Atypical protein kinase C (aPKCs) serve key functions in embryonic development by regulating apical-basal polarity. Previous studies have shed light on their roles during adulthood, especially in the development of Alzheimer's disease (AD). Although the crystal structure of PKCι has been resolved, an agonist of aPKCs remains to be discovered. In the present study, by using the Discovery Studio program and LibDock methodology, a small molecule library (K66-X4436 KINA Set) of compounds were screened for potential binding to PKCι. Subsequently, the computational docking results were validated using affinity selection-mass spectrometry, before in vitro kinase activity was used to determine the …

Ontogenetic Drivers Of Morphological Evolution In Monitor Lizards And Allies (Squamata: Paleoanguimorpha), A Clade With Extreme Body Size Disparity, Carlos J. Pavón-Vázquez, Damien Esquerré, J. Scott Keogh

Publications and Research

Background

Heterochrony, change in the rate or timing of development, is thought to be one of the main drivers of morphological evolution, and allometry, trait scaling patterns imposed by size, is traditionally thought to represent an evolutionary constraint. However, recent studies suggest that the ontogenetic allometric trajectories describing how organisms change as they grow may be labile and adaptive. Here we investigated the role of postnatal ontogenetic development in the morphological diversification of Paleoanguimorpha, the monitor lizards and allies, a clade with extreme body size disparity. We obtained linear and geometric morphometric data for more than 1,600 specimens belonging to …

Does Breastfeeding Account For The Association Between Maternal Sensitivity And Infant Cognitive Development In A Large, Nationally Representative Cohort?, P. Nina Banerjee, Karen E. Mcfadden, Jacqueline D. Shannon, Leslie L. Davidson

Publications and Research

Background: Previous research has established that exposure to high maternal sensitivity is positively associated with advances in infant cognitive development. However, there are many fixed and modifiable factors that influence this association. This study investigates whether the association between maternal sensitivity and infant cognitive development in the first year of life is accounted for by other factors, such as breastfeeding, maternal depressive symptoms, maternal alcohol use, infant birth weight or demographic covariates.

Methods: Using data from the Early Childhood Longitudinal Study-Birth (ECLS-B) Cohort, a nationally representative sample of U.S. born children, multi-variable regression analyses was used to examine whether breastfeeding, …

Emotion Recognition With Audio, Video, Eeg, And Emg: A Dataset And Baseline Approaches, Jin Chen, Tony Ro, Zhigang Zhu

Publications and Research

This paper describes a new posed multimodal emotional dataset and compares human emotion classification based on four different modalities - audio, video, electromyography (EMG), and electroencephalography (EEG). The results are reported with several baseline approaches using various feature extraction techniques and machine-learning algorithms. First, we collected a dataset from 11 human subjects expressing six basic emotions and one neutral emotion. We then extracted features from each modality using principal component analysis, autoencoder, convolution network, and mel-frequency cepstral coefficient (MFCC), some unique to individual modalities. A number of baseline models have been applied to compare the classification performance in emotion recognition, …

The Rise And Fall Of Dopamine: A Two-Stage Model Of The Development And Entrenchment Of Anorexia Nervosa, Jeff A. Beeler, Nesha S. Burghardt Jan 2022

The Rise And Fall Of Dopamine: A Two-Stage Model Of The Development And Entrenchment Of Anorexia Nervosa, Jeff A. Beeler, Nesha S. Burghardt

Publications and Research

Dopamine has long been implicated as a critical neural substrate mediating anorexia nervosa (AN). Despite nearly 50 years of research, the putative direction of change in dopamine function remains unclear and no consensus on the mechanistic role of dopamine in AN has been achieved. We hypothesize two stages i n AN– corresponding to initial development and entrenchment– characterized by opposite changes in dopamine. First, caloric restriction, particularly when combined with exercise, triggers an escalating spiral of increasing dopamine that facilitates the behavioral plasticity necessary to establish and reinforce weight-loss behaviors. Second, chronic self-starvation reverses this escalation to reduce or impair …

Glycocalyx Mechanotransduction Mechanisms Are Involved In Renal Cancer Metastasis, Heriberto Moran, Limary M. Cancel, Peigen Huang, Sylvie Roberge, Tuoye Xu, John M. Tarbell, Lance L. Munn

Publications and Research

Mammalian cells, including cancer cells, are covered by a surface layer containing cell bound proteoglycans, glycoproteins, associated glycosaminoglycans and bound proteins that is commonly referred to as the glycocalyx. Solid tumors also have a dynamic fluid microenvironment with elevated interstitial flow. In the present work we further investigate the hypothesis that interstitial flow is sensed by the tumor glycocalyx leading to activation of cell motility and metastasis. Using a highly metastatic renal carcinoma cell line (SN12L1) and its low metastatic counterpart (SN12C) we demonstrate in vitro that the small molecule Suberoylanilide Hydroxamic Acid (SAHA) inhibits the heparan sulfate synthesis enzyme …

The protozoan parasite, Trypanosoma cruzi, causes severe morbidity and mortality in afflicted individuals. Approximately 30% of T. cruzi infected individuals present with cardiac pathology. The invasive forms of the parasite are carried in the vascular system to infect other cells of the body. During transportation, the molecular mechanisms by which the parasite signals and interact with host endothelial cells (EC) especially heart endothelium is currently unknown. The parasite increases host thrombospondin-1 (TSP1) expression and activates the Wnt/β-catenin and hippo signaling pathways during the early phase of infection. The links between TSP1 and activation of the signaling pathways and …

Who’S Afraid Of Bob Jones?: 'Fundamental National Public Policy' And Critical Race Theory In A Delicate Democracy, Lynn D. Lu

Publications and Research

In Summer of 2021, Republican legislators across the United States introduced a host of bills to prohibit government funding for schools or agencies that teach critical race theory (“CRT”), described by the American Association of Law Schools not as a single doctrine but a set of “frameworks” to “explain and illustrate how structural racism produces racial inequity within our social, economic, political, legal, and educational systems[,] even absent individual racist intent.” Characterizing such an explicitly race-conscious analysis of legal and social institutions as “divisive,” opponents of CRT, such as former Vice President Mike Pence, labeled it “nothing short of state-sponsored …

A Study Protocol To Prepare An Rbd Protein For Vaccine Against Covid-19, Z.M.G. Sarwar Jahangir, Arleta Helena Marnik

Publications and Research

Background: SARS-CoV-2 pandemic is a global threat to humans and the world’s economy. Effective and safe vaccines against this virus are essential to control and eradicate the pandemic. The currently applied vaccines carry SARS-CoV-2 spike-protein mRNA/cDNA. These vaccines go through several cellular processes in the recipients for producing antigens. On the contrary, the SARS-CoV-2 RBD (receptor binding domain)-protein is an antigen. It will directly stimulate antibody production against SARS-CoV-2. Hence, we propose to produce SARS-CoV-2 RBD-protein as a fast acting, effective and safe vaccine.

Methods: We propose to reconstruct a plasmid carrying three types of DNA sequences: RBD cDNA, FP …

The Imaginal Lexicon Of Aging Studies, William Ebenstein Jan 2022

The Imaginal Lexicon Of Aging Studies, William Ebenstein

Publications and Research

Aging Studies focuses on the cultural, humanistic and experiential aspects of aging insofar as these are already embedded in literature, art, language and everyday life. The lexicon of this multi-disciplinary field has evolved in response to shifts in cultural attitudes, social roles, and conceptual frameworks. At the same time, many age-related words have links to poetic imagery and ancient myths, and to evocative etymologies that conjure an historical context. The article looks at the ongoing and surprisingly difficult effort to find the right word to call an older person. It reviews the language and literary genres associated with longevity. And, …

Toward Raising The Child And The Village: Analyzing The Barriers To Implementing Community-Based Curriculum In New York City High Schools, Ife Damon

Publications and Research

As automation increases and single-skill set jobs become non-existent, employees can no longer only possess the technical knowledge of their industry but must also be well versed in a variety of essential skills that artificial intelligence cannot acquire. It is evident from the research that the United States’ traditional approach to teaching and learning has not kept up with the demands of the changing labor market because young adults are graduating from high school lacking the 21st-century skills needed for contemporary careers. High school educators can develop these skills within students by implementing community-based learning (CBL) activities into the curriculum. …

Biomedical Applications Of Lanthanide Nanomaterials, For Imaging, Sensing And Therapy, Qize Zhang, Stephen O'Brien, Jan Grimm

Publications and Research

The application of nanomaterials made of rare earth elements within biomedical sciences continues to make significant progress. The rare earth elements, also called the lanthanides, play an essential role in modern life through materials and electronics. As we learn more about their utility, function, and underlying physics, we can contemplate extending their applications to biomedicine. This particularly applies to diagnosis and radiation therapy due to their relatively unique features, such as an ultra-wide Stokes shift in the luminescence, variable magnetism and potentially tunable properties, due to the library of lanthanides available and their multivalent oxidation state chemistry. The ability to …

Infant Motor Development Predicts The Dynamics Of Movement During Sleep, Aaron Demasi, Melissa N. Horger, Anat Scher, Sarah E. Berger Jan 2022

Infant Motor Development Predicts The Dynamics Of Movement During Sleep, Aaron Demasi, Melissa N. Horger, Anat Scher, Sarah E. Berger

Publications and Research

The characteristics of infant sleep change over the first year. Generally, infants wake and move less at night as they grow older. However, acquisition of new motor skills leads to temporary increases in night waking and movement at night. Indeed, sleep-dependent movement at night is important for sensorimotor development. Nevertheless, little is known about how movement during sleep changes as infants accrue locomotor experience. The current study investigated whether infant sleep and movement during sleep were predicted by infants' walking experience. Seventy-eight infants wore an actigraph to measure physical activity during sleep. Parents reported when their infants first walked across …

Importin-Mediated Pathological Tau Nuclear Translocation Causes Disruption Of The Nuclear Lamina, Tdp-43 Mislocalization And Cell Death, Robert F. Candia, Leah S. Cohen, Viktoriya Morozova, Christopher Corbo, Alejandra D. Alonso Jan 2022

Importin-Mediated Pathological Tau Nuclear Translocation Causes Disruption Of The Nuclear Lamina, Tdp-43 Mislocalization And Cell Death, Robert F. Candia, Leah S. Cohen, Viktoriya Morozova, Christopher Corbo, Alejandra D. Alonso

Publications and Research

Tau is a cytosolic protein that has also been observed in the nucleus, where it has multiple proposed functions that are regulated by phosphorylation. However, the mechanism underlying the nuclear import of tau is unclear, as is the contribution of nuclear tau to the pathology of tauopathies. We have previously generated a pathological form of tau, PH-tau (pseudophosphorylation mutants S199E, T212E, T231E, and S262E) that mimics AD pathological behavior in cells, Drosophila, and a mouse model. Here, we demonstrated that PH-tau translocates into the nucleus of transiently transfected HEK-293 cells, but wildtype tau does not. We identified a putative …

What Can Aphasia Tell Us About How The First-Acquired Language Is Instantiated In The Brain?, Mira Goral Jan 2022

What Can Aphasia Tell Us About How The First-Acquired Language Is Instantiated In The Brain?, Mira Goral

Publications and Research

Recent neurolinguistic theories converge on the hypothesis that the languages of multilingual people are processed as one system in the brain. One system for the multiple languages is also at the core of a translanguaging framework of multilingualism—a framework that focuses on each speaker’s complete linguistic repertoire rather than on the separate languages they know. However, evidence from neuroimaging studies suggests at least some nonoverlapping activations of the first-acquired language (L1) and other (non-L1) languages of multilingual people, especially when the age of acquisition and/or levels of proficiency differ across the languages. Neurolinguistic studies of acquired language disorders have demonstrated …

Satellite Evidence Of Canopy-Height Dependence Of Forest Drought Resistance In Southwestern China, Peipei Xu, Wei Fang, Tao Zhou, Hu Li, Xiang Zhao, Spencer Berman, Ting Zhang, Chuixiang Yi

Publications and Research

The frequency and intensity of drought events are increasing with warming climate, which has resulted in worldwide forest mortality. Previous studies have reached a general consensus on the size-dependency of forest resistance to drought, but further understanding at a local scale remains ambiguous with conflicting evidence. In this study, we assessed the impact of canopy height on forest drought resistance in the broadleaf deciduous forest of southwestern China for the 2010 extreme drought event using linear regression and a random forest (RF) model. Drought condition was quantified with standardized precipitation evapotranspiration index (SPEI) and drought resistance was measured with the …

An Optimal Lysis Time Maximizes Bacteriophage Fitness In Quasi-Continuous Culture, Sherin Kannoly, Abhyudai Singh, John J. Dennehy Jan 2022

An Optimal Lysis Time Maximizes Bacteriophage Fitness In Quasi-Continuous Culture, Sherin Kannoly, Abhyudai Singh, John J. Dennehy

Publications and Research

Optimality models have a checkered history in evolutionary biology. While optimality models have been successful in providing valuable insight into the evolution of a wide variety of biological traits, a common objection is that optimality models are overly simplistic and ignore organismal genetics. We revisit evolutionary optimization in the context of a major bacteriophage life history trait, lysis time. Lysis time refers to the period spanning phage infection of a host cell and its lysis, whereupon phage progenies are released. Lysis time, therefore, directly determines phage fecundity assuming progeny assembly does not exhaust host resources prior to lysis. Noting that …

Contesting Imagined Communities: Minor Feelings, Opacity, And Spectrality In Ling Ma’S Severance, Christine Marks Jan 2022

Contesting Imagined Communities: Minor Feelings, Opacity, And Spectrality In Ling Ma’S Severance, Christine Marks

Publications and Research

This article foregrounds pandemic fiction’s subversive and visionary potential through the example of Ling Ma’s dystopian pandemic novel Severance (2018). Emphasizing fiction’s capacity for symbolic intervention in a pandemic era, I argue that the text undermines tropes of nation-building through strategic employment of minor feelings, opacity, and spectral liminality. My analysis focuses on the Chinese American protagonist’s conflicted relationship with imagined communities forged in the conventional “outbreak narrative” (Wald) as her unhomely positionality places her at odds with unifying ideological responses to the catastrophic upheaval of the pandemic. At the intersection of pandemic and diasporic literature, Ma’s novel constitutes a …

A Horor Theory For Introspective Consciousness, Adriana Renero, Richard Brown Jan 2022

A Horor Theory For Introspective Consciousness, Adriana Renero, Richard Brown

Publications and Research

Higher-order theories of consciousness typically account for introspection in terms of one’s higher-order thoughts being conscious, which would require a third-order thought—i.e., a thought about a thought about a mental state. In this work, we offer an alternative account of introspection that builds on the recent Higher-Order Representation of a Representation (HOROR) theory of phenomenal consciousness. According to HOROR theory, phenomenal consciousness consists in having the right kind of higher-order representation. We claim that this theory can be extended to introspection by recognizing that there is a distinctive kind of consciousness—i.e., introspective consciousness—which can be accounted for as the theory …

Organizational Practices And Second-Generation Gender Bias: A Qualitative Inquiry Into The Career Progression Of Us State-Level Managers, Maria J. D’Agostino, Helisse Levine, Meghna Sabharwal, Ai C. Johnson-Manning Jan 2022

Organizational Practices And Second-Generation Gender Bias: A Qualitative Inquiry Into The Career Progression Of Us State-Level Managers, Maria J. D’Agostino, Helisse Levine, Meghna Sabharwal, Ai C. Johnson-Manning

Publications and Research

Ely and Meyerson’s gendered organizations framework reconceptualizes traditional gender differences defined by biology and lack of structural opportunities, to a complex set of social relations in the workplace. We apply this framework to second-generation gender bias to further understand impediments to women’s career progression in the public sector workplace. In-depth interviews of state-level administrators in U.S. public sector agencies indicate that “narratives” perpetuate second-generation gender bias that is deeply ingrained in organizational practices and policies, especially for women and women of color. This framework can be applied to future studies examining the gendered nature of organizations in different workplace settings. …

The Naming Of Homo Bodoensis By Roksandic And Colleagues Does Not Resolve Issues Surrounding Middle Pleistocene Human Evolution, Eric Delson, Chris Stringer

Publications and Research

Roksandic et al. (2022) proposed the new species name Homo bodoensis as a replacement name for Homo rhodesiensis Woodward, 1921, because they felt it was poorly and variably defined and was linked to sociopolitical baggage. However, the International Code of Zoological Nomenclature includes regulations on how and when such name changes are allowed, and Roksandic et al.'s arguments meet none of these requirements. It is not permitted to change a name solely because of variable (or erroneous) later use once it has been originally defined correctly, nor can a name be modified because it is offensive to one or more …
