Phenazines, the water-soluble pigments produced naturally by bacteria with distinct antimicrobial properties, have been implicated in the biological control of several plant diseases. Pseudomonas chlororaphis strain O6, obtained from the rhizosphere of wheat, is known to produce at least three different phenazines, which include phenazine-1-carboxylic acid, 2-hydroxyphenazine-1-carboxylic acid, and 2-hydroxyphenazine. A field trial conducted in the fall of 1994 to test the biocontrol properties of O6 indicated that this strain inhibited wheat seed germination, which led me to hypothesize that phenazines cause phytotoxicity. The Development Of A Bluetongue Virus Diagnostic Assay Using Heterologously Expressed Structural Protein Vp7 And Nonstructural Protein Ns2, Mary Anne Wise

The S1 gene of bluetongue virus that encodes major inner core protein, VP7, was cloned and expressed in the Thiofusion Expression Vector System. The VP7-thioredoxin protein was then partially purified. The nonstructural protein NS2 having previously been cloned into the GST Gene Fusion Vector System was also expressed and purified as NS2-GST fusion protein. These two viral proteins were used together in an immunologic detection assay. This assay was able to distinguish between bluetongue virus infected, vaccinated and noninfected sheep. Assayed serum samples of infected sheep, cattle, bison, and goats showed detectable antibodies to both VP7 and NS2, whereas vaccinated …

Adoptive immunotherapy using in vitro expanded tumor-infiltrating lymphocytes (TIL) continues to be the subject of research and clinical trials for the treatment of cancer. However, the culture methods used to expand large numbers of TIL are limited and often ineffective. In this study using murine TIL, the level of in vitro expansion, expression of adhesion molecules and infiltration into tumors after infusion was evaluated. A comparison was made between lymphocyte culture fluid (LCF), a bovine plasma-based medium, and a bovine serum-based medium (SCM) as the culture media. Because plasma-based media lack the lymphocyte inhibitory factors associated with serum-based media, it …

A Comparison Of Deterministic And Stochastic Population Models, Clarissa Smith

In this thesis, the author considers the exponential and the predator-prey population models. A comparison of the deterministic and stochastic versions of each is made. The author shows that on average a stochastic model is precisely its corresponding deterministic model. The first model considered is the exponential model. A solution is provided for both the deterministic and the stochastic versions. A predator-prey model is also analyzed. A solution of the stochastic predator-prey model is shown to be intractable. Analysis of this model is performed through numerical simulation.
Sensitizing Resin Acids In Aerosol Derived From Rosin Flux Used In Electronics Soldering: Molecular Mechanistic Studies In Resin Acid/Protein Hapten Formation, Philip A. Smith

Exposure to rosin and its resin acid constituents is associated with dermal and pulmonary sensitization. Methodologies to detect resin acid compounds in aerosol from heated rosin flux included air filtration, filter extraction, and gas chromatography/mass spectrometry. Various resin acids were seen in aerosol collected under field conditions, and in the laboratory. The predominant resin acid recovered was dehydroabietic acid, with lower levels of abietic acid and oxidized resin acids, which included 7-oxodehydroabietic, 15-hydroxydehydroabietic, and 7-hydroxydehydroabietic acids. Mass balance experiments with a soldering iron and liquid rosin flux showed most of the nonvolatile mass in unheated flux to be filtrable following …

Studies concerning the microinvertebrate communities of differing pool sizes were conducted at Chaney Lake, an ephemeral, karst wetland located in Bowling Green, Kentucky. As the lake begins to dry in the early summer, smaller, isolated ponds begin to form. Several ponds of various sizes were marked for analysis. Microinvertebrate samples and physiochemical readings were taken at each of the sites and were counted in the lab. T-tests were performed on the samples to determine if the microinvertebrate densities differed between ponds that dried later in the season as opposed to those that dried earlier. Several investigators have demonstrated an increased resistance to the protozoal parasite Trypanosoma cruzi (the causative agent of Chagas' disease) in highly susceptible C3H mice when maintained at an environmental temperature of 36°C. This increased resistance is manifested as delay in onset of parasitemia, decreased parasitemia, decreased pathology, and increased longevity with eventual clearing of the parasite from the bloodstream. In addition, this enhanced resistance has been shown to be an immunologic phenomenon and not due to the direct effects of elevated temperature on the parasite itself. To ascertain the possible contributions of humoral vs. cellular immunity to the above phenomenon, …

An exploratory analysis of national beauty pageants for children was conducted through the administration of a questionnaire and through observation. The population surveyed was mothers with daughters sixteen years old or under. Questionnaires were distributed at six national pageants in five states with a total of 134 respondents. The questionnaire probed a variety of areas concerning the child's involvement, expenses incurred, beauty enhancements worn, and general demographic information concerning the contestant's family. Attitudinal statements concerning the role of women in society and the importance of beauty in society were investigated. A comparison was made between the attitudes of the pageant …

The dominant paradigms of the past decade involve the preservation of biodiversity through the use of landscape corridors and buffer zones, as well as the integration of economic growth through sustainable use. The International Biosphere Reserve (IBR) program, as defined in the Seville Strategy, encourages locations already involved in conservation to continue to protect biodiversity and foster harmonious relationships between humans and their environment through sustainable development. An excellent example of an IBR is the Mammoth Cave Area Biosphere Reserve (MCABR) in south-central Kentucky. This study evaluated Na/K-ATPase alpha 1- and alpha 3-subunit isoform polypeptide expression and localization during bovine preattachment development. Na/K-ATPase cation transport activity from the one-cell to blastocyst stage was also determined by measuring ouabain-sensitive 86Rb+ uptake. Both alpha1- and alpha 3-subunit polypeptides were detected by immunofluorescence to encircle the entire cell margins of each blastomere of inseminated zygotes, cleavage stage embryos, and morulae. Immunofluorescent localization of alpha1-subunit polypeptide in bovine blastocysts revealed an alpha1 immunofluorescence signal confined to the basolateral membrane margins of the trophectoderm and encircling the cell periphery of each inner cell mass (ICM) cell. The explosive growth in genetic technology will quickly make possible an unprecedented number of tests for genetically based conditions. A necessary condition for the use of such tests without risk of harm to the patient is that they are “accurate”. However, most discussions of test accuracy in the literature have equivocated between two importantly different meanings of the word. In particular, it must be kept in mind that a high analytical accuracy does not imply a high diagnostic accuracy. Questions about the diagnostic accuracy of genetic tests loom large at present given our limited knowledge of the complex etiology of …

Lack of legitimacy of land tenure institutions in the tropical Peten, Guatemala, contributes to the tenure insecurity that encourages rapid colonization, deforestation and forest conversion to agriculture. The author identifies historical periods of delegitimation of land tenure institutions and analyzes social, political and property rights trends which reduced thee effectiveness of property rights in the Peten. Three case studies present the complexities and chaos of land tenure institutions. For most of the 20th century the mechanistic, linear world of Newtonian science has functioned both as metaphor for thinking about organizational dynamics and as the mental model around which organizations have developed. Recently, the postclassical sciences have yielded an alternate understanding of the world, one fundamentally different from that of the Newtonian model. This paper explores a world view emerging from complexity studies and quantum physics, contrasts it with the dominant image derived from classical physics, and draws implications for organizational leadership. The lens of postclassical science challenges our fundamental ontology and reframes our views of organizational life in …

In these Holmes Lectures, delivered a century after the publication of Oliver Wendell Holmes's great essay The Path of the Law, Judge Posner argues for an essentially Holmesian conception of the proper relations among modern normative moral philosophy ("academic moralism"), morality, and law. Academic moralism, he argues, lacks either the intellectual cogency or the emotional power to change people's beliefs or behavior; the power to do so resides in "moral entrepreneurs," which academic moralists emphatically are not. Academic moralism's lack of cogency disqualifies it to guide judicial decisionmaking even - in fact, especially - in cases involving controversial moral issues, …

The Reproductive Biology And Ecology Of Penstemon Harringtonii (Scrophulariacea): Implications For Conservation, Romaine L. Nielson

I examined the reproductive biology of Harrington's Penstemon, a rare endemic of the Colorado Plateau, to obtain information important to conservation efforts for this species. I applied four different hand-pollination treatments to P. harringtonii's flowers and found that it is partially self-compatible but sets more fruits when cross-pollinated by insects. Plant placement in new populations may be important because pollinators are more attracted to dense floral resources. Thus, the fruit set of plants within a 450 cm2 area was compared with those at least 3 m apart. Unexpectedly, aggregated plants produced 16.1 fruits per plant compared to 21.0 …

DNA markers for Aegilops caudata C-genome chromosomes will be useful in linkage studies of disease resistance and alien gene introgression, e.g., these markers can be tools in gene mapping and plant breeding. However, the individual chromosomes have not been characterized at the molecular level in Ae. Caudata. The objective of this study was to identify and characterize DNA markers for Ae. caudata chromosomes using the randomly amplified polymorphic DNA (RAPD) technique. The management of a rare, edaphic endemic plant species requires an understanding of the ecological setting and processes that contribute to its restriction. Included in the ecological processes is a set of biotic characters associated with the reproductive ecology, including its breeding system, pollination vectors, and germination requirements. The geologic and soil setting of the plant must also be characterized in order to understand the plant's restriction to the substrate. These issues were addressed by this study of the rare endemic Penstemon debilis O'Kane and Anderson (Scrophulariaceae). Most sternorrhynchan insects harbor endosymbiotic bacteria in specialized cells (bacteriocytes) near the gut, which provide essential nutrients for hosts. In lineages investigated so far with molecular methods (aphids, mealybugs, whiteflies), endosymbionts apparently have arisen from independent infections of common host ancestors and cospeciated with their hosts. Some endosymbionts also exhibit putatively negative genetic effects from their symbiotic association. In this study, the identity of endosymbionts in one major sternorrhynchan lineage, psyllids (Psylloidea), was investigated to determine their position in eubacterial phylogeny and relationship to other sternorrhynchan endosymbionts. The exact pathogenesis of autism is not known, but experimental evidence shows that there are genetic, immunologic, and even microbial factors involved in its development. A potential genetic and immunologic link to the development of autism is the presence of the HLA-DRβ1*0101 and *0401 genes in autistic children or their mothers. These genes have been found 5 to 6 times more often in autistic children or their mothers than in normal subjects. It has also been shown that people expressing these genes might be tolerant to self-peptides from the third hypervariable region of the HLA-DRβ1 protein. The third hypervariable region …
