The purpose of this thesis research was to study the natural history of the northern spring salamander Gyrinophilus p. porphyriticus in West Virginia. The objectives of this study were to determine the environmental characteristics of the habitat of Gyrinophilus p. porphyriticus, to determine its reproductive biology, and then examine non-reproductive aspects of the natural history of the species. Study sites were located at the Westvaco Wildlife and Ecosystem Research Forest (WWERF) in Randolph County, West Virginia. Four sites on three streams were divided into two 25m transects. In February of 1999, the chief of the U.S. Forest Service, Michael Dombeck, placed a moratorium on road building on most roadless areas. In October, President Clinton put forth an initiative to prohibit road building on 40 million acres of roadless area. Such modifications in Forest Service land management decisions is not new as suggested by Char Miller in this look back at early grazing decisions by Pinchot. To be proactive and reactive at the same time in relation to changing social pressures and political realties may be the legacy of the agency.

Locomotion In Sturgeon: Function Of The Pectoral Fins, C. D. Wilga, G. V. Lauder

Pectoral fins are one of the major features of locomotor design in ray-finned fishes and exhibit a well-documented phylogenetic transition from basal to derived clades. In percomorph fishes, the pectoral fins are often used to generate propulsive force via oscillatory movements, and pectoral fin propulsion in this relatively derived clade has been analyzed extensively. However, in the plesiomorphic pectoral fin condition, exemplified by sturgeon, pectoral fins extend laterally from the body in a generally horizontal orientation, have been assumed to generate lift to balance lift forces and moments produced by the heterocercal tail, and are not oscillated to generate propulsive …

Phylogenetic Relationships In Nuphar (Nymphaeaceae): Evidence From Morphology, Chloroplast Dna, And Nuclear Ribosomal Dna, Donald Padgett, D. H. Les, G. E. Crow

The genus Nuphar consists of yellow-flowered waterlilies and is widely distributed in north-temperate bodies of water. Despite regular taxonomic evaluation of these plants, no explicit phylogenetic hypotheses have been proposed for the genus. We investigated phylogenetic relationships in Nuphar using morphology and sequences of the chloroplast gene matK and of the internal transcribed spacer (ITS) regions of nuclear ribosomal DNA. Two major lineages within Nuphar are consistently resolved with the morphological and molecular data sets. One lineage comprises New World taxa and the other represents a primarily Old World lineage. Integrins and vascular cell adhesion molecule-1 (VCAM-1) are required for normal placental development. In this study, integrin subunits α4, αv, β1, and β3, and VCAM-1 were investigated for expression in uteroplacental units (gestation day [g.d.] 6 and 8) and placentas (g.d. 10, 12, 14, 16, and 18) of Swiss-Webster mice. All subunits and VCAM-1 mRNA (identified by reverse transcriptase polymerase chain reaction [RT-PCR]) and protein (detected by immunofluorescence) were present in all tissues throughout gestation. VCAM-1 was expressed strongly in the ectoplacental cone and trophoblast giant cells, α4 was expressed strongly by trophoblast giant cells and moderately by spongiotrophoblast and …

Screening Pinus Sylvestris Grown For The Production Of Christmas Trees For Resistance To Western Gall Rust Peridermium Harknessii Using Different Sources Of Aeciospores, Todd A. Burnes, Jennifer Juzwik, Robert A. Blanchette

Results showed a moderate to high susceptibility of Pinus sylvestris to western gall rust Peridermium harknessii from Pinus sylvestris in Michigan and Pinus banksiana in Minnesota. In general, Pinus sylvestris seed sources were more susceptible to aeciospores collected from Pinus sylvestris than aeciospores collected from Pinus banksiana.

Effect Of Human Disturbance On Small Mammal Communities In Itasca State Park, Minnesota, Nancy L. Staus, Kathleen Conforti, Leslie N. Clapper, Jennifer A. Longhenry, C. C. Schoenbauer, Michael S. Rentz, John R. Tester

Journal of the Minnesota Academy of Science

We determined effects of different levels of human disturbance on small mammal richness and relative abundance from live-trapping data obtained in Itasca State Park in northwestern Minnesota. We developed a quantitative measure of human disturbance based on disturbance units and trapped small mammals on three study sites, each reflecting a different level of disturbance. Our data revealed that small mammal diversity decreased with increasing human disturbance. Amount of ground cover and litter depth also appeared to be important in explaining differences in the demographic patterns of small mammals among sites.

Cloning And Sequencing Of A Cellobiohydrolase Gene From Trichoderma Harzianum Fp108, Patrick Guilfoile, Ron Burns, Zu-Yi Gu, Matt Amundson, Fu-Hsian Chang

A cbh1 cellobiohydrolase gene was cloned and sequenced from the fungus Trichoderma harzianum FP108. The cloning was performed by PCR amplification of T. harzianum genomic DNA, using PCR primers whose sequence was based on the cbh1 gene from Trichoderma reesei. The 3' end of the gene was isolated by inverse PCR; attempts to clone regions upstream of the 5' end of the gene were unsuccessful. Sequence comparisons suggest that this gene is closely related to cbb1 genes from other Trichoderma species. In particular, all catalytically important amino acids in the protein sequence deduced from the T harzianum cbb1 gene are …

Metamorphosis In The Summer Flounder Paralichthys Dentatus: Changes In Gill Mitochondria-Rich Cells, Alex M. Schreiber, Jennifer Specker

Salinity tolerance changes during larval development and metamorphosis in the summer flounder (Paralichthys dentatus) and other teleosts. The physiological mechanisms responsible for osmoregulation during these early stages of development are not well understood. This study characterized changes in ultrastructure, intracellular membranes and immunoreactive Na+/K+-ATPase of mitochondria-rich cells (MRCs) in the gills of summer flounder during metamorphosis. Gill ultrastructure at the start of metamorphosis revealed only one type of MRC, which had weak reactivity to osmium and lacked a well-defined apical pit. Fibrillin-1, the main component of 10-12 nm microfibrils of the extracellular matrix, is synthesized as profibrillin and proteolytically processed to fibrillin. The putative cleavage site has been mapped to the carboxy-terminal domain of profibrillin-1, between amino acids arginine 2731 and serine 2732, by a spontaneous mutation in this recognition site that prevents profibrillin conversion. This site contains a basic amino acid recognition sequence (R-G-R-K-R-R) for proprotein convertases of the furin/PACE family. In this study, we use a mini-profibrillin protein to confirm the cleavage in the carboxy-terminal domain by both fibroblasts and recombinantly expressed furin/PACE, PACE4, PC1/3 and PC2. Over the Rim is the first book about an important but little-known expedition sent by Brigham Young to explore southern Utah. Led by Mormon apostle Parley P. Pratt, the party traveled from Salt Lake City south across the rim of the Great Basin to the Virgin River near future St. George. They brought back to Mormon leaders their first detailed portrait of the country to the south that the church planned to settle. Skewed Sex Ratio In An Estuarine Lobster (Homarus Americanus) Population, Winsor H. Watson Iii, William Hunting Howell

A total of 19,485 lobsters were caught sites in the estuarine and coastal waters of New Hampshire from 1989 to 1992, and their size and sex were determined. The sex ratio of lobsters caught farthest from the coast, in Great Bay, was heavily skewed in favor in males. Sex ratios in other estuarine and river sites were also skewed toward males, and there was a tendency for the number of males per female to decline as one moved down the estuary toward the coast, where the sex ratio was nearly 1:1.
