Ciliary neurotrophic factor, a member of the interleukin-6 family of cytokines, promotes neuronal survival and axonal sprouting in various populations of neurons including sensory neurons, motor neurons, and magnocellular neurons (MCN's) of the supraoptic nucleus (SON). Unilateral lesion of the hypothalamo-neurohypophysial tract (HNT) results in an initial period of axonal degeneration followed by a restoration of axonal density in the neurohypophysis by 30 days post-lesion. The restoration of axonal density is attributed to collateral sprouting from MCN's of the intact contralateral SON. Caveolae are cholesterol-rich plasma-membrane (PM) microdomains which foster wide-range of signal-transduction events including Ca2+-influx. Caveolae contain Caveolin1 (Cav1) as their major structural-protein. Transient receptor potential canonical 1 (TRPC1) channels constitute an integral-component of store-operated Ca2+ channels (SOCC) which initiate store-operated Ca2+ entry (SOCE), in response to endoplasmic reticulum (ER) Ca2+-store depletion. Recently, Stromal interaction molecule 1 (STIM1) was identified as the ER Ca2+-sensor which, following store-depletion, forms clusters and translocate to peripheral-ER at sub-plasma membrane region and communicates with PM-SOCC activating SOCE. Lipid peroxidation is a free radical-mediated process characterized by addition of molecular oxygen to polyunsaturated fatty acids (PUFA). Lipid peroxidation occurs basally, resulting in products that participate in normal signaling. One product is trans-4-hydroxy-2-nonenal (HNE). HNE is a product of lipid peroxidation of n-6 PUFA, such as arachidonic acid, and has the ability to mofify proteins, deplete glutathione, and induce apoptosis. Brain is a tissue that consumes large amounts of oxygen and contains an abundance of PUFA, which suggests a high degree of susceptibility of lipid peroxidation. Selenium Determinations, Nanoparticle Sorbent Analysis And Nanoparticle Catalysts For Chemiluminescence, Sandra Grace Hazelton

Three projects in chemical analysis are presented. In the first, calibration-free Se(IV) determinations were accomplished using an electrochemical flow cell and stripping potentiometry at an unmodified gold electrode. The method was optimized prior to analysis of water and wheat samples. A detection limit of approximately 0.5 ppb Se(IV) was observed. Method accuracy was good and precision was acceptable (%RSD < 10). In the second project, stripping analysis was also employed, this time to analyze metal ion adsorption on carboxylic acid-modified silica nanoparticles. The metals analyzed were Cu(II), Cd(II), and Pb(II). By using anodic stripping voltammetry, adsorbed metal ions could not be detected, thus the need to separate the sorbent from the free metal ions was eliminated. All three metal ions in this study adsorbed in a nearly one-to-one ratio with the carboxylic acid groups at high pH (∼1000 μmol g-1). The effect of pH on adsorption behavior was also investigated. The third project also employed silica nanoparticles and a flow-through detection system. Co(III)-triethanolamine-doped silica nanoparticles were synthesized and used as catalysts for the luminol-H2O2 chemiluminescence reaction. It was found that the chemiluminescent signal increased linearly with the concentration of nanoparticles in bulk solution. The detection limit for the nanoparticles was 1.5 × 10-12 mol L-1 . Luminol-H2O2 chemiluminescence had superior precision when catalyzed by nanoparticles as compared with the free cobalt(III) complex. It was demonstrated that the nanoparticles met the criteria required for a biosensor label.

Multiple sclerosis is an autoimmune disease which can affect several areas of functioning including physical and psychological functioning. Many individuals experience difficulties with depression and neuropsychological capabilities. Additionally, social networks often appear to be impacted by the disease. Previous research has shown that some of the physical functioning as well as the depressive symptomatology in multiple sclerosis can be ameliorated by improvements in social support. The present study used archival data to explore the impact of social support on neuropsychological function in 112 adults with multiple sclerosis. Transient receptor potential canonical 3 (TRPC3), a non-selective Ca 2+-permeable cation channel, is a member of the TRP superfamily of ion channels. The mode of activation of TRPC3 is governed by a cascade of Phospholipase C (PLC) mediated signaling events, following G-protein coupled receptor (GPCR) or tyrosine kinase receptor activation, resulting in endoplasmic reticulum (ER) Ca2+ store depletion. The purpose of this qualitative study was to describe perceptions held by Native American students currently residing on a reservation, of the experiences they encountered while attending a non-reservation school. Data for the study was drawn from five participants who attended a high school nearby, but not on, a Native American reservation. Using a phenomenological approach, participants were interviewed three times. The audio recorded interviews were transcribed, coded, and analyzed.Three themes emerged from the data. Theme one: Participants faced numerous challenges stemming from home and school environments. Chief among these were issues related to living on a reservation, and issues …

The purpose of this study was to explore the tensions that existed between my co-teaching partner and me while working together during the last four years. Additionally, I studied how my partner, the special educator, and I, the general educator, negotiated the tensions that came up during our collaboration. Using a narrative approach to share our stories about our teaching, I investigated how we worked together and contributed to our co-teaching relationship. Objective: Determine change in eating competence (EC) and factors related to EC in students enrolled in basic nutrition courses at a major private university. Design: Eating competence was measured by administering the ecSatter Inventory (ecSI) both before and after class intervention. Additional data on eating disorder prevalence, food security, and general demographics were also collected in the same structured survey. Significance identified with p value <0.01. Setting/Participants: This survey was administered to 566 students enrolled in basic nutrition courses at a major private university in the western United States. Analysis: ANOVA was used to determine relationships between ecSI end scores and participant characteristics. ANCOVA was used to determine relationships between change in ecSI scores over time and participant characteristics. Results: Enrollment in NDFS 100 is associated with an overall increase in eating competence (EC). However, students with current eating disorders had a significant decrease in EC during enrollment. Current or past eating disorders and low or very low Food Security Status were associated with lack of EC. Females and younger participants were not eating competent at the course end. Enrollment in NDFS 201 was not associated with significant change in EC. Conclusions: Basic nutrition instruction improves EC among students without eating disorders. Low food security status and presence of an eating disorder may be a barrier to improving EC.

In this dissertation, fluorescent silica nanoparticles (NPs) have been successfully developed to improve the performance of bioanalysis. In the first step, the sizes of silica NPs were precisely controlled by changing the organic solvent in the reverse microemulsion method. The size of the NPs can be continuously tuned from 20 to 100 nm. Then, multicolored fluorescent silica NPs were developed with controllable fluorescence properties by manipulating the properties of the dopants. Given potential risk factors in the lives of adolescents, parents are usually motivated to monitor and protect their adolescents. There is a need to better understand what combinations of parental dimensions and practice best influence an adolescent's propensity to disclose or conceal personal information with their parents. This paper examines how parenting dimensions (warmth, psychological control, and harsh punishment) and the parenting practice of solicitation influence an adolescent's propensity to disclose or conceal information. Three different approaches were used to prepare dual mode nanoparticles using a previously described reverse micelle technique. Superparamagnetic Fe@FeOx core shell nanoparticles were chosen to be the magnetic component for all three dual mode systems. In the first type of particles, 2- amino-1,3- propane diol (APD) was used as a functionalization ligand to stabilize the surface of the particles and its functional amino group also provided a binding site for the attachment of a fluorescent probe. Aquatic bacteria respond to changing environmental conditions through a variety of mechanisms including changes in abundance, shifts in community composition and variable activity states. In the tidal-freshwater James River, variation in bacterial abundance was linked to nutrient availability and autochthonous production with highest bacterial densities associated with low-nutrient, high-chlorophyll a conditions. Laboratory experiments revealed that bacterial growth rates were nutrient limited at the low-nutrient site, while co-limitation (nutrients, glucose, light) was apparent at the high nutrient site. Despite large differences in abundance, community composition was similar based on TRFLP and 16S rDNA pyrosequencing. Community similarity was lower among rRNA libraries …

This study examined the thermodynamic electrochemistry of the process of proton exchange membrane electrolysis of a 6 kW electrolyzer manufactured by Proton Energy Systems. Each modeling parameter was examined based on the definition and the theories which characterize their behavior using experimental data generated at the University of North Dakota hydrogen laboratory. The exchange current densities, membrane resistance and charge transfer coefficients were examined at both electrodes for their temperature dependence. While most of the temperature studies revealed an Arrhenius behavior, the exchange current density at the anode showed a deviation. This leads me to believe that stacking cells together …

This study is a description of male and female secondary students' experiences of safety in public schools. Gender differences in reported victimization and perceptions of school safety have been noted. The National Center for Educational Statistics ([NCES], 2006) reported that boys were the victims of violent acts in the schools more often than girls. Their unifying principle is the measurement of electrical impedance for the detection and analysis of biological materials. The purpose of this project was to investigate the mechanism of action of acupuncture on a critical neural substrate involved in alcoholism. Specifically, this study evaluated the effects of stimulation of the acupuncture Shenmen (HT7) point on inhibitory GABA neurons in the ventral tegmental area (VTA), a midbrain structure implicated in drug and alcohol abuse, and ethanol self-administration. Phosphodiesterase Type 5 (PDE5) inhibitors are cardioprotective against ischemia/reperfusion (I/R) injury. However, it remains uncertain if I/R affects PDE5. We hypothesized that generation of reactive oxygen species (ROS) during I/R leads to upregulation of PDE5, which contributes to pathological changes following acute myocardial infarction (AMI). Adult male ICR mice were subjected to 30 minutes of in vivo or ex vivo I/R. To examine the role of ROS, a subset of hearts were perfused with 100 µM hydrogen peroxide (H2O2). Tropical forests provide important habitat for a tremendous diversity of plant and animal species. However, limitations in measuring and monitoring the structure and function of tropical forests has caused these systems to remain poorly understood. Remote-sensing technology has provided a powerful tool for quantification of structural patterns and associating these with resource use. Satellite and aerial platforms can be used to collect remotely sensed images of tropical forests that can be applied to ecological research and management. The first one is an aqueous reduction method using sodium borohydride and sodium citrate. The leading causes of liver disease are Hepatitis C virus infection and chronic alcohol abuse. Alcohol accelerates liver disease in HCV but the mechanisms are poorly understood. The identification of molecular gene expression profiles on human liver tissue was performed using microarrays. Samples were obtained from alcoholic-cirrhotic, HCV-cirrhotic, HCV/alcohol-cirrhotic and control non-cirrhotic liver tissue. Probe set expression summaries were calculated using RMA. Probe set level linear models were fit where probe set expression was modeled by HCV status, alcohol status, and the interaction between HCV and Alcohol. This study explores the notion of racial empathy and whether or not the nonadopted white siblings in transracially adoptive families develop the ability to empathize within the context of race. Studies suggest that adoptees are at risk for a number of psychopathological behaviors. To understand the etiology of this risk, 150 socially housed rhesus macaques were studied, including 107 infants reared with their biological mothers and 43 infants reared with unrelated adoptive mothers. Mother-infant behaviors were recorded across the first 6 months of life. Analyses were performed using a hierarchical linear mixed model. All reported results were tested at p<0.05. Adopted infants were observed on average to approach and leave their mothers more frequently, explore the environment and locomote longer, exhibit more anxiety-like behavior, spend less time being held to their mother's breast, and were rejected by their mothers more when compared to nonadopted infants, indicating they are more likely responsible for maintaining the relationship. They also direct and receive more noncontact aggression on average to other social group members, and showed evidence of higher anxiety exhibiting high levels of anxiety-like self-directed behavior when compared to nonadopted infants. Also, results indicate that adopted infants have significantly lower levels of the CSF serotonin metabolites 5-hydroxyindoleacetic acid when compared to nonadopted infants.

This is a study of how a teacher's understanding of the nature of science (NOS) correlates to student understanding of the nature of science. Participants are in semester long seventh grade science classes in a suburban school district. Seven strands of the nature of science were identified in the literature. Four strands were analyzed in this study. Teachers were ranked according to their understanding of the nature of science and compared to their corresponding students' average gain. Malaise traps are a common tool for collecting insects used by many researchers. Although there have been variations in the models and materials used for Malaise traps, the potential for sampling bias due to mesh gague has been explored inadequately. This study compared coarse and fine mesh Townes model Malaise traps in two habitats on the Grand Staircase-Escalante National Monument. The two habitats next to the Lick Wash trailhead were defined by dominant vegetation type – sagebrush and grasses or Piñon-Juniper.
