Dendritic Cells In Transplantation - Friend Or Foe?, Robert I. Lechler, Wan F. Ng, Ralph M. Steinman

The existence of two sets of dendritic cells in transplantation (of donor and recipient origin) poses unique problems in alloimmunity and tolerance. Donor DCs are potent stimulators of the direct pathway, in which recipient T cells respond to peptides presented on donor MHC products or to the donor MHC molecules themselves. Reduced DC function in the direct pathway is used to explain the acceptance of certain allografts that have been depleted of passenger leukocytes. Reciprocally, purified donor DCs powerfully stimulate the rejection of these grafts as well as the mixed leukocyte reaction by purified allogeneic T cells in culture. Donor …

Recombinant canarypox virus vectors containing human immunodeficiency virus type 1 (HIV-1) sequences are promising vaccine candidates, as they replicate poorly in human cells. However, when delivered intramuscularly the vaccines have induced inconsistent and in some cases transient antigen-specific cytotoxic T-cell (CTL) responses in seronegative volunteers. An attractive way to enhance these responses would be to target canarypox virus to professional antigen-presenting cells such as dendritic cells (DCs). We studied (i) the interaction between canarypox virus and DCs and (ii) the T-cell responses induced by DCs infected with canarypox virus vectors containing HIV-1 genes. T Cell Studies In A Peptide-Induced Model Of Systemic Lupus Erythematosus, Magi Khalil, Kayo Inaba, Ralph M. Steinman

We have previously reported that immunization with a peptide mimetope of dsDNA on a branched polylysine backbone (DWEYSVWLSN-MAP) induces a systemic lupus erythematosus-like syndrome in the nonautoimmune BALB/c mouse strain. To understand the mechanism underlying this breakdown in self tolerance, we examined the role of T cells in the response. Our results show that the anti-foreign and anti-self response induced by immunization is T cell dependent and is mediated by I-Ed-restricted CD4+ T cells of the Th1 subset. The role of the rodent temporal cortex in relational perception was investigated. Eighteen Long Evans rats were separated into three surgical groups (sham, partial striate, and Te2/3) and trained on a preoperative and postoperative discrimination task. The rats were then presented a series of relational judgement trials consisting of both between category judgments and within category judgments. It was hypothesized that the rats with Te2/3 lesions would perform as well as controls on the preoperative and postoperative discrimination task, but show a considerable behavioral dysfunction on the tests of visual relational perception. In this article, the literary and archaeological evidence for burial practices that can be associated with the English expression ‘to be disposed of like rubbish’ are discussed. These disposal methods (átaphon rhíptesthai) include the exposure of corpses to carrion animals, to the elements, as well as the disposal of corpses into the sea, pits or natural fissures without burial rites. They also include cases in which graves were dug up in order to throw their contents out. The effect of space and furnishings, as measured by the Early Childhood Environment Rating Scale – Revised (ECERS-R), on the School Readiness Composite (SRC) of the Bracken Basic Concept Scale –Revised (BBCS-R) was investigated. The data analyzed was collected as part of the West Virginia Educare Initiative and included the SRC from 65 preschool students, ranging in age from 2 years, 6 months to 5 years, 0 months. The first subtest score on the ECERS-R, space and furnishings, was also calculated for each child’s classroom. The purpose of this study was to determine whether Durkheim’s concept of anomie applies to National Football League players. This study was an attempt to discover if and why NFL players are more likely to commit deviant acts. Why do these players that seemingly have everything throw their lives away by committing crimes? This study attempts to address these questions by linking Durkheim’s classic anomie theory with the deviance in the NFL.

Strip mining causes major disturbances of the natural environment. One such disturbance is the creation of valley fills, which often fill in the headwaters of small streams. Runoff from these valley fills can cause heavy siltation as well as acid and heavy metal deposition downstream. One way to combat this problem is through the construction of sedimentation ponds, which slow down the flow of water so that sediment can settle out and water chemistry can be altered before the water is discharged into the stream. The presence of Escherichia coli (E. coli) indicates recent fecal contamination from either animal or human sources. To solve this growing problem one must identify the source(s) of such contamination by creating a database of DNA profiles of£. coli isolates. Hence, tracking the source(s) of E. coli can help link water contamination to animal source(s) of contamination. The central Appalachian region of the United States receives some of the highest inputs of nitrogen (N) due to acidic deposition in the nation. It is believed that these high could levels contribute to a decline in forest soils within the next 50 to 70 yrs. This study examines factors that influence spatial variability in N-treated and untreated watersheds of the Fernow Experimental Forest, Parsons, West Virginia. The chemistry of transport, storage, energy production, and reproduction in physiological systems is controlled by the actions and interactions of proteins. A species full complement of these amazing machines is often referred to as its proteome. The new science of proteomics utilizes a set of powerful and evolving tools to characterize the nature of the proteome, and further to fully elucidate the functional attributes of the proteins and their associations.

Downstream impacts of contour surface mining and valley fill construction were evaluated utilizing both water chemistry and EPA approved Rapid Bioassessment Protocol III. Wiley Branch was sampled in February of 1999 before mining perturbation. Five downstream stations were established with benthic macroinvertebrates and water samples collected seasonally for a period of sixteen months. Water chemistry parameters, pH, total aluminum, iron, and manganese, total suspended solids, total dissolved solids, sulfates, alkalinity, acidity and specific conductivity fluctuated but generally remained within acceptable governmental limits. In the Skn model of autoimmunity, pathogenic T cells sensitized to Skn epidermal antigens (Skn-immune spleen cells or SIS cells) elicited disease in the form of skin lesions when adoptively transferred into shaved immunoincompetent syngeneic adult recipients (‘lesion-forming animals’) while concomitant injection of immunoregulatory normal spleen cells at twice the concentration (2 x normal spleen cells or 2XNS cells) of SIS cells significantly lessened lesion development (‘lesion-controlling animals’) by Day 7 post cell transfer. In a time-course RT-PCR cytokine analysis of skin from lesion-forming and lesion-controlling animals, IL-7 mRNA was significantly elevated in lesion-controlling skin on Day 7 while no …

Research was conducted on 60 subjects from mental health agencies in order to determine that childhood anxiety and depression can be differentiated. The 60 subjects were given the CDI and the RCMAS self-report measures in order to be placed in groups. This allowed the subjects to be divided into 4 groups: Anxious, Anxious/Depressed, Depressed and Not Anxious/Not Depressed. The 4 groups were then given 2 additional self-report measures, the STAIC and FSSC -R to measure whether Anxious and the Anxious/Depressed children exhibited more fears and anxious traits than did the Depressed and Not Anxious/Not Depressed children. Very few genera of vertebrates demonstrate variation in body size, color, pattern, and life history traits to the extreme that is demonstrated in the genus Desmognathus. Exact distribution of Desmognathus, as well as other genera of salamanders, is very difficult to outline because of hybridization between certain desmognathine species. Tooth morphology, tail keeling, head and body shape, and geographic location are factors used in the separation among species in this genus. The Black Mountain salamander, Desmognathus welteri, is a relatively large brownish-green salamander that possesses morphological characters associated with a semi-aquatic lifestyle. The history of this species in the state of West Virginia is an interesting one. Due to taxonomic confusion and the lack of a thorough investigation of the species in the state, little was known about the species until recently. Surveys for Desmognathus welteri in southern West Virginia revealed five new populations. This makes 17 localities for the species in the state.
