The amyloid beta peptide particularly the 40 and 42 amino acid residues are the responsible for plaque formation in Alzheimer's disease (AD) patients. Extra cellular plaque formation has been recognized after incessant investigations along with the formation of intracellular tau protein tangles as the hallmarks of AD. Furthermore, the plaque formation has been linked mostly as a cause of the disease and the tangles mostly as a consequence. Our investigation is focused on studying the formation of AD plaques. This study is an exploration of Latina/o-White hybrid identity for constructions and negotiations of hybridity as performed in the lives of individuals and as rearticulated in discourse. These discourses are drawn from interviews with nine individuals, stories of my own life, and three published memoirs. Despite these different forms, all the selfidentified Latina/o-White hybrid individuals speak to the difficulty of imagining and enacting a hybrid identity within today’s discourse on race and ethnicity. This study articulates these difficulties as lived experience, theory, and performance come together to argue for and against hybridity as a model for contemporary identity. The project …

Sol-gel capillary microextraction (CME) is a new direction in solvent-free extraction and preconcentration of trace analytes. CME presents significant interest in environmental, pharmaceutical, petrochemical, biomedical, agricultural, food, flavor, and a host of other important areas. Sol-gel CME utilizes advanced material properties of organic-inorganic hybrid sol-gel polymers to perform efficient extraction and enrichment of target analytes from a variety of matrices. In this dissertation, two novel sol-gel coatings were developed for CME: (a) sol-gel benzyl-terminated dendrimer coating, and (b) sol-gel polytetrahydrofuran (poly-THF) coating. A Future With Hope: The Social Construction Of Hope, Help, And Dialogic Reconciliation In A Community Children's Mental Health System Of Care, Christine S. Davis

This research examines the social construction of hope in a community mental health system of care. Groopman (2004) defines hope as the elevating feeling we experience when we see a path to a better future. A year-long ethnographic study of a children's mental health system of care team found that members of the mental health care team construct hope for themselves and for the family they're helping by cycling through the dialectical tensions of hegemony and equality, marginalization and normalization, relating and othering, empowerment and disempowerment, and control and emotionality. They reconcile these tensions in dialogic moments of empathy toward …

Electroporation is a methodology for the introduction of drugs and genes into cells. This technique works by reducing the exclusionary nature of the cell membrane [125, 129, 186, 189]. Electroporation has successfully been used in electrochemotherapy and electrogenetherapy [57, 68, 86, 87, 110, 112, 131]. The two major components of electroporation are an induced transmembrane potential and the motion of the deliverable through a compromised cell membrane into the target cell [38, 55, 62, 114, 131]. Characterization of micron and sub-micron size particles requires the simultaneous measurement of the joint particle property distribution (JPPD). The JPPD is comprised of particle size, shape, orientation, composition, optical properties, and surface properties. Measurement of each of the particle properties independently is a difficult task and it has been only partially successful. To determine as many particle properties as possible using optical methods it is necessary to simultaneously measure all aspects of the interaction of the incident light with the particles of interest. A novel method of controlling miniature sensors using Handspring Visor Prism PDA has been implemented. A generic motherboard was developed to map the data and address lines from the Visor onto a Complex Programmable Logic Device (CPLD) to provide basic electrical signals to the sensor board. The sensor board housed the sensor and contained application specific circuitry. The PDA, the motherboard, and the sensor board completed the control mechanism for the sensor. In situ measurements were used to study the bio-optical properties of marine waters within the Gulf of Paria (GOP, Venezuela) and in the Southeastern Caribbean Sea (SEC) as they are affected by the seasonal discharge of the Orinoco River plume. The main purpose of this study was to determine the impact of colored dissolved organic matter (CDOM) (also known as Gelbstoff), phytoplankton, and total suspended matter (TSM) in the color of the Orinoco River plume. The Key deer (Odocoileus virginianus clavium) population has increased from less than 80 individuals in the 1950s to approximately 700 by 2000. Over two-thirds of the Key deer population resides on two islands out of a potential 26 islands within their range. The skewed distribution resulted in high deer densities on Big Pine, No Name, and Big Munson keys. The objective of the study was to evaluate the effects of Key deer herbivory in mangrove, buttonwood, hammock and pineland habitats between islands with low, medium, and high densities of deer. The focus of this study is Lily Bart and how she maneuvers in the cold, competitive world of upper class New York. To create a framework for my investigation, I draw upon naturalistic readings of the story which portray Lily as an outsider or "other" in her society. Lily's ethical principles lead to her destruction. Her marriage problem is just an example of her rejection of the life that her society expects her to lead. Prescribed self-management behaviors have been found to be important factors affecting the rates of morbidity and mortality in multiple medical conditions including chronic diseases such as diabetes, a condition that disproportionately affects high health risk populations such as African Americans.

Polyacrylamides (PAMs) are a class of polymers formed from acrylamide alone or copolymerized with other monomers. PAMs have been used in drinking water treatment as flocculants or coagulants in the highest volume among all the polymer types. In potable water treatment processes, polyacrylamides are often exposed to oxidants (e.g. chlorine and permanganate) and UV irradiation from sunlight or artificial sources. N-Thiolated β-lactams (1) represent a promising new group of compounds with potent inhibition effects on bacteria, like Bacillus anthracis and methicillin resistant Staphylococcus aureus, and onco-systems, like breast cancer and leukemia. Originally developed as part of a synthetic pathway to bicyclic lactams, N-thiolated β-lactams have been shown in this laboratory to possess intriguing biological activities. Leachate collection systems consist of a series of pipes installed beneath the waste at the base of a landfill. The liquid drains toward a central location where it is pumped and then treated, discharged, or recirculated. In some landfills, solid precipitates form in the collection system resulting in clogging and malfunctions of the drainage system. The formation of the precipitates is linked to the chemical and biological stability of the leachate generated within the landfill. To control the formation of precipitates and prevent clogging of leachate collection systems, it is important to understand factors that influence leachate characteristics.

Biosensor development has the potential to meet the need for rapid, sensitive, and specific detection of pathogenic bacteria from natural sources. An antibody-based fiber-optic biosensor assay to detect low levels of Vibrio vulnificus in estuarine waters following an enrichment step was developed. The principle of the sensor is based on an immuno-sandwich assay where an anti-V. vulnificus polyclonal capture antibody preparation was first immobilized on a polystyrene fiber-optic waveguide using a biotin-avidin association. Despite existing mad cow disease surveillance efforts in the United States, in place since the 1980s, a cow that tested positive for mad cow disease was granted entrance into the U.S. in December, 2003. The cow that tested positive, according to witnesses, displayed no symptoms that are synonymous with advanced bovine spongiform encephalopathy, BSE. This occurrence had detrimental effects on the U.S. beef export market, as many countries banned American beef. Estimates of the damage inflicted reach into the billions of dollars. It has been noticed for a long time that humans can identify others based on their biological movement from a distance. However, it is only recently that computer vision based gait biometrics has received much attention. In this dissertation, we perform a thorough study of gait recognition from a computer vision perspective. We first present a parameterless baseline recognition algorithm, which bases similarity on spatio-temporal correlation that emphasizes gait dynamics as well as gait shapes. The psychological impacts of infertility have been well documented in the literature, providing evidence to support that at least some women who confront infertility are at risk for heightened distress and depressive symptoms. In response to this accumulated evidence, it has been argued that psychoeducational interventions may provide an important component to the treatment of infertility. The work detailed in this dissertation has an overarching theme of modulating microglia activation in both in vivo and in vitro models relevant to AD. The premise is that understanding microglia function in this context may lead to a better understanding of AD pathogenesis and thus to effective therapeutic interventions. In chapter 3, we employ a well-defined model of microglia activation whereby the intraperitoneal delivery of LPS results in CNS microglia activation and TNF-α production. Oocyte development in Xenopus laevis spans six morphologically distinct stages (stage I-VI), and is associated with a decrease in protein O-GlcNAc levels. As a first step in elucidating the role of O-GlcNAc in developing oocytes, initial efforts were focused on isolation and identification of fifteen modified proteins that decrease during oocyte development. Stage I oocytes due to their high amounts of these proteins, were used as starting material for purification. Particle analysis using multiwavelength UV-visible spectroscopy provides the potential for extracting quantitative red blood cell information, such as hemoglobin concentration, cell size, and cell count. However, if there is a significant presence of hypochromism as a result of the concentrated hemoglobin (physiological value of 33%), successful quantification of red cell values would require a correction.

Managers of Biscayne National Park are proposing a new General Management Plan (GMP) in order to update the recreational and commercial use of resources in the park. A Fishery Management Plan (FMP) is also being drafted simultaneously in conjunction with the Florida Fish and Wildlife Conservation Commission in order to address concerns associated with management of fish stocks within the park.

The purpose of the present study was to examine the effect of varying the amount of participant engagement on alcohol expectancy and drinking outcomes during a social/sexual expectancy challenge based on Darkes and Goldman's (1993, 1998) protocol. This study was also intended to provide a test of the efficacy of administering an alcohol/placebo expectancy challenge outside of a live drinking scenario through video presented as part of a computerized intervention.

Jay B. Starkey Wilderness Park, located in southwestern Pasco County, Florida, contains nearly 7,689.06 hectares (19,000 acres) and includes 18 natural communities. A floristic inventory was conducted on approximately 404.69 hectares (1,000 acres) within Jay B. Starkey Wilderness Park which included 11 community types. A comparison of those 11 communities in the study area with the rest of the park shows that the dominant community types do occur within the study site, making it representative of the entire park. Microcalcification (MC) detection is an important component of breast cancer diagnosis. However, visual analysis of mammograms is a difficult task for radiologists. Computer Aided Diagnosis (CAD) technology helps in identifying lesions and assists the radiologist in making his final decision.
This work is a part of a CAD project carried out at the Imaging Science Research Division (ISRD), Digital Medical Imaging Program, Moffitt Cancer Research Center, Tampa, FL. A CAD system had been previously developed to perform the following tasks: (a) pre-processing, (b) segmentation and (c) feature extraction of mammogram images.
