Background: Genetic genealogy is an emergent topic in sociology. To date, over 41 million individuals worldwide have tested with the top four direct-to-consumer genetic testing (DTCGT) companies: AncestryDNA, FamilyTreeDNA, 23andMe, and MyHeritageDNA (Glynn 2022). These tests provide consumers with ethnicity estimates, a list of DNA cousin matches, or both. Despite the popularity of DTCGT, there is a lack of data on consumer experiences with genetic relative-finder services (GRF) in the sociological literature. Bone morphogenetic proteins (BMPs) are the largest subfamily of the Transforming Growth Factor (TGF)-ß superfamily. BMPs are renowned for their pivotal role in cellular functions such as cell growth, apoptosis, and differentiation. Yet, BMPs have also been attributed to the maintenance of adult tissue homeostasis. BMPs elicit physiological function at the cell membrane via a heterotetrameric complex composed of two Type I and two Type II serine-threonine kinase BMP receptors. Activation of BMP transmembrane receptors results in activation of either canonical (Smad-dependent) and/or non-canonical (Smad-independent) signaling. Cellular migration assays, particularly scratch assays, are pivotal in biomedical research for cell motility, wound healing, and metastasis. However, conventional scratch assays suffer from variability due to operator dependence, non-uniform scratch geometry, and a modified microenvironment that impacts reproducibility. This thesis presents the development and validation of a low-cost, 3D-printed device designed to standardize cellular migration assays. The device utilizes a spring mechanism and a biocompatible elastomer to create a uniform cellular exclusion zone without harming the surrounding cells. This dissertation investigates the synthesis and characterization of 3D printed polymer composites, with engineered porosity and both natural and synthetic additives, using fused deposition modeling (FDM) and selective laser sintering (SLS) for potential biomedical applications. The structural, mechanical, tribological, and surface behavior of these composites were evaluated with respect to their suitability for orthopedic load-bearing implants and scaffolds. In the first phase, PEEK-based composites reinforced with 10 or 20 wt% carbon fibers or 20 wt% E-glass fibers were fabricated using FDM with varying infill densities of 30-100% to introduce porosity. Traditional models in psychiatric research often impose assumptions of causal homogeneity, treating population-level associations as reflective of uniform underlying mechanisms. This dissertation challenges that assumption by introducing statistical and machine learning frameworks designed to detect and model causal heterogeneity in the development of psychopathology. Central to this approach is the advancement of finite mixture structural equation modeling (FM-SEM) to identify latent subgroups characterized by distinct, and sometimes opposing, causal pathways.

Montane regions in the Neotropics are hotspots of endemism and biodiversity, yet their species are especially vulnerable to the impacts of climate change and geographic isolation. This thesis investigates the evolutionary and ecological dynamics of three bird species endemic to the Darién Highlands of eastern Panama: Myadestes coloratus, Chlorospingus tacarcunae, and Margarornis bellulus. Through an integrative approach combining ecological niche modeling, morphometric analysis, and genomic data from ultraconserved elements (UCEs) and mitochondrial DNA, the study explores the historical biogeography, current distribution patterns, population structure, and phenotypic differentiation of these montane taxa. Black adolescents are at an increased risk of experiencing racial discrimination due to a developmental period marked by increased time away from parents and caregivers and increased time with peers (Fisher et al., 2000). The spongy moth (Lymantria dispar) is an invasive forest pest that has caused significant ecological damage across the United States. Its invasion front is shaped by a number of factors, including temperature in both the northern and southern regions. With ongoing climate change, areas that were previously uninhabitable may become increasingly favorable for moth population establishment and expansion, while other areas may experience thermal stress limiting persistence. Lyme disease (LD) is the most common arthropod-borne disease in North America and is caused by the spirochete, Borreliella burgdorferi. LD spirochetes are maintained in an enzootic cycle of Ixodes species ticks and a vertebrate reservoir. The secondary messenger molecule c-di-GMP plays a crucial role in adaptive responses during the enzootic cycle. PlzA is the sole c-di-GMP-binding protein identified to date that is encoded by all Borreliella burgdorferi isolates. Lyme disease, caused by the spirochete Borreliella burgdorferi, is the most common vector-borne infection in North America. A critical determinant of early mammalian infection is outer surface protein C (OspC), a highly variable lipoprotein that is indispensable for spirochete survival but whose precise function remains unresolved. OspC has been proposed to mediate diverse roles, including complement evasion, adhesion, tissue dissemination, and antiphagocytic activity, supporting the view that it is a multifunctional virulence factor. Structural studies have identified two putative ligand-binding domains (LBD1 and LBD2), with LBD2 forming a solvent-exposed crown at the membrane-distal end of the OspC dimer.

This clinical laboratory study utilizes subjective and behavioral economic measures to assess the abuse liability of own-brand cigarettes and six currently authorized ENDS products, examining how device characteristics, nicotine flux, and liquid flavor influence outcomes among adults who smoke cigarettes. Research on tobacco product standards to limit the abuse liability of electronic nicotine delivery systems (ENDS) has primarily focused on nicotine concentration and flavor. Other liquid characteristics, such as protonated nicotine ratio and sweetener additives, are less understood but may serve as regulatory targets. This clinical laboratory study examined the effects of ENDS protonated nicotine ratio and sweetener presence on nicotine delivery, use behavior, and subjective effects.

I examined a potential source of variation in phylogenetic inference, investigating whether handling single-nucleotide polymorphisms (SNPs) through different methods during reference-guided genome assembly of pigeon and dove species impacts phylogenetic reconstruction and inference. Specifically, I created a custom consensus base calling tool that handles allelic variation through two different approaches, either ignoring heterozygous sites by denoting them as an ambiguous character “N,” or using a pseudo-random choice between supported base variants. Emotion dysregulation, or responses to emotions that do not meet the needs of the situational demands or social standards, has been noted as an early risk factor for later psychopathology. Maternal postpartum depression (PPD) has been shown to increase children’s risk of developing maladaptive emotion regulation (ER) strategies. One potential mechanism linking PPD to these outcomes is affect incongruence, or a mismatch between biological and emotional responses to stress. This study explored the relationship between supervisor support and job satisfaction, offering work-life balance and perceived employability as key mechanisms through which this relationship occurs. It also explored gender differences in the relationship between work-life balance and job satisfaction, and perceived employability and job satisfaction. Participants were full-time employees in the U.S, recruited via Prolific, who completed an online survey. While supervisor support was found to have positively influenced job satisfaction through perceived employability, the role of work-life balance was less clear. The temporal styloid process in humans serves as an attachment point for the styloglossus, stylohyoid, and stylopharyngeus, which are involved in tongue movement and deglutition. I hypothesized that the length of the ossified styloid process in humans is the result of selection for increased musculature that would support tongue movements involved in speech. This hypothesis assumes that the relative length of the ossified temporal styloid process is unique to humans.
