Free Solution Electrophoresis Measurements And Their Theoretical Relationships To Net Protein Valence, Jennifer A. Durant Jan 2003

Free Solution Electrophoresis Measurements And Their Theoretical Relationships To Net Protein Valence, Jennifer A. Durant

Doctoral Dissertations

Biochemical solutions are composed to a large degree of charged molecules (proteins, DNA, dissolved salts, etc) which can interact and therefore exert a force on each other. Analytical determination of the valence of biological macro-ions contributes to our understanding of the forces involved in both intra- and inter-molecular interactions. This dissertation uses two free solution techniques, membrane confined electrophoresis (MCE) and capillary electrophoresis (CE), to study valence in the context of a series of charge mutants of T4 lysozyme and RNase Sa. The use of these mutants allows attribution of the changes in electrophoretic behavior entirely to changes in charge. …

Introducing Performance Into The Communication Curriculum For Engineers: Breaking The Mould, Constance Matilda Israel Jan 2003

Introducing Performance Into The Communication Curriculum For Engineers: Breaking The Mould, Constance Matilda Israel

Doctoral Dissertations

Education in South Africa has, thankfully, not escaped the winds of change. The introduction of Outcomes-based Education (OBE) has generated much debate. How have technikons responded to the educational transformation impelled by sociopolitical change? I refer here specifically to the Communication curricula for engineers at Mangosuthu Technikon.

This research explores the effects in my Communication classroom of introducing performance, through games and a dramatic text. My intention is to subvert the potential dangers of OBE, given its behaviourist and prescriptive thrust, and a possible reversion to the transmission model in its application at Mantec. I argue that gaps in the …

Living The Stereotype: Connections Between Male Behavior And Male Images, Andrew Paul Smiler Jan 2003

Living The Stereotype: Connections Between Male Behavior And Male Images, Andrew Paul Smiler

Doctoral Dissertations

This project examined some linkages and discrepancies between theories that describe the acquisition of gender typical attributes in childhood with theories that describe the maintenance of those attributes in adulthood. This perspective included the idea that there are several well known ways of enacting masculinity, related to well known stereotypes (e.g., jock, business, sensitive new age guy), and highlighted within sex variability. Because a distinction between biological sex and gender was made, the sample was not restricted to males despite focusing on the masculine. One focus of the project was the consistency with which an individual enacts a particular stereotypical …

"Everybody Get Together": The Sixties Counterculture And Public Space, 1964--1967, Jill Katherine Silos Jan 2003

"Everybody Get Together": The Sixties Counterculture And Public Space, 1964--1967, Jill Katherine Silos

Doctoral Dissertations

Historians and cultural analysts have traditionally considered the sixties counterculture an apolitical phenomenon by historians and other analysts. Yet concentrated examination of the public activities of the counterculture in San Francisco from 1963 to 1967 reveals that they were engaged in the creation of a public political culture that challenged the power of civil authorities to regulate the uses of parks, streets and sidewalks. In doing so, the counterculture constituted a distinct community with a political agenda.

This thesis is demonstrated through an analysis of the development of an ethos toward public space in the Beat movement and Merry Prankster …

Spatial Properties Of Photophobia, James Michael Stringham Jan 2003

Spatial Properties Of Photophobia, James Michael Stringham

Doctoral Dissertations

The squint response corresponding to photophobia (light-induced discomfort) was assessed via electromyography as a function of stimulus size, retinal locus (superior, inferior, temporal, nasal), and retinal eccentricity in three subjects. These results were compared to the same subjects' psychophysically-determined spatial distributions of macular pigment, to examine in detail the attenuation of photophobia by macular pigment. Results indicate that the photophobia response is preferentially biased to prevent the fovea from exposure to potentially damaging short-wavelength light. In this regard, macular pigment appears to act as a spatially-integrated filter, serving to attenuate photophobia to a great extent. Photophobia and macular pigment therefore …

Conceptualizing And Measuring The Life Space And Its Relation To Openness To Experience, Marc A. Brackett Jan 2003

Conceptualizing And Measuring The Life Space And Its Relation To Openness To Experience, Marc A. Brackett

Doctoral Dissertations

The Life Space is divided into four domains that surround personality, which broadly encompass a person's biological foundations, owned possessions, interactions and daily activities, and group memberships. In Study 1, a revised measure of the Life Space (the College Student Life Space Scale) was developed. Factor analysis of the items within each domain resulted in 96 meaningful and reliable factor-based scales that provided a rich description of college students' personal surroundings and everyday behavior. A second-order (hierarchical) factor analysis of the first-order scales resulted in seven global Life Space dimensions. In Study 2, the first- and second-order Life Space scales …

Calcium -Dependent Protein Kinases Are Myristoylated And Associated With Different Membranes In Arabidopsis Thaliana, Sheen Xun Lu Jan 2003

Calcium -Dependent Protein Kinases Are Myristoylated And Associated With Different Membranes In Arabidopsis Thaliana, Sheen Xun Lu

Doctoral Dissertations

In plants, calcium-dependent protein kinases (CDPKs) are the predominant calcium-stimulated kinases and are known to be involved in many cellular processes. CDPK enzymatic activity previously has been detected in many locations in plant cells, including the membrane fraction. However, little is known about the subcellular locations of individual CDPKs or the mechanisms involved in targeting them to those locations. Arabidopsis contains 34 genes that are predicted to encode CDPKs and 28 of the predicted CDPK proteins have potential myristoylation motifs at their amino termini. Myristate is a 14-carbon saturated fatty acid that is attached co-translationally to the amino-terminal glycine of …

Chemical Sensors Based On Swellable Molecularly Imprinted Polymer Microspheres In A Hydrogel, Wenzhe Fan Jan 2003

Chemical Sensors Based On Swellable Molecularly Imprinted Polymer Microspheres In A Hydrogel, Wenzhe Fan

Doctoral Dissertations

A novel design of chemical sensors based on swellable molecularly imprinted polymer microspheres is presented. These sensors were successfully used to measure norephedrine and theophylline in water and in buffer at neutral pH.

A new method was developed for preparing swellable norephedrine templated poly (N-isopropylacrylamide-acrylic acid) (poly NIPA-AAc) particles by suspension polymerization in a perfluorocarbon solvent using ultrasonic emulsion technique. The particles were crosslinked with 5% of N, N'-methlene-bis-acylamide (MBA) using 20% fluorinated surfactant as a stabilizer. The resulting particles were around 6 mum in diameter. HYPAN and poly (vinyl alcohol) (PVA) hydrogels were used to make hydrogel membranes containing …

Characterization Of Prokaryotic Diversity In A Chlorinated Solvent-Contaminated Bedrock Aquifer Using Denaturing Gradient Gel Electrophoresis, Ribosomal Dna Sequencing, And Real Time Pcr, Walid Naser

Doctoral Dissertations

A culture-independent molecular phylogenetic approach was implemented to investigate spatial and temporal variations in microbial diversity and abundance in a chlorinated solvent-contaminated bedrock aquifer. Core and groundwater samples were collected from several boreholes and included the following samples: (1) groundwater that was associated with the core, (2) open fractures and (3) partially mineralized sealed fractures (PMSF) exposed by striking the core with a surface-sterilized geology hammer. DNA representing the attached and unattached microbial communities was extracted from these samples, and the 16S rRNA gene was amplified with primers specific for Bacteria, Archaea, sulfate reducers, Geobacteraceae family, Dehalorespirers (Dehalococcoides sp. and …

Intimacy With The Natural World: A Humanistic Perspective, Gabriel F. Bautista Jan 2003

Intimacy With The Natural World: A Humanistic Perspective, Gabriel F. Bautista

Doctoral Dissertations

Thomas Berry's "intimacy with the natural world" is not an alternative to the anthropocentric-biocentric dispute, which belong to the same rationality. The frontier story and environmentalism are both part of the same rationality because they ignore people. While anthropocentrism is focused on the utilization of natural resources, biocentrism is focused on nature's conservation. The frontier and environmental narrative provides the explicit genealogy of intimacy-alienation, which are always in counterpoint with each other. Further still, if intimacy means only "intimacy with the natural world", then it is a hypostatization of "wilderness" that indicates more alienation. Nevertheless, in my interpretation, intimacy has …

An Overview: The Difference Slavery Made: A Close Analysis Of Two American Communities, Edward L. Ayers

History Faculty Publications

Using digital media, we wanted to give readers full access to a scholarly argument, the historiography about it, and the evidence for it. Our early models of the article contained neat squares and lines and carefully arranged explanations of the links from one part to another. Through two sets of readings by peer reviewers and presentations to a range of audiences, we have revised our presentation and our argument while maintaining the original purpose of the article. This essay introduces the electronic article and explains its development, as well as our intentions for it.

Usability Testing At Florida International University Libraries: What We Learned, Sarah J. Hammill

E-JASL: Electronic Journal of Academic and Special Librarianship (1999-2009, Volumes 1-10)

The Florida International University Libraries’ Web site’s new look was launched in Fall 2001. As a result of the new look, a group formed to undertake a usability study on the top page of the site. The group tested three target groups to determine the usability of the top page. The study pointed out some revisions for the top page; however, more importantly, it suggested areas for future research.

Review Of The Lifted Veil, Tim Heath Jan 2003

Review Of The Lifted Veil, Tim Heath

The George Eliot Review Online, ISSN: 2831-5375

It was perhaps Herbert Spencer in his Autobiography (1904) who first disseminated the myth that, though George Eliot will remain for posterity one of our finest novelists, her weakness was that she could not construct effective, dramatic plots. By implication, her stories would thus not be effective on the stage, without the commentary of her wise, omniscient narrator. Perhaps in consequence there have been fewer dramatic adaptations of George Eliot's work than those of other nineteenth-century novelists, including Jane Austen and Dickens - this despite, amongst others, an extremely distinguished silent film based on Romola, starring Lillian Gish, in 1924. …

Address Of Wreath-Laying In Poets' Corner, Westminster Abbey 20 June 2002, Margaret Wolfit

The George Eliot Review Online, ISSN: 2831-5375

Dear George Eliot

You don't know me - but I have been a great admirer of yours for a long time now, since I was at school in fact. I must have been very young when I first had parts of your novels read to me. Later when I was at boarding school I studied Silas Mamer for a literature exam - I can't recall my reaction to other authors at the time - but I know I wrote a letter home saying I thought George Eliot must be a wonderful person because my mother kept the letter.

After leaving …

Internationalizing American Higher Education: A Call To Thought And Action, Deborah Dezure Jan 2003

Internationalizing American Higher Education: A Call To Thought And Action, Deborah Dezure

To Improve the Academy: A Journal of Educational Development

In the wake of the World Trade Center disaster, many faculty developers are asking themselves what they do to promote international peace and understanding. But even before these events, there has been an indication that there was a pressing need to focus on global competencies as an important part of higher education for the 21st century. The purpose of this essay is threefold: 1) to summarize the research on the status of internationalization on American campuses, 2) to make the case for the active involvement of faculty developers in internationalizing higher education, and 3) to offer strategies with which we …

Establishing A Teaching Academy: Cultivation Of Teaching At A Research University Campus, Patricia Kalivoda, Jodef Broder, William K. Jackson

To Improve the Academy: A Journal of Educational Development

The University of Georgia (UGA) has worked hard over the last 22 years to increase the respect and reward for teaching through the faculty development programs of the office of instructional support and development and through the establishment of two campus-wide teaching awards. Looking for a means to extend a celebration of teaching beyond one-time recognition or one-time participation, the university established a campus-wide teaching academy. The purpose of this chapter is to chronicle the evolution of the teaching academy that was founded at the University of Georgia in 1999. The mission, goals, membership, funding, and programs and activities of …

Accommodating Students With Disabilities: Professional Development Needs Of Faculty, Sheryl Burgstahler Jan 2003

Accommodating Students With Disabilities: Professional Development Needs Of Faculty, Sheryl Burgstahler

To Improve the Academy: A Journal of Educational Development

Faculty members play an important role in making academic programs accessible to postsecondary students with disabilities. However, instructors do not always possess the knowledge, experiences, and attitudes that result in the most inclusive environment for these students. A literature review was conducted to explore what faculty members need to know about accommodating students with disabilities in their courses and how they can best gain this knowledge. These results were used to develop a comprehemive set of training options that can be used with postsecondary instructors nationwide. The content of these options focuses on legal issues, accommodation strategies, and resources. Modes …
