Characterization Of Salmonella Serovars By Pcr–Single-Strand Conformation Polymorphism Analysis, S. Nair, Kwai Lin Thong, T. Pang, M. Altwegg

PCR-restriction fragment length polymorphism (PCR-RFLP) and PCR–single-strand conformation polymorphism (PCR-SSCP) analyses were carried out on the 1.6-kb groEL gene from 41 strains of 10 different Salmonella serovars. Three HaeIII RFLP profiles were recognized, but no discrimination between the serovars could be achieved by this technique. However, PCR-SSCP analysis of the groEL genes of various Salmonella serovars produced 14 SSCP profiles, indicating the potential of this technique to differentiate different Salmonella serovars (interserovar differentiation). Moreover, PCR-SSCP could differentiate strains within a subset of serovars (intraserovar discrimination), as three SSCP profiles were produced for the 11 Salmonella enterica serovar Enteritidis strains, and …

In 1996, the Council for Agricultural Science and Technology (CAST) published a report entitled Integrated Animal Waste Management. One of the recommendations in that report was to “change animal diets to decrease nutrient outputs” (CAST 1996, 1). Since that time, concentration of animal production units has continued, public concern about the environmental effects of animal manure has increased, and the Environmental Protection Agency (EPA) has proposed more restrictive requirements for concentrated animal feeding operations (CAFO regulations).

A dynamic model is proposed in order to simulate sandhopper (Talitrus saltator) populationdynamics under various scenarios. It was built under the Stella simulation software and so far it has attainedthe calibration and verification stages. We discuss the motivations for, and software design concepts underpinning, the development of a regional water quality model. The Environmental Management Support System (EMSS) was developed to predict daily fluxes of runoff, total suspended sediment, total nitrogen and total phosphorous through a large-scale river network. It was built using a custom environmental modelling framework called Tarsier, founded on the Borland C++ Builder rapid application development environment. Three autonomous models are integrated within the EMSS, but are loosely coupled so that alternative models could be retrofitted into the system if desired. Model development is a difficult and timely process involving various steps to create, check and improve the model. Typically all kinds of software tools are used to facilitate this process. Central to model development is the model definition and we show that, apart from calculating model results, it can be used to tailor some of the tools used to the model at hand. This speeds up model development and might lead to better models.

Natural resource management decisions are complicated by multiple property rights, management objectives, and stakeholders with varying degrees of influence over the decision making process. Managers must consider all opinions when they develop management policies. Often this type of information is not understood until after a decision has been made, which can result in wasted time and effort. Few farmers and ranchers adopt agricultural software such as decision support systems (DSS). While numerous decision aids are available, most are too difficult for producers to use, exclude components (e.g., economic budgeting, weeds, multicriteria decision analysis) necessary for meaningful use on farms and ranches, and usually suffer from poor understanding by scientists of producer needs and how they process information. In this paper, we present a conceptual framework for integrating ecological models into a process for management of complex ecological systems. This conceptual framework, that we call Adaptive Ecological Risk Analysis, is an iterative process based upon ecological modeling, the development and implementation of management alternatives, and the evaluation of their effectiveness. Adaptive Ecological Risk Analysis depends heavily on the use of ecological models to perform both prospective and retrospective risk assessments and to analyze the outcomes of alternative management strategies. EGPE is an object-oriented model, which estimates the environmental distribution ofcontaminants within different environmental compartments and the specific risk associated with thesesdistributions and final concentrations. This model evaluates environmental impacts related to: a) the use ofsludge/manure as fertiliser, and b) direct discharges of slurry or effluents from treatment plants to waterstreams. An agent based system (HOOFS) has been developed in order to examine the interactions between foraging animals and a spatially explicit description of the environment, including vegetation growth processes. HOOFS makes use of the agent-based approach to develop a model of foraging in which animal decision making processes are explicitly modelled. The model has evolved from one where foraging decisions are based on a simple biased random walk model with an information weighting parameter, through one in which animals decide between direct walking towards an object as against continuous foraging, ending with a model where memory is used. Application of a new graphic multi-criterion model-based approach to integrated assessment and screening of environmental strategies is described. The approach is based on integration of mathematical models and visualization of the Pareto-efficient (non-dominated) frontier. In the case of linear integrated models, the visualization is based on application of the Interactive Decision Maps (IDM) technique that visualizes the frontier in the form of multiple animated decision maps provided on-line. An approach is proposed for evaluating water management strategies in river basins. Reasonably large sets of criterions and objectives are manipulated during a three-phase process by appropriate shrinking and enlarging related decision hierarchies. In the first phase an unrestricted set of management interests are grouped into (1) long-term, and (2) middle- and short-term decision context by creating decision hierarchies and evaluating them by the Analytic Hierarchy Process (AHP). After refining the hierarchies by deleting dominated elements, management plans are introduced at their fingertips and so enlarged hierarchies are repeatedly evaluated by AHP in the second phase. In the past, models have focused on one environmental compartment, a single organism, or specific process. All of these topics are limited in scope and often fail to take into account the more complex interactions of the real world. We have endeavored to change this practice by combining environmental transport models with an animal movement model and a physiologically based toxicokinetics (PBTK) model that has an embedded effects model.
