Clinically Applied Anthropology: A Syndemic Intervention., Jason W. Wilson

This dissertation demonstrates that a critical, clinically applied anthropology is possible by testing a hypothesis that a syndemic intervention, and use of a structural vulnerability assessment tool, can achieve improved healthcare outcomes. A critical, clinically applied anthropology integrates social scientists into healthcare delivery, alongside biomedical providers, through the co-creation of new diagnostic and patient care pathways that utilize anthropological methods (ethnographically informed care, syndemics, thematic/mixed methods data analysis) and advance anthropological theory (biomedicine and culture, structural violence, structural competency/vulnerability, ontology, assemblage theory, and entanglements) to decrease healthcare inequities.

The intrinsically disordered transcription factor and tumor suppressor p53 binds to promoter response element DNA upon cellular stress and activates genes associated with cell cycle arrest, senescence, and apoptosis. Disruption of sequence specific binding to target gene promoters is heavily implicated in human health, where a majority of cancers contain mutations localized to the DNA binding domain (DBD) of p53. p53 DNA binding is regulated by posttranslational modifications, associations with cellular factors, and by an autoinhibitory intramolecular interaction. NRF2 is a redox-responsive transcription factor the directs the antioxidant program and several critical metabolic processes. Mutations in NRF2 or its negative regulator KEAP1 occur in up to one third of non-small cell lung cancers (NSCLCs) and are often associated with resistance to therapy and poor outcomes. In the present studies, murine alleles of the Keap1 and Nrf2 mutations found in human NSCLC were developed and I comprehensively investigated their impact on tumor initiation and progression. I observed that chronic Nrf2 stabilization by Keap1 loss-of-function or Nrf2 activating mutation was not sufficient to cause lung tumor initiation, even when p53 …

With gene-based immunotherapies on the rise to treat a multitude of diseases, the ability to genetically modify cells in a rapid and efficient fashion is necessary to keep up with demand. Gene electrotransfer (gene transfer by electroporation) is an appealing transfection method due to its low cost, high efficiency compared to other physical forms of gene delivery, and low toxicity compared to liposomal or viral vectors.

Introduction: Globally, 17 million adolescent girls—ages 15-19-- give birth every year, which affects educational and professional outcomes such as school completion, future employment, and economic empowerment. In the past two decades, a lot of different peptidomimetics, such as β-peptides, oligoureas, azapeptides, peptoids, aromatic oligoamides, and others, have been developed. These peptidomimetics with distinctive scaffolds have shown potential in protein surface recognition and mimicry, PPI regulation, catalysis, etc. Moreover, these bioactive peptidomimetics demonstrate much better bioavailability and biostability than the bioactive peptides. The evolution of hemochorial placentas in which the fetal trophoblast is bathed in maternal blood has vastly improved fetal development and survival in primates and rodents, due to the enhanced efficiency of nutrient, gas and waste exchange between the mother and the fetus. However, the extensive contact between the fetal tissues and maternal blood in these placentas also poses an increased risk of vertical transmission of pathogens such as viruses from the mother to the fetus. This work details the effort toward structure-activity-relationship (SAR) studies and synthesis of the small molecule modulators of two non-traditional drug targets. The first part of this manuscript (chapters 1–2) will discuss the collaborative work and synthesis of an inhibitor of Slingshot Homology 1 (SSH1) with the potential therapeutic application toward Alzheimer’s Disease (AD) treatment. The second part (chapters 3–6) will discuss the synthetic efforts toward modulation of a protein target known as STING, or stimulator of interferon genes, with the therapeutic application toward autoimmune disease or anti-tumor activity.

However, exposures to environmental chemicals are not evenly distributed throughout childhood. Understanding the distribution of organisms is an important priority for society as we live through ecosystem transformations that threaten the well-being of all organisms. I have approached the dynamic and complex issue of studying biodiversity by using open-access seabird data collected along the Americas since the 1960s. I explained how these data have changed over time and space, how certain species populations could have shifted over time, and possible correlations between this potential geographic change and select environmental variables.

However, this region is both distant and dangerous and is therefore largely under-sampled and understudied. Endothelial cells (ECs) form the inner lining of blood capillary walls. Our laboratory has developed a serum-free, defined Factor system that allows for human ECs to sprout, form lumens and tube networks, recruit pericytes, and induce capillary basement membrane deposition in 3D extracellular matrices. We have shown that the Factor system drives morphogenesis through the activation of multiple signaling pathways; however, we do not know if all factors in the Factor system are needed to induce maximal EC morphogenesis. A Gait Extraction System (GES) was developed to investigate the accuracy of wearable sensors by producing gait parameters, to be compared against a gold standard motion capture system. Two inertial measurement units (IMUs) were placed in the lower limb region, specifically in the shank region of each leg. Headwater wetlands are connected to one another and to downstream waters by dynamic hydrologic flowpaths, functioning as integrated hydrologic networks at the watershed scale. Headwater wetlands perform a variety of hydrologic lag, sink, and source functions, including flow generation and propagation, thereby contributing to the natural flow regimes of downgradient waters. Vulnerable populations have been shown to be disproportionately impacted by natural disasters, such as hurricanes. Hurricane Michael, a Category 5 storm, in October of 2018 hit the rural and socioeconomically vulnerable area of the Florida Panhandle, causing 50 fatalities in Florida, massive destruction to area including the healthcare infrastructure, and led a prolonged recovery period. The purpose of the research was to examine long-term impacts to health in Florida Panhandle after Hurricane Michael; changes to mortality trends, changes to health of survivors, and changes to their access to health care using a mixed-methods, sequential, explanatory study design.

Bloom syndrome (BS) is an extremely rare autosomal recessive genetic disorder caused by defects in the BLM gene. Patients with BS display a broad phenotypic spectrum including abnormally short stature, microcephaly, sun sensitivity, insulin resistance, perpetual oxidative stress, and a ~300-fold greater propensity for developing cancer than the general population. As such, BS patients typically do not live past their 20’s. The BLM gene encodes the BLM helicase, a member of the highly conserved RecQ family of 3’ to 5’ DNA helicases. Quinones are compounds containing cyclohexadiene diones and can be found in a variety of biological systems. Quinones participate in redox cycling to initiate the formation of free radicals which contributed to their overall biological activity. When quinones are reduced by either biological or chemical reductants, they can become unstable and undergo autoxidation back to their quinoidal form. One way to avoid autoxidation is the use of protected hydroquinones. This research aligned with protocols established by the Florida Board of Governors in 2016 to form a new funding model tied to the institution's performance. The literature review highlighted a disparity between students' expectations and the reality of college which could negatively impact persistence, and therefore funding to the institution. This dissertation investigates the multifaceted utility of γ-AA peptides in various biological contexts, highlighting their capability in mimicking bioactive proteins, facilitating novel drug discovery, and obstructing protein-protein interactions. Initially, we introduced the design of an ingenious γ-AA peptide scaffold that replicates the hydrolysis capabilities of natural enzymes, with the intention to overcome their stability limitations. Notably, the designed catalyst exhibited excellent esterase activity with the coordination of Zn2+. Subsequently, optimization of a linear γ-AA peptide compound through a dimeric strategy resulted in P3-1, a potent inhibitor of amyloid-β 42. Lung cancer is the first cause of cancer-related deaths in both men and women with an overall five-year survival rate of 28%. Although immune checkpoint blockers (ICBs) are currently FDA-approved for the treatment of non-small cell lung cancer (NSCLC), only 17-20% of patients achieve durable responses by the induction of immunologic memory. The lack of response in most patients can be attributed to the tumor-intrinsic or tumor-extrinsic immune resistance mechanisms. Cancer impacts millions of Americans every year, with an estimated 1.9 million new cases and 600,000 deaths predicted to occur in 2023 [1]. Medical professionals and scientists have developed several techniques and procedures to combat this disease including gene therapies, surgeries, and chemotherapies. Several of these techniques incorporate the use of pulsed electric fields to deliver molecular material to treat, or directly destroy, the cells of the tumor. These pulsed electric field techniques are known as gene electrotransfer, electrochemotherapy, and irreversible electroporation. Synthesis Of Constrained N-Heterocyclic Peptidomimetics And Development Of Strain-Release Reagents, Kyle P. Pedretty

Part 1:The initial portion of this dissertation delves into the enhancement of β-sheet secondary structure stability through modification of the peptide backbone while examining the conformational characteristics of peptides featuring N-amino substituents. The investigation consists of two major studies, presented in separate sections. The first study outlines various approaches to achieve stable β-hairpin folds by introducing N-amino substituents to peptide strands. A novel method utilizing on-resin Mitsunobu cyclization between α-hydrazino acid residues and a serine or homoserine side chain was utilized to incorporate pyrazolidinone and tetrahydropyridazinone dipeptide constraints.
