The Role Of The Dna Helicase Rrm3 Under Replication Stress, Julius Muellner
The Role Of The Dna Helicase Rrm3 Under Replication Stress, Julius Muellner
USF Tampa Graduate Theses and Dissertations
DNA replication needs to be strictly monitored to ensure proper duplication of the genome. During DNA synthesis, the DNA replication machinery encounters multiple obstacles such as incorrect dNTP incorporation, RNA-DNA hybrids, modification of DNA nucleobases, collision with the transcription-replication machinery. These endogenous and exogenous sources of DNA damage may result in single-strand breaks (SSBs) and double-strand DNA breaks (DSBs), thereby impeding fork progression. To prevent stalling of DNA synthesis a multitude of DNA repair pathways are specifically designed to deal with these kinds of blockages. One of the proteins involved in ensuring DNA replication fork progression is the DNA helicase …
Scale-Up Of Perovskite Oxides For Conversion Of Co2 To Co Through Reverse Water-Gas Shift Chemical Looping, Hanzhong Shi
Scale-Up Of Perovskite Oxides For Conversion Of Co2 To Co Through Reverse Water-Gas Shift Chemical Looping, Hanzhong Shi
USF Tampa Graduate Theses and Dissertations
CO2 conversion to CO has the benefit of further conversion to hydrocarbon-based fuels and chemicals. Due to CO2 being an important greenhouse gas, its conversion to CO and an increase of the carbon cycling will help reduce the greenhouse effect. Reverse water-gas shift chemical looping (RWSG-CL) is a two-step process that can be employed for the conversion of CO2 to CO. The first step involves partial reduction of oxygen by hydrogen to form oxygen vacant oxide, and the second step is the conversion of CO2 to CO over these partially reduced materials to regain their original structures. The RWSG-CL process …
Role Of Hla-Drb1 Fucosylation In Anti-Melanoma Immunity, Daniel K. Lester
Role Of Hla-Drb1 Fucosylation In Anti-Melanoma Immunity, Daniel K. Lester
USF Tampa Graduate Theses and Dissertations
Melanoma is the one of the most lethal skin malignancies due to its ability to rapidly metastasize and evade the immune system. One factor that influences melanoma’s ability to metastasize and evade the immune system is the tumor microenvironment. The tumor microenvironment is a complex ecosystem that consists of melanoma cells interacting with different proteins and cell types such as cytokines, extra cellular matrix proteins, and various immune cells. While different immune cells can have various implications in the tumor microenvironment, some tumor infiltrating lymphocytes (TIL) have the potential to suppress these tumors. Recent therapeutic strategies aim to enhance TILs …
Bioinformatic Analysis Predicts Microglial Dysfunction In Murine Aging, Amadu Idrisa Jalloh
Bioinformatic Analysis Predicts Microglial Dysfunction In Murine Aging, Amadu Idrisa Jalloh
USF Tampa Graduate Theses and Dissertations
Age-related disease is a growing concern as the global geriatric population increases. Neurodegenerative diseases scale unfavorably in prevalence with aging and inflict disastrous consequences to human health and well-being. These disorders are challenging to investigate because they arise from complex molecular origins. The neuroimmune system is a common factor among these diseases and microglia play an important role in maintaining homeostasis in the central nervous system. Aging progressively impairs microglia by decreasing their ability to adapt and respond to noxious environmental stimuli or injury. Microglial dysfunction aggravates neurodegenerative pathology when microglia are unable to regulate neuroinflammation effectively. We investigated aging …
Diet And Salivary Microbiome On Cardiovascular Risk And Glycemic Control In Participants With And Without Type 1 Diabetes: The Cacti Study, Tiantian Pang
USF Tampa Graduate Theses and Dissertations
Introduction: The prevalence of type 1 diabetes mellitus (T1DM) has been increasing drastically in the United States. People with T1DM are at a higher risk of developing diabetes-related complications, including cardiovascular disease (CVD). Diet is one of the most modifiable factors and healthy eating could reduce CVD risk among individuals with T1DM. The current 2015-2020 Dietary Guidelines for Americans focus on overall healthy eating patterns rather than individual foods and nutrients. A healthy dietary pattern or high-quality diet can be measured by diet quality indexes, and generally contains minimal ultra-processed foods (UPF). Although substantial evidence has linked poor diet quality …

Histone Deacetylase 8 Is A Novel Therapeutic Target For Mantle Cell Lymphoma And Preserves Natural Killer Cell Cytotoxic Function, January M. Watters
USF Tampa Graduate Theses and Dissertations
This study demonstrates for the first time that HDAC8 function is critical for MCL survival, and abrogating its activity in human primary NK cells does not interfere with NK IgG antibody directed ADCC therapies ex vivo. Human NK cells, isolated from healthy donors, are highly resistant to HDAC8 inhibitor treatment with PCI-34051. Even at the highest concentration, 20uM, no toxicity was observed. Conversely, MCL cell lines representative of the aggressive MCL subtype, classical MCL, were especially sensitive to PCI-34051, an HDAC8 selective inhibitor, treatment. Blocking HDAC8 activity and/or abrogating expression through shRNA silencing induced significant DNA damage, hyperacetylation of SMC3, …

The current project seeks to illuminate the diets of the earliest farming societies in central Northern Italy. Neolithic peoples who first began settling in the Central Po Valley sometime at the end of the seventh and beginning of the sixth millennium BCE forever changed the landscape from one of expanding sub-continental forests to one of intensive agricultural production and anthropogenic influence. A total of 109 individual burials from 24 separate infrastructure project excavations of 17 sites surrounding the modern city of Mantua, Italy were analyzed utilizing a biochemical approach and a bioarchaeological explanatory theoretical framework based within embodiment and life …
Synergistic Effect Of Temperature And Acid Catalyst To Enhance L-Ald, Prabhsimran Singh
Synergistic Effect Of Temperature And Acid Catalyst To Enhance L-Ald, Prabhsimran Singh
USF Tampa Graduate Theses and Dissertations
Liquid Atomic layer deposition (LALD) is an emergent technology used for depositing thin films. Extensive research has been done on this method, designing different techniques and modifications for the deposition of films. Wastage of alkoxide precursors, extra purging cycle, and extensive time for the reaction when added stoichiometrically are some of the drawbacks which shift the focus from metal alkoxide as precursors. In this thesis, modifications for the L-ALD method for the complete reaction of alkoxide precursors when added stoichiometrically were achieved. Temperature and catalyst’s synergistic effects were utilized for the modification. This modified method was applied for the doping …
Dreaming To Get Out The “Sunken Place”: Fantasy, Film, And The Inner-White- I(Eye), Jordan Battle
Dreaming To Get Out The “Sunken Place”: Fantasy, Film, And The Inner-White- I(Eye), Jordan Battle
USF Tampa Graduate Theses and Dissertations
Since its inception during the transatlantic slave trade, blackness within the collective unconscious of Western society has been sutured to negative stereotypes, images, and feelings. In modern discourse on race and racism, the unconscious manifestations of anti-black racism adopted from cultural impositions, the impact of enslavement and colonization on the psyche of the subjugated, and the importance of the mind in revolutionary efforts are all too often undertheorized. Through an Afropessimist framework, this thesis uncovers the origination of the black imago in Western society, how and why it is habitually recreated, and how black theorists, artists, political activists, and others …

Apkcs Role In Neuroblastoma Cell Signaling Cascades And Implications Of Apkcs Inhibitors As Potential Therapeutics, Sloan Breedy
USF Tampa Graduate Theses and Dissertations
Neuroblastoma (NB) is a cancer that develops in the neuroblasts. It is the most common cancer in children under the age of 1 year, accounting for approximately 6% of all cancers. The prognosis of NB is linked to both age and degree of cell differentiation. This results in a range of survival rates for patients, with outcomes ranging from recurrence and mortality to high survival rates and tumor regression. Our previous work indicated that PKC-ι promotes cell proliferation in NB cells through the PKC-ι/Cdk7/Cdk2 cascade. We report on two atypical protein kinase inhibitors as potential therapeutic candidates against BE(2)-C and …

Time Series Analysis Of Pseudo-Nitzschia Species Composition, Domoic Acid, And Environmental Conditions In The Gulf Of Maine From 2013-2020, Christina Chadwick
USF Tampa Graduate Theses and Dissertations
Phytoplankton are abundant in both fresh and marine waters and can form harmful algal blooms (HABs) by producing potent toxins that have devastating effects on human, wildlife, and economic health. The HAB-forming diatom genus, Pseudo-nitzschia, occurs globally and approximately half of the 59 currently identified species can produce the neurotoxin, domoic acid (DA). DA accumulates in shellfish and finfish and can cause Amnesic Shellfish Poisoning (ASP) in humans and DA Poisoning (DAP) in several species of wildlife, resulting in severe neurological dysfunction and mortality. Along with these health threats, DA toxicity is a major economic threat for shellfish and fish …
Identifying Barriers And Facilitators To Family Communication Regarding Genetic Testing For Hereditary Cancer, Andrea K. Shields
Identifying Barriers And Facilitators To Family Communication Regarding Genetic Testing For Hereditary Cancer, Andrea K. Shields
USF Tampa Graduate Theses and Dissertations
Cascade testing, whereby at-risk family members are tested for known pathogenic or likely pathogenic (P/LP) variants in high risk cancer genes, provides the opportunity for changes in medical management (e.g., increased surveillance to detect cancer early or preventative surgery to reduce cancer risk). However, the rates of cascade testing are low which suggests that one-time communication may be insufficient to prompt action among family, and an increase in understanding of how to promote ongoing communication about genetic testing within families is needed. We surveyed individuals with P/LP variants in inherited cancer genes and conducted multiple linear regression with forward analysis …

Pathogenic variants (PV) in the gene LMNA cause autosomal dominant inherited “laminopathies” that can affect multiple different organs, most specifically the heart. Current resources for LMNA patients are sparse and disjointed, leaving a need for a comprehensive resource catering to the wants and needs of the patients. A needs assessment of LMNA support resources was completed through reviewing published literature and existing support resources and conducting 11 semi-structured interviews with individuals who have a PV in LMNA. The Social Support Theoretical Model and thematic analysis of interview transcripts were used to identify discrepancies between the support that affected individuals receive …
Comparing Mortality In Response To Simulated Climate Change In The Harvester Ant, Pogonomyrmex Badius, And The Fire Ant, Solenopsis Invicta, Haley Depner
USF Tampa Graduate Theses and Dissertations
With the rapid transport of plants, produce, and goods out of its South American home, the fire ant, Solenopsis invicta, has invaded continents on a global scale. In the United States alone, S. invicta is responsible for an estimated $1 billion in damages annually. In contrast to the invasive fire ant, Florida’s native harvester ant, Pogonomyrmex badius, is a keystone seed disperser, providing seed relocation benefits to Florida’s sandhill wildfire-adapted habitats. Seed relocation reduces seedling competition for nutrients with the parent plant, reduces predation of seeds by rodents, increases nutrient loads associated with ant mound soils, and protects seeds from …

Modeling Leachate Treatment Processes In Adsorbent-Amended Hybrid Constructed Wetland, Ishfaqun Nisa
USF Tampa Graduate Theses and Dissertations
Due to their efficiency in removing ammonia nitrogen, Constructed Wetlands (CW) are currently considered a very useful method for on-site leachate treatment. In recent years, the use of adsorbent media in CWs, such as zeolite and biochar, to improve pollutant removal efficiency has been explored and these studies indicate that incorporating zeolite can improve the nitrification process, while the addition of biochar can enhance the denitrification process and the removal of organic compounds. However, studies on mechanistic/process models simulating performance of adsorbent-amended CWs for treating landfill leachate are limited. This study presents simulation results for both unamended and adsorbent-amended hybrid …

Rehabilitation Technologies To Abate Infiltration In Sanitary Sewers, Steve Youssef
USF Tampa Graduate Theses and Dissertations
In urban coastal environments, sewer infrastructure is plagued by infiltration which seeps into aging and compromised pipes. Infiltration increases pumping costs, reduces treatment efficiency, and may trigger release of partially treated wastewater at water reclamation facilities. To restore pipes and abate infiltration, local utilities rely on structural and non-structural trenchless rehabilitation technologies. These methods are commonly used throughout the United States and the world due to their numerous benefits compared to traditional open-cut construction. However, limited data is available regarding their efficacy in reducing infiltration in sewer systems. In this study, we analysed infiltration in a small urban sewershed in …
