General Guidelines For The Establishment & Evaluation Of Invasive Species Early Detection & Rapid Response Systems Jan 2003

General Guidelines For The Establishment & Evaluation Of Invasive Species Early Detection & Rapid Response Systems

National Invasive Species Council

Preventing the introduction of invasive species is the first line of defense against invasions. However, even the best prevention efforts will not stop all invasive species introductions. Early detection and rapid response (ED&RR) efforts increase the likelihood that invasions will be addressed successfully while populations are still localized and population levels are not beyond that which can be contained and eradicated. Once populations are widely established, all that might be possible is the partial mitigation of negative impacts. In addition, the costs associated with ED&RR efforts are typically far less than those of long-term invasive species management programs.

The charge …

Aerosol Exposure Of White-Tailed Deer (Odocoileus Virginianus) To Mycobacterium Bovis, Mitchell V. Palmer, W. Ray Waters, Diana L. Whipple Jan 2003

Aerosol Exposure Of White-Tailed Deer (Odocoileus Virginianus) To Mycobacterium Bovis, Mitchell V. Palmer, W. Ray Waters, Diana L. Whipple

Michigan Bovine Tuberculosis Bibliography and Database

Tuberculosis due to Mycobacterium bovis affects both captive and free-ranging Cervidae in the United States. Various animal models have been developed to study tuberculosis of both humans and animals. Generally, tuberculosis is transmitted by aerosol and oral routes. Models of aerosol exposure of large animals to M. bovis are uncommon. In order to develop a reliable method of aerosol exposure of white-tailed deer (Odocoileus virginianus) to M. bovis, 12 healthy white-tailed deer, aged 8–10 mo, were infected by aerosol exposure to 2x105 to 1x106 colony forming units (CFU) (high dose, n=4) of M. bovis or …

Feral Swine - Are They A Disease Threat To Livestock In The United States?, Gary W. Witmer, Robert B. Sanders, Arnold C. Taft

Michigan Bovine Tuberculosis Bibliography and Database

Feral swine populations provide both benefits and liabilities to citizens of the United States. Their expanding range and increasing densities, however, have raised concern over the adverse environmental and agricultural effects and the increased risk of disease transmission between feral swine and livestock. We discuss the role of feral swine in the transmission of wildlife diseases and, in particular, in diseases of national significance to the livestock industry. We also discuss available management tools and strategies for reducing feral swine populations, minimizing damage or disease occurrences and eradicating populations when deemed appropriate. Finally, we note areas of research that may …

Cystic Echinococcosis In The Arctic And Sub-Arctic, Robert L. Rausch Jan 2003

Cystic Echinococcosis In The Arctic And Sub-Arctic, Robert L. Rausch

Harold W. Manter Laboratory of Parasitology: Faculty Publications

The northern biotype of Echinococcus granulosus occurs throughout the holarctic zones of tundra and taiga, from eastern Fennoscandia to the Bering Strait in Eurasia and in North America from arctic Alaska approximately to the northern border of the United States. The cycle of the cestode is complex in taiga at lower latitudes, because of the greater diversity of potential hosts. In the Arctic and Subarctic, however, four patterns of predator/prey relationships may be discerned. Two natural cycles involve the wolf and wild reindeer and the wolf and elk (moose), respectively. Where deer of the two species coexist, both are prey …

Harold W. Manter Laboratory of Parasitology: Faculty Publications

We report the use of six oligoprobes designed from intergenic spacer region sequences to identify fourth-stage larvae (L4) of the tribe Cyathostominae. Oligoprobes were designed for identification of the following species: Cylicocyclus ashworthi, Cylicocyclus nassatus, Cylicocyclus insigne, Cyathostomum catinatum, Cylicostephanus goldi, and Cylicostephanus longibursatus. A seventh probe was designed as a positive control to identify all these members of the Cyathostominae. The intergenic spacer region was amplified by PCR using conserved primers. Initially, three oligoprobes were used in Southern blot analysis. To facilitate high-throughput identification, these and a further four oligoprobes were developed for use in a PCR–ELISA. …

Tafenoquine is a promising new 8-aminoquinoline drug that may be useful for malaria prophylaxis in nonpregnant persons with normal glucose-6-phosphate dehydrogenase (G6PD) function. A randomized, doubleblind, placebo-controlled chemoprophylaxis trial was conducted with adult residents of northern Ghana to determine the minimum effective weekly dose of tafenoquine for the prevention of infection by Plasmodium falciparum. The primary end point was a positive malaria blood smear result during the 13 weeks of study drug coverage. Relative to the placebo, all 4 tafenoquine dosages demonstrated significant protection against P. falciparum infection: for 25 mg/week, protective efficacy was 32% (95% confidence interval [CI], …

Functional Implications In Apoptosis By Interferon Inducible Gene Product 1-8d, The Binding Protein To Adenovirus Preterminal Protein, Insil Joung, Peter C. Angeletti, Jeffrey A. Engler

Nebraska Center for Virology: Faculty Publications

Adenovirus (Ad) precursor to the terminal protein (pTP) plays an essential roles in the viral DNA

replication. Ad pTP serves as a primer for the synthesis of a new DNA strand during the initiation

step of replication. In addition, Ad pTP forms organized spherical replication foci on the nuclear

matrix (NM) and anchors the viral genome to the NM. Here we identified the interferon inducible

gene product 1-8D (Inid) as a pTP binding protein by using a two-hybrid screen of a HeLa cDNA

library. Of the clones obtained in this assay, nine were identical to the Inid, a 13-kDa polypeptide …

The diseases and virulence genes associated with Shiga toxin–producing Escherichia coli (STEC) are characterized incompletely. We analyzed, by polymerase chain reaction, 82 STEC isolates collected prospectively in Montana and profiled associated illnesses by patient chart review. All E. coli O157:H7 contained stx2-group genes, as well as eae, iha, espA, and ehxA; 84% contained stx1. Non-O157:H7 STEC less frequently contained stx1( P = .046 ), stx2 (P < .001), iha (P < .001), eae, and espA (P = .039 for both), were isolated less often from patients treated in emergency departments (P = .022), and …

Hermone, Jonathan L. Vennerstrom, Connor F. Mcgrath, Douglas J. Lane, Edward A. Sausville, Daniel W. Zaharevitz, Rick Gussio, Sina Bavari

United States Army: Publications

Botulinum neurotoxins (BoNTs) are among the most lethal biological substances to have been weaponized and are listed as biodefense category A agents. Currently, no small molecule (non-peptidic) therapeutics exist to counter this threat; hence, identifying and developing compounds that inhibit BoNTs is a high priority. In the present study, a high-throughput assay was used to identify small molecules that inhibit the metalloprotease activity of BoNT serotype A light chain (BoNT/A LC). All inhibitors were further verified using a HPLC-based assay. Conformational analyses of these compounds, in conjunction with molecular docking studies, were used to predict structural features that contribute to …

Protracted Protection To Plasmodium Berghei Malaria Is Linked To Functionally And Phenotypically Heterogeneous Liver Memory Cd8+ T Cells1, Dmitri Berenzon, Robert J. Schwenk, Lisa Letellier, Mimi Guebre-Xabier, Jackie Williams, Urszula Krzych

United States Army: Publications

We previously demonstrated that protection induced by radiation-attenuated (y) Plasmodium berghei sporozoites is linked to MHC class I-restricted CD8+ T cells specific for exoerythrocytic-stage Ags, and that activated intrahepatic memory CD8+ T cells are associated with protracted protection. In this study, we further investigated intrahepatic memory CD8+ T cells to elucidate mechanisms required for their maintenance. Using phenotypic markers indicative of activation (CD44, CD45RB), migration (CD62L), and IFN-y production, we identified two subsets of intrahepatic memory CD8+ T cells: the CD44highCD45RBlowCD62LlowCD122low phenotype, representing the dominant effector …

Biological Sciences Faculty Publications

Both soluble and cell-mediated components are involved in the innate immune response of arthropods. Injection of Borrelia burgdorferi, the Lyme disease agent, results in the secretion of defensin into the hemolymph of the ixodid tick, Dermacentor variabilis. The presence of the peptide is observed as early as 15 min post-challenge and remains present through 18 h post-challenge. As observed in insects and soft ticks, the transcript for defensin is detected as early as 1 h post-challenge in D. variabilis. RT-PCR resulted in an amplicon of 624 bp with a 225 bp region that translates to a 74 …
