A Novel Family Of Mobile Genetic Elements Is Limited To The Germline Genome In Tetrahymena Thermophila, Jeffrey D. Wuitschick, Jill A. Gershan, Andrew J. Lochowicz, Shuqiang Li, Kathleen M. Karrer

In the ciliated protozoan Tetrahymena thermophila, extensive DNA elimination is associated with differentiation of the somatic macronucleus from the germline micronucleus. This study describes the isolation and complete characterization of Tlr elements, a family of approximately 30 micronuclear DNA sequences that are efficiently eliminated from the developing macronucleus. The data indicate that Tlr elements are comprised of an ~22 kb internal region flanked by complex and variable termini. The Tlr internal region is highly conserved among family members and contains 15 open reading frames, some of which resemble genes encoded by transposons and viruses. The Tlr termini appear to …

This work is an analytical and computational study of the nonlinear interfacial instabilities found in core-annular flows in the presence of surfactants. Core-annular flows arise when two immiscible fluids (for example water and oil) are caused to flow in a pipe under the action of ail axial pressure gradient. In one typical type of flow regime, the fluids arrange themselves so that the less viscous (e.g. water) lies in the region of high shear near the pipe wall, with the more viscous fluid occupying the core region. The Sharp Wave-associated Ripple is a high-frequency, extracellular recording observed in the rat hippocampus during periods of immobility. During the ripple, pyramidal cells synchronize over a short period of time despite the fact that these cells have sparse recurrent connections. Additionally, the timing of synchronized pyramidal cell spiking may be critical for encoding information that is passed on to post-hippocampal targets. Both the synchronization and precision of pyramidal cells is believed to be coordinated by inhibition provided by a vast array of interneurons. We examine the stability of a thin two-dimensional liquid film with a regular electric field applied in a direction parallel to an initially flat bounding fluid interface. We study the distinct physical effects of surface tension, van der Waals and electrically induced forces for a viscous incompressible fluid. The film is assumed to be sufficiently thin, and the surface tension and electrically induced forces are large enough that gravity can be ignored to the leading order. This dissertation has developed, implemented and tested a novel computer based system (AUTOALIGN) that incorporates an algorithm for the alignment of functional Magnetic Resonance Image (fMRI) time series. The algorithm assumes the human brain to be a rigid body and computes a head coordinate system on the basis of three reference points that lie on the directions correspondent to two of the eigenvectors of inertia of the volume, at the intersections with the head boundary. A non-invasive polarized light reflection measurement method to measure the stretch of soft tissue, such as skin, is described. The technique utilizes changes in the reflectivity of polarized light intensity as a monitor of skin stretch. Measurements on in-vitro pigskin and invivo human skin show that the reflectivity of polarized light intensity increases linearly with stretch over a range. The changes in diffusive reflectivity properties of skin result from the alterations that take place in the roughness across the thickness of the skin layers due to stretch. An Analysis Of Field Data For Evidence Of Anaerobic Degradation Of Pah And Btex Plumes At Manufactured Gas Plant Sites, Georgene Mortimer

Field data (including geology, hydrogeology, soil chemistry, groundwater chemistry, and aquifer characteristics) were analyzed at eight MGP sites located in the New Jersey Coastal Plain. These data were used to calculate biodegradation rate constants, which were then compared with laboratory microcosm data and published values. Isopleths were drawn from the field data to indicate the dominant biodegradation mechanisms at each of the sites, and stoichiometric calculations were used to determine the total capacity of the electron acceptors present to degrade the plume contaminants. The objective of the proposed work is to acquire knowledge needed for the development and deployment of manufacturing processes utilizing the enormous technological potential of water ice. Material removal by blasting with ice media such as particles, pellets and slugs was investigated. The ice media was accelerated by entrainment in a fluid stream (air, steam, liquid water, supercritical C02), impact of rotating blades, fluid expansion, etc. The ice-airjet has to replace sand blasting and the ice-waterjet has to replace the abrasive waterjet. BACKGROUND: Alternative mRNA splicing of alpha(i2), a heterotrimeric G protein alpha subunit, has been shown to produce an additional protein, termed salpha(i2). In the salpha(i2) splice variant, 35 novel amino acids replace the normal C-terminal 24 amino acids of alpha(i2). Whereas alpha(i2) is found predominantly at cellular plasma membranes, salpha(i2) has been localized to intracellular Golgi membranes, and the unique 35 amino acids of salpha(i2) have been suggested to constitute a specific targeting signal. RESULTS: This paper proposes and examines an alternative hypothesis: disruption of the normal C-terminus of alpha(i2) produces an unstable protein that fails to localize to plasma …

A research consortium comprised of representatives from several universities and national laboratories has been established as part of this on-going project to generate internal and external dose conversion coefficients for radionuclides produced in spallation neutron sources. Information obtained from this multi-year study will be used to support the siting and licensing of future accelerator-driven nuclear initiatives within the U.S. Department of Energy complex, including the Spallation Neutron Source (SNS) and Accelerator Production of Tritium (APT) Projects. This thesis will explore the ethical dimensions of global agriculture from the productionist paradigm to the land ethic in the context of the sustainable agriculture promoted in the fair trade, shade grown, and organic coffee movements. Much of the sustainable development debate in the environmental ethics literature has centered on justifications (from anthropocentric and nonanthropocentric worldviews) for preserving nature. Since agriculture is an artifact of human sedentary civilization, traditional nonanthropocentric arguments do not suffice because of the tendency to focus exclusively on natural systems. Cardiac hypertrophy is the remodeling or enlargement of the heart exhibited by individuals suffering from heart disease. Cardiac hypertrophy involves two mechanisms, mechanical stresses and humoral changes in the cardiac tissue. One such humoral change involves the Renin Angiotensin System (RAS). The primary fimction of RAS is maintenance of hemodynamics of the body mediated through an effector protein, Angiotensin 11. However, RAS has been shown to play a significant role in the pathophysiology of the heart; contributing to hypertension, cardiac hypertrophy and myocardial infarction. A new image processing method is described for measuring the 3-D coordinates of a complex, biological surface. One of the problems in stereo vision is known as the accuracy-precision tradeoff problem. This thesis proposes a new method that promises to solve this problem. To do so, two issues are addressed. First, stereo vision instrumentation methods are described. This instrumentation includes a camera system as well as camera calibration, rectification, matching and triangulation. Second, the approach employs an array of cameras that allow accurate computation of the depth map of a surface by propagation of correspondences through pair-wise camera views.

A Computational Model Of Spasticity Based On A Decoupling Of The Alpha And Gamma Efferents, Bruno A. Mantilla

It is generally accepted that spasticity results from changes in the excitability of the stretch reflex. This change lowers the threshold of the motoneurons of the spinal cord where the integration of a signal from velocity/position sensors is processed and then fed back to the contracting muscle (alphaextrafusal and gamma-intrafusal fibers). The stretch reflex depends on the initial length of the muscle, the stretch velocity and voluntary activity. The exact sequence of the triggering events remains unknown, is poorly understood and as a result is controversial. The clinical classification scales are mainly subjective and by definition, inaccurate.

Evidence For A Compound In Comstock-Kellog Glands Modulating Premating Behavior In Male Desert Locust, Schistocerca Gregaria, Peter G.N. Njagi, B. Torto

The Comstock-Kellog glands in adult females of certain acridid species, including the desert locust, Schistocerca gregaria have been implicated as a source of volatiles that play a role in mating behavior. A dichloromethane extract of the glands was analyzed for metabolites by gas chromatography, coupled gas chromatography-electroantennographic detection, and mass spectrometry. Coupled gas chromatography-electroantennographic detector (GC-EAD) analysis revealed a component that elicited an electroantennogram response from the antenna of adult male S. gregaria. The compound was identified by GC-MS as pentanoic acid. The levels of the compound in the gland extract varied with age of female locust; it was present …
