Synthesis And Characterization Of Polyionic Hydrogels, Pooja Desai Jul 2008

Synthesis And Characterization Of Polyionic Hydrogels, Pooja Desai

Theses and Dissertations

In this study we describe novel polyionic dendrimer – PEG hydrogels for drug delivery. Hydrogels have a crosslinked insoluble network of polymer chains, which have found many applications including drug delivery and tissue regeneration. Dendrimers provide an ideal platform for drug delivery as they possess a well-defined highly branched nanoscale architecture with many reactive surface groups. Their highly clustered surface groups allow for targeted drug delivery and high drug payload to enhance therapeutic effectiveness. This study presented a new type of polyionic hydrogels based on dendrimers with potential applications in drug delivery and tissue engineering. Polyethylene glycol (PEG) with various …

Male Survivors: Walking The Tightrope Of Masculinity, Luke Rogers Jul 2008

Male Survivors: Walking The Tightrope Of Masculinity, Luke Rogers

Theses and Dissertations

Sexual assault of men is a serious social problem and has drawn well deserved attention from many different researchers as well as the general public. Even though there is much concern and interest in this troubling crime, limited research has been conducted on the long-term effects of sexual abuse on male survivors. Using data collected by the Virginia Department of Health, this study investigated the effects of sexual abuse of men. Specifically, this inquiry examined specific emotional and physical conditions as potential consequences of sexual victimization among men and the extent to which those conditions vary between male victims and …

Modeling Pure Vasogenic Edema In The Rat Brain, Charles Nottingham Jul 2008

Modeling Pure Vasogenic Edema In The Rat Brain, Charles Nottingham

Theses and Dissertations

Targeted drug delivery to the brain is difficult to achieve using conventional techniques, largely due to the blood-brain barrier’s (BBB) impediment to drug diffusion into the brain parenchyma. In response, development of convection-enhanced delivery (CED) offers the ability to circumvent the BBB and target specific areas of the brain. Predictability of infusate movement in pathological brain states during CED will maximize the effectiveness of this treatment, and therefore modeling of infusate movement must be characterized. Previous work from our lab effectively modeled CED in rats using the middle carotid artery occlusion model of cytotoxic edema. However, previous models examined for …

Physiological And Molecular Characterization Of Genetic Competence In Streptococcus Sanguinis, Alejandro Rodriguez Jul 2008

Physiological And Molecular Characterization Of Genetic Competence In Streptococcus Sanguinis, Alejandro Rodriguez

Theses and Dissertations

The ability of bacteria to assimilate free DNA from the environment is known as competence. Though many studies have focused on competence regulation in Streptococcus pneumoniae and Streptococcus gordonii, Streptococcus sanguinis has yet to be examined. Physiological characterization of competence in S. sanguinis strain SK36 and its comC mutant, JFP41, led to the genome-wide transcriptional analysis of cells induced to competence via addition of competence-stimulating peptide (CSP). A total of 128 genes were induced at least 2-fold, 74 of which were classified as either “early” or “late” based on their induction patterns. Expression patterns were verified using qRT-PCR. This study …

Deriving Operational Principles For The Design Of Engaging Learning Experiences, Richard Heywood Swan Jul 2008

Deriving Operational Principles For The Design Of Engaging Learning Experiences, Richard Heywood Swan

Theses and Dissertations

The issue of learner engagement is an important question for education and for instructional design. It is acknowledged that computer games in general are engaging. Thus, one possible solution to learner engagement is to integrate computer games into education; however, the literature indicates that pedagogical, logistical and political barriers remain. Another possible solution is to derive principles for the design of engaging experiences from a critical examination of computer game design. One possible application of the derived design principles is that instruction may be designed to be inherently more engaging. The purpose of this dissertation was to look for operational …

Comparison Of Guided And Open Inquiry Instruction In A High School Physics Classroom, Brett M. Guisti

Theses and Dissertations

This study compared two levels of inquiry in high school physics classrooms over a year-long course. One class fit well the definition of guided-inquiry and the other matched common descriptions of open-inquiry. Four sections of introductory physics at Lone Peak High School in Highland, Utah were randomly divided into two sections for each treatment. The majority of students in all classes were sophomores with relatively few juniors and seniors. The guided-inquiry classes followed the Modeling Instruction Program developed at Arizona State University, while the open-inquiry classes were be based on an approach used by Wolff-Michael Roth, at the University of …

Differential Expression And Functional Characterization Of Alpha3 Beta2 Neuronal Nicotinic Acetylcholine Receptors, John Hideo Mizukawa Jul 2008

Differential Expression And Functional Characterization Of Alpha3 Beta2 Neuronal Nicotinic Acetylcholine Receptors, John Hideo Mizukawa

Theses and Dissertations

Neuronal nicotinic acetylcholine receptors (nAChRs) are expressed in both the periperhal and central nervous systems, and are involved in pre-, post-, and non-synaptic control of neuronal activation. In the brain, these receptors play an important role in a variety of physiological processes such as cognition, development, learning, and memory formation. Malfunction of these receptors have been implicated in neurodegenerative diseases like Alzheimer's disease (AD), schizophrenia, and Parkinson's disease. To date, 17 different nAChR subunits, including α2-α7 and β2-β4, have been cloned that can form homo- and/or hetero-pentameric ionotropic receptors. The unique combinations of subunit pentamers manifest in distinct functional receptors. …

Assessing Multivariate Heritability Through Nonparametric Methods, Benjamin Alan Carper Jul 2008

Assessing Multivariate Heritability Through Nonparametric Methods, Benjamin Alan Carper

Theses and Dissertations

The similarities between generations of living subjects are often quantified by heritability. By distinguishing genotypic variation, or variation due to parental pairings, from phenotypic variation, or normal intraspecies variation, the heritability of traits can be estimated. Due to the multivariate nature of many traits, such as size and shape, computation of heritability can be difficult. Also, assessment of the variation of the heritability estimate is extremely difficult. This study uses nonparametric methods, namely the randomization test and the bootstrap, to obtain both a measure of the extremity of the observed heritability and an assessment of the uncertainty.

Arthropod And Plant Communities As Indicators Of Land Rehabilitation Effectiveness In A Semi-Arid Shrub-Steppe, Eric T. Gardner

Theses and Dissertations

We describe a case study evaluating the ecological impact of Bromus tectorum L. (cheatgrass) invasion following fire disturbance and the effectiveness of revegetation in improving ecological integrity in a degraded semi-arid shrub steppe system. The effectiveness of rehabilitation efforts was assessed from measurements of arthropod richness, vegetation and arthropod community composition, and ground cover characteristics in three habitats: undisturbed, burned and weed-infested (B. tectorum), and burned and rehabilitated with native and non-native vegetation. Arthropods were collected in each habitat using pitfall traps. Differences in arthropod richness were compared using rarefaction curves. Non-metric multidimensional scaling, and non-parametric multivariate statistical procedures including …

Music And The Modern Maya: A Reception Study Of Rock-Maya Music In Guatemala, Malcolm Miguel Botto Jul 2008

Music And The Modern Maya: A Reception Study Of Rock-Maya Music In Guatemala, Malcolm Miguel Botto

Theses and Dissertations

The current global flows of people, capital, technology, images and ideas--a phenomenon described as "mediascapes" by Arjun Appadurai (1996), traverse the most isolated Maya communities in Guatemala. These flows have recently influenced the creation of hybrid media products among the Maya. Among them we find an emerging indigenous musical genre called "Rock-Maya." I use reception analysis methods to document the encoding and decoding of this new indigenous medium of communication. Through qualitative interviews I attempt to show how K'iche'-Maya youth appropriate, what Motti Regev (1997) calls, the rock aesthetic to promote a sense of K'iche'-Maya youth identity in a modern …

Investigating And Modeling Possible Mechanisms By Which Healthy Cell Membranes Become Resistant To Hydrolysis By Secretory Phospholipase A2, Jennifer Nelson Jul 2008

Investigating And Modeling Possible Mechanisms By Which Healthy Cell Membranes Become Resistant To Hydrolysis By Secretory Phospholipase A2, Jennifer Nelson

Theses and Dissertations

Secretory phospholipase A2 (sPLA2) behaves differently toward the membranes of healthy cells compared to those of damaged or dying cells. The enzyme catalyzes rapid and sustained hydrolysis of compromised cells consistent with a simple catalytic mechanism. In contrast, when healthy cells are incubated with sPLA2, they become resistant to hydrolytic attack as manifest by three unusual observations: First, hydrolysis is transient and represents only a small fraction of the total membrane phospholipid content. Second, subsequent addition of sPLA2 fails to generate additional product. Third, the apparent potency of the enzyme to cause the membrane to be refractory is much greater …

Evaluation Of Downy Mildew (Peronospora Farinosa F.Sp. Chenopodii) Resistance Among Quinoa Genotypes And Investigation Of P. Farinosa Growth Using Scanning Electron Microscopy, Leilani Kitz Jul 2008

Evaluation Of Downy Mildew (Peronospora Farinosa F.Sp. Chenopodii) Resistance Among Quinoa Genotypes And Investigation Of P. Farinosa Growth Using Scanning Electron Microscopy, Leilani Kitz

Theses and Dissertations

Quinoa (Chenopodium quinoa Willd.) is a pseudocereal native to the Andean region of South America and a staple crop for subsistence farmers in the altiplano of Bolivia and Peru. Downy mildew is the most significant disease of quinoa caused by the pathogen Peronospora farinosa f.sp. chenopodii Byford. This disease greatly impacts quinoa crops with yield losses up to 99%. As fungicides are expensive for farmers, the development of resistant cultivars appears to be the most efficient means for controlling downy mildew. The quinoa germplasm bank contains high amounts of genetic diversity, some of which exhibit mildew resistance. Methods for evaluating …

A Comparative Analysis Of Hydrolysis Kinetics By Spla2 Isoforms During Apoptosis In S49 Cells, Erin Dalene Olson Jul 2008

A Comparative Analysis Of Hydrolysis Kinetics By Spla2 Isoforms During Apoptosis In S49 Cells, Erin Dalene Olson

Theses and Dissertations

Secretory Phospholipase A2 (sPLA2) represents a diverse class of roughly 20 enzymes, 12 of which have been identified in humans. These isoforms can be distinguished based on their tissue distribution, structure, and regulation. These differences in structure between the isoforms lead to the question does the enzyme's ability to respond to physical changes in the membrane during apoptosis governed by structure. S49 cell apoptosis was initiated by treatment with either the glucocorticoid dexamethasone (6–48 h) or with the calcium ionophore, ionomycin. The rates of hydrolysis were compared with each treatment condition for various concentrations of snake venom and human groups …

Studies On Transformation Of Tomato(Solanum Lycopersicum L.) And Arabidopsis Thaliana Using Chimerical Constructs Of Varying Tospoviral Origin, Joshua Nathaniel Cobb

Theses and Dissertations

Pathogen derived resistance (PDR) is a recent breakthrough where plant hosts can be made to be resistant to viral infections through transformation with conserved viral genes. Given the severity of Tospovirus diseases worldwide (particularly in tomato), PDR has the potential to garner large yield returns where pathogen populations have overcome the established resistance. Tomato breeding lines FLA7804, FLA8044, and the research line MP1 were used in transformation experiments with potions of the Tomato spotted wilt virus (TSWV) N-gene, and two other chimerical viral nucleocapsid gene constructs from, Impatiens necrotic spot virus (INSV), and Groundnut ringspot virus (GRSV). We conducted 19 …

Religious Networks As A Sociolinguistic Factor: The Case Of Cardston, Benjamin Joseph Chatterton Jul 2008

Religious Networks As A Sociolinguistic Factor: The Case Of Cardston, Benjamin Joseph Chatterton

Theses and Dissertations

Religious affiliation and its inherent membership in an associated social network as a sociolinguistic factor is examined in the community of Latter-day Saints (LDS) in Cardston, Alberta. Building on Meechan's 1998 findings that the LDS community in the area used Canadian Raising in a different set of phonotactic environments than the surrounding non-LDS English speakers, the study aims to determine if the LDS community uses other Canadian speech features differently or less frequently and if any Utah features (defined as Utah English in the literature, being the language of LDS English speakers in Utah) have continued from the settling of …

The Role Of Microrna-155 And Microrna-146a As Putative Oncomirs In The Tumor Progression Of Prostate Cancer, Jennifer Hoyt Jul 2008

The Role Of Microrna-155 And Microrna-146a As Putative Oncomirs In The Tumor Progression Of Prostate Cancer, Jennifer Hoyt

Theses and Dissertations

Prostate cancer is the most common cancer occurring in males. The identification of novel microRNAs (miRs) that contribute to tumor progression represents prospective treatment targets. miRs are small non-coding RNAs important in gene regulation with specific tissue expression patterns. Each miR is thought to affect the expression of hundreds of different RNA targets. Two putative oncomiRs, miR-155 and miR-146a, were shown to be differentially expressed in the human derived, prostate cell sublines M12 and F6. Quantification of endogenous miR expression showed high levels in the metastatic M12 cell line versus low in its weakly tumorigenic F6 variant. The restoration of …

Comparative Cytotoxicity Of An Fda-Approved Cancer Drug To Extracts Of Atriplex Confertifolia On Human Breast And Cervical Cancer Cells, Christopher James Capua

Theses and Dissertations

The severity and number of people affected by cancer is a worldwide problem with millions of people affected annually. The search for treatment and cures of cancer continues to be a global effort. As part of this global effort, many natural products have been tested against cancer cell lines, most from plants located in tropical regions. However, this study reports that extracts of Atriplex confertifolia, a native North American plant, has significant bioactivity against human breast cancer cell lines MCF-7, 435, and 231, and HeLa cells (cervical cancer cells). The bioactivity of A. confertifolia extracts of these cells lines was …

Late Homestead Period Householding At Benmore And Tintic Junction: Comparing Rural And Sub-Rural Communities In Tooele And Juab Counties, Utah, Jennifer Aurora Beard Jul 2008

Late Homestead Period Householding At Benmore And Tintic Junction: Comparing Rural And Sub-Rural Communities In Tooele And Juab Counties, Utah, Jennifer Aurora Beard

Theses and Dissertations

Historical archaeologists are turning more and more attention to the study of capitalism in post-Industrialist nations. Rhoda Halperin's concept of householding considers networks of families or other groups that operate outside of the mainstream capitalist economy. The concept is most often applied in anthropological contexts, but may be a useful tool in the study of homesteading in the American West. At Benmore, a small homesteading community in southern Rush Valley, Tooele County, Utah, 20 families sought to survive by dry farming in a marginal environment. The enthusiasm of such residents as Israel Bennion, whose journal provides deep insight into the …

Genetic Analysis Of The Role Of Smpb In Establishing The Reading Frame On Tmrna, Talina Christensen Watts Jul 2008

Genetic Analysis Of The Role Of Smpb In Establishing The Reading Frame On Tmrna, Talina Christensen Watts

Theses and Dissertations

Ribosomes translate the genetic information encoded by mRNA into proteins. Defective mRNAs can cause stalling of translating ribosomes. The molecule tmRNA (transfer-messenger RNA) rescues stalled ribosomes in eubacteria. Together with its protein partner SmpB, tmRNA mimics a tRNA by entering the ribosomal A site and linking an alanine residue to the growing polypeptide chain. The ribosome then abandons the defective mRNA template and resumes translation on tmRNA, adding ten more amino acids to the nascent polypeptide. As a result of tmRNA action, stalled ribosomes are released and recycled, the defective mRNA is destroyed, and the aborted protein product is tagged …

The Effect Of A Pseudopalate On Voiceless Obstruent Production: A Spectral Evaluation Of Adaptation, Karie Lindsay Dean Jul 2008

The Effect Of A Pseudopalate On Voiceless Obstruent Production: A Spectral Evaluation Of Adaptation, Karie Lindsay Dean

Theses and Dissertations

Many studies in speech communication have provided valuable findings concerning the kinematic nature of speech articulation. This type of research often involves introducing an oral device to the vocal tract such as lingual pellets, magnets, and different forms of pseudopalates to track the movement and placement of the articulators. This study examined the effect of an electropalatography (EPG) pseudopalate on the production of five voiceless obstruents (/p, t, k, s/ and /sh/). Acoustic recordings from 20 adult speakers with typical speech production were made during three different speaking conditions: prior to pseudopalate placement, immediately after placement, and following 20 minutes …

Ion Permeation Through Membrane Channels: Molecular Dynamics Simulations Studies, Morad Mustafa Jul 2008

Ion Permeation Through Membrane Channels: Molecular Dynamics Simulations Studies, Morad Mustafa

Theses and Dissertations

Molecular dynamics simulation was used to study ion permeation through different membrane proteins embedded in a lipid bilayer (DMPC) with different saline solutions. The potential of mean force (PMF) for ion transport was obtained by umbrella sampling simulations. A revised MacKerell force field for tryptophan residues was studied using gramicidin A (gA) channel as a test model. The revised force field contribution to the Na+ PMF was consonant with the prediction from the experimental results, but in stark contrast to the prediction of the CHARMM force field, version 22, for the tryptophan side-chain. A new grid-based correction map algorithm by …

Competitive Exclusion Of Cyanobacterial Species In The Great Salt Lake, Hillary Christine Roney Jul 2008

Competitive Exclusion Of Cyanobacterial Species In The Great Salt Lake, Hillary Christine Roney

Theses and Dissertations

Under Gause's principle two species cannot indefinitely occupy the same niche. The north and south arms of Great Salt Lake are separated by a rail causeway, resulting in salinity differences and color variation between the arms. Farmington Bay is also separated from the south arm by a vehicular causeway to Antelope Island. These causeways allow examination of competitive exclusion of cyanobacteria in the hypersaline environment of the Great Salt Lake. Cyanobacterial distributions partially map salinity, with Aphanothece halophytica proliferating in the north arm, and Nodularia spumigena in the south arm. I hypothesized that cyanobacterial species abundant north of the railway …

Promotion Of Tumor Cell Death Through The Induction Of, Tuyen Nguyen Jul 2008

Promotion Of Tumor Cell Death Through The Induction Of, Tuyen Nguyen

Theses and Dissertations

Microtubule poisons have proven to be effective in the treatment of a variety of malignancies. Although taxol-based derivatives promote microtubule stabilization, there is continuing interest in compounds that, like colchicines, act as microtubule destabilizing agents. Previous work from this laboratory showed that the novel microtubule poison, JG- 03-14, was active against breast tumor cells, promoting autophagic cell death. In the current work, we studied the influence of JG-03-14 on p53 wild type HCT116 colon carcinoma cells. A crystal violet sensitivity assay indicated that JG-03-14 induced growth inhibition, with 75% suppression of growth evident at a concentration of 500 nM. Time …

Using A Chelator-Buffered Nutrient System To Study Phosphorus, Manganese And Zinc Interactions In Russet Burbank Potato, Steven A. Barben

Theses and Dissertations

Potato production requires high phosphorus (P) application with potential negative environmental or nutritional consequences for potato as well as for subsequent crops. Impacts of high available P on yield and plant nutrition of species in potato cropping rotations are inadequately understood, and could result in antagonistic interactions with cationic micronutrients such as zinc (Zn) and manganese (Mn). Three hydroponic experiments were conducted with Russet Burbank potato to elucidate P and Zn relationships and associated interactions with other nutrients. In the first experiment, P solution concentration was constant at 256 µM while Zn concentration varied: 0.1, 2, 6, 18, 54, 162 …

Effect Of Ca And Ph On Disease Severity Of Pink Rot Phytophthora Erythroseptica In Russett Norkotah Potato Solanum Tuberosum, Jared H. Benson Jul 2008

Effect Of Ca And Ph On Disease Severity Of Pink Rot Phytophthora Erythroseptica In Russett Norkotah Potato Solanum Tuberosum, Jared H. Benson

Theses and Dissertations

Phytophthora erythroseptica Pethyb. is a devastating fungal pathogen of potato (Solanum tuberosum). The pathogen causes a disease known as pink rot. Pink rot results in necrosis and decay of tubers; and is responsible for major losses pre and post harvest. Attributes of the disease are progressing toward epidemic proportions. To help prevent such dramatic outbreaks of the disease, understanding the factors associated with incidence will provide opportunities to control the pathogen. A link between pH and disease severity has been observed. We studied the effects of pH and Ca to determine their influence on disease development. Low pH and Ca …

A Compliant Threshold Acceleration Sensor Integrated With Radio Frequency Identifiable Tags, Benjamin L. Todd Jul 2008

A Compliant Threshold Acceleration Sensor Integrated With Radio Frequency Identifiable Tags, Benjamin L. Todd

Theses and Dissertations

Fully compliant bistable mechanisms have been proposed to be used as threshold accelerometers. The advantages to using these devices are that they require no external power to operate and maintain their sensing state. Using this characteristic the devices can be integrated with passive radio frequency identification tags (RFID). This allows for the sensing package to lay dormant with no maintenance needed until the sensor is read by the RFID reader. This thesis presents a successfully fabricated and integrated threshold accelerometer with a passive RFID tag. This in turn has been successfully read with an RFID reader and shown to act …

Dialectic, Perspective, And Drama, Ethan Mckay Sproat Jun 2008

Dialectic, Perspective, And Drama, Ethan Mckay Sproat

Theses and Dissertations

This project is by and large a project of elucidation: it may add something to studies of Kenneth Burke, but I doubt it adds much to Kenneth Burke's studies. This thesis begins and ends with analyses of Burke's famous motto Ad Bellum Purificandum (or Toward the Purification of War). The Introduction focuses on "war" while the Conclusion focuses on "purification." In short, purified war is a dialectical activity which actively and perpetually pits divergent perspectives against each other. Such an activity keeps the conflictual nature of divergent perspectives in verbal and symbolic arenas rather than physical ones. Burke owes this …

Pygmy Rabbit (Brachylagus Idahoensis) Habitat Use, Activity Patterns And Conservation In Relationship To Habitat Treatments, Janet E. Lee Jun 2008

Pygmy Rabbit (Brachylagus Idahoensis) Habitat Use, Activity Patterns And Conservation In Relationship To Habitat Treatments, Janet E. Lee

Theses and Dissertations

This study examined activity patterns and habitat use of pygmy rabbits (Brachylagus idahoensis) in mechanically treated and untreated areas in south-central Utah 2005-2008. We monitored fecal pellet plots in continuous sagebrush habitat as well as along treatment edges to record deposition and leporid presence over timed periods. Pygmy rabbit use of big sagebrush was higher than black-tailed jackrabbits (Lepus californicus) and mountain cottontail rabbits (Sylvilagus nuttallii ) (P< 0.01) relative to treated areas (P <0.01). We also compared pygmy rabbit use of areas with continuous sagebrush to residual sagebrush in a sample of mechanically treated areas. Our results suggest a treatment effect with higher (P <0.01) average counts of pygmy rabbit pellets in areas with continuous sagebrush compared to sagebrush strips and islands within treated areas. Before the big sagebrush biotype inhabited by pygmy rabbits is treated to reduce the occurrence and dominance of big sagebrush, we recommend managers consider two options. The first is no treatment, thus preserving, as is, the critical habitat of the pygmy rabbit and other sympatric big sagebrush obligate species of wildlife. The second option cautiously introduces the first prescription of habitat treatment ever recommended in relationship to the pygmy rabbit. This prescription includes recommended widths of the treated areas, seed mixes, widths of the preserved intact big sagebrush habitat for pygmy rabbits as well as suggested grazing systems for domestic livestock. Activity patterns of pygmy rabbits at their burrow were documented through the use of remote cameras. Photographs were analyzed for temporal and seasonal patterns of activity. Our results suggested that time of day was important in the activity level of pygmy rabbits while season was not. Pygmy rabbits were active during all time periods of the day but the greatest levels of activity occurred at night. Numerous other wildlife species were recorded by our remote cameras including other species of leporids, birds, rodents, reptiles and terrestrial predators.
