveys conducted within the study area and stomach contents of commercially caught whales. Counting Seals: Estimating The Unseen Fraction Using A Covariate And Capture-Recapture Model, John R. Moran

We evaluated the influences of environmental covariates on the proportion of harbor seals (Phoca vitulina richardsi) ashore on Tugidak Island using a series of photographic capture-recapture experiments and ground based counts. We used general linear models to examine the significance of combinations of covariates including quadratics terms. Time of day, tide height, rate of tide change, surf, and wind speed significantly influenced the number of seals ashore during the molting period. The model including all significant covariates best explained the probability of seals being hauled out. We observed a decline in the local population using the haulout suggesting that seasonal …

Evaluating The Hooking Injury And Immediate Physiological Response Of Wild Rainbow Trout To Capture By Catch-And-Release Angling, Julie M. Meka

Rainbow trout from the Alagnak River watershed, Alaska, were captured by angling to determine the types of terminal gear contributing to hooking injury and the physiological response to angling based on concerns over high incidences of hooking injuries and the physiological impact of multiple recaptures on individual fish. Landing and hook removal times were recorded for a portion of fish captured, and plasma cortisol, glucose, ions (sodium, chloride, potassium), and lactate were evaluated in fish following capture to document physiological changes in relation to capture duration. Seasonal Distribution Of Steller Sea Lions (Eumetopias Jubatus) In Relation To High-Quality Ephemeral Prey Species In Southeastern Alaska, Jamie Neil Womble

Energetic demands are high for sea lions during spring when females are pregnant and lactating and males are preparing for extended fasting during the breeding season. Therefore, I predicted that the distribution of sea lions in spring would be influenced by the distribution of spring-spawning aggregations of high-energy Pacific herring (Clupeapallasii) and eulachon (Thaleichthys pacificus) in southeastern Alaska. Monthly aerial surveys at 23 Steller sea lions haul outs revealed that haulout use was seasonally dynamic. Some sea lion haulouts were only occupied during spring. Other haulouts exhibited pronounced increases in the number of sea lions during certain seasons. Sea lion …

Effects Of Smolt Length And Emigration Timing On Marine Survival And Age At Maturity Of Wild Coho Salmon (Oncorhynchus Kisutch) At Auke Creek, Juneau Alaska, Judith L. Lum

Coho salmon Oncorhynchus kisutch smolt were collected during the 1993-1997 emigrations at Auke Creek near Juneau, Alaska. Each day emigrants were separated into four size categories: small <90 mm), medium (90 - 110 mm), large (111 - 125 mm), and extra large (> 125 mm), tagged with a sequentially coded-wire tag, and released at tidewater. Tags from returning adults and jacks were collected and decoded in 1993-1998. Most survivors originated from the large and extra large categories, 40.5% and 43.1%, respectively. Large smolts contributed 28.9% to smolt-to-jack returns, significantly less than the smolt-to-jack survival contributed by extra large smolts, 67.8%. Smolt year, emigration date, and smolt length were significant in determining the length of returning jacks. …

Acute Exposures Of Salmonid Embryos To Total Dissolved Solids, Barbi Jean Failor-Rounds

Two exposure methodologies are described here utilizing embryonic and juvenile life stages of several species of salmonids. Specific life stages of the fish were exposed to solutions of varying total dissolved solids (TDS) modeled after the measured produced water from the Red Dog Mine in Kotzebue, Alaska. Embryonic and juvenile coho salmon (0. kisutch) were exposed for 96 hours to determine acute response to TDS. Following exposure, fish were grown out to button up to assess delayed effects. Results from the 96-hour study suggest fertilization is the most sensitive developmental stage of salmon exposed to TDS. Six fish species were …

Maturity, Fecundity, Growth, And Sustained Yield Of Coastal Cutthroat Trout At Florence Lake, Southeast Alaska, M. Birch Foster

The resident coastal cutthroat trout Oncorhynchus clarki clarki population in Florence Lake, Southeast Alaska was sampled from July through October, 1997 to assess its maturity, fecundity, growth and sustained yield. Maturing female cutthroat have significant gonad development between mid September and late October. A gonadosomatic index threshold was established for female cutthroat trout. A logistic model for maturity estimated asymptotic percentages by age and length: 92% and 100% for males and 86% and 80% for females, indicating presence of skip spawning. Male cutthroat trout matured earlier and at smaller length than females, but females matured more rapidly. An allometric model …

Alaska Native Attachment: A Qualitative Study With Four Athabascan Participants, Lester R. Keller

Attachment between caregiver and child is an affectional, nurturing bond that develops through the provision of sensitive, constantly available, and responsive care for the child. The attachment bond evolves around diverse interactive experiences that encourage the development of cognitive-emotional schemata and the internalization of a cognitive-emotional working model of relationships. Different cultural experiences encourage the development of different cognitive-emotional schemata. Using a semi-structured interview, behavior, values, and the developmental endpoint associated with attachment was collaboratively explored with four Athabascan research participants, and concepts that emerged were compared and contrasted with those articulated by mainstream attachment theory within Western psychology.
