Transgenic Bovine Nuclear Transfer Embryos From Adult Somatic Cell Lines*, Sezen Arat, S. Jacek Rzucidlo, Steven L. Stice

This study was performed to examine whether adult fibroblast cells can be manipulated genetically and whether clonal lines derived from those cells can support embryonic development. A primary adult cell line was established from the ear of an aged cow. Adult cells with a plasmid containing the enhancer green fluorescence protein (EGFP) gene, and non-transfected cells were used for cloning. Green fluorescence expression was observed in 35/49 (71.4%) adult clones. The developmental rates of embryos were significantly lower for cell lines expressing EGFP (11.4%) than for non-transfected cells (20.1%, P < 0.05). However, there was no decrease in nuclear transfer (NT) developmental rates (21.5%) when donor nuclei from EGFP transfected cell lines not expressing EGFP but retaining neomycin-resistance gene expression were used as donor nuclei. The NT embryos from transfected and non-transfected cell lines had similar morphology and cell numbers. The results indicated that adult cells can complete clonal propagation including transfection and selection and can be used to produce transgenic NT embryos.

Retained And Trash Fish Catches Of Beach-Seining In The Aegean Coast Of Turkey, Okan Akyol

In this study, findings of the qualities and quantities of commercial and trash catches from beach-seining in coastal waters of the Aegean Sea are presented. A total of 62 finfish and eight invertebrate taxa were identified in catches sampled during May, June, July and September 1996, with 42 taxa being retained and 28 taxa being genuine trash catches. The estimated total ratio of commercial to genuine trash catch (kg) was 1:0.273. The fish and invertebrate species caught from beach seine fishery were typical of those that inhabit coastal embayments in the Aegean Sea. The Application Of A Phosphorus Budget Model Estimating The Carrying Capacity Of Kesikköprü Dam Lake, Serap Pulatsü

The aim of this study was to estimate the carrying capacity of Kesikköprü Dam Lake, Ankara, where cage farms for the intensive culture of rainbow trout are located. For this purpose Dillon and Rigler's phosphorus budget model was applied in a series of steps and the carrying capacity of the lake was found to be 3335 tonnes per year. This estimated value was about 10 times higher than the present production level of the lake. It seems possible to orientate the fish culture in cages in inland waters by using the phosphorus budget model recommended in this study.

The aim of this study was to investigate the findings of clinical, haemotological, biochemical, histopathological, mycological and urine analyses and to evaluate the efficiency of itraconazole treatment in dogs experimentally infected with Aspergillus fumigatus. In this study, 25 healthy male dogs were used. The animals were divided into 3 groups: group I (control), group II (infected) and group III (treatment) consisting of 5, 10 and 10 dogs, respectively. This study is concerned with the feeding behavior of pikeperch (Sander lucioperca (L., 1758)) living in Hirfanlı Dam Lake constructed on the Kızılırmak River. There were 326 pikeperch caught between August 1996 and July 1997. The fork length and weight of the pikeperch caught ranged between 105 and 529 mm and 12 and 1520 g in the general population. Pikeperch are carnivorous and piscivorous and they eat their own species when nutrients are lacking. The smallest fish-swallowing length was determined to be 145 mm. Ten motile Aeromonads, including one Aeromonas hydrophila ATCC 7512, were used. Of the strains, five were isolated from gulls (one A. hydrophila strain and four A. caviae strains) and four from rainbow trout (three A. hydrophila strains and one A. caviae strain). Whole cell protein profiles of these strains were analyzed by SDS-PAGE. Forty-five pairs were formed in comparing the protein profiles of each strain. Of the pairs, 17 (37.8%) were different, 18 (40%) were unclear and 10 (22.2%) were identical. This study was intended to establish whether there is any relation between immune deficiency and cryptosporidiosis by using an animal model. Forty Rattus norvegicus rats obtained from the Experimental Animal Laboratories of the Ege University Faculty of Medicine were divided into two groups. The first group (n = 20) received corticosteroids (dexamethasone, 1.5 mg, subcutaneously, twice weekly) for eight weeks for immunosuppression, while the control group (n = 20) received no treatment. Longissimus dorsi muscle obtained from beef carcasses was used in this study. Meat cores 2.54 cm in diameter were marinated in 0.34, 0.68 and 1.02 M sodium chloride (NaCl) and 0.05, 0.1 and 0.15 M calcium chloride (CaCl_2) solutions at 5 °C for 72 h. Warner Bratzler Shear (WBS) values were significantly different between samples. The samples marinated with CaCl_2 held less water than those marinated in NaCl. Cooking losses were lower in the control samples than in the marinated samples. Differential scanning calorimetry was employed to determine the amount of bound water in the meat samples. Rainbow trout reared in fresh water were exposed to a sublethal concentration of mancozeb (1/2 of LC_{50} = 1.1 mg/l) at 24 h intervals, for 3 weeks in tanks containing 780 l water under natural light conditions with a constant flow (1.5 l min^{-1}) of aerated dechlorinated tap water at 9-11 °C and with no recirculation. Approximately 2 cc venous blood was drawn from each subject (after weekly exposure) for the determination of hematological parameters, using heparin as an anticoagulant. The objective of this study was to demonstrate the specific blood velocity parameters called peak systolic velocity (PSV), end diastolic velocity (EDV) and the resistive index (RI) in the internal ophthalmic artery (IOA) and the long posterior ciliary artery (LPCA) with color Doppler imaging (CDI) in clinically normal dogs. Eight healthy dogs were used in the study. Color Doppler ultrasonography was performed on both eyes. PSV, EDV and RI measurements were obtained for both eyes. In a 9-month-old male German Shepherd, degenerative arthritis and incongruity of the elbow joint were observed due to premature closure of the distal radial growth plate. An obvious shortening was present in the radius and ulna, when compared to the contralateral leg. Corticotomy was carried out on the radius and an Ilizarov apparatus was applied to the leg by passing Kirschner wires through the proximal and distal fragments of the radius. Liver Antioxidant Systems And Lipid Peroxidation In Sea Bass (Dicentrarchus Labrax)Adapted To Fresh Water, Aysel Şahan, Ergül Belge Kurutaş, Suat Di̇kel

Changes in the antioxidant systems of the liver tissues of sea bass (Dicentrarchus labrax) adapted to low salinity levels (0 ) were investigated. Malondialdehyde levels, as a specific indicator of oxidant stress, were determined. The results indicated highly significant differences among the test and control groups (P < 0.05). Some histopathological destruction, liver weight and hepatosomatic index values were determined at high levels and, moreover, superoxide dismutase, glutathione peroxidase (GPX), glucose-6phosphate-dehydrogenase and malondialdehyde activities increased, while reducted glutathione levels decreased in comparison with the control group. An increase in the malondialdehyde level was determined to be an indicator of lipid peroxidation.

Osmotic fragility and fatty acid composition in erythrocytes of mirror carp (Cyprinus carpio) kept at 8 °C, 16 °C and 28 °C were determined. Osmotic fragility in erythrocytes (EOF) increased significantly (0.19-0.24%) with the low water temperature. Significant declines in the quantity of saturated fatty acid (44.49-38.40%) and monounsaturated fatty acids (26.30-13.99%), and significant increases in that of polyunsaturated fatty acid (29.21-47.59%) were determined (P < 0.05). It was concluded that low water temperature caused a tolerable stress in mirror carps.

The possible pharmacological and toxicological effects of Pistacia terebinthus L. in experimentally hypercholesterolemic rabbits were investigated in this study. A total of 28 rabbits were divided into four groups, and these groups were fed diets containing standard pellets (Group 1), cholesterol (2%) (Group II), P. terebinthus L. (1 g/kg body weight) and cholesterol (2%) (Group III) and P. terebinthus L. (1 g/kg body weight) (Group IV) for 14 weeks. When the data were statistically evaluated over the weeks, body weight gains, showed no marked difference among the groups although liver weight was 58% higher in the cholesterol control group (P < 0.01) and 20.3% higher in P. terebinthus L. and cholesterol group than in the control group. In the comparison of the cholesterol-fed groups, Group III exhibited a marked decrease (P < 0.01) in total cholesterol and triglyceride evels and very low density lipoprotein (VLDL) and low density lipoprotein (LDL) levels by the 14th week and a 1.3-times higher (P < 0.001) high density lipoprotein (HDL) value. In the group fed cholesterol and P. terebinthus L., clotting and bleeding times increased by 13% and 12%, respectively, compored to the cholesterol control group. On account of the histopathological findings, it was confirmed that P. terebinthus L. restrained the progression of the atherosclerotic lesions in the thoracic artery and of hydropic degeneration and fatty changes in the liver. No toxic effect could be attributed to P. terebinthus L. from an investigation of the biochemical data. The results of this study show that P. terebinthus L. has a hypolipidemic effect in hypercholesterolemic rabbits.

Karyotype analysis was performed in Chalcalburnus tarichi specimens by investigating the number and structures of their chromosomes. The fish used in this study were caught with fishing nets from Lake Van and taken to the laboratory. The fish were injected i.p. with 0.01 ml/g body weight doses of a 0.6% solution of a colchicine for 190 min. As a result of the metaphase investigation we determined that C. tarichi had 2n = 50 chromosomes. This study was performed on the blood samples of 7 Brown cows collected during the estrus, metestrus, diestrus and proestrus phases of the estrous cycle. We investigated whether or not there was a difference between these stages of the cycle with regard to hematologic parameters. Mean hematologic parameters determined during the period of the estrous cycles were found to be within the normal range. This study was conducted to investigate the effects of some factors on litter size fertility and pregnancy period in Morkaraman sheep as well as birth weight and survival rate in Morkaraman, Dorset Down x Morkaraman (F_1) crossbreed lambs. In the present study, 151 Morkaraman sheep of different ages, and 110 purebred and 46 crossbreed (Dorset Down x Morkaraman F_1) lambs were used as the research material. The effect of the genotype and birth weight on pregnancy period were significant (P < 0.01). However, the effects of birth type, sex of the lamb and age of the sheep were not. The effects of birth type, genotype, sex of the lamb and age of the sheep on birth weight were significant (P < 0.01). The effects of genotype, sex of the lamb and age of the sheep on the number of lambs per gestation were not significant. The effects of birth type on the survival rate of the lambs were significant (P < 0.01); on the other hand, the effects of genotype, sex of the lamb and age of the sheep on survival rate were not.

This study was carried out to investigate milk yield traits and some environmental factors affecting milk yield traits in Brown Swiss cattle reared at the Bahri Dağdaş International Agricultural Research Institute. Between 1987 and 1998, 730 lactation records obtained from 273 cows were evaluated. The least-squares means of measured milk yield, 305-day milk yield, dry period and lactation duration were 3631 kg, 3332 kg, 97.1 days and 313.8 days, respectively. Both year and age statistically affected (P < 0.01-0.001) milk yield and lactation duration, but the calving season was not affected (P > 0.05). The effect of the year on dry period was also significant (P < 0.01), but the effect of age and calving season were not significant (P > 0.05).

This study was carried out to estimate the phenotypic and genetic parameters for the gestation length and birth weight of Brown Swiss cattle reared at the Bahri Bağdaş International Agricultural Research Institute. The experimental material consisted of 891 calving records and 666 gestation length records between 1989 and 2000. The least-squares means of birth weight and gestation length were 35.6 kg and 284 days, respectively. Except for the effect of the dam's age on gestation length, all other factors investigated in this study had significant effects on both gestation length and birth weight (P < 0.05-0.001). The estimations of heritability and repeatability for birth weight were 0.10 and 0.15, respectively. Estimated repeatability for gestation length was 0.14.

Pomphorhynchus laevis is an acanthocephala which lives in the intestines of fish. These acanthocephala are characterised by a cylindrical proboscis, and an extremely long neck with a bulbous anterior expansion. In this study, the surface structures of P. laevis were examined with a scanning electron microscope. On the proboscis of this parasite 18 hook rows, each consisting of 12 hooks, were observed. The anterior hooks were smaller than the posterior hooks of the proboscis. The recurved hooks were within pockets on the proboscis. This research was carried out to determine the possibilities of improving the feed value of lentil straw with steam and acid treatment. Lentil straw (Lens culinaris M.) was used as feed material and three 3-year-old, Kıvırcık rams fitted with rumen cannula were used as the animal material. Lentil straw was impregnated with different concentrated H_2SO_4 solutions, and then autoclaved at different periods and temperatures. Biochemical composition, condition indices, water temperature, salinity, seston, particulate organic matter and chlorophyll-a were monitored for 1 y. Wet meat volumetric condition index ranged from 23.96% to 47.43%, with a mean of 29.71 ± 1.99% . Mean dry weight condition index was 5.07 ± 0.3% and ranged from 3.94% to 8.17% . Mean moisture, ash, lipid, protein and carbohydrate values were 82.99 ± 0.43%, 6.7 ± 0.22% , 8.7 ± 0.58% , 60.21 ± 1.47%, and 24.23 ± 1.72%, respectively. A study was carried out to determine the effects of yeast based (Biosacc^TM) and mixture of bacteria-mold-yeast based (Protexin^TM) dietary probiotics on live weight, feed consumption, feed efficiency, egg production, egg weight, egg shell weight, and some blood parameters of quails raised under constant heat stress conditions. In this study, four male Kıvırcık yearlings with permanent rumen cannula were used. The animals were fed according to a 4 x 4 Latin square design over 20-day periods (15-day adaptation period, 5-day collection period). The animals were fed four different ration types. Respiratory diseases due to infectious agents are an important problem in the broiler chicken industry. One of the most important infectious agents of chickens is Ornithobacterium rhinotracheale, which has recently been isolated in Turkey and around the world. In this report, O. rhinotracheale infection was detected in broiler chickens submitted to the poultry disease laboratory with respiratory problems, increased mortality and retarded growth.

Haemoglobin and transferrin types were investigated in Çine type sheep. Separation of transferrin (Tf) and haemoglobin (Hb) types was carried out using horizontal starch-gel and polyacrylamide gel electrophoresis, respectively. Three Hb A and 59 Hb B types, and 5 transferrin alleles (A, B, M, D, E) were isolated from Çine type sheep. Eight Tf phenotypes consisting of 3 homozygote types (Tf AA, Tf MM, Tf DD) and 5 hetorozygote types (Tf AM, Tf AD, Tf AE, Tf BD, Tf MD) were detected.

This study examined the relationship between child, respondent, parent/guardian, and household characteristics and the preferences of the availability of sick care, a small group size, a reasonable cost, and a caregiver who shares similar beliefs about raising children in the selection of nonparental arrangements. Groundwater phytotechnology systems are currently being used to control the migration of contaminant plumes and remove groundwater that flows into containment areas. Trees in such systems function by reducing recharge and using groundwater via transpiration. A stand of deep-rooted trees can act as a biological "pump", removing a substantial amount of water from the saturated zone. This article addresses the role of the genetic tie in the parent-child relationship through three lenses. First, we argue that the legal system recognizes children's rights not by treating children as autonomous actors, but by identifying the individuals and institutions most likely to promote children's interests and encouraging their success. Second, we examine the existing empirical and socio-biological literature that considers the importance of biological relationships, and concludes that it demonstrates not a single set of answers, but a set of tradeoffs. Competition policy is on the WTO agenda for the Doha Round, but it is unlikely that it will result in any substantive international standards; the goal, instead, seems to be to agree on core principles to guide the development of national law, including transparency, non-discrimination, and procedural fairness, perhaps extending to special and differential treatment for developing countries. While there is much to commend these principles, this paper takes a deliberately contrarian view, arguing that core principles are not at all where WTO competition policy should begin.
