Novel synthases and the corresponding nucleic acids encoding such synthases are disclosed herein. Such synthases possess an active site pocket that includes key amino acid residues that are modified to generate desired terpenoid reaction intermediates and products. Synthase modifications are designed based on, e.g., the three-dimensional coordinates of tobacco 5-epi-aristolochene synthase with or without a substrate bound in the active site.

The purpose of this study was to determine the validity of a new assessment tool for Alaska Native clients with alcoholism. A sample of 23 Yup'ik clients at a regional treatment center were interviewed using the Drinker Inventory of Consequences for Alaska Natives (DrInC-AN), an adaption of the Drinker Inventory of Consequence (DrInC), the Alcohol Use Disorders Identification Test (AUDIT) and the Yup'ik Cultural Practices and Traditions (YCPT). These clients were selected, on a voluntary basis during the intake interview to the treatment center. An understanding of the excitation mechanism of the ionospheric molecules during auroral activity is of vital importance for the overall ionospheric understanding including its interaction with the magnetosphere. In this thesis we study two emissions originating from the excited nitrogen molecule ion. The first negative (0,1) emission at 4278 Å originating from the B state, and Meinel (2,0) emission at 7852 Å originating from the A state during moderate to strong aurora have been observed with an imaging spectrograph at Poker Flat, Alaska. A prolonged winter shutdown of the Trans-Alaska Pipeline (TAPS) could cause the fluid in the pipeline to form a gel with sufficient strength to prevent restart. With the economic viability of a North Slope Gas-To-Liquid (GTL) plant improving, understanding the effect the addition of GTL products to TAPS will have on the gel strength of pipeline fluids is essential to quantify the risk of such a project. The present work deals with trace formulas for a half-line Schrödinger operator with long-range potentials. These formulas relate the potential with some scattering data. We generalize some relevant results by Buslaev; Faddeev; Gesztesy, Holden, Simon, and others to the case of square integrable potentials. The relation between the number of the trace formulas and the number of integrable derivatives of the potential is also given.

Disinfection byproducts (DBP) are formed during the reaction of chemical disinfectants with natural organic material (NOM). DBPs are potential carcinogens and are regulated by the US Environmental Protection Agency (USEP A) under the Disinfectant/DBP (D/DBP) rule. High concentrations of NOM in drinking water sources used by Alaska's communities often result in the formation of DBPs during treatment. Since surface water sources in the Arctic are frozen for 6-9 months of the year, communities are often forced to store raw water for treatment during the winter or treat and store enough drinking water during the summer to last through the winter. …

Stratigraphy, Major Oxide Geochemistry, And ⁴⁰Ar/³⁹Ar Geochronology Of A Tephra Section Near Tok, Alaska, Janet Schaefer

Stratigraphy, major oxide geochemistry, and ⁴⁰Ar/³⁹Ar geochronology are used to describe a tephra section near Tok, Alaska, north ofthe Wrangell volcanic field. This tephra section contains seven type II tephras and two type I tephras. Hornblende in a tephra near the base of the section was dated at 627.5 +/- 47.7 ka using the ⁴⁰Ar/³⁹Ar method of single-step total laser fusion. This hornblende-rich tephra is here named the Tetlin Tephra. The age and composition of the Tetlin Tephra indicates that it is likely the product of an explosive phase of activity of Mount Drum volcano, where petrologically similar domes were …

Geology And Gold Mineralization Of The Richardson District, East-Central Alaska, Garth E. Graham

The Richardson district contains multiple granitic units intruding gneiss and schist. The Bald Knob prospect contains gold-bearing quartz veins with the assemblage Bi-̊ Au-̊ hedleyite and high methane fluid inclusions. These features indicate very low fs₂-f₀₂ conditions, lower than any reported for 90 Ma interior Alaska gold systems, including the nearby Democrat and Buckeye prospects. The Bald Knob and Democrat Lode prospects returned ⁴⁰Ar/³⁹Ar ages of 1̃04 and 9̃0 Ma, respectively. Peraluminous dikes possess collisional tectonic signatures and interpreted age of 114 Ma. Younger dikes and 3 km² granite pluton possess arc-type compositions and yield ⁴⁰Ar/³⁹Ar ages of 9̃0 Ma. …

A simultaneous inversion of a large dataset produced robust estimates of regional attenuation for the Lg phase in interior Alaska. Data were collected from summer 1999 through summer 2001 from the Broadband Experiment Across the Alaska Range (BEAAR). Spectra calculated from this dataset were used in a nonlinear simultaneous least squares inversion to determine source factors, path effects and site amplification terms. The average regional attenuation of the Lg phase in interior Alaska was found to be: LgQ(f)=166f⁰·⁵⁸ Variation in regional attenuation has two main characteristics: variation on small spatial scales caused by small crustal heterogeneities, and increased excitation of …

The net mass balance on Gulkana Glacier has been measured since 1966 by the glaciological method, in which seasonal balances are measured at three index sites and extrapolated over large areas of the glacier. Systematic errors accumulate through time in this method; therefore the geodetic balance, in which errors are independent of time, was calculated for comparison to and possible calibration of the glaciological method. Digital elevation models (DEMs) of the glacier in 1974, 1993, and 1999 were prepared and geodetic balances computed, giving -6.0+̲0.5 m of water equivalent (weq) from 1974 to 1993 and -11.8+̲0.5 m weq from 1974 …

Snowshoe hares, Lepus americanus, are a 'keystone' prey species in northern boreal forests and experience population fluctuations of 8-11-years. Despite intense responses of both vegetation and predators to changes in hare densities, landscape-scale comparisons of hare populations in Alaska have been limited to qualitative descriptions. We conducted capture-recapture studies of snowshoe hares at 5 locales in the Tanana valley, from Tok in the east to Clear in the west from 1999 to 2002. Snowshoe hare densities were highest in 1999 (x=6.36 ha⁻¹, SE=0.63) and declined thereafter. We were unable to detect declines in apparent survival during declining densities in our …

Arctic ground squirrels (Spermophilus parryii kenicotti) hibernate underground; experiencing burrows temperatures of -5 to -25C̊. Hibernation consists of 7-9 months of torpor interrupted every 2-3 weeks by 10-20 h arousal episodes with high body temperatures and metabolic rates. Metabolic rates and body temperatures were measured during arousal episodes under temperate and arctic conditions. No difference in cost of arousal episodes was detected. Yet, when the cost of thermogenesis during torpor was included, proportional cost of arousal episodes decreased from 86 to 27% comparing temperate to arctic conditions. The relationship between metabolic rate and body temperature was examined during the recooling …

This study gathered baseline limnological data to investigate the thermal structure, water quality, phytoplankton, and zooplankton of Lake Clark, Alaska. Results indicate Lake Clark is oligotrophic and mixes biannually, but stratification is weak and thermoclines are deep. Longitudinal gradients were seen in measurements of temperature, suspended solids, turbidity, light penetration, algal biomass, and zooplankton density. Wind and tributary inputs determine the thermal regime. Glacially-influenced tributaries drive turbidity and light gradients by introducing suspended solids to the inlet end of the lake. Suspended solids likely create the algal biomass gradient by limiting the light available for photosynthesis in the inlet basin. …

A dynamic actin cytoskeleton in central nervous system (CNS) neurons is pivotal for regeneration. Following acute CNS trauma, the proinflammatory cytokines TNF[alpha] and IL-1[beta] become expressed in cells and induce Rac 1-mediated actin filament reorganization. Also, Rac 1 regulates a NAD(P)H oxidase activity that generates superoxide (·O₂). This study's objective was to determine whether TNF[alpha] and/or IL-1[beta] induce a Rac 1-dependent actin filament reorganization in SH-SY5Y neuroblastoma cells and in chick spinal cord neurons via the signaling intermediate, superoxide. I studied woody browse distribution, production, removal, species composition, twig size, moose diets, and predicted daily intake of resident and migratory moose in the Tanana Flats and adjacent Alaska Range Foothills, Alaska, 1999-2000. Density of moose in these areas was high (1.1 moose/km²). Moose were experiencing density-dependent effects on reproduction and growth, exhibited by low adult twinning rate (6%) and absence of pregnant yearlings, yet 17.5 kg higher 10-month-old calf body weights in the migratory segment. Of all willow, poplar, and paper birch plants sampled, 74% had a broomed architecture, which I attributed to heavy use by moose. Using a …
