Killing Of Mycobacterium Avium Subspecies Paratuberculosis Within Macrophages, John P. Bannantine, Judith R. Stabel

Background: Mycobacterium avium subspecies paratuberculosis (M. paratuberculosis) is a facultative intracellular pathogen that resides within host macrophages during infection of ruminant animals. We examined survival of M. paratuberculosis infections within cultured macrophages to better understand the interplay between bacterium and host.

Results: Serial plating of M. paratuberculosis infected macrophage lysates on Herold's egg yolk medium showed that mycobacterial replication takes place between 0 and 24 hours post-infection. This initial growth phase was followed by a steady decline in viability over the next six days. Use of the rhizomatous perennial forage legume kura clover (Trifolium ambiguum M. Bieb.) has been limited by slow establishment. Mature kura clover responds to liming on some acid soils, but the soil pH required for vigorous growth of young plants is unknown. A factorial greenhouse experiment was conducted with two kura clover cultivars (Rhizo and Endura) and one cultivar of birdsfoot trefoil (Lotus corniculatus L., Norcen) planted in three soil types (Sartell loamy fine sand, Hubbard loamy sand, and Sanborg clay loam) amended with Ca(OH)₂ to obtain six soil pH levels. Traps baited with the synthetic aggregation pheromone of the boll weevil (Anthonomus grandis Boheman) are often used to monitor population fluctuations, distribution, and behavior. However, many factors generate variability in daily captures, making interpretation of trapping data difficult. Previous studies have shown that wind speed in the microenvironment around a trap can greatly affect numbers captured on a given day. It is possible that variation in air movement may also generate variation in trap captures through its effects on the pheromone plume. The history is recounted of developing the sterile insect technique to eradicate the screwworm, Cochliomyia hominivorax (Coquerel), from the Caribbean island of Curacao, Florida and the southeastern U.S. Observations of screwworm biology and challenges faced in conducting these eradication projects are described by the author who worked on all aspects of the research and field operations. Eradication was first demonstrated on Curacao, essentially a 170 mi² outdoor laboratory. The population dynamics of the wild screwworm was determined and overflooding ratios and dispersal patterns essential for population suppression were defined. Eradication was achieved with minimal resources by attacking the pest …

There are many exotic animal pathogens throughout the world that, if introduced into the United States, could have a significant detrimental impact on the health of livestock, agricultural economy, the environment, and public health. Many of these pathogens are arthropod-borne and potential vectors are readily available in the United States. A number of these arthropod-borne pathogens are discussed here as examples that illustrate the potential risk and the consequences of inadvertent introductions. Plants may be more sensitive to carbon dioxide (CO₂) enrichment at subambient concentrations than at superambient concentrations, but field tests are lacking. We measured soil-water content and determined xylem pressure potentials and δ¹³C values of leaves of abundant species in a C3/C4 grassland exposed during 1997-1999 to a continuous gradient in atmospheric CO₂ spanning superambient concentrations (200-560 µmol mol^-1).

Apoptosis is a highly conserved energy-requiring program for non-inflammatory cell death that is important in both normal physiology and disease. Numerous techniques have been used to study apoptosis. In the nervous system, apoptosis is necessary for normal development, but it also occurs in many acute and chronic pathologic conditions. This review places commonly used markers of apoptosis and their detection in the context of what is now known about the process of apoptosis. We review the potential role of apoptosis in nervous system and neurodegenerative disorders (Alzheimer's disease, Parkinson's disease, and amyotrophic lateral sclerosis). This paper provides a selective examination of the current stress management literature and its practices. The author introduces strategies that can be utilized by commercial pilots to handle stressful events in the often-challenging flight environment and establishes a foundation for further empirical research in the area of human stress control in aviation settings. The purpose of this research was to examine Mexican-American 8th grade students' attributions of success and failure in school-related contexts. The relationship between and among these attributions, socio-cultural variables and academic achievement was also explored and analyzed. Attribution theory formed the theoretical foundation for the study.

The study utilized three instruments as the primary sources of self-reported data: the Texas Assessment of Academic Skills (TAAS ), Student Achievement Responsibility Survey ( SARS ) (Ryckman, 1985), and the Short Acculturation Scale for Hispanic Youth ( SASH-Y ) (Barona & Miller, 1994). The primary focus of this study is on illegal weapons trade in Romania and the Caucasus region of the former USSR. Illegal weapons traffic in these regions is linked with both organized crime and terrorist activities. The main source of weapons for conflicts in the Caucasus region is corruption in Russian military circles.

The value of coral reefs for developing countries and areas where tourism is on the rise make the management of the reef a high priority. Akumal, Mexico is no exception. For management to be effective, information for accurately evaluating the condition of the reef is critical. Crabs play an important role in Elkhorn Slough. The three main species found in the slough, that are native to it, and that I will be looking at are Pachygrapsus crassipes, the striped shore crab, Hemigrapsus oregonensis, the yellow shore crab, and Cancer antennarius, the Pacific Rock Crab. There is one non native species of crab that is found in the slough, the European green crab, Carcinus maenas. Not much is known about the population sizes of any of these four crab species. We report the thermal stability of wild type (WT) and 14 different variants of human copper/zinc superoxide dismutase (SOD1) associated with familial amyotrophic lateral sclerosis (FALS). Multiple endothermic unfolding transitions were observed by differential scanning calorimetry for partially metallated SOD1 enzymes isolated from a baculovirus system. We correlated the metal ion contents of SOD1 variants with the occurrence of distinct melting transitions. Altered thermal stability upon reduction of copper with dithionite identified transitions resulting from the unfolding of copper-containing SOD1 species.
