Ancient Antibiotics : Tetracycline In Human And Animal Bone From The Dakhleh Oasis, Egypt, Corey Maggiano

HIM 1990-2015

Two decades ago archaeologists in northern Africa discovered evidence that an antibiotic was somehow included in diet of ancient peoples, possibly affecting the health of the population. It has been proposed that the causative organisms are Streptomyces aureofaciens - ubiquitous, mold-like, tetracycline-producing bacteria that could have contaminated grain products. Upon consumption, tetracyclines are incorporated into developing or remodeling bone, remaining observable under ultraviolet light for thousands of years. The current project focuses on an analysis of Roman-Egyptian human and animal bone from the Dakhleh Oasis in southwestern Egypt (100 BC to AD 360). Confocal Laser Scanning Microscopy (CLSM) is used …

Protein Farnesyltransferase And Protein Prenylation In Plasmodium Falciparum, Debopam Chakrabarti, Thiago Da Silva, Jennifer Barger, Steve Paquette, Hetal Patel, Shelley Patterson, Charles M. Allen

Faculty Bibliography 2000s

Comparison of the malaria parasite and mammalian protein prenyltransferases and their cellular substrates is important for establishing this enzyme as a target for developing antimalarial agents. Nineteen heptapeptides differing only in their carboxyl-terminal amino acid were tested as alternative substrates of partially purified Plasmodium falciparum protein farnesyltransferase. Only NRSCAIM and NRSCAIQ serve as substrates, with NRSCAIM being the best. Peptidomimetics, FTI-276 and GGTI-287, inhibit the transferase with IC50 values of 1 and 32 nm, respectively. Incubation of P. falciparum-infected erythrocytes with [H-3]farnesol labels 50- and 22-28-kDa proteins, whereas [H-3]geranylgeraniol labels only 22-28-kDa proteins. The 50-kDa protein is shown to be …

Faculty Bibliography 2000s

To identify human proteins that bind to the Smac and caspase-9 binding pocket on the baculoviral inhibitor of apoptosis protein (IAP) repeat 3 (BIR3) domain of human XIAP, we used BIR3 as an affinity reagent, followed by elution with the BIR3 binding peptide AVPIA microsequencing, and mass spectrometry. The mature serine protease Omi (also known as HtrA2) was identified as a mitochondrial direct BIR3-binding protein and a caspase activator. Like mature Smac (also known as Diablo), mature Omi contains a conserved LAP-binding motif (AVPS) at its N terminus, which is exposed after processing of its N-terminal mitochondrial targeting sequence upon …

Down-Regulation Of Cdc6, A Cell Cycle Regulatory Gene, In Prostate Cancer, Liza D. Robles, Andra R. Frost, Monica Davila, Alan D. Hutson, William E. Grizzle, Ratna Chakrabarti

Faculty Bibliography 2000s

CDC6 plays a critical role in regulation of the onset of DNA replication in eukaryotic cells. We have found that Cdc6 expression is down-regulated in prostate cancer as detected by semiquantitative reverse transcriptase-PCR of prostate cell lines and laser-captured microdissected prostate tissues. This result was substantiated by immunohistochemical analysis of paraffin-embedded tissue sections and immunoblot analysis of benign (BPH-1) and adenocarcinomatous prostatic cells. Furthermore, a 100-fold reduction in the transcription efficiency of the Cdc6 promoter-luciferase construct was noted in the metastatic PC3 cells compared with that in BPH-1 cells. Concentration of the E2F and Oct1 transcription factors that have putative …

Method Of Adjustments Versus Method Of Constant Stimuli In The Quantification Of Accuracy And Precision Of Rendered Depth In Head-Mounted Displays, J. P. Rolland, C. Meyer, K. Arthur, E. Rinalducci Jan 2002

Method Of Adjustments Versus Method Of Constant Stimuli In The Quantification Of Accuracy And Precision Of Rendered Depth In Head-Mounted Displays, J. P. Rolland, C. Meyer, K. Arthur, E. Rinalducci

Faculty Bibliography 2000s

The utilization of head-mounted displays (HMDs) in high-end applications such as medical, engineering, and scientific visualization necessitates that the position of objects be rendered accurately and precisely. Accuracy and precision of rendered depth for near-field visualization were measured in a custom-designed bench prototype HMD. Experimental results were compared to theoretical predictions established from a computational model for rendering and presenting virtual images by Robinett and Rolland (1992). Such a theoretical model provided the necessary graphics transformations required so that rendered virtual objects be perceived at the rendered depth in binocular HMDs, Three object shapes of various sizes were investigated under …

Field Detection And Monitoring Of Explosives, Jehuda Yinon Jan 2002

Field Detection And Monitoring Of Explosives, Jehuda Yinon

Faculty Bibliography 2000s

Detection of explosives in situ is of major importance in several applications: finding hidden explosives in airport luggage and in mail; screening of personnel for concealed explosives; environmental monitoring of explosives-contaminated sites; and, detection of buried landmines. Mobile and hand-held detectors have been developed for the various applications. They can be sub-divided into three classes: vapor and particle detectors; radiation detectors; and, biochemical detectors. An overview of the various explosives detectors and their principles of operation is presented. (C) 2002 Published by Elsevier Science B.V. All rights reserved.

Parameters Affecting Partitioning Of 6 Pcb Congeners In Natural Sediments, Eid A. Alkhatib, Carl Weigand Jan 2002

Parameters Affecting Partitioning Of 6 Pcb Congeners In Natural Sediments, Eid A. Alkhatib, Carl Weigand

Chemistry & Physics Faculty Publications

Polychlorinated biphenyls (PCBs) released by bottom sediments were determined by experiments in which the sediments were artificially resuspended using sediment contaminated with PCBs in a particle entrainment simulator (PES). Sediment cores, spiked with PCBs, were collected from the Housatonic River in Connecticut and run in the PES at simulated shear stresses from 0 to 0.6 N m(-2). Experimental results from these simulations have shown that mean concentration of PCBs in the solid phase for sites with high volatile organic carbon (VOC) were significantly greater than samples with low VOC; the reverse was true for the water phase. In addition, on …

Primaquine-Induced Hemolytic Anemia: Formation And Mechanism Of Action Of The Hemotoxic Metabolite 6-Methoxy-8-Hudroxylamoniquinoline (Maq-Noh), Laura Jane Campbell-Bolchoz Jan 2002

Primaquine-Induced Hemolytic Anemia: Formation And Mechanism Of Action Of The Hemotoxic Metabolite 6-Methoxy-8-Hudroxylamoniquinoline (Maq-Noh), Laura Jane Campbell-Bolchoz

MUSC Theses and Dissertations

Primaquine is an important antimalarial drug that is often dose-limited in therapy by the onset of hemolytic anemia. It is well accepted that this toxicity is due to the action of its metabolites and not the parent compound. However, the toxic species have not been identified and little is known about the mechanism underlying primaquine-induced red cell injury. Previous studies to identify the hemotoxic metabolites have focused on the redox active phenolic derivatives of primaquine and have shown that a number of these compounds are capable of decreasing red cell GSH levels and oxidizing oxyhemoglobin to methemoglobin. However, these derivatives …

Environmental Justice In An Era Of Devolved Collaboration , Sheila R. Foster Jan 2002

Environmental Justice In An Era Of Devolved Collaboration , Sheila R. Foster

Faculty Scholarship

Environmental decision-making is undergoing a profound shift. Traditional forums and processes are being displaced by mechanisms emphasizing local, "place-based" decision-making. These emerging decision-making mechanisms are orchestrated through collaborative processes featuring stakeholders from both the public and private sectors. This transformation is evident in a number of recent governmental initiatives, including those by the Environmental Protection Agency ("EPA"), most notably its Community-Based Environmental Protection ("CBEP") initiative. Other federal agencies, particularly those with land or species management responsibilities, have similarly advocated a greater role for local decision-makers and collaborative problem-solving. This Article examines the points of convergence and divergence between devolved collaboration …
