Saccharomyces cerevisiae vacuole inheritance requires two low molecular weight activities, LMA1 and LMA2. LMA1 is a heterodimer of thioredoxin and protease B inhibitor 2 (I^B2). Here we show that the second low molecular weight activity (LMA2) is monomeric I^B2. Though LMA2 / I^B2 was initially identified as a protease B inhibitor, this protease inhibitor activity is not related to its ability to promote vacuole fusion: ( i ) Low M r protease B inhibitors cannot substitute for LMA1 or LMA2, ( ii ) LMA1 and LMA2 promote the fusionof vacuoles from a strain that …

Stocking rate, and the resulting level of grass utilization (or grass weight removed) by grazing livestock, has more impact on grass productivity than any other single factor within the range manager’s control. Determining the level of utilization for a pasture is one of the most important measurements that a manager can make when monitoring grazing Management over the years. The photo guide and procedures presented in this publication are intended to make grass utilization estimates both rapid and useful. The terms “utilization,” “use,” and “degree of use” all have similar meanings.

The cDNA for soybean leghemoglobin a (Lba) was cloned from a root nodule cDNA library and expressed in Escherichia coli. The crystal structure of the ferric acetate complex of recombinant wild-type Lba was determined at a resolution of 2.2 Å. Rate constants for O₂, CO and NO binding to recombinant Lba are identical with those of native soybean Lba. Rate constants for hemin dissociation and auto-oxidation of wild-type Lba were compared with those of sperm whale myoglobin. At 37°C and pH 7, soybean Lba is much less stable than sperm whale myoglobin due both to a fourfold …

Molecular Cloning And Functional Analysis Of Polyphosphoinositide-Dependent Phospholipase D, Pldβ, From Arabidopsis, Kirk Pappan, Wensheng Qin, Jim Dyer, Ling Zheng, Xuemin Wang

A novel plant phospholipase D (PLD; EC 3.1.4.4) activity, which is dependent on phosphatidylinositol 4,5-bisphosphate (PIP2) and nanomolar concentrations of calcium, has been identified in Arabidopsis. This report describes the cloning, expression, and characterization of an Arabidopsis cDNA that encodes this PLD. We have designated names of PLDβ for this PIP2- dependent PLD and PLDα for the previously characterized PIP2-independent PLD that requires millimolar Ca2+ for optimal activity. The PLDβ cDNA contains an open reading frame of 2904 nucleotides coding for a 968-amino acid protein of 108,575 daltons. Expression of this PLDβ cDNA clone in Escherichia coli results in the …

Native And Multimeric Vitronectin Exhibit Similar Affinity For Heparin: Differences In Heparin Binding Properties Induced Upon Denaturation Are Due To Self-Association Into A Multivalent Form, Ping Zhuang, Anthony I. Chen, Cynthia B. Peterson

For many years, the concept that the heparin-binding sequence is sequestered within vitronectin and exposed upon denaturation of the protein has guided experimental design and interpretation of related structure- function studies on the protein. To evaluate binding of heparin to both native and denatured/renatured vitronectin, methods for monitoring binding in solution have been developed. A fluorescence method based on changes in an extrinsic probe attached to heparin has been used to evaluate heparin binding to native and denatured/renatured vitronectin. This approach indicates that there are not major differences in intrinsic heparin-binding affinities between native and renatured protein and invalidate the …
