Global Cns Gene Transfer For A Childhood Neurogenetic Enzyme Deficiency: Canavan Disease., Paola Leone, Christopher G Janson, Scott J Mcphee, Matthew J During

Department of Neurosurgery Faculty Papers

The neurogenetic prototypic disease on which we chose to test our gene therapy strategy is Canavan disease (CD). CD is an autosomal recessive leukodystrophy associated with spongiform degeneration of the brain. At present the disease is uniformly fatal in affected probands. CD is characterized by mutations in the aspartoacylase (ASPA) gene, resulting in loss of enzyme activity. In this review, recent evidence is summarized on the etiology and possible treatments for CD. In particular, we discuss two gene delivery systems representing recent advances in both viral and liposome technology: a novel cationic liposome-polymer-DNA (LPD) complex, DCChol/DOPE-protamine, as well as recombinant …

Systematics Of The Eucestoda: Advances Toward A New Phylogenetic Paradigm And Observations On The Early Diversification Of Tapeworms And Vertebrates, Eric P. Hoberg, Scott Lyell Gardner, Ronald A. Campbell

Harold W. Manter Laboratory of Parasitology: Faculty Publications

Evolutionary relationships of the Eucestoda have received intense but sporadic attention over the past century. Since 1996, the landscape has dramatically changed with respect to our knowledge of the phylogenetic relationships among the tapeworms. The 2nd International Workshop for Tapeworm Systematics (IWTS) held in Lincoln, Nebraska in October of that year provided the catalyst for development of novel hypotheses for inter-and intra-ordinal phylogeny. The working-group structure of the 2nd IWTS and results of phylogenetic studies are briefly introduced in the present manuscript. Higher-level phylogenies derived from parsimony analysis of independent data bases representing comparative morphology or molecular sequences were largely …

Evidence Supporting The Recent Origin And Species Status Of The Timberline Sparrow, John Klicka, Robert M. Zink, Jon C. Barlow, W. Bruce Mcgillivray, Terry J. Doyle

Ornithology Program (HRC)

The Timberline Sparrow (Spizella taverneri), although originally described as a species, is currently classified as a subspecies of the more widespread Brewer's Sparrow (S. breweri). We investigated the taxonomic status and recent evolutionary history of these species by comparison of both morphological and molecular characters. Morphometric comparisons using 6 external and 18 skeletal measurements show that S. taverneri specimens from two widely separated populations (Yukon and southwestern Alberta, Canada) are indistinguishable with respect to size yet are significantly larger (by 3%) than representatives of several breweri populations. Analysis of 1,413 base pairs of mitochondrial DNA (mtDNA) for 10 breweri and …

Implementation Of An Activity-Based Costing System In An Air Force Laboratory Environment, Jayson M. Wrona, Martin Memminger

Theses and Dissertations

The purpose of this study was to examine the initial implementation of an Activity Based Costing (ABC) system within the United States Air Force's Research Laboratories (AFRL). We were attempting to ascertain what the initial purposes for implementing ABC within AFRL were, then determine whether or not those goals were being attained. We also attempted to evaluate the current system's appropriateness for achieving those initial purposes. Finally, we provide suggested changes to the model and areas for future research. We noted problems with AFRL's current ABC system. We were able to condense the problem into two main categories. First, when …

Skin: A Work Of Fiction, Kellie Wells Aug 1999

Skin: A Work Of Fiction, Kellie Wells

Dissertations

In Skin, a polyphonous novel, a community of characters similarly metaphysically vexed and similarly grappling with issues of selfhood and identity intersect and catalyze in one another moments of recognition. These brief self-reckonings suggest to the characters that though the quest to discern their place in the universe is a plagued one, it is not without value as it at least results in faint glimmers of enlightenment. Ultimately, how ever, these flickerings of understanding inch the characters no closer to the big certainties they seek, and they therefore must determine that the process itself, or, more to the point, …

Thanatos-Eros, Being-Non Being: Psychoanalytic - Existential Connection, Ryan D. Miller Aug 1999

Thanatos-Eros, Being-Non Being: Psychoanalytic - Existential Connection, Ryan D. Miller

Student Work

This Thesis examines and shows the relationship between Sigmund Freud's Death Instinct and Life Instinct Theories and their relation to the Existential concepts of Being and Non- Being. These intertheoretical concepts were interpreted in order for practitioners of Psychotherapy to be better acquainted with both concepts; so they may be better used in the mental health professions. A discussion of the two concepts as seen in Existential Psychotherapy and Freudian Psychoanalytic thought was presented and expanded upon and the relationship between the two was explained.

Trait Anxiety As A Moderator Of Problem Structuring Effects On Solution Generation, Judith A. Wightman Aug 1999

Trait Anxiety As A Moderator Of Problem Structuring Effects On Solution Generation, Judith A. Wightman

Student Work

This study investigated the effects of problem structuring and anxiety on the quantity and quality of solutions generated for ill-structured, complex problems. Trait anxiety, the tendency to feel anxious across a wide variety of situations, has been shown to impair problem solving performance in certain conditions. Trait anxiety was examined as a possible moderator of the relationship between problem structuring and solution generation. Participants were 184 undergraduate psychology students. Participants completed a trait anxiety measure (State-Trait Anxiety Inventory; Spielberger, 1983) and generated solutions to an ill-structured problem, with varying levels of structuring (no objectives, one-objective-at-a-time, conflicting objectives). The quantity and …

Signaling Mechanisms For Muscarinic Receptor-Mediated Coronary Vasoconstriction In Isolated Rat Hearts, Yi Zhang Aug 1999

Signaling Mechanisms For Muscarinic Receptor-Mediated Coronary Vasoconstriction In Isolated Rat Hearts, Yi Zhang

Electronic Theses and Dissertations

The signaling mechanisms for muscarinic receptor-mediated vasoconstriction in coronary resistance arteries were studied in KCl-arrested isolated rat hearts perfused at a constant flow rate. The cholinergic agonists acetylcholine and bethanechol were given by bolus injection or constant infusion. The coronary vascular resistance was monitored by measuring the changes in perfusion pressure. The selective muscarinic agonist bethanechol caused a similar vasoconstrictor response as ACh, but with less potency and efficacy. Bolus injection of bethanechol evoked a phasic vasoconstriction in a dose-dependent manner, while infusion of bethanechol evoked a tonic vasoconstriction without producing tachyphylaxis. Coronary vascular responses to bethanechol were further examined …

‘Supernatural, Or At Least Romantic': The Ancient Mariner And Parody, Steven Jones Aug 1999

‘Supernatural, Or At Least Romantic': The Ancient Mariner And Parody, Steven Jones

English: Faculty Publications and Other Works

This essay looks through the lens of parody at one of Coleridge's most characteristically "romantic" works, his famous ballad of the supernatural, The Rime of the Ancient Mariner. Along with the other so-called "Mystery Poems"—"Christabel" and "Kubla Khan"—this is among his most significant generic contributions to the developing idea of Romanticism, the kind of work that comes through a kind of synecdoche to stand for the whole movement as it was conceived.

Associations Of Egg Production With The Major Histocompatibility Complex In Broiler Breeder Hens, Becky Jean Tarleton

Graduate Theses, Dissertations, and Problem Reports (ETD)

This study documents effects of the major histocompatibility complex (MHC) on reproductive traits in a flock of Cobb X Cobb broiler breeder hens. The objective was to determine if the MHC affects reproduction of non-congenic broiler breeder hens. Genomic DNA was isolated from 35 hens. Restriction fragment length polymorphism (RFLP) analysis was performed on each sample, using a chicken class II MHC probe (Xu et al. 1989). Restriction endonuclease digestion using PvuII resulted in polymorphisms in all birds. Five polymorphic bands were identified. The largest bands, 7.5 kb and 5.4 kb, were present in one and four hens respectively. The …

Influence Of Selection For Breast Muscle Mass On Ph And Metabolism Of Supracoracoideus Muscle From Male And Female Turkey, John Kenneth Yost Aug 1999

Influence Of Selection For Breast Muscle Mass On Ph And Metabolism Of Supracoracoideus Muscle From Male And Female Turkey, John Kenneth Yost

Graduate Theses, Dissertations, and Problem Reports (ETD)

The objective of this study was to determine if selection for increased breast muscle mass has affected the supracoracoideus muscle's ability to regulate pH during physical activity in turkeys. The lightweight male line (LM) had the greatest breast muscle mass as a percent of body weight (p < 0.0001), a lower average pH (6.68; p < 0.0001), and greater Lactate Dehydrogenase (293 mmol NADH min-1mug-1 ; p < 0.0001) and Glyceraldehyde Phosphate Dehydrogenase (0.4452 mmol NADH min-1mug-1; p < 0.05) activities than the HM and FL lines. The LM line had the highest ratio (2.33:1) (p < 0.05) of adult to neonatal myosin. Hens had greater breast muscle mass as a percent of body weight than males (p < 0.0001), and a lower average pH (6.62; p < 0.05). This study showed that genetic selection for breast muscle mass resulted in an increased ratio of adult to neonatal myosin and an increased anaerobic capacity, which may predispose birds to a lower ultimate pH during physical activity.

Effect Of Sward Height And Concentrate Supplementation On Forage Dry Matter Intake, Digestibility And Grazing Time Of Spring-Calving Beef Cows, Ondieki J. Gekara Aug 1999

Effect Of Sward Height And Concentrate Supplementation On Forage Dry Matter Intake, Digestibility And Grazing Time Of Spring-Calving Beef Cows, Ondieki J. Gekara

Graduate Theses, Dissertations, and Problem Reports (ETD)

To establish the effect of sward height (SH) and concentrate supplementation on performance of grazing cattle, twenty-four cross bred Angus beef cow-calves were randomly assigned to two SH treatments (4--8 or 8--11 cm) with three supplement levels: 6.24, 3.12 or 0 kg/d. The study was conducted in three periods (P): May (P1), June/July (P2) and August (P3) in 1996. There was no SH x supplement level x period or SH x supplement level interaction (P > .10) on forage DMI, DMD or grazing time (GT). Cows on low SH had greater (P < .08) DMI and GT (P < .01) than cows on high SH. Forage DMI, DMD and GT decreased (P < .05) with supplementation. Grazing efficiency declined with supplementation and advancing grazing season (P < .01). Cows consumed more forage DM (P < .01) in P1 and P2 than P3. Forage DMI and grazing efficiency decreased with supplementation suggesting that supplemented cattle can optimally be grazed at SH < 8 cm.

Forecasting Electricity Demand Using Regression And Monte Carlo Simulation Under Conditions Of Insufficient Data, Kathleen Ann Cullen Aug 1999

Forecasting Electricity Demand Using Regression And Monte Carlo Simulation Under Conditions Of Insufficient Data, Kathleen Ann Cullen

Graduate Theses, Dissertations, and Problem Reports (ETD)

The problem studied is that of a summer peak residential energy demand model for Appalachian Power Company's service area in West Virginia. By restricting the forecast to a region smaller than the state, serious data problems result due to insufficient data to obtain reliable forecasts.;Regression analysis and Monte Carlo Simulation are the two methods used to forecast energy demand. Both methods incorporate risk into the analysis in different ways. Regression analysis yields a measure of the reliability of the coefficients of the variables and of the reliability of the forecast. The resulting forecast and confidence limits of the forecast values …

Occurrence, Etiology And Management Of Ringwomb In Ewes, Nancy Jean Kerr Aug 1999

Occurrence, Etiology And Management Of Ringwomb In Ewes, Nancy Jean Kerr

Graduate Theses, Dissertations, and Problem Reports (ETD)

Pedigrees of ewes with Ringwomb were studied to determine the etiology, incidence and treatment of Ringwomb. Purebred Suffolk and crossbred Hampshire and Shropshire ewes aged from one to seven years were observed over a period of five lambing seasons. The study was conducted on 24 clinical cases of ringwomb occurring in one flock. Ringwomb or incomplete cervical dilation accounted for an incidence of 20.5% (24/117) of all ewes lambing. The ewes affected were all daughters, granddaughters, or great-granddaughters of one particular sire used on the farm as a service sire. Because ringwomb does not typically affect ewes in two consecutive …

Unconstrained Instantaneous Center Integration (Ici) Algorithms, Nathan Eggleton Aug 1999

Unconstrained Instantaneous Center Integration (Ici) Algorithms, Nathan Eggleton

Theses

This paper presents several unique "instantaneous center integration" (ICI) algorithms for use in studying unconstrained vehicle motion using computer simulations and animations. Specifically, these algorithms are most appropriate for the case when high angular rates are present, such as would occur during tight cornering and/or possibly during an accident. The three ICI algorithms presented are the "pseudo-exact", first order approximation and the second order approximation. These ICI algorithms are capable of producing valid, accurate, and efficient simulation results, which describe the behavior of a vehicle under these conditions. The "pseudo-exact" is the most accurate, yet the most computationally expensive. The …

Use Of Extractable Lipofuscin For Age Determination Of Blue Crab Callinectes Sapidus, Se-Jong Ju, David H. Secor, H. Rodger Harvey

OES Faculty Publications

The blue crab Callinectes sapidus is an economically and ecologically important species in many temperate estuaries, yet stock assessments have been limited to length-based methods for demographic analyses. We evaluated the potential of age pigments (lipofuscins) sequestered in neural tissue of eye-stalks and brains to estimate the age of blue crabs collected from Chesapeake Bay and Chincoteague Bay. The rate of lipofuscin accumulation was determined using crabs of known age reared in the laboratory. Age pigments were extracted from neural tissues (eye-stalk or brain), quantified, and normalized to protein content to allow comparisons across tissue types and crab sizes. Field-collected …

The Relationship Between Homicidal Fantasies And Dangerousness, James C. Peterson

All-Inclusive List of Electronic Theses and Dissertations

Aggressive behavior is a serious problem in the United States and has been studied from a variety of perspectives. Psychologists in many different settings are frequently asked to assess the risk of future dangerous behavior by individuals. Due to their important role, research into the prediction of dangerousness has dramatically increased in the past ten years. However, to date, research has offered few reliable predictors that can be used in such assessments. This exploratory study was designed to examine whether homicidal fantasies can be used to reliably discriminate between violent and non-violent individuals. Undergraduate male students (N=l42) completed a questionnaire …
