Relationships Of Chute-Side Measurements To Carcass Measurements, James C. Macdonald, Terry J. Klopfenstein, Galen E. Erickson, Casey Macken, Jeffrey Folmer

Nebraska Beef Cattle Reports

Three data sets were compiled to determine the relationship of weight, performance, hip height and ultrasound-measured fat thickness to hot carcass weight and fat thickness. Weight is generally the best predictor of relative differences in carcass weight at any time in the production system. Hip heights do not predict relative differences in carcass weight. Although the combination of hip height and weight is a more precise indicator of carcass weight than is hip height alone, generally this combination is inferior to weight alone. Only ultrasound-measured fat thickness predicted relative differences in fat thickness. Prediction of relative differences in carcass weight …

Rck Domain Model Of Calcium Activation In Bk Channels, Alexander R. Pico Jan 2003

Rck Domain Model Of Calcium Activation In Bk Channels, Alexander R. Pico

Student Theses and Dissertations

Potassium ion channels are ubiquitously expressed from bacteria to mammals where they are involved in various processes ranging from the regulation of osmotic pressure in a single cell to the electrical response in muscle fibers to the generation of action potentials in neurons. The BK channel family (BK for Big K+ conductance) is an interesting subfamily of K+ channels responsive to both membrane voltage and intracellular calcium ion. The unique, high-affinity Ca2+ sensitivity of BK channels is critical to their physiological function in various cell types. The mechanism by which Ca2+ activates BK channel gating, however, …

Protein Semi-Synthesis In Vivo, Izabela Giriat Stankiewicz Jan 2003

Protein Semi-Synthesis In Vivo, Izabela Giriat Stankiewicz

Student Theses and Dissertations

Incorporation of chemical probes into proteins is a powerful way to elucidate biological processes and to engineer novel function. This thesis describes the ligation of synthetic molecules to target proteins in an intracellular environment, and the use of semi-synthetic proteins for whole animal studies. In the first approach a cellular protein is genetically tagged with one half of a split-intein. The complementary half is linked in vitro to the synthetic probe and this fusion is delivered into cells using a transduction peptide. Association of the intein halves in the cytosol triggers protein transsplicing, resulting in the ligation of the probe …

Death By Any Other Name: The Federal Government's Inconsistent Treatment Of Drugs Used In Lethal Injections And Physician-Assisted Suicide, Colin Miller

Journal of Law and Health

While the FDA is under no legal obligation to regulate the drugs used in executions, these recent developments certainly create a moral imperative requiring review. This paper will argue that the federal government cannot consistently refrain from regulating lethal injection drugs while arguing for prosecution of those prescribing drugs to be used by patients in assisted suicide. Part II will look at the opinions in Chaney and the factors behind the FDA's decision not to regulate the drugs used in executions. Part III will look at Oregon's Death with Dignity Act and its authorization by the Supreme Court. Parts IV-VI …

RNA silencing in plants likely exists as a defense mechanism against molecular parasites such as RNA viruses, retrotransposons, and transgenes. As a result, many plant viruses have adapted mechanisms to evade and suppress gene silencing. Tombusviruses express a 19 kDa protein (p19), which has been shown to suppress RNA silencing in vivo and bind silencing-generated and synthetic small interferingRNAs (siRNAs) in vitro. Here we report the 2.5 A° crystal structure of p19 from the Carnation Italian ringspot virus (CIRV) bound to a 21 nt siRNA and demonstrate in biochemical and in vivo assays that CIRV p19 protein acts as molecular …

Are Hummingbirds Facultatively Ammonotelic? Nitrogen Excretion And Requirements As A Function Of Body Size, Todd J. Mcwhorter, Donald R. Powers, Carlos Martinez Del Rio

Faculty Publications - Department of Biological & Molecular Science

Most birds are uricotelic. An exception to this rule may be nectar-feeding birds, which excrete significant amounts of ammonia under certain conditions. Although ammonia is toxic, because it is highly water soluble its excretion may be facilitated in animals that ingest and excrete large amounts of water. Birdpollinated plants secrete carbohydrate- and water-rich floral nectars that contain exceedingly little protein. Thus, nectarfeeding birds are faced with the dual challenge of meeting nitrogen requirements while disposing of large amounts of water. The peculiar diet of nectar-feeding birds suggests two hypotheses: (1) these birds must have low protein requirements, and (2) when …

Hypothalamic Proopiomelanocortin Neurons Are Glucose Responsive And Express Katp Channels, Nurhadi Ibrahim, Martha A. Bosch, James L. Smart, Jian Qiu, Marcelo Rubinstein, Oline K. Rønnekleiv, Malcolm J. Low, Martin J. Kelly Jan 2003

Hypothalamic Proopiomelanocortin Neurons Are Glucose Responsive And Express Katp Channels, Nurhadi Ibrahim, Martha A. Bosch, James L. Smart, Jian Qiu, Marcelo Rubinstein, Oline K. Rønnekleiv, Malcolm J. Low, Martin J. Kelly

Faculty Publications - Department of Biological & Molecular Science

Hypothalamic proopiomelanocortin (POMC) neurons are critical for controlling homeostatic functions in the mammal. We used a transgenic mouse model in which the POMC neurons were labeled with enhanced green fluorescent protein to perform visualized, whole-cell patch recordings from prepubertal female hypothalamic slices. The mouse POMC-enhanced green fluorescent protein neurons expressed the same endogenous conductances (a transient outward K current and a hyperpolarization-activated, cation current) that have been described for guinea pig POMC neurons. In addition, the selective -opioid receptor agonist DAMGO induced an outward current (maximum of 12.8 1.2 pA), which reversed at K equilibrium potential (EK), in the majority …

The Anorexigenic Fatty Acid Synthase Inhibitor, C75, Is A Nonspecific Neuronal Activator, Kanji A. Takahashi, James L. Smart, Hongyan Liu, Roger D. Cone

Faculty Publications - Department of Biological & Molecular Science

C75, a recently derived compound that potently suppresses feeding and induces weight loss, has been proposed to act mainly by inhibiting fatty acid synthase (FAS) in central neurons that control feeding. For example, normal, fasting- associated, hypothalamic increases in neuropeptide Y (NPY)/Agouti-related protein (AGRP) expression and decreases in proopiomelanocortin (POMC)/cocaine and amphetamine regulated transcript (CART) expression were reported to be blocked by C75. Using loose-patch extracellular recording in acute slices, we tested the effect of C75 on anorexigenic POMC neurons and orexigenic NPY neurons of the hypothalamic arcuate nucleus, which were identified by promoter-driven GFP expression, as well as on …

Intracellular Dynamics Of Lead And Calcium In Lead-Exposed Cultured Neurons Of Aplysia Californica (Mollusca: Opisthobranchia), Catherine T. Tamse Jan 2003

Intracellular Dynamics Of Lead And Calcium In Lead-Exposed Cultured Neurons Of Aplysia Californica (Mollusca: Opisthobranchia), Catherine T. Tamse

Open Access Dissertations

The neurotoxin lead (Pb2+) is a continuing health and environmental problem. However, its neurotoxic mechanism remains unclear. Since calcium (Ca2+) is vital in cellular processes and with Pb2+ reported to interfere with Ca2+-mediated processes, the hypothesis that Pb2+ subverts Ca2+ homeostasis by entering cells through Ca2+ transport pathways and displacing intracellular Ca2+ ([Ca2+] i) was investigated. A multi-technique approach tested this hypothesis. Using cultured Aplysia bag cell neurons, changes in transmembrane Ca2+ flux after Pb2+ exposure were directly measured with the Ca2+-selective self-referencing electrode. Cumulative concentrations of extracellular Pb2+ produced a corresponding and significant (P < 0.05) increase in Ca 2+ efflux, independent of time exposure. Ca2+ channel blockers Co2+, La3+, and verapamil inhibited Pb2+-induced flux. The sarco/endoplasmic reticulum Ca 2+-ATPase pump inhibitors, thapsigargin and cyclopiazonic acid, both elicited a Ca2+ efflux reflecting inhibition of ER pumps and [Ca2+]i release. Furthermore, Pb2+ evoked a moderate and sustained elevation in thapsigargin-stimulated Ca 2+ efflux. For morphological evidence that Pb2+ can mobilize [Ca 2+]i, potassium pyroantimonate (KPA) cytochemistry of bag cells were analyzed. 2% KPA resulted in widespread electron-dense precipitates. 10 mM EGTA + KPA or 200 μM Pb2+ + KPA both produced marked reductions of precipitates in neurons. X-ray microanalysis confirmed antimony (Sb) in precipitates but could not separate the overlapping Ca2+-Sb spectral peaks. However, reduced precipitates in EGTA-incubated cells indicate presence of Ca2+ in intracellular precipitates observed. No apparent pathological changes were correlated to Pb2+ exposure. The effects of Pb2+ on [Ca2+]i using the fluorescent Ca2+ indicator, chlortetracycline (CTC), was examined. Control neurons incubated in 10 μM CTC demonstrated extensive yellow-green fluorescence while reduced fluorescence intensities were seen in cells exposed to 200 μM Pb2+. Multi-sized punctate fluorescence were observed along the perimeter and sometimes, throughout the soma. No neuronal autofluorescence nor neuritic fluorescence in control and Pb2+-exposed cells were detected. Average fluorescence intensities between control and Pb2+-exposed neurons were statistically significant (P < 0.0005). Collectively, these results are consistent with the hypothesis that Pb 2+ perturbs Ca2+ homeostasis via mobilization of [Ca 2+]i. Pb2+ operates through Ca2+ transport pathways as shown by changes in transmembrane Ca 2+ flux, in KPA precipitate reactions, and in CTC fluorescence.

On Democracy As A Shar'i Moral Presumption: Response To Khaled Abou El Fadl, Anver M. Emon Jan 2003

On Democracy As A Shar'i Moral Presumption: Response To Khaled Abou El Fadl, Anver M. Emon

Fordham International Law Journal

Khaled Abou El Fadl has done more in his Article than simply argue the possibility of democracy in the Muslim world. He reminds us that Islam is not a monolithic idea lacking varying contours, possibilities, and dispositions. Whether in talking about an Islamic legal system or an Islamic political system, the most frequent mistake is assuming that there is an ideal vision of such systems that precludes debate, difference, and the continued search for possibilities. Another common mistake is to presume that the limits of an “Islamic political system” are inherent in the existing practice of so-called Muslim States. To …

Why Does The American Constitution Lack Social And Economic Guarantees?, Cass R. Sunstein Jan 2003

Why Does The American Constitution Lack Social And Economic Guarantees?, Cass R. Sunstein

Public Law and Legal Theory Working Papers

Why does the American Constitution lack contain social and economic guarantees, which appear in most contemporary constitutions? This essay explores four possible answers: chronological, cultural, institutional, and realist. The chronological explanation emphasizes the fact that in the late eighteenth century, social and economic rights simply were not on the viewscreen for constitution-makers. The point is correct, but as a complete account, the chronological explanation fails for the simple reason that constitutional meaning changes over time. The institutional explanation emphasizes that Americans typically see constitutional rights not as mere goals or aspirations, but as pragmatic instruments for judicial enforcement. The difficulty …

Land Preservation, John R. Nolon Jan 2003

Land Preservation, John R. Nolon

Elisabeth Haub School of Law Faculty Publications

This exploration of local preservation law and practice recognizes the critical influences of federal, state, and regional governments. Local governments are instruments of state governments; municipalities derive their authority from state statutes. Extensive control over land use has been given to localities, but important powers have been retained. State governments are the partners of federal agencies that are charged with protecting environmental resources that are subject to federal jurisdiction. The land use system that has evolved over the last century in this country is intensely intergovernmental and interdependent. While this system was not designed as a whole and is often …
