A Novel Abi-Domain Protein Controls Virulence Determinant Production In Staphylococcus Aureus, Stephanie Michelle Marroquin

A major factor in the success of Staphylococcus aureus as a pathogen is its vast arsenal of virulence determinants and, more importantly, the tight and precisely- timed regulation of these factors. Here we investigate the product of the S. aureus gene, SAUSA300_1984, encoding a putative transmembrane protein. This as yet uncharacterized protein belongs to the Abi (abortive infection) family, which are commonly annotated as CAAX-proteases, and are significantly understudied in prokaryotes. In S. aureus the disruption of SAUSA300_1984 results in a drastic reduction of proteolytic and hemolytic activity, as well as diminished pigmentation. Over the next thirty years, Alzheimer’s disease rates will increase alongside global aging. To handle the anticipated increase in demand, knowledgeable and skilled dementia caregivers are in need throughout the long-term care spectrum. Online training programs have emerged as a viable and convenient platform to educate both formal and informal caregivers. The first and second study systematically reviewed online dementia training programs and evaluated the CARES® Dementia Basics Training Program among formal and informal caregivers.

Individuals with stroke have neuromuscular weakness or paralysis on one side of the body caused by some muscles disengaging and overexciting other muscles. Hyperextension of the knee joint and complete lack of plantar flexion of the ankle joint are common symptoms of stroke. This thesis encompasses the simulation of hemiparetic function using both a knee orthosis with variable impedance, specifically in terms of stiffness and damping, and the Proprioception Interference Apparatus.

According to the World Resources Institute, Barbados is one of seven Caribbean countries ranked as being the most water stressed territories in the world. Prevailing drought conditions for the period 2010-2016 further compromised its water security while confirming predictions of a drier regional Caribbean climate. The simultaneous failing of at least 50-year-old water infrastructure at many points in the distribution network adds to these water stress conditions, and presents a financial burden to the local water utility in the form of lost revenues, and increased energy consumption for pumping.

Across avian systems, plumage ornamentation is often considered a signal of individual fitness, condition, sex, or status, and varies due to genetics or environmental sources. In species with structural coloration, plumage variation results from differences in the amount of energy allocated to feather growth during molt, presenting a unique opportunity to study the link between individual quality and ornamentation. Nutrient inputs into the environment greatly impact urban ecosystems. Appropriate management strategies are needed to limit eutrophication of surface water bodies and contamination of groundwater. In many existing urban environments, retrofits or complete upgrades are needed for stormwater and/or wastewater infrastructure to manage nutrients. However, sustainable urban nutrient management requires comprehensive baseline data that is often not available. A Framework for Urban Nutrient (FUN) Management for Geographic Information Systems (GIS) was developed to specifically address those areas with limited data access. Research over the last few decades has consistently found that fathers are not routinely included in the provision of child welfare services. The current study examined whether ambivalent sexism on the part of child welfare workers was related to their beliefs about involving fathers. Ambivalent sexism theory posits that gender stereotypes include subjectively positive beliefs in addition to hostile beliefs that both serve to perpetuate patriarchal systems. Participants (N = 490) were currently front-line child welfare workers in the United States who completed an online survey assessing ambivalent sexism and beliefs about father involvement in child welfare cases. Social supports are linked in public health research to improved birth outcomes. This study explored the relationship of social supports, stress and birth outcomes among pregnant Latinas in Pinellas County, Florida. A sample of 411 Healthy Start women at risk of poor birth outcomes participated in this study (99 Latinas, 142 Black, and 158 White). Study methods included ANOVA, Principal Component Analysis, multivariable regression, logistic regression, and structural equation modeling to identify significant associations between social support scores, stress scores, demographics and health risk factors with infant birth weight, preterm and small for gestational age by ethnic group. There is a paucity of data on dry forests, the most threatened biome in the tropics. The Nandamojo is a tropical dry forest watershed in Western Costa Rica that has been impacted by varying degrees of human induced modifications. This research was conducted to examine the influence of land use and channel characteristics on invertebrate communities within a sub basin of the Nandamojo watershed. This study analyzes how spatial organization impacts science communication at the St. Petersburg Science Festival in Florida. Through map analysis, qualitative interviews, and a close reading of evaluation reports, the author determines that sponsorship, logistics, exhibitor ambience, and map usability and design are the factors most affecting the spatial performance of science. To mitigate their effects, technical communicators can identify these factors and provide the necessary revisions when considering how science is communicated to the public.

Food insecurity and environmental factors including neighborhood deprivation may influence gestational weight gain. Au(I) catalysis has recently emerged as a powerful tool for the realization of a broad range of organic transformations. Despite this rapid development, attaining selectivity and maintaining catalyst stability remain significant challenges. Rational ligand design, such as the employment of NHC or TA ligands, has been used to confront these issues. This thesis focuses on the use of Au(I) catalysts bearing these ligands for the selective hydroalkynylation of enol ethers. By employing a TA-Au stabilized catalyst, [(OAr)3PAu(TA-H)]OTf, the intermolecular hydroalkynylation of enol ethers, a substrate that is well-known to promote decomposition of the gold cation, was efficiently achieved. …

Current water policy in four US cities was compared to a matrix of recommended sustainability themes that have been presented in the literature to determine the extent of which these themes have been implemented into water policy. This paper argues that the interactive problem-solving workshops created by political scientist John Burton and applied to the Israeli-Palestinian conflict by social psychologist Herbert Kelman, while not, as yet, resulting in a just and permanent peace agreement, are effective in resolving intractable conflict, and, if persistently used, can significantly help to produce such an agreement. This is done by closely examining two books of Burton and a series of articles by Kelman to describe their process; the characteristics of intractable conflict are also reviewed from the work of social psychologist Daniel Bar-Tal. Electric field mediated gene delivery modalities have preferable safety profiles with the ability to rapidly transfect cells in vitro and in vivo with high efficiency. However, the current state of the art has relied on trial and error studies that target the average cell within a population present in treated tissue to derive electric pulse parameters. Myelodysplastic syndromes (MDS) are genetically diverse hematopoietic stem cell malignancies that share a common phenotype of cytological dysplasia, ineffective hematopoiesis and aberrant myeloid lineage maturation. Apoptotic cell death potentiated by inflammatory cytokines has been considered a fundamental feature of MDS for over two decades. An organism's body size entails both physiological and ecological costs. Furthermore, as a parameter in analyzing organisms, it represents a fundamental and essential morphometric character. Reductions in size following mass extinction is a commonly observed phenomenon in the fossil record. This study examines the evolutionary significance of this phenomenon termed the: 'Lilliput Effect' by proposing that it represents a rapid evolutionary response to altered selection pressures during a mass extinction. This primary hypothesis is evaluated against two additional hypotheses of size reduction: 1) stunted growth as a response to stressed ecosystems, and/or 2) mass extinctions are size selective.

This thesis offers a critical examination of what I call the “Apatow aesthetic” in order to analyze the social processes of growing up in contemporary neoliberal network society. While doctors, psychologists and social scientists still proffer a model of mid- 20th century human development centered around a chronologically-determined life cycle, the Apatow aesthetic imagines a non-linear reality where traditional life events and social practices don’t always correspond to specific age groups. Of the considerable challenges researchers face in the control and elimination of malaria, the development of antimalarial drug resistance in parasite populations remains a significant hurdle to progress worldwide. Atovaquone is used in combination with proguanil (Malarone) as an antimalarial treatment in uncomplicated malaria, but is rendered ineffective by the rapid development of atovaquone resistance during treatment. Our ability to control malaria has been challenged by increasing antimalarial resistance. Plasmodium falciparum undergoes dormancy in the blood stages which is hypothesized to be a means by which they are able to survive under drug pressure. This helps select for resistant parasites which grow following removal of drug. The mechanisms behind dormancy and the subsequent recrudescence are not fully understood but translating knowledge from related organisms which undergo a similar phenomenon might shed some light. A perennial goal of environmental education is to produce a scientifically literate citizenry capable of negotiating and resulting complex environmental problems. Popular methods of environmental education instruction tend to overemphasize scientific content knowledge and neglect to consider ethical and moral aspects of the problem. This qualitative study examines the longitudinal association between an experiential environmental education course infused with SSI instruction and students’ environmental behaviors. The results indicate that several students’ conceptualizations of contentious environmental issues change after completing the course and specifically. Receptor interacting protein kinase 1 (RIPK1) is an enzyme acting downstream of tumor necrosis factor alpha to control cell survival and death. RIPK1 expression has been reported to cause drug resistance in cancer cells; but so far, no published studies have investigated the role of RIPK1 in vitamin D action. In the present study, we investigated whether RIPK1 played any role in 1,25-dihydroxyvitamin D3 (1,25D3)-induced growth suppression. In our studies, RIPK1 decreased the transcriptional activity of vitamin D receptor (VDR) in luciferase reporter assays independently of its kinase activity, suggesting a negative role of RIPK1 in 1,25D3 action. Using an applied anthropological approach focused on language, this study investigates the relationship between linguistic, racial, and ethnic identities and school resource access in the context of migration. This project examines how these identities are established, experienced, reified, and resisted by various school actors. Exposing power at its roots through a multi-level analysis, this research informs on how people negotiate socialization into particular identities, propelling them toward positions in school and society of varying opportunity.

Ovarian cancer is the most lethal gynecological cancer, with over 200,000 women diagnosed each year and over half of those cases leading to death. These poor statistics are related to a lack of early symptoms and inadequate screening techniques. This results in the cancer going undetected until later stages when the tumor has metastasized through a process that requires the epithelial to mesenchymal transition (EMT). Multifunctional nanocomposites are promising for a variety of applications ranging from microwave devices to biomedicine. High demand exists for magnetically tunable nanocomposite materials. My thesis focuses on synthesis and characterization of novel nanomaterials such as polymer nanocomposites (PNCs) and multi-walled carbon nanotubes (MWCNTs) with magnetic nanoparticle (NP) fillers.

PANcreatic DERived factor (PANDER, FAM3B) is a member of a superfamily of FAM3 proteins that are uniquely structured and strongly expressed from the endocrine pancreas and co-secreted with insulin. Unique animal models available to our lab have indicated that PANDER can induce a selective hepatic insulin resistant (SHIR) phenotype whereby insulin signaling is blunted yet lipogenesis is increased. The Effects Of Synthetic And Dietary Therapeutics On Learning, Memory, Motor Coordination, And Seizure In An Angelman Syndrome Mouse Model, Stephanie Lynn Ciarlone

Angelman syndrome (AS) is a rare genetic and neurological disorder presenting with severe developmental delay, ataxia, epilepsy, and lack of speech. AS is associated with a neuron-specific loss of function of the maternal UBE3A allele, a gene encoding an E3 ubiquitin ligase. Currently, no cure exists for this disorder; however, recent research using an AS mouse model suggests that pharmacological intervention is plausible, and can alleviate some of the detrimental phenotypes reported in AS patients.

Although there is no curative treatment for AS, seizure medication and behavioral therapies are most commonly prescribed in order to minimize symptoms. However, these options …
