Morphological Analysis Of Microvessels Using High Voltage Electron Microscopy (Hvem), Ibm Personal Computer And 3-D Reconstruction Software, Wieslaw Slawomir Krajewski

The mesenteric arterioles (outer diameter < 100 µ) from a normotensive and a hypertensive dog were perfused in situ, excised, post-fixed and embedded in Spurr medium block. Thick serial cross sections (0.5 µ) of the vessels were obtained from hand formed mesas and placed on Formvar coated slot grids.

The cross sections were subjected to High Voltage Electron Microscopy (HVEM) and five or more electronmicrographs were taken to cover the individual cross sections. The prints of electronmicrographs were linearly enlarged (3X) and made into photomontages of sections. The membrane contours in the montages were traced on transparent overlays, producing polygons which in turn were traced on GTCO digitizing pad. All polygons were labeled, color coded and stored in IBM PC's filing system. The vessels were reconstructed from up to 100 whole cross sections and subjected to cell compartmentalvolume analysis. Low power laser induced fluorescence of dihematoporphyrin ether (DHE, photofrin 2) is used to detect cancer. The goal was to determine the effect of low power argon laser light and DHE on C-1300 murine neuroblastoma cells (MNB, NB41A3). The study groups were as follows: Control, untreated cells (C), DHE only at concentrations of 1 (D1) and 5 (D5) µg/ml, Argon laser 2 light only at exposure times of 1 minute (1.8 J/cm2 , L1) 2 and 5 minutes (9.0 J/cm2 , L5), combined treatment of Light at 1, 5 minutes, and DHE at 1, 5 µg/ml.

Primary process level communication is discussed as the underlying mechanism in the expressive therapy process . The suitability of expressive therapy methods is examined through informal interviews with parents and teachers, observations by the program facilitator , and the products the children created . Measurement Of The Thermal Conductivity Of Thin Solid Films With A Thermal Comparator, Charles A. Amsden

A direct-reading thermal comparator was used to measure the thermal conductivities of several thin solid films. This new application of the thermal comparator was based on heat flow modelling using the thermal constriction resistance, generalized here for the case of a film on the surface of an infinite half-space.

Four dielectric optical coating materials were tested, and found to have thermal conductivities significantly lower than those for the same material in bulk form.

The flow concept gives dataflow languages the advantage of representing dataflow programs in graphical forms. This thesis presents a graphical programming system that supports the editing and simulating of dataflow programs. The system is implemented on an AT&T UnixTM PC.

Hierarchical data structures for image representation have been widely explored in recent years. These data structures are based on the principle of "recursive decomposition" of an image region. The most commonly mentioned picture data structure for two-dimensional data is referred to as a "quadtree". The purpose of this thesis is to investigate the use of a general quadtree scheme as a means of representing symmetric images. Specifically, images are generated according to the rules of selected two-dimensional plane symmetry groups.

Approaches To The Synthesis Of Dentin Bonding Agents Containing Vinyl And Carbamate Functional Groups, Steven W. Amato

The synthesis of four new monomers containing vinyl and carbamate functional groups was attempted. The desired products were N-m-methacryloxyphenyl-O-phenyl carbamate, N-p-methacryloxy-O-phenyl carbamate, m-N-phenylcarbamato-2-methacryloxyethoxy benzoate, and p-N-phenylcarbamato-2-methacryloxyethoxy benzoate. The carbamate functional group was derived by the Lewis Acid catalyzed reaction of meta-aminophenol, para-aminophenol, meta-aminobenzoic acid, and para-aminobenzoic acid with diphenyl carbonate. The vinyl function was derived, in the case of the aminophenols, by reaction with methacrylyl chloride, and in the case of the aminobenzoic acids, with 2-hydroxyethyl methacrylate.

Deuterium Isotope Effects On In Vitro Dealkylation Of 1,1-Dimethylhydrazine And 1,1-Dimethylnitrosamine Via Hepatic Enzymes, Robert Sloane

Boiling 1,1-dimethylnitrosamine or unsymmetrical dimethylnitrosamine several times with 25% NaOD in D2O promoted the complete exchange of the nitrosamine’s alpha protons for deuterium. The deuterated nitrosamine was reduced by LiAlH4, in anhydrous diethyl ether to the corresponding 1,1-dimethylhydrazine-d6. An in-process production of the oxalate salt was necessary due to the air sensitivity of the free base hydrazine. After recrystallization from absolute ethanol and extensive drying, the oxalate salt was evaluated for purity by mixed melting point, high performance liquid chromatography and thin layer chromatography. Fibrinogen and thrombin were combined to make a "glue" for adherence in microvascular anastomoses and split-thickness autologous skin grafts in rats. The powdered human fibrinogen and bovine thrombin were mixed with sterile water and each applied drop-wise through separate syringes in equal concentrations. A gelatinous clot formed in 10 seconds, and was completely coagulated in 45 seconds. In part I, one-half of the group had their grafts sutured, and one-half had their grafts glued in place.

Blood circulation at the microcirculatory level plays an important role in the inflammatory process. Adhesion and emigration of white blood cells are among the major microcirculatory responses to inflammation. It has been recently suggested that platelet activating factor (PAF) may be a significant mediator in the local inflammatory response. This research was undertaken to characterize and quantitate the effects of PAF on the adhesion of white cells to the wall of postcapillary venules of 7-80um diameter.

Studies were conducted to characterized airborne particulate extractable organic matter (EOM) with respect to chemical composition and biological activity. It has been well established that the nitrated polycyclic hydrocarbons (nitroPAH) are environmental contaminants. To identify nitro-PAH in ambient air sample, a technique was developed involving introduction of a reducer column into an reversephase high performance liquid chromatography (HPLC) system. Strong fluorescence signals were achieved from nitro-PAH and detected together with u.v. signals for the qualitative and quantitative analysis of nitro-PAH. 2-Nitrotriptycene was synthesized according to published procedures and used as an internal standard. A mathematical model was proposed for the isometric contraction of smooth muscle (i.e., vascular, urterine, & gastrointestinal) that accounts for the experimental observations on tension transients and pCa2+-tension curves. The model is based on the elementary reaction kinetics involved in the process; it uses the Ca2+-dependent myosin phosphorylation regulatory mechanism in its interpretation. The model is composed of four parts : (a) the activation of myosin light chain kinase; (b) the phosphorylation of myosin light chains; (c) the actin-myosin Mg2+ATPase cross-bridge reaction cycle; and (d) the development of isometric tension.
