Glutamate Transmission In The Nucleus Accumbens Mediates Relapse In Cocaine Addiction, Jennifer L. Cornish, Peter W. Kalivas

Elevated dopamine transmission in the nucleus accumbens is thought to be a primary mediator of addiction to cocaine. However, repeated exposure to cocaine is associated with the recruitment of glutamate transmission. This poses the possibility that the behaviors characterizing cocaine addiction, such as craving-induced relapse, may not be preferentially mediated by dopamine transmission. An animal model of relapse was used to demonstrate that glutamate, and not dopamine transmission in the nucleus accumbens, is a primary mediator of cocaineinduced reinstatement of drug-seeking behavior. We identified hyaluronan synthase-2 (Has2) as a likely source of hyaluronan (HA) during embryonic development, and we used gene targeting to study its function in vivo. Has2-/- embryos lack HA, exhibit severe cardiac and vascular abnormalities, and die during midgestation (E9.5-10). Heart explants from Has2-/- embryos lack the characteristic transformation of cardiac endothelial cells into mesenchyme, an essential developmental event that depends on receptor-mediated intracellular signaling. This defect is reproduced by expression of a dominant-negative Ras in wild-type heart explants, and is reversed in Has2-/- explants by gene rescue, by administering exogenous HA, or by expressing activated Ras. Publishing online is an increasingly prevalent means for scholars to test their ideas. But what of its challenges? Focusing on an ill-fated Web site dedicated to the polemic French writer Louis-Ferdinand Céline, and on a proposed hypertextual edition of his most multilinear and multisequential work, this paper asks how to reconcile the need of academics with the bullheadedness of publishers who resist the renegotiation of copyright and the marketplace it (once) enabled.

An outbreak of encephalitis occurred in New York City in late August 1999, the first caused by West Nile virus in North America. Histopathologic and immunopathologic examinations performed on human autopsy materials helped guide subsequent laboratory and epidemiologic investigations that led to identification of the etiologic agent.

High School Teachers' Perceptions Of School-Related Violence: Effects On Fear Of Victimization And Perceived Risk, Brandon Ungetheim

Using a sample of 204 high school teachers from nine different counties in Kentucky, this study examined the predictors of both teachers1 fear and perceived risk of victimization at school in an attempt to learn more about this adult population. The predictors that were analyzed on both fear and perceived risk of victimization are as follows: age, sex, school location (metropolitan/nonmetropolitan), victimization experience, indirect victimization experience, and perceived seriousness of school violence. Results indicate that, sex, school location, victimization experience, and perceived seriousness of school violence were all significant predictors of both teachers' fear and perceived risk of victimization. Cyclooxygenase-2 (COX2) is a primary inflammatory mediator that converts arachidonic acid into precursors of vasoactive prostaglandins, producing reactive oxygen species in the process. Under normal conditions COX2 is not detectable, except at low abundance in the brain. This study demonstrates a distinctive pattern of COX2 increases in the brain over time following traumatic brain injury (TBI). Quantitative lysate ribonuclease protection assays indicate acute and sustained increases in COX2 mRNA in two rat models of TBI. In the lateral fluid percussion model, COX2 mRNA is significantly elevated (>twofold, p < 0.05, Dunnett) at 1 day postinjury in the injured cortex and bilaterally in the hippocampus, compared to sham-injured controls. In the lateral cortical impact model (LCI), COX2 mRNA peaks around 6 h postinjury in the ipsilateral cerebral cortex (fivefold induction, p < 0.05, Dunnett) and in the ipsilateral and contralateral hippocampus (two- and six-fold induction, respectively, p < 0.05, Dunnett). Increases are sustained out to 3 days postinjury in the injured cortex in both models. Further analyses use the LCI model to evaluate COX2 induction. Immunoblot analyses confirm increased levels of COX2 protein in the cortex and hippocampus. Profound increases in COX2 protein are observed in the cortex at 1-3 days, that return to sham levels by 7 days postinjury (p < 0.05, Dunnett). The cellular pattern of COX2 induction following TBI has been characterized using immunohistochemistry. COX2-immunoreactivity (-ir) rises acutely (cell numbers and intensity) and remains elevated for several days following TBI. Increases in COX2-ir colocalize with neurons (MAP2-ir) and glia (GFAP-ir). Increases in COX2-ir are observed in cerebral cortex and hippocampus, ipsilateral and contralateral to injury as early as 2 h postinjury. Neurons in the ipsilateral parietal, perirhinal and piriform cortex become intensely COX2-ir from 2 h to at least 3 days postinjury. In agreement with the mRNA and immunoblot results, COX2-ir appears greatest in the contralateral hippocampus. Hippocampal COX2-ir progresses from the pyramidal cell layer of the CA1 and CA2 region at 2 h, to the CA3 pyramidal cells and dentate polymorphic and granule cell layers by 24 h postinjury. These increases are distinct from those observed following inflammatory challenge, and correspond to brain areas previously identified with the neurological and cognitive deficits associated with TBI. While COX2 induction following TBI may result in selective beneficial responses, chronic COX2 production may contribute to free radical mediated cellular damage, vascular dysfunction, and alterations in cellular metabolism. These may cause secondary injuries to the brain that promote neuropathology and worsen behavioral outcome.

George Nicholas Sanders' involvement in regional, national, and international affairs in the mid-nineteenth century significantly shaped the history and unique character of America, as perceived by both Americans and Europeans. Sanders influenced the course of national political events not by idealistic leadership but by active participation. No one has yet written a biography on George N. Sanders, even though he played a prominent role in the annexation of Texas, Young America, presidential elections, diplomatic affairs, and in the Confederacy. Historians often ignored or slighted him because he tended to wield his influence behind-the-scenes. Hence, Sanders' true significance was often masked …

The purpose of this seminar is to acquaint participants with the ever-changing characteristics, often conflicting issues of causation, and various proposed solutions to the myriad of problems associated with gangs in schools. Special attention is given to the evolving nature of gangs in K-12 educational institutions (e.g., new types of juvenile groups developing and their associated behavior). Traditional subjects such as gang recruitment, initiation, and criminal activity are examined by discussing the many ways they manifest themselves in the school setting. Human Conflicts with Wildlife: 2002 Symposium
Since the late 1980s, results of oral vaccination trials in several states have provided growing evidence that this vaccination method may be effective for controlling the spread of rabies in raccoons (Procyon lotor). This study examines the economic feasibility of using oral vaccination on a larger scale than previous trials. We analyze the benefits and costs associated with a hypothetical barrier that would stretch from Lake Erie to the Gulf of Mexico, combining natural geographic features provided by the Appalachian Mountains with oral vaccination zones.
