The purpose of this study is to attempt to analyze and interpret an outbuilding located in Ste. Genevieve, Missouri, a town established by French settlers in the 1780s. First the functional activities associated with the outbuilding will be determined and examined to best understand where and how the structure fit within the larger property. The activities taking place within/outside the outbuilding--especially those linked to domestic chores--will theoretically paint a clearer picture of who was living and/or working within the structure and who was not. Inexpensive label-free detection of biomarker panels in serum could revolutionize earlycancer diagnosis and treatment. Such detection capabilities may be possible with dynamicnanochannels in conjunction with electrical impedance measurement. In Dr. Greg Nordin's lab I designed, fabricated and tested several iterations of these sensors with polydimethyl-siloxane microfluidics. The final design yielded a dynamic nanochannel array sensor thatshowed a 140% impedance change when exposed to 14µM bovine serum albumin in phos-phate buffered saline. For the geometry and noise limits of the tested device, simulationsindicated that a minimum detectable concentration of 20pM with specifically bound strep-tavidin should be possible. RNA degradation plays a fundamental role in maintaining cellular homeostasis, along with being a part of normal regulatory mechanisms, whether it occurs as a surveillance mechanism eliminating aberrant mRNAs or during RNA processing to generate mature transcripts. 3’-5’ exoribonucleases are essential mediators of RNA decay pathways, and one such evolutionarily conserved enzyme is polynucleotide phosphorylase (PNPase). The human homologue of this fascinating enzymatic protein (hPNPaseold-35) was cloned a decade ago in the context of terminal differentiation and senescence through a novel ‘overlapping pathway screening’ approach. Background: A cross-sectional design was employed to ascertain the relationship between dairy consumption and insulin resistance (IR) in 272 middle-aged, nondiabetic women. Methods: Participants kept a seven-day weighed food record to report their diets, including consumption of dairy foods. IR was assessed using the homeostatic model assessment (HOMA), using the following formula: fasting plasma insulin (µU/ml) x fasting plasma glucose (mg/dL)/405. The Bod Pod was used to examine body fat percentage, and accelerometry over a seven-day period was used to assess physical activity. The major objective of the research described in this dissertation is the development of new laser vaporization methods for the synthesis of metal and semiconductor nanoparticles, graphene, B- and N-doped graphene, and metal and semiconductor nanoparticles supported on graphene. Visual system development requires the formation of precise circuitry in the dorsal lateral geniculate nucleus (dLGN) of the thalamus. Although much work has examined the molecular mechanisms by which retinal axons target and form synapses in dLGN, much less is known about the mechanisms that coordinate the formation of non-retinal inputs in dLGN. These non-retinal inputs represent ~90% of the terminals that form in dLGN. Despite decades of intense research and a consensus view in the field, the work presented herein provides strong evidence that the primary mechanism of amphetamine action should be re-evaluated. Rather than depleting vesicular dopamine stores and promoting non-exocytotic efflux through the dopamine transporter in vivo, recent evidence demonstrates that amphetamine augments phasic dopamine signaling instead. This signaling modality is critical for reinforcement learning and is dependent on intact vesicular stores. The new findings support over-activation of phasic dopamine signaling as a common mechanism in the addiction process.

Nitric oxide (NO) is an important mediator of angiogenesis and is primarily produced endogenously through the action of nitric oxide synthase (NOS). An alternate pathway for NO production is the conversion of nitrite to NO, which depends on the presence of hemoglobin (Hb) and hypoxic conditions. The angiogenic effects of topically applied sodium nitrite on two vascular beds in the ex ovo chicken embryonic model of angiogenesis were assessed. Methods for the rapid detection and differentiation of the Burkholderia pseudomallei complex comprising B. pseudomallei, B. mallei, and B. thailandensis, have been the topic of recent research due to the high degree of phenotypic and genotypic similarities of these species. B. pseudomallei and B. mallei are the causative agents of melioidosis and glanders, respectively. B. pseudomallei and B. mallei are recognized by the CDC as tier 1 select agents. Although B. thailandensis is generally avirulent in mammals, this species displays very similar phenotypic characteristics to that of B. pseudomallei. Writing about personal traumatic experiences is associated with beneficial effects on physical and psychological symptoms compared with writing about emotionally neutral events. However, not everyone benefits from trauma writing to the same extent. The present study hypothesized that the effectiveness of trauma writing may be moderated by emotion regulation, as indexed by respiratory sinus arrhythmia (RSA). Research also shows that greater physiological reactivity is predictive of better trauma writing outcomes. Given the importance of physiological output in emotional processing, response training was developed and found to increase appropriate physiological reactivity. Nanofluidic devices are structures having at least one dimension in the submicron range, which is of the same order of magnitude as the sizes of biomolecules and bioparticles such as proteins and viruses. As a result, size-selective separations are important applications for nanofluidics. Well-defined micro or nano device structures fabricated via micromachining have greatly reduced sample consumption and enabled separations in a parallel fashion, promising significant speed and resolution advantages over conventional size separation techniques, such as gel electrophoresis and size exclusion chromatography. Inwardly rectifying potassium (Kir) channels are critically important for regulating resting membrane potential in excitable cells, a job underscored by the severe pathophysiology associated with channel dysfunction. While all Kir channels require the activating lipid PIP2, many of these channels have diverse modulatory factors that couple to PIP2-dependent gating. Channels in the Kir3 (GIRK) family, in particular, have several co-activating elements, including G-protein betagamma subunits, ethanol, and sodium. Unmanned Aerial Vehicles (UAVs) are increasingly becoming economical platforms for carrying a variety of sensors. Building flight plans that place sensors properly, temporally and spatially, is difficult. The goal of sensor-driven planning is to automatically generate flight plans based on desired sensor placement and temporal constraints. We propose a simple taxonomy of UAV-enabled sensors, identify a set of generic sensor tasks, and argue that many real-world tasks can be represented by the taxonomy. Attentional bias to sleep related stimuli by poor sleepers has been reported as a factor in initiating and maintaining sleep disturbance. In this study, three paradigms (Flicker, Posner, and Stroop) were used in comparison to further investigate the role of attentional biases in Primary Insomnia. Participants (n=117) were split into good sleepers and poor sleepers using a median split with the Pittsburgh Insomnia Rating Scale (PIRS). This study examined Jeffrey Young's Schema Theory (1990; 2003) and its influence in understanding the etiological factors related to depressive symptoms. It was hypothesized that there would be significant interactions between parenting and temperament traits. It was also hypothesized that The Defectiveness and Insufficient Self Control EMSs would mediate the relationship between the significant parenting/temperament interactions and depressive symptoms. Participants were administered the Young Schema Questionnaire Short form-3 to measure the endorsement of schemas; the Beck Depression Inventory to measure symptoms of depression; the Young Parenting Inventory to measure perceived parenting; and the Adult Temperament Questionnaire to measure temperament traits. …

The present study collected data from 200 undergraduate students (106 male, 94 female) and sought to examine the relationship between levels of stress, cognitive processes related to stress management (Locus of Control- LOC; Sense of Coherence; SOC), and quality of life (QOL) across varying levels of risk for developing schizophrenia. Results indicated that high-risk individuals are significantly more likely to have an external locus of control (F=4.121, p=.018) supporting hypothesis 1. Counter to our second hypothesis, locus of control was not directly associated with QOL. However, LOC was correlated with both stress and level of risk for schizophrenia which (in …

Anaplasma phagocytophilum and A. marginale are the etiologic agents of human granulocytic anaplasmosis and bovine anaplasmosis, respectively. Both diseases can be severe, even fatal, and protective vaccines for each are lacking. We recently identified A. phagocytophilum outer membrane protein A (ApOmpA) as being critical for cellular invasion and is expressed during infection of mammalian but not tick cells. Disrupting ApOmpA-host cell interactions significantly inhibits A. phagocytophilum entry into host cells. ApOmpA and its A. marginale ortholog, AM854 (A. marginale OmpA; AmOmpA) exhibit 44% amino acid identity. The ApOmpA invasin domain is highly conserved between both proteins. In this study, we …

The inflammatory nature of the tumor microenvironment provides a cytokine and chemokine rich proliferative environment. Much of the responsibility of this environment is due to the production of Reactive Oxygen Species (ROS). These studies examined the proliferative rich tumor environment from a new perspective of Nitric Oxide Synthase (NOS) dysregulation. NOS’s have the ability to become uncoupled and generate superoxide in lieu of nitric oxide (NO). exploring the ethics of appropriative writing and the reparative potential of homophonic translation. The opening essay examines the ethics of appropriation- based poetry and introduces the concept of what I call "appositional writing," a term to describe ethically-minded works of poetry that make use of appropriative writing methods. The next three parts of this dissertation are each appositional writing projects that make use of homophonic translation as the primary method of composition. The purpose of this exploratory research was to investigate if change in sexual risk-taking behavior between high school and college can be predicted as a function of the interaction of person and situation traits. College, conceptualized as a backspace (situation) construct, and personality traits (sensation-seeking, neuroticism, extraversion) conducive to deviant behavior were examined. While significant risky behavior was reported for all students (n = 252), hierarchical regression analyses revealed that higher levels of the specified personality traits in combination with on-campus backspace residential status was not predictive of greater change in engagement in sexual risk-taking behavior. My work has focused on a variety of different types of diamond-based, core-shell particles. These particles are formed with inert cores and poly(allylamine)/nanodiamond shells. Their intended purpose is to form an LC stationary phase that is stable from pH 1 – 14 and at elevated temperatures. At the beginning of my studies, the particles that had been made in the Linford laboratory were pH stable, but irregular and had poor mechanical stability. Yet, little is known about how patients perceive SDM and whether patient-defined SDM is associated with patient outcomes. An important goal of nanoscience is the self-assembly of nanoscale building blocks into complex nanostructures. DNA is an important and versatile building block for nanostructures because of its small size, predictable base pairing, and numerous sequence possibilities. I use DNA origami to design and fold DNA into predesigned shapes, to assemble thin, branched DNA nanostructures as templates for nanoscale metal features. Using a PCR-based scaffold strand generation procedure, several wire-like nanostructures with varying scaffold lengths were assembled. The investigator in the study compared traditional middle and high school teachers to alterative teachers regarding the perceived organizational support, self-efficacy, and retention. The purpose of the study was to determine if there was a difference in the level of self-efficacy between alternative school teachers and traditional school teachers. The researcher also examined the relationships between Perceived Organization Support and teacher self-efficacy between alternative school teachers and traditional school teachers.
