Acute myeloid leukemia (AML) is an aggressive hematological malignancy with various molecular and cytogenetic subtypes. Treatment options for older adult patients are limited due to high toxicity of conventional chemotherapy. The B-cell leukemia/lymphoma 2 inhibitor venetoclax is effective in combination with hypomethylating agents or low-dose cytarabine, but ∼30% of patients do not respond to the initial combination treatment. Thus, alternative combinations are needed to sensitize AML cells to venetoclax and overcome resistance mechanisms. Here, we report that targeting histone lysine-specific demethylases induces a ferroptosis-like phenotype driven by oxidative stress in various AML subtypes. In both patient samples and cell lines, …

Rationale: Cryotherapy and its diagnostic analogue, cryobiopsy, are emerging techniques via bronchoscopy with a wide range of clinical applications. While the use of cryotherapy and cryobiopsy has been established in adult pulmonary medicine, pediatric data describing the indications, procedural specifics, outcomes, and complications are limited.
Objective: To describe the indications, procedural specifics, outcomes, and complications of cryotherapy and cryobiopsy use in pediatric bronchoscopy.

Objective: To identify proteomic signatures underlying 3-year progression from impaired fasting glucose (IFG) to diabetes.
Research design and methods: We examined IFG progression in the Atherosclerosis Risk in Communities (ARIC) study from visit 2 (1990-1992) to visit 3 (1993-1995). We tested associations of 4,955 plasma proteins (SomaScan version 4.0) with ∼3-year progression from IFG (FG 100-125 mg/dL without diabetes) to diabetes (diagnosis, medication, or FG ≥126 mg/dL) using logistic regression models adjusted for demographics, cardiometabolic risk factors, and baseline glucose with Bonferroni correction (P < 10-5). We explored biological pathways enriched among the top proteins and calculated improvements in prediction (ΔAUC and net reclassification using 3-year risk thresholds of 6% and 15% in 80% training and 20% internal validation subsamples). We validated results in the Multi-Ethnic Study of Atherosclerosis (MESA) cohort.

Objective: To evaluate the content and structure of OCD-specific training within United States osteopathic (D.O.) and allopathic (M.D.) medical schools. Although many physicians encounter patients with obsessive-compulsive disorder (OCD) in clinical practice, limited familiarity with the full range of OCD presentations, including typical and atypical forms, may contribute to misdiagnosis and delays in accurate diagnosis and treatment.

BACKGROUND: Injuries to the Achilles tendon, such as tears or ruptures, are commonly observed among elite athletes. These injuries can be debilitating for professional soccer players, leading to prolonged recovery and potential performance declines.
PURPOSE: To investigate the annual incidence, return-to-play (RTP) duration, and postinjury performance outcomes of Major League Soccer (MLS) players who sustained Achilles tendon injuries from 2010 to 2021.

Background: Eosinophilic gastritis currently has no approved treatments and is postulated to be driven by type 2 inflammation. Dupilumab blocks type 2 cytokines IL-4 and IL-13 and has efficacy in multiple diseases characterised by type 2 inflammation, including eosinophilic oesophagitis. We aimed to assess the efficacy and safety of dupilumab in patients with eosinophilic gastritis.
Methods: DEGAS was a proof-of-concept, phase 2, multicentre, randomised controlled trial consisting of a 12-week, double-blind, placebo-controlled period, followed by a 24-week open-label extension period. The present study analyses the narrative treatment of suicide in Spanish youth fiction television series produced between 2015 and 2024, aiming to identify representation trends in relation to the recommendations for responsible media coverage and their implications for media literacy. The methodology underpinning this study encompasses eight fiction TV series and combines quantitative research, derived from data collection and analysis, with a qualitative component, focusing on the interpretation and analysis of the results. Findings indicate that the representation lacks due caution in depicting the method and location of suicide, which increases the risk of imitation. Although the narratives present alternatives …

Introduction: Periprosthetic joint infection (PJI) is a rare but devastating complication following total ankle arthroplasty (TAA). Acute infections are commonly managed with debridement, antibiotics, and implant retention (DAIR), in which irrigation plays a critical role in reducing bacterial burden. Although irrigation protocols have been investigated in total hip and total knee arthroplasty, evidence specific to infected TAA remains unclear. This systematic review evaluated the available literature regarding irrigation practices in the surgical management of infected TAA.

Why do decision makers value the same risky opportunity differently when the underlying probability of success is unchanged? We study this question in ecosystem investment decisions, where success requires that none of the critical components fail, causing component risks to compound. We conducted a high-powered preregistered replication and extension of Adner and Feiler's key study (N = 3,600). Participants valued a $100,000 opportunity that required six independent components to succeed, each with a 75% chance of success, yielding an overall success probability of ≈18%. In this unregulated nonpayment proceeding, the court granted the tenant's motion for summary judgment and dismissed the petition for lack of a lease agreement. The court distinguished a prior Appellate Term holding, finding that the regulatory status of the unit made the prior decision inapplicable. The court also rejected the landlord's argument that RPL § 226-c's language about 'continuance of existing terms' creates an automatic lease renewal after expiration, holding that such an interpretation conflicts with RPL § 232-c and that payment of rent after a lease expires does not create an involuntary leasehold by statute.

OBJECTIVES: Patent ductus arteriosus (PDA) is one of the most common heart conditions in preterm infants, and indomethacin treatment is used for PDA closure. However, its efficacy varies among patients. This pilot study aimed to identify novel metabolic biomarkers to predict effective indomethacin response in infants with PDA.
METHODS: Preterm infants with PDA < 34 weeks gestational age were recruited and treated with indomethacin. Plasma and urine samples were collected before and 2-7 days after initial treatment. Longitudinal targeted oxylipin and untargeted metabolomics analysis were performed on plasma and urine samples.
RESULTS: Among 17 infants, 12 were responders and 5 non-responders to indomethacin treatment. Before the treatment, four plasma oxylipins: 12,13-DiHOME, 9,10-DiHOME, 15,16-DiHODE, and 9,10-DiHODE were significantly higher in responders compared to non-responders (P < 0.05). Following treatment, these oxylipins decreased in responders but remained stable in non-responders. Urinary 12,13-DiHOME showed an opposite trend, with lower baseline concentrations in the responders.

Oral history interview with John Barry, who has served as Executive Director of the Southern Tier AIDS Program (STAP) since 2008. He was born in Endicott, New York in 1966, grew up in Vestal, and attended high school at Seton Catholic Central in Binghamton before serving in Germany with the United States Army during the early 1980s. When he returned to Broome County in 1987, some of his friends had been affected by HIV/AIDS and, while earning a degree at SUNY Binghamton, Barry became involved in mutual aid efforts to help people affected by the disease. The rapid emergence of metallo-b-lactamase-mediated antibiotic resistance has created an urgent need for new inhibitor discovery strategies. In this work, a machine-learning-guided workflow was developed to generate and prioritize potential inhibitors targeting NDM-1. A SMILES-based variational autoencoder was first pretrained on a broad molecular dataset to learn general chemical syntax and latent molecular representations. The model was then fine-tuned on an 8-hydroxyquinoline-enriched dataset to bias molecular generation toward zinc-binding chemical space relevant to metallo-β-lactamase inhibition. Generated compounds were processed through structural filtering and docking-based evaluation to create training data for downstream predictive modeling. As artificial intelligence (AI) systems increasingly assume roles with social, educational, and emotional significance, understanding the psychological drivers behind individuals' readiness to delegate such roles to AI is crucial. Drawing on Self-Determination Theory (SDT), this study examines how the satisfaction of basic psychological needs (autonomy, competence, and relatedness) predicts individuals' readiness to delegate socially significant roles to AI across four domains (education, healthcare, mental health, and companionship) and 35 nations. Using data from over 35,000 participants in the 2023 Global Digital Wellbeing Survey, we applied Bayesian multilevel multivariate modelling to assess both global and culture-specific motivational associations. Este estudio realiza un análisis comparativo y retrospectivo de las estrategias de comunicación utilizadas para promocionar las exposiciones temporales de Cristóbal Balenciaga entre 2015 y 2022. Los elementos utilizados en sus planes de comunicación se identificaron mediante entrevistas semiestructuradas con los directores de comunicación de 16 espacios expositivos internacionales y con el director de comunicación del Cristóbal Balenciaga Museoa.
