The aim of this study is the three-dimensional structural characterization of the genome packaging inside viral capsids via cryo-electron microscopy and three-dimensional reconstruction. The genome of some single-stranded viruses can be densely packaged within their capsid shells. Several stretches of the genome are known to adopt stable secondary structures, however, to date, little is known about the three-dimensional organization of the genome inside their capsid shells. Two techniques have been developed to facilitate the structural elucidation of genome packaging: the asymmetric random-model method, and the symmetry-mismatch, random model method. In recent years there has been a growing interest in the relationship between religion and the environment. The purpose of this project is to explore the environmental practices and attitudes of Christian churches in two Virginia Communities. The two communities; the Northern Neck and Eastern Shore are located on the Chesapeake Bay and have a shared history of dependence on the Bay. The results of the dissertation demonstrate the prevalence of environmental programs in the population, the nature of these programs and the respondents’ attitudes towards a host of environmental issues. Top-loading This study compared the effects on oxygen (O2) transport of four fluids: 5.9% human serum albumin (HSA) a non-O2 carrying iso-oncotic solution (volume control);HBOC-201 (Hemopure, Biopure Corp., Cambridge, MA); MP50, HBOC-201 with a P50 of 18 mmHg; and lastly LP50, an HBOC-201 with P50 of 17 mmHg and higher viscosity (4 cP). It has been proposed that HBOC with a higher O2 affinity and a viscosity closer to that of whole blood will cause less vasoconstriction and thus a lower MAP than Hemopure (P50 = 40 mmHg and 2.2 cP). Marital discord contributes to the development and continuation of depression and to the recurrence of depressive episodes for those in troubled relationships. Early research suggests that family therapy may reduce the severity and frequency of depressive episodes through modification of family interactional patterns. This would result in a reduction in the cost of treating depression. This study summarizes the literature linking family dynamics with depression. Knowledge transferred during meetings is often ephemeral in nature and thus must be captured if it is to be retained. Ideally, a capture solution should be able to 1) accommodate any number of screens without sacrificing image quality and 2) allow dynamic access to a complete media capture while the capture is taking place. This project analyzed gender and color-specific differences in event-related potentials (ERPs). Previous studies have shown that males process color differently than females. In a recent study, sex differences were found in ERPs during a visual object recognition task. There were higher EEG amplitudes in females (especially P300) than males. Significant sex and color-specific differences have been found in diseases involving altered dopamine (DA) machinery. This thesis analyzes the linguistic attitudes of natives from Santiago, Chile regarding three linguistic phenomena present in Chilean Spanish of Santiago: the fricative realization of the Spanish /tʃ/, the assibilated realization of /tɾ/ and the use of the definite article before the first names of people. The social factors of age, sex and socioeconomic status are acknowledged as possible factors contributing to the linguistic attitudes of the 64 participants interviewed.

Self assembled monolayers (SAMs) are widely used for surface modification. Alkylsilane monolayers are one of the most widely deposited and studied SAMs. My work focuses on the preparation, patterning, and application of alkysilane monolayers. 3-aminopropyltriethoxysilane (APTES) is one of the most popular silanes used to make active surfaces for surface modification. To possibly improve the surface physical properties and increase options for processing this material, I prepared and studied a series of amino silane surfaces on silicon/silicon dioxide from APTES and two other related silanes by chemical vapor deposition (CVD). A new technique, called nanoinjection, is being developed to insert foreign DNA into a living cell. Such DNA transfection is commonly used to create transgenic organisms vital to the study of genetics, immunology, and many other biological sciences. In nanoinjection, DNA, which has a net negative charge, is electrically attracted to a micromachined lance. The lance then pierces the cell membranes, and the voltage on the lance is reversed, repelling the DNA into the cell. While age-related sex chromosomal aneuploidy is a well-characterized phenomenon, the relationship between autosomal loss and age remains unclear. The emergence of the specific and highly sensitive fluorescence in situ hybridization (FISH) technology has enabled investigators to study interphase cells, thereby overcoming problems inherent with the study of metaphase spreads for acquired aneuploidy assessment. Despite all the advantages of this technique, there are some limitations that could be misleading when scoring interphase autosomal aneuploidy. The assessment of ventilatory efficiency (VE/VCO2 slope) is increasingly being utilized to complement traditional cardiorespiratory fitness testing during graded exercise. The purpose of this study was to compare cardiorespiratory responses and ventilatory efficiency in obese children during a progressive exercise test to volitional fatigue performed on a treadmill and a cycle ergometer. Fifteen obese male (N=3) and female (N=12) adolescents aged 10 to 18 years were recruited in the study and completed both the treadmill and cycle ergometer trials. The bacterial species belonging to the order Mycoplasmatales have highly truncated genomes, and are thus ideal for studying genome evolution patterns. Fourteen members (twelve species) of this order were selected for study of genome evolution based on gene function and phylogeny. A database was constructed that consisting of the set of genes that are common to all of these species, and these genes were further subdivided based on their functions. To deal with this problem, maize generates a wide variety of responses to attack by pests, from activation of woundresponse pathways to the release of volatile compounds. We demonstrate the success of recurrent neural networks in gene network inference and expression prediction using a hybrid of particle swarm optimization and differential evolution to overcome the classic obstacle of local minima in training recurrent neural networks. We also provide an improved validation framework for the evaluation of genetic network modeling systems that will result in better generalization and long-term prediction capability. Success in the modeling of gene regulation and prediction of gene expression will lead to more rapid discovery and development of therapeutic medicine, earlier diagnosis and treatment of adverse conditions, and vast advancements in life science research.

The present study examined the relationships of depression, social support, and socio-demographic factors on health behaviors of mothers with preterm infants hospitalized in the neonatal intensive care unit (NICU). In addition, the study also assessed the moderation effect of social support on the relationship between depression and health behaviors. Eighty-nine mothers with hospitalized infants in the central Richmond area participated in the study. Analyses found that mother’s education level and her marital status to be significantly associated with her health behaviors. I examined a system of 378-riverine rock pools situated along the James River in Richmond, Virginia, to uncover the factors influencing their macroinvertebrate diversity and community composition. I characterized (total = 60) the physical characteristics, water quality, hydrology, and community assemblages in July 2009 for a subset of pools. I examined whether selected environmental factors (pH, nearest neighbor distance, depth, surface area and inundation height) were related to macroinvertebrate diversity and community composition. The objective of this research was to evaluate the impact of current body armor design on physical performance during simulated law enforcement activities. Twenty participants completed three trials of 13 individual activities representative of routine law enforcement activities. Three body armor configurations were evaluated: baseline (i.e., no armor), concealable body armor, and external body armor. Dependent variables included task completion time, heart rate, and center of pressure (COP). Repeated measures ANOVAs were used to test the dependant variables, with Tukey’s HSD post hoc tests used where appropriate. Ethanol fermentation of highly concentrated aflatoxin-contaminated corn (Zea mays) was conducted on a lab scale to determine if aflatoxin concentrated in the distilled ethanol and/or dry distillers grain end-products. Alliquots of fermented mash, distilled ethanol, stillage, and dry distillers grain (DDG) were analyzed via LC-MS/MS and immunoassay detection methods for aflatoxin. Results indicate that aflatoxin does not greatly concentrate during fermentation in the DDGs and is undetectable in distilled ethanol. Addition of binders, MTB-100®, to aflatoxin-contaminated DDGs showed great reduction in aflatoxin concentrations when analyzed via LC-MS/MS. Also, an experiment investigating detoxification of aflatoxin using Clorox® was conducted. Results obtained …

Prescribed fire and imazapyr are two silviculture tools used to control hardwood midstory competition in intensively managed, mid-rotation pine (Pinus spp.) stands but also may support conservation of biodiversity in the southeastern United States. Therefore, I investigated select measures of biodiversity response, small mammals, reptiles, amphibians, carabid beetles, songbirds, and vegetation communities, to fire and imazapyr treatments in intensively managed, mid-rotation pine stands of east-central Mississippi. This work focused on decay and dimensional stability of southern pine lumber and environmental issues associated with emissions released during kiln drying of southern pine. In one study decking boards were treated with a 3% resin acid solution to increase the dimensional stability. The boards were placed on a roof and weathered for two years. The increased water repellency reduced moisture gain following summer rainstorms by one-third compared to untreated matched samples, this significantly reduced splitting by half and cupping by one-third.
