Investigating The Novel Role Of Dextromethorphan In Mitigating Oxidative Stress Induced Changes In Retinal Pigment Epithelial Cells (Rpe), Zeinab A. Mohamed
Investigating The Novel Role Of Dextromethorphan In Mitigating Oxidative Stress Induced Changes In Retinal Pigment Epithelial Cells (Rpe), Zeinab A. Mohamed
USF Tampa Graduate Theses and Dissertations
Oxidative stress contributes to the degeneration of retinal pigment epithelial (RPE) cells and photoreceptors that lead to the dry form of age-related macular degeneration. Activation of N-Methyl-D-aspartate (NMDA) receptors contributes to the increase of free radicals that induce oxidative stress-induced changes. Our study investigates if Dextromethorphan, an NMDA receptor antagonist, reverses oxidative stress-induced changes in RPE. We used Paraquat and H2O2 as oxidative stress-inducing agents to induce oxidative damage in human ARPE-19 cells. To illustrate the change in cell viability and measure the apoptosis of ARPE-19 cells from oxidative damage, we performed MTT assay and Annexin V-DAPI flow cytometry analysis. …

Passive Nitrifying Biofilters For Onsite Treatment Of Saline Domestic Wastewater, Daniel Arnulfo Delgado
Passive Nitrifying Biofilters For Onsite Treatment Of Saline Domestic Wastewater, Daniel Arnulfo Delgado
USF Tampa Graduate Theses and Dissertations
The infrastructure to support the growing world population has polluted many water bodies with nitrogen from urban and agricultural runoff and failing wastewater systems, such as improperly sited and maintained septic systems. Septic systems present a low energy and maintenance option to treat domestic wastewater and is a treatment option that can be done far from a centralized grid. This further reduces cost, maintenance, and energy as it reduces the need to build and maintaining sewage piping and the infrastructure to move the water longer distances. However, these systems tend to fail when their upkeep is left to homeowners that …

Bait-And-Kill: Targeting A Novel Heme Biochemical Pathway In Hundreds Of Cancers, Christopher G. Marinescu
Bait-And-Kill: Targeting A Novel Heme Biochemical Pathway In Hundreds Of Cancers, Christopher G. Marinescu
USF Tampa Graduate Theses and Dissertations
Cancer cells are characterized by their ability to grow and proliferate in rates that far exceed normal cells by modifying their iron/heme metabolisms to levels higher than normal. This imbalance of heme biosynthesis can lead to cancer cells having a flux of heme intermediates to the point where they enter a state called heme-overdrive. Heme-overdrive is a process unique to a variety of cancers but absent in normal tissues. With this enhanced production, heme can act as an epigenic regulator for signaling proliferation (18). Through the novel strategy called ‘bait-and-kill,’ cancer cells will be coerced into a state of heme …

USF Tampa Graduate Theses and Dissertations
Biodiversity is essential to the ecosystem functions responsible for generating the multi-billion-dollar industries that the US relies on for food, storm mitigation, tourism and recreation, but is facing increasing natural and anthropogenic threats. High-resolution diversity data covering large spatial areas would enhance current ecology knowledge and better address conservation concerns in the Gulf of Mexico. However, acquiring and handling data of this nature is resource intensive and has not yet been established. In this study, high spatial resolution data was collected from gastropod living and death assemblages collected over 10 years from the West Florida Shelf (WFS). A multivariate regression …
Drug Candidates Targeting Hsp90/Cdc37 For The Treatment Of Neurodegenerative Disease, Ryan Martin
Drug Candidates Targeting Hsp90/Cdc37 For The Treatment Of Neurodegenerative Disease, Ryan Martin
USF Tampa Graduate Theses and Dissertations
Neurodegenerative disease refers to the death of specific populations of neurons that canresult in dementia, loss of voluntary muscle motility, and other abnormalities. Two major neurodegenerative diseases are Alzheimer’s disease (AD) and Parkinson’s disease (PD), which are linked to abnormalities in Tau and a-synuclein proteins, respectively. Currently, there is no cure for these diseases. Our group and others have shown that chaperone proteins play a critical role in the processing of Tau and a-synuclein. The inhibition of the Hsp90/Cdc37 chaperone complex is shown to affect the levels of these two proteins. Hence, we hypothesized that a drug molecule, conglobatin (Cb), …

Nest Site Choice As A Potential Climate Response In The Gopher Tortoise (Gopherus Polyphemus), A Threatened Species, Meghan Craft
USF Tampa Graduate Theses and Dissertations
Climate warming is expected to continue in coming decades, posing a threat to species worldwide. While some species may be able to shift their ranges in the pursuit of cooler temperatures, a large portion of species may have to find ways to adapt to warmer conditions in situ. Oviparous reptiles may be able to buffer against rising temperatures by altering nest site choice. In this study, I quantified nest site choice and its thermal consequences in a population of the gopher tortoise (Gopherus polyphemus) in southwestern Florida. I found marked variation in the openness of selected nest sites, …

¿De Dónde Eres?: Negotiating Identity As Third Culture Kids, Sophia Margulies
¿De Dónde Eres?: Negotiating Identity As Third Culture Kids, Sophia Margulies
USF Tampa Graduate Theses and Dissertations
Sparked by David C. Pollock's (1988) concept of "third culture kids" (children of government officials and military personnel), this thesis uses autoethnography to examine "white-passing" Latinx identity and gender passing as it relates to individuals who identify as transgender to understand what it means to "pass" within these communication contexts. I situate the study at the intersections of queer, trans, and Latinx theories. Ultimately, I argue that the communicative and identity practices inherent to the liminal spaces in which third culture kids perform create the conditions for performances as transnational subjects. What the contexts of place and home are like, …

Glutathione-S-Transferase Mu 1 (Gstm1) Gene Augmentation Attenuated Oxidative Stress Induced Changes In Retinal Pigment Epithelium Cells (Rpe), Riddhi Sandeep Vichare
USF Tampa Graduate Theses and Dissertations
Purpose: Loss of Retinal Pigment Epithelial (RPE) cells and photoreceptors leads to Age-related macular degeneration (AMD), one of the leading causes of blindness in the elderly worldwide. Oxidative stress-mediated damage is one of the factors that impair the survival of RPE and photoreceptors in dry-AMD. We sought to determine the role of human Glutathione S-Transferase Mu-1 (GSTM1) in protecting RPE cells from oxidative stress-induced changes.
Method: We cloned human GSTM1 cDNA with P2A tag into a lenti-plasmid vector and later made a lentivector to express GSTM1. After that, we infected human retinal pigment epithelial cells (ARPE-19), screened stable ARPE-19 cells …

Transparent self-cleaning coatings with superhydrophobic and superhydrophilic behavior are of great interest in a wide range of industrial applications such as automotive, solar panels, windows, and optical devices. Each has its uses, properties, and characteristics. For the use of these coatings on solar panels, certain features are important in maintaining the efficiency of the panels in different weather conditions and reducing the operation and maintenance cost, such as self-cleaning, high transparency, antifouling, anti-fogging, and anti-icing. Herein, we have investigated the fabrication of self-cleaning coatings using two simple methods and ways to improve their characteristics. In the first method, a hydrophobic …
Effect Of Temperature On Growth And Gene Expression In The Marine Diatom, Thalassiosira Pseudonana, John M. Foster
Effect Of Temperature On Growth And Gene Expression In The Marine Diatom, Thalassiosira Pseudonana, John M. Foster
USF Tampa Graduate Theses and Dissertations
Diatoms are unicellular, eukaryotic, photosynthetic algae responsible for the production of 20% of our atmospheric oxygen. Diatoms contribute to several important biogeochemical functions. Diatoms contribute to the carbon cycle, sequestering carbon and forming oxygen as a product of photosynthesis. Moreover, by converting abiotic forms of energy such as sunlight into organic compounds (i.e., sugars, starches and lipids), primary producers including diatoms, plants, and phytoplankton feed organisms at higher trophic levels. Consequently, changes in temperature, light intensity, nutrients, salinity and other stress factors that affect primary producers can generate a potentially catastrophic ripple effect at higher trophic levels In this study …

The Impact Of Bahiagrass (Paspalum Notatum) On Florida Sand Skink (Plestiodon Reynoldsi) Movements, Chandler A. R. Eaglestone
USF Tampa Graduate Theses and Dissertations
The Florida Sand Skink (Plestiodon reynoldsi) is a federally threatened lizard precinctive to scrub habitat, a pyrogenic habitat that experiences fires infrequently. Plestiodon reynoldsi spends most of its time right under the sand surface, moving, or “sand swimming,” through the granular substrate found in Florida’s central ridges. While the distribution of this species has been greatly reduced, agricultural development of P. reynoldsi habitat has led to potentially suitable fragmented scrub occurring near bahiagrass (Paspalum notatum) pastures. Bahiagrass is widely used as a pasture grass in sandy soils of the southeast United States, because of its deep …
Novel Co2 Transporters From Autotrophic Bacteria From Extreme Environments, Sarah E. Schmid
Novel Co2 Transporters From Autotrophic Bacteria From Extreme Environments, Sarah E. Schmid
USF Tampa Graduate Theses and Dissertations
Autotrophic organisms are responsible for introducing carbon into food webs. These organisms take up dissolved inorganic carbon (DIC) and use it as their major carbon source, converting it to biomass using a variety of carbon fixing pathways (e.g., the Calvin-Benson-Bassham cycle). In nature, DIC concentrations can be low or temporally heterogeneous, so autotrophic organisms adapt with CO2 concentrating mechanisms (CCMs) to facilitate the acquisition and fixation of this dissolved gas. CCMs consist of membrane-spanning DIC transporters, which elevate intracellular DIC concentrations, which in turn are harvested by carboxysomes, proteinaceous microcompartments that contain the carboxylase Ribulose-1,5-bisphosphate carboxylase-oxygenase (RubisCO) and carbonic anhydrase. …

USF Tampa Graduate Theses and Dissertations
Plasmonic nanomaterials have become a strong contender for improving the efficiency of photocatalytic degradation of air pollutants. This study demonstrates an easy and scalable fabrication method, employing electron beam evaporation and rapid thermal annealing, for producing plasmonic photocatalysts. Samples were made by either coating silver on, or layering silica-protected (i.e., silica-coated) or unprotected (i.e., uncoated) silver beneath, the photocatalyst (either zinc oxide or titanium dioxide). Stability and catalytic performance for gas-phase toluene degradation was assessed by monitoring total volatile organic compound (TVOC) concentration versus ultraviolet-A (UV-A) illumination time in a recirculating batch reactor (plate-type flow-through). Samples were characterized using a …
The General Psychopathology Factor (P) From Adolescence To Adulthood: Disentangling The Developmental Trajectories Of P Using A Multi-Method Approach, Alexandria M. Choate
The General Psychopathology Factor (P) From Adolescence To Adulthood: Disentangling The Developmental Trajectories Of P Using A Multi-Method Approach, Alexandria M. Choate
USF Tampa Graduate Theses and Dissertations
Considerable attention is directed towards studying co-occurring psychopathology through the lens of a general factor (p-factor). However, the developmental trajectories and stability of the p-factor have yet to be fully understood. Study 1 first examined the explanatory power of dynamic mutualism theory — an alternative framework positing the p-factor to be a product of lower-level symptom interactions rather than the inherent cause of them. Predictions of dynamic mutualism were tested using three distinct statistical approaches including: longitudinal bifactor models, random-intercept cross-lagged panel models (RI-CLPMs), and network models. Next, given prior suggestions that borderline personality disorder (BPD) could be a marker …
