Spotted Fever Group Rickettsiae, Rickettsia Parkeri And "Candidatus Rickettsia Andeanae", Associated With The Gulf Coast Tick, Amblyomma Maculatum Koch, Flavia Araujo Girao Ferrari

Theses and Dissertations

The public health and veterinary importance of Gulf Coast ticks, Amblyomma maculatum Koch (1844) have become more apparent during the last several decades. In addition, new records of this three-host ixodid tick presently show a geographic distribution throughout much of the southern United States. Rickettsia parkeri, a spotted fever group rickettsia (SFGR) that is commonly found infecting the Gulf Coast tick, was only recently recognized as a human pathogen. Over the last decade, more than 20 human cases of disease caused by R. parkeri have been recognized in the Americas, all of which were similar in presentation to mild Rocky …

Remote Sensing Of Cyanobacteria In Turbid Productive Waters, Sachidananda Mishra Aug 2012

Remote Sensing Of Cyanobacteria In Turbid Productive Waters, Sachidananda Mishra

Theses and Dissertations

Cyanobacterial algal bloom is a major water quality issue in inland lakes, reservoirs, and estuarine environments because of its scum and bad odor forming and toxin producing abilities. Health risks from cyanobacterial toxin can vary from skin irritations to fever, intestinal problems, and neurological disorders. Terminations of blooms also cause oxygen depletion leading to hypoxia and widespread fish kills. Adding to the problem, many species of cyanobacteria produce odorous compounds such as geosmin and 2-methylisoborneol (MIB) that cause “earthy-muddy” and “musty” odor in drinking water, which is also a serious issue in aquaculture and drinking water industry. Therefore continuous monitoring …

In Vitro Catalytic Activity And Inhibition Study Of Prnb From Burkholderia Ambifaria, Qi Ge

Theses and Dissertations

PrnB is a heme-containing enzyme, which catalyzes the ring rearrangement reaction of 7-chlorotryptophan to produce 3-(3-Chloro-2-nitrophenyl)pyrrole. This thesis describes the initial isolation and characterization of PrnB, the second enzyme associated with the pyrrolnitrin biosynthetic pathway in Burkholderia ambifaria. Additionally, alternative peroxidase reactivity was used to study how amino-acids bind to the substrate binding pocket of PrnB. The peroxidase activity of PrnB was measured using three different peroxidase activity assays at various pH values. The peroxidase data was compared to similar studies with the classic peroxidase, Horseradish peroxidase (HRP). Generally, PrnB showed weak peroxidase reactivity. However this weak reactivity was an …

Theses and Dissertations

Fresh tannic acid (5%) (FTA), heated to 121C for 15 min (PTA), and the combination of FTA and phosphates (FPH) were vacuum tumbled with catfish fillets and compared to water tumbled fillets (CTL). Fillets treated with FTA or PTA had higher whiteness whereas all treated fillets had higher chroma values than CTL, resulting in a deeper yellow color fillet. Microbial shelf life (CTL, log CFU/g, APC) of FTA and PTA treated fillets was extended by two days, to11.8 d at 4C, but FPH fillets did not reach over 5 log CFU/g for 15 d of storage. Fat from FTA and …

Addressing Sample Bias And Representativeness At The Kinlock Site (22su526) A Freshwater Mussel Shell Ring In The Mississippi Delta, Joseph Alan Mitchell Aug 2012

Addressing Sample Bias And Representativeness At The Kinlock Site (22su526) A Freshwater Mussel Shell Ring In The Mississippi Delta, Joseph Alan Mitchell

Theses and Dissertations

Applied zooarchaeology provides baselines which can be used in modern conservation biology to better understand how faunal communities have changed over time. This goal can only be accomplished, however, by first accounting for the multiple biases present within the archaeological record, and how they may affect sample representativeness. Taxonomic analysis was conducted on freshwater mussel shell from the late prehistoric (ca. A.D. 700 - 1200) Kinlock site, Sunflower County, Mississippi. Species-area curves and biodiversity indices demonstrate that random sampling of surface clusters of shell, up to about 4,000 valves, provides an adequate picture of the overall surface assemblage. Comparison of …

Alternate Stable States In Ecological Systems, Sarath Sasi Aug 2012

Alternate Stable States In Ecological Systems, Sarath Sasi

Theses and Dissertations

In this thesis we study two reaction-diffusion models that have been used to analyze the existence of alternate stable states in ecosystems. The first model describes the steady states of a logistic growth model with grazing in a spatially homogeneous ecosystem. It also describes the dynamics of the fish population with natural predation. The second model describes phosphorus cycling in stratified lakes. The same equation has also been used to describe the colonization of barren soils in drylands by vegetation. In this study we discuss the existence of multiple positive solutions, leading to the occurrence of S-shaped bifurcation curves. We …

The Analysis Of The Expression And Phenotype Of The At3g02400 Gene In Arabidopsis Thaliana, Will Gray Aug 2012

The Analysis Of The Expression And Phenotype Of The At3g02400 Gene In Arabidopsis Thaliana, Will Gray

Theses and Dissertations

The involvement of several genes in DNA repair in animals and bacteria, and the induced expression of At3G02400 by Mitomycin C, suggest that this gene is involved in DNA repair in plants. I have tested this hypothesis by studying the regulation of its expression and identifying the function of this gene. To study the expression of this gene, I used a reporter line and bio-computing tools. To identify the function of the gene, I have isolated two T-DNA mutants and screened Targeting Induced Local Lesions in Genomes (TILLING) mutants. T-DNA mutagenesis is the process of inserting transfer DNA into the …

Sediment And Phosphorus Dynamics Behind Weirs In Agricultural Drainage Ditches, Elizabeth Louise Usborne Aug 2012

Sediment And Phosphorus Dynamics Behind Weirs In Agricultural Drainage Ditches, Elizabeth Louise Usborne

Theses and Dissertations

Low grade rip rap weirs installed in agricultural surface drainage ditches manage downstream eutrophication by slowing water flow, allowing sediments time to settle out of the water column and phosphorus (P) to sorb to soil. A laboratory experiment was conducted in microcosm chambers to simulate increased hydraulic residence time caused by weirs and two field studies were conducted to compare experimental data with field data and determine sediment deposition rates. One field study monitored weirs monthly after installation and the other measured weirs of varying ages. Weirs retained significantly more water and sediment than controls. Longer inundation times led to …

Manganese Oxidation, Pseudomonas, And Potential Mercury Remediation, Kendra L. Wright Aug 2012

Manganese Oxidation, Pseudomonas, And Potential Mercury Remediation, Kendra L. Wright

Theses and Dissertations

East Fork Poplar Creek (EFPC) in Oak Ridge, TN was highly contaminated with elemental mercury in the 1950 and 1960. The area is still experiencing the effects of mercury contamination, and researchers are searching for ways to remediate the EFPC. One possible mechanism for bioremediation is the use of biogenic Mn oxides to remove heavy metals from water systems. Native Pseudomonas bacteria species were isolated from EFPC in order to examine biogenic Mn oxides production and bioremediation of Oak Ridge slurries. Pseudomonas isolates did produce Mn oxides which bound to mercury, and mercury bound to organic matter significantly decreased. However, …

Effectiveness Of Phosphorus Fertilizers In Hydroponics And Glasshouse Settings With Moderate And High Organic Matter Soils, Jeffrey Sean Christian Summerhays

Theses and Dissertations

Phosphorus (P) is poorly soluble in most soils and, thus, has poor plant uptake efficiency. AVAIL® and Carbond P (CBP) are new fertilizer products shown to increase P use efficiency (PUE) and increase crop yields when grown in P limiting soils. Carbond P has specifically been seen to increases P uptake and crop yields in soils low in P, although effectiveness in regards to soil organic matter is unknown. The objectives of these studies were to determine if the mode of action for these products is related to physiological response, to determine if Carbond P is toxic to plant roots …

A Confidence-Prioritization Approach To Data Processing In Noisy Data Sets And Resulting Estimation Models For Predicting Streamflow Diel Signals In The Pacific Northwest, Nathaniel Lee Gustafson Aug 2012

A Confidence-Prioritization Approach To Data Processing In Noisy Data Sets And Resulting Estimation Models For Predicting Streamflow Diel Signals In The Pacific Northwest, Nathaniel Lee Gustafson

Theses and Dissertations

Streams in small watersheds are often known to exhibit diel fluctuations, in which streamflow oscillates on a 24-hour cycle. Streamflow diel fluctuations, which we investigate in this study, are an informative indicator of environmental processes. However, in Environmental Data sets, as well as many others, there is a range of noise associated with individual data points. Some points are extracted under relatively clear and defined conditions, while others may include a range of known or unknown confounding factors, which may decrease those points' validity. These points may or may not remain useful for training, depending on how much uncertainty they …

Schmidtea Mediterranea Planaris As A Novel Animal Model For Investigating The Dynamics Of Nervous System Development In Fetal Alcohol Spectrum Disorder, Jesse Lowe Aug 2012

Schmidtea Mediterranea Planaris As A Novel Animal Model For Investigating The Dynamics Of Nervous System Development In Fetal Alcohol Spectrum Disorder, Jesse Lowe

Theses and Dissertations

Fetal Alcohol Spectrum Disorder (FASD) is a prevalent developmental disease that is caused by excess in utero exposure to ethanol. The importance of timing and dosage of ethanol has been elucidated by previous investigations conducted with humans, rodents, and other model organisms, but challenges associated with determining functional deficiencies have necessitated the introduction of a novel animal model for investigating the dynamics of FASD. In this thesis, we propose that the freshwater flatworm planarian species, Schmidtea mediterranea, has the capability to fulfill this need. Planaria have the ability to completely regenerate fragments into intact, functional animals, and in these studies, …

Discovery And Biophysical Characterization Of Allosteric Inhibitors Of Factor Xia (Fxia), Malaika Argade

Theses and Dissertations

Thrombosis is one of the leading causes of mortality and morbidity that is associated with myocardial infarction, stroke and pulmonary embolism. Anti-thrombotic agents which intend to reduce the occurrence and severity of thrombosis usually target the enzymes of the coagulation cascade. FXIa, a 160 kDa homodimer is gaining popularity of late as a potential target for anti-thrombotic agents due to its relative safety. A number of inhibitors which target the active site of FXIa have been reported but to our knowledge there have been no inhibitors which act via an allosteric mechanism. The aim of this project was to screen …

Characterizing The Role Of Hspb2 In Cardiac Metabolism And Muscle Structure Using Yeast And Mammalian Systems, Jonathan Paul Neubert Aug 2012

Characterizing The Role Of Hspb2 In Cardiac Metabolism And Muscle Structure Using Yeast And Mammalian Systems, Jonathan Paul Neubert

Theses and Dissertations

HspB2 is a small heat shock protein encoded on human chromosome 11. Less than 1000 base pairs away from HSPB2 and situated in a head-to-head orientation lies the gene encoding another small heat shock protein, CRYAB. Because they are uncommonly close to one another they share regulatory elements. In addition, they share protein homology as sHSPs, suggesting that they perhaps perform aimilar functions. SHSPs such as HspB2 and CryAB are traditionally thought to provide protective effects to cells in response to a variety of stress inducers. In response to stress they form complexes around misfolded proteins or proteins in danger …

Gas Phase Characterization Of Supramolecules Using Cross-Sectional Areas By Fticr And Sustained Off-Resonance Irradiation Collision Induced Dissociation Techniques In A Fourier Transform Ion Cyclotron Resonance Mass Spectrometer, Fan Yang Aug 2012

Gas Phase Characterization Of Supramolecules Using Cross-Sectional Areas By Fticr And Sustained Off-Resonance Irradiation Collision Induced Dissociation Techniques In A Fourier Transform Ion Cyclotron Resonance Mass Spectrometer, Fan Yang

Theses and Dissertations

In my dissertation, I use a Fourier Transform Ion Cyclotron Resonance Mass Spectrometer (FTICR-MS) to investigate supramolecules. Cross-sectional areas by Fourier transform ICR (CRAFTI), a novel technique for measurements of collision cross sections by FTICR, is demonstrated for the first time. The CRAFTI method measures the total "dephasing cross section" for removal of the ions from the coherent packet in the FTICR cell, including contributions not only from momentum transfer but also from reactive collisions including those leading to collisional dissociation. Experimental CRAFTI collision cross sections correlate linearly with theoretically computed results and with results obtained using ion mobility measurements. …

Translocation Mortality And Local, Regional, And Continental Diet Of The Northern River Otter (Lontra Canadensis), Casey Craig Day

Theses and Dissertations

The northern river otter (Lontra canadensis) is a semi-aquatic carnivore whose range extends throughout most of the United States and Canada. The northern river otter experienced a severe range contraction post-European settlement, but due to widespread management has in recent decades begun to recover much of its former range and habitat. We translocated 27 river otters from Utah and Idaho to the Provo River, Utah from November 2009 through January 2012 in a reintroduction effort to restore the northern river otter to its native range. Of these 27 otters, 6 died as a result of effects related to the translocation. …

Α2-Adrenoceptor And 5-Ht3 Serotonin Receptor Ligands As Potential Analgesic Adjuvants, Genevieve Alley Aug 2012

Α2-Adrenoceptor And 5-Ht3 Serotonin Receptor Ligands As Potential Analgesic Adjuvants, Genevieve Alley

Theses and Dissertations

There continues to be a need for more effective analgesics. The α2-adrenoceptor (AR) agonist clonidine is an analgesic whose use is severely limited by undesirable side effects. meta-Chlorophenylguanidine (MD-354), an agent developed in our laboratory, selectively potentiates the antinociceptive effects of clonidine in a biphasic manner. Mechanistic studies suggest that both 5-HT3 receptor and α2-AR mechanisms are involved. To further evaluate mechanisms underlying the analgesia-potentiating effect of clonidine by MD-354, pharmacological studies using more established 5-HT3 receptor agonists: meta-chlorophenylbiguanide (mCPBG) and centrally-acting SR57227A, and non-selective α2-adrenoceptor ligand TDIQ, administered alone and in combination with clonidine, were conducted in mouse antinociceptive …

Investigating Synergy Between Ribonucleotide Reductase Inhibitors And Cmv Antivirals, Sukhada Bhave Aug 2012

Investigating Synergy Between Ribonucleotide Reductase Inhibitors And Cmv Antivirals, Sukhada Bhave

Theses and Dissertations

Cytomegalovirus (CMV) infections remain a significant problem in congenitally infected infants and immunocompromised individuals. Modest antiviral activities of currently approved drugs coupled with dose-limiting toxicities restrict effectiveness and promote development of resistance. The potential for ribonucleotide reductase (RR) inhibitors hydroxyurea (HU), Didox, and Trimidox to synergize, through reduction of nucleotide pools, with the deoxynucleotide analog Ganciclovir (GCV) was examined. A yield reduction assay that utilizes luciferase expressed by a recombinant virus as a surrogate measure of viral infectious units was developed and used to determine effective dose ranges for each drug. RR inhibitors exhibited intrinsic anti-CMV activities on their own …

Mass Spectroscopic Identification And Quantification Of Protein Carbonyls, Zafer Ugur Aug 2012

Mass Spectroscopic Identification And Quantification Of Protein Carbonyls, Zafer Ugur

Theses and Dissertations

It is well established that free radical mediated oxidative stress plays a critical role in aging and age-related diseases. Among the post-translational protein modifications, carbonylation has attracted a great deal of attention due to its irreversible and irreparable nature. Despite the fact that protein carbonylation is associated with a series of physiological and pathological processes, there are still issues to be clarified such as why certain proteins are more vulnerable to modification, what are the locations of the protein modifications, and how does the nature of the oxidant affect the preferred site of modification. In this study, we will seek …

Flow Valve Diagnostics For Label-Free, Quantitative Biomarker Detection: Device Fabrication, Surface Modification, And Testing, Danielle Scarlet Mansfield Aug 2012

Flow Valve Diagnostics For Label-Free, Quantitative Biomarker Detection: Device Fabrication, Surface Modification, And Testing, Danielle Scarlet Mansfield

Theses and Dissertations

Diseases are often diagnosed by detection of disease-specific biomarkers in fluid samples. However, many state-of-the-art detection methods require a lab with complex machinery, trained operators, and/or lengthy analysis time. In contrast, point-of-care (POC) devices are brought to the patient's location, they are easy to use, and results are obtained almost immediately. Many current POC devices are too difficult to be used without a skilled assistant, and although many are able to detect analytes above a threshold value, they give little or no quantitative information. This work presents the development of polymer-based microfluidic devices capable of sensing and quantifying biomarkers in …

Characterization Of The Cellular And Organellar Dynamics That Occur With A Partial Depletion Of Mitochondrial Dna When Arabidopsis Organellar Dna Polymerase Ib Is Mutated, John D. Cupp Aug 2012

Characterization Of The Cellular And Organellar Dynamics That Occur With A Partial Depletion Of Mitochondrial Dna When Arabidopsis Organellar Dna Polymerase Ib Is Mutated, John D. Cupp

Theses and Dissertations

Plant mitochondrial genomes are large and complex, and the mechanisms for maintaining mitochondrial DNA (mtDNA) remain unclear. Arabidopsis thaliana has two DNA polymerase genes, polIA and polIB, that have been shown to be dual localized to mitochondria and chloroplasts but are unequally expressed within primary plant tissues involved in cell division or cell expansion. PolIB expression is observed at higher levels in both shoot and root apexes, suggesting a possible role in organelle DNA replication in rapidly dividing or expanding cells. It is proposed that both polIA and polIB are required for mtDNA replication under wild type conditions. An Arabidopsis …

Synthesis And Biological Evaluation Of Various Derivatives Of A Broad-Spectrum Anticancer Nucleoside, Jadd R. Shelton

Theses and Dissertations

Recently the Peterson lab discovered a promising anticancer adenosine derivative-- 2´,3´-bis-O-tert-butyldimethylsilyl-5´-deoxy-5´-[N-(methylcarbamoyl)amino]-N6-(N-phenylcarbamoyl)adenosine. This compound showed selective toxicity against human colon cancer cells in vitro with LC50's = 6--10 µM. It was hypothesized that the lead compound exerted its cytotoxic effects by interacting with a protein kinase. A systematic Structure Activity Relationship (SAR) was undertaken in an attempt to increase the kinase-binding affinity of the lead compound. Many regions of the lead compound were examined: the N6-phenyl urea moiety, the 5´-N-methyl urea group, the 2´,3´-bis-O-TBS groups, the nucleobase, and the ribose sugar. Results of these studies produced some promising new derivatives. In …

The Forgotten Medium: The Impact Of Reading Violent Literature On Aggressive Thoughts, Mckay Robert Stevens

Theses and Dissertations

Media violence in television, film, video games, and music has been linked to increased aggression. Only in recent years have researchers begun to investigate the impact that reading violent literature can have on individuals. The present study exposed individuals to either a violent or nonviolent story and measured aggressive cognitions. No significant effect was found for story type on aggressive cognitions but a gender effect for aggressive word completions was found. Discussion centers on possible differences between media types as well as future suggestions for investigations into violent literature.
