β-Glucuronidase Assay For The Detection Of Escherichia Coli In Foods, Seref Tagi Jan 2001

β-Glucuronidase Assay For The Detection Of Escherichia Coli In Foods, Seref Tagi

Open Access Dissertations

Enzyme assays were developed for the measurement of β-D-glucuronide (GUS) as a rapid detection method for E. coli in food. A fluorescent assay utilizing 4-methylumbelliferyl-β-D-glucuronide as substrate was optimized and tested with growth cultures of E. coli. A relationship between E. coli and the level of GUS activity was established. A chemiluminescent assay using a 1,2-dioxetene derivative as substrate was compared to the fluorescent assay, and was found to be 10x more sensitive. Anti-E. coli GUS antibodies were covalently immobilized on magnetic beads and used in a 30 min immunocapture method. GUS was recovered from E. coli grown at 37°C …

Characterization Of Interfacial Fracture Using Strain Fields, Vittorio Ricci Jan 2001

Characterization Of Interfacial Fracture Using Strain Fields, Vittorio Ricci

Open Access Dissertations

An experimental study was conducted to develop and introduce strain gage techniques as an effective and simple alternative experimental method to investigate interfacial fracture in isotropic-isotropic and isotropic-orthotropic bimaterials. The study looked to develop the requisite strain field equations and experimental techniques to obtain static and dynamic interfacial fracture parameters using strain gages and, in the static case, comparing to several optical experimental methods. In the first part of the study, the strain gage technique was compared to three widely used experimental techniques--photoelasticity, coherent gradient sensing (CGS), and caustics--as applied to the stationary interfacial crack problem. These experimental methods were …

Multivalent Metadata : Exploiting The Layers Of Meaning In Digital Resources, Alison M. Anderson Jan 2001

Multivalent Metadata : Exploiting The Layers Of Meaning In Digital Resources, Alison M. Anderson

Theses : Honours

The rapid growth of the World Wide Web was due in part to the simplicity of the Hypertext Markup Language (HTML). It is anticipated that the next generation of web technology, coined the Semantic Web, by Tim Berners-Lee (1989, p. 1), will be driven by the Extensible Markup Language (XML). The XML suite of technologies provides a framework for the application of metadata, and hence semantic information, to web resources. Advantages of a semantic web include improved sharing and reuse of resources, enhanced search mechanisms and knowledge management. The knowledge or meaning contained in digital information may vary according to …

Guidelines For Development Of Courses For Delivery Over The Iowa Communications Network, Gary F. Hasman Jan 2001

Guidelines For Development Of Courses For Delivery Over The Iowa Communications Network, Gary F. Hasman

CCAC Theses and Dissertations

This paper examines the quality of education as it relates to the Iowa Communications Network (ICN). It reviews the literature to determine a working definition of quality that was used to create a list of characteristics desirable in teachers who use technology. A list of such teachers was solicited from three administrative committees of the ICN and from the directors of the state's 15 area education agencies. Four teachers were selected from the list and their approach to creating programs for delivery over the ICN was examined. Personal interviews were used to discover commonalities among the four teachers' approaches to …

Tyrosine And Phenylalanine Catabolism Bylactobacillus Cheese Flavor Adjuncts, S. Gummalla, Jeff Broadbent Jan 2001

Tyrosine And Phenylalanine Catabolism Bylactobacillus Cheese Flavor Adjuncts, S. Gummalla, Jeff Broadbent

Nutrition, Dietetics, and Food Sciences Faculty Publications

Bacterial metabolism of Tyr and Phe has been associated with the formation of aromatic compounds that impart barny-utensil and floral off-flavors in cheese. In an effort to identify possible mechanisms for the origin of these compounds in Cheddar cheese, we investigated Tyr and Phe catabolism by Lactobacillus casei and Lactobacillus helveticus cheese flavor adjuncts under simulated Cheddar cheese-ripening (pH 5.2, 4% NaCl, 15°C, no sugar) conditions. Enzyme assays of cell-free extracts indicated that L. casei strains catabolize Tyr and Phe by successive, constitutively expressed transamination and dehydrogenation reactions. Similar results were obtained with L. helveticus strains, except that the dehydrogenase …

Comparison Of Student Performance And Attitude In A Lecture Class To Student Performance And Attitude In A Telecourse And A Web-Based Class, William J. Ryan Jan 2001

Comparison Of Student Performance And Attitude In A Lecture Class To Student Performance And Attitude In A Telecourse And A Web-Based Class, William J. Ryan

CCAC Theses and Dissertations

Increasing numbers of students are returning to school and choosing alternatives to the lecture method of instruction. Using technology to reach students is a solution colleges and universities are evaluating and implementing with the goal of increasing enrollment and reducing the cost of instruction. This research examines the impact two technology-based delivery systems have on the student's learning experience compared with an equivalent classroom-based instructional method.

Academic institutions are being asked to respond to the rapid changes faced by the communities they serve especially as current workers return to join new students in obtaining knowledge and skills needed in today's …

First Person Singular (Plural), Mishel Filisha Jan 2001

First Person Singular (Plural), Mishel Filisha

Honors Capstones

First Person Singular (Plural) is a script and performance piece that explores female/feminine identity. The script, set for five female performers, is comprised of eight first person monologues, one group interactive sketch and an additional monologue that runs intermittently throughout the piece. The semi-autobiographical monologues discuss issues of particular concern to women such as rape, body image and the delegitimation of the feminine in Western religious tradition. Other monologues discuss topics of broad social concern such as acknowledging narrative bias, the silencing of democratic voice, and the importance of exploring one’s personal history and intellectual histories. Each monologue stands alone …

Changing Horses In Mid-Stream: The Copyright Office's New Rule Makes Broadcasters Pay For "Streaming" Their Signals Over The Internet, Samuel Fifer, Gregory R. Naron Jan 2001

Changing Horses In Mid-Stream: The Copyright Office's New Rule Makes Broadcasters Pay For "Streaming" Their Signals Over The Internet, Samuel Fifer, Gregory R. Naron

Vanderbilt Journal of Entertainment & Technology Law

With traditional, over-the-air broadcasts, radio stations do not have to start paying license fees to rights holders if they get a bigger transmitter or technology allows their signal to reach more listeners. That much, not even the RIAA will dispute. So why should it matter if stations use the Internet to reach additional listeners? The law shouldn't be interpreted to discourage radio stations from using new Internet technology to do the same thing they're doing now--especially since Congress itself expressly said it did not want to "[hamper] ... new technologies" or "impose new and unreasonable burdens" on broadcasters. And make …
