Positive Rate-Dependent Action Potential Prolongation By Modulating Potassium Ion Channels, Candido Cabo Jun 2022

Positive Rate-Dependent Action Potential Prolongation By Modulating Potassium Ion Channels, Candido Cabo

Publications and Research

Pharmacological agents that prolong action potential duration (APD) to a larger extent at slow rates than at the fast excitation rates typical of ventricular tachycardia exhibit reverse rate dependence. Reverse rate dependence has been linked to the lack of efficacy of class III agents at preventing arrhythmias because the doses required to have an anti-arrhythmic effect at fast rates may have pro-arrhythmic effects at slow rates due to an excessive APD prolongation. In this report we show that, in computer models of the ventricular action potential, APD prolongation by accelerating phase 2 repolarization (by increasing IKs) and decelerating …

Borders: A Story Of Political Imagination, Miriam Ticktin Jun 2022

Borders: A Story Of Political Imagination, Miriam Ticktin

Publications and Research

This article traces three different political imaginaries about borders, suggesting that the dominant imaginary—the one of border walls, driven by a fear of invasion—is only one way to live in the world. The goal is to make space in our political imaginations to rethink how we live together, including thinking beyond nation-states as containers that keep people in or out. By first showing how the vision of invasion is built and maintained with intersecting transnational technologies and ideologies, I open the way to thinking otherwise. Second, I trace the counterpolitics of borders developed by artists and activists, resisting borders and …

Medical Avoidance Among Marginalized Groups: The Impact Of The Covid‑19 Pandemic, Amani R. Holder‑Dixon, Olivia R. Adams, Tianna L. Cobb, Alison J. Goldberg, Rachel A. Fikslin, Mora A. Reinka, Amanda N. Gesselman, Devon M. Price

Publications and Research

Medical avoidance is common among U.S. adults, and may be emphasized among members of marginalized communities due to discrimination concerns. In the current study, we investigated whether this disparity in avoidance was maintained or exacerbated during the onset of the COVID-19 pandemic. We assessed the likelihood of avoiding medical care due to general-, discrimination-, and COVID-19-related concerns in an online sample ( N = 471). As hypothesized, marginalized groups (i.e., non-White race, Latinx/e ethnicity, non-heterosexual sexual orientation, high BMI) endorsed more general- and discrimina - tion-related medical avoidance than majoritized groups. However, marginalized groups were equally likely to seek COVID-19 …

Knee Pain From Osteoarthritis: Pathogenesis, Risk Factors, And Recent Evidence On Physical Therapy Interventions, Jean-Philippe Berteau Jun 2022

Knee Pain From Osteoarthritis: Pathogenesis, Risk Factors, And Recent Evidence On Physical Therapy Interventions, Jean-Philippe Berteau

Publications and Research

For patients presenting knee pain coming from osteoarthritis (OA), non-pharmacological conservative treatments (e.g., physical therapy interventions) are among the first methods in orthopedics and rehabilitation to prevent OA progression and avoid knee surgery. However, the best strategy for each patient is difficult to establish, because knee OA’s exact causes of progression are not entirely understood. This narrative review presents (i) the most recent update on the pathogenesis of knee OA with the risk factors for developing OA and (ii) the most recent evidence for reducing knee pain with physical therapy intervention such as Diathermy, Exercise therapy, Ultrasounds, Knee Brace, and …

The Cost Of Living In Larger Primate Groups Includes Higher Fly Densities, Jan F. Gogarten, Mueena Jahan, Sébastien Calvignac-Spencer, Colin A. Chapman, Tony L. Goldberg, Fabian H. Leendertz, Jessica M. Rothman Jun 2022

The Cost Of Living In Larger Primate Groups Includes Higher Fly Densities, Jan F. Gogarten, Mueena Jahan, Sébastien Calvignac-Spencer, Colin A. Chapman, Tony L. Goldberg, Fabian H. Leendertz, Jessica M. Rothman

Publications and Research

Flies are implicated in carrying and mechanically transmitting many primate pathogens. We investigated how fly associations vary across six monkey species (Cercopithecus ascanius, Cercopithecus mitis, Colobus guereza, Lophocebus albigena, Papio anubis, and Piliocolobus tephrosceles) and whether monkey group size impacts fly densities. Fly densities were generally higher inside groups than outside them, and considering data from these primate species together revealed that larger groups harbored more flies. Within species, this pattern was strongest for colobine monkeys, and we speculate this might be due to their smaller home ranges, suggesting that movement patterns may influence fly–primate associations. Fly associations increase with …

Weakly-Supervised Tumor Purity Prediction From Frozen H&E Stained Slides, Matthew Brendel, Vanesa Getseva, Majd Al Assaad, Michael Sigouros, Alexandros Sigaras, Troy Kane, Pegah Khosravi, Juan Miguel Mosquera, Olivier Elemento, Iman Hajirasouliha Jun 2022

Weakly-Supervised Tumor Purity Prediction From Frozen H&E Stained Slides, Matthew Brendel, Vanesa Getseva, Majd Al Assaad, Michael Sigouros, Alexandros Sigaras, Troy Kane, Pegah Khosravi, Juan Miguel Mosquera, Olivier Elemento, Iman Hajirasouliha

Publications and Research

Background

Estimating tumor purity is especially important in the age of precision medicine. Purity estimates have been shown to be critical for correction of tumor sequencing results, and higher purity samples allow for more accurate interpretations from next-generation sequencing results. Molecular-based purity estimates using computational approaches require sequencing of tumors, which is both time-consuming and expensive.

Methods

Here we propose an approach, weakly-supervised purity (wsPurity), which can accurately quantify tumor purity within a digitally captured hematoxylin and eosin (H&E) stained histological slide, using several types of cancer from The Cancer Genome Atlas (TCGA) as a proof-of-concept.

Findings

Our model predicts …

Gender Equity Commission Priorities: An Archival Study And Prospects For The Future, Maria D' Agostino, Nicole M. Elias

Publications and Research

This research explores the priorities of the gender equity commission in New York City over forty-five years. Archival commission data was organized thematically to understand the history of gender equity and suggest future possibilities for gender equity beyond New York City. In our historical analysis, we see an expansion of the definition of gender and an adoption of an intersectional approach to gender. We identify four historical gender priorities: sexual harassment and violence, pay equity and economic advancement, health and safety, and gender recognition and celebration. To address systemic issues of gender inequity, we recommend local level administrators embed an …

Imaging Of Transmetallation And Chelation Phenomena Involving Radiological Contrast Agents In Mineral-Rich Fruits, Subhendra Sarkar, Zoya Vinokur, Bleidis Buitrago, Lin Mousa, Hayley Sanchez, Analia Basilicata, Jodi-Ann Douglas, Seanetta Reddock

Publications and Research

Exogenous heavy metals or non-metallic waste products, for example lanthanide or iodinated contrast media for radiological procedures, may interfere with the biochemical pools in patients and in common food sources, creating an excess buildup of exogenous compounds which may reach toxic levels. Although the mechanisms are unknown, our experiments were designed to test if this toxicity can be attributed to “transmetallation” or “chelation” reactions freeing up lanthanides or chelated transition metals in acidic fruits used as phantoms representing the biologically active and mineral-rich carbohydrate matrix. The rapid breakdown of stable contrast agents have been reported at a lower pH. The …

X-Ray And Mri Theory For Mineral-Rich Fruits Affected By Heat Waves During Climate Change, Daler Djuraev, Somdat Kissoon, Robert O'Brien, Nino Jvarishvil

Publications and Research

Conventional Nutrition facts data relays the amounts of different minerals in apples. This information from such analysis may not connect these minerals with their natural biochemical or functional state for health or disease. These minerals may also be part of different particle sizes that affects X-ray transmission, absorption and scatter. The minerals may also redistribute within the tissue over time with or without heat shock. Current Medical Imaging procedures (Xray and MRI) do not localize such minerals. Our work offers a way to increase detection sensitivity of transition metals in various apple varieties non-invasively. This can constitute a step in …

Large-Herbivore Nemabiomes: Patterns Of Parasite Diversity And Sharing, Georgia C. Titcomb, Johan Pansu, Matthew C. Hutchinson, Kaia J. Tombak, Christina B. Hansen, Christopher C. M. Baker, Tyler R. Kartzinel, Robert M. Pringle

Publications and Research

Amidst global shifts in the distribution and abundance of wildlife and livestock, we have only a rudimentary understanding of ungulate parasite communities and parasite-sharing patterns. We used qPCR and DNA metabarcoding of fecal samples to characterize gastrointestinal nematode (Strongylida) community composition and sharing among 17 sympatric species of wild and domestic large mammalian herbivore in central Kenya. We tested a suite of hypothesis-driven predictions about the role of host traits and phylogenetic relatedness in describing parasite infections. Host species identity explained 27 – 53% of individual variation in parasite prevalence, richness, community composition and phylogenetic diversity. Host and parasite phylogenies …

Importin-Mediated Pathological Tau Nuclear Translocation Causes Disruption Of The Nuclear Lamina, Tdp-43 Mislocalization And Cell Death, Robert F. Candia, Leah S. Cohen, Viktoriya Morozova, Christopher Corbo, Alejandra D. Alonso May 2022

Importin-Mediated Pathological Tau Nuclear Translocation Causes Disruption Of The Nuclear Lamina, Tdp-43 Mislocalization And Cell Death, Robert F. Candia, Leah S. Cohen, Viktoriya Morozova, Christopher Corbo, Alejandra D. Alonso

Publications and Research

Tau is a cytosolic protein that has also been observed in the nucleus, where it has multiple proposed functions that are regulated by phosphorylation. However, the mechanism underlying the nuclear import of tau is unclear, as is the contribution of nuclear tau to the pathology of tauopathies. We have previously generated a pathological form of tau, PH-tau (pseudophosphorylation mutants S199E, T212E, T231E, and S262E) that mimics AD pathological behavior in cells, Drosophila, and a mouse model. Here, we demonstrated that PH-tau translocates into the nucleus of transiently transfected HEK-293 cells, but wildtype tau does not. We identified a putative …

Exploration Of Chemical Space With Partial Labeled Noisy Student Self‑Training And Self‑Supervised Graph Embedding, Yang Liu, Hansaim Lim, Lei Xie

Publications and Research

Background Drug discovery is time-consuming and costly. Machine learning, especially deep learning, shows great potential in quantitative structure–activity relationship (QSAR) modeling to accelerate drug discovery process and reduce its cost. A big challenge in developing robust and generalizable deep learning models for QSAR is the lack of a large amount of data with high-quality and balanced labels. To address this challenge, we developed a self-training method, Partially LAbeled Noisy Student (PLANS), and a novel self-supervised graph embedding, Graph-Isomorphism-Network Fingerprint (GINFP), for chemical compounds representations with substructure information using unlabeled data. The representations can be used for predicting chemical properties such …

A Role For Myosin Ii Clusters And Membrane Energy In Cortex Rupture For Dictyostelium Discoideum, Emmanuel Asante-Asamani, Daniel Grange, Devarshi Rawal, Zully Santiago, John Loustau, Derrick Brazill Apr 2022

A Role For Myosin Ii Clusters And Membrane Energy In Cortex Rupture For Dictyostelium Discoideum, Emmanuel Asante-Asamani, Daniel Grange, Devarshi Rawal, Zully Santiago, John Loustau, Derrick Brazill

Publications and Research

Blebs, pressure driven protrusions of the cell membrane, facilitate the movement of eukaryotic cells such as the soil amoeba Dictyostelium discoideum , white blood cells and cancer cells. Blebs initiate when the cell membrane separates from the underlying cortex. A local rupture of the cortex, has been suggested as a mechanism by which blebs are initiated. However, much clarity is still needed about how cells inherently regulate rupture of the cor- tex in locations where blebs are expected to form. In this work, we examine the role of mem- brane energy and the motor protein myosin II (myosin) in facilitating …

Eukaryotic Initiation Factor 4f Promotes A Reorientation Of Eukaryotic Initiation Factor 3 Binding On The 5 And The 3 Utrs Of Barley Yellow Dwarf Virus Mrna, Paul Powell, Usha Bhardwaj, Dixie Goss

Publications and Research

Viral mRNAs that lack a 5 m 7 GTP cap and a 3 poly-A tail rely on structural elements in their untranslated regions (UTRs) to form unique RNA-protein complexes that regulate viral translation. Recent studies of the barley yellow dwarf virus (BYDV) have revealed eukaryotic initiation factor 3 (eIF3) plays a significant role in facilitating communication be- tween its 5 and 3 UTRs by binding both UTRs simultaneously. This report uses in vitro translation assays, fluorescence anisotropy binding assays, and selective 2 -hydroxyl acylation analyzed by primer ex-tension (SHAPE) footprinting to identify secondary structures that are selectively interacting with eIF3. …

Acknowledging The Relevance Of Elephant Sensory Perception To Human–Elephant Conflict Mitigation, Robbie Ball, Sarah L. Jacobson, Matthew S. Rudolph, Miranda Trapani, Joshua M. Plotnik Apr 2022

Acknowledging The Relevance Of Elephant Sensory Perception To Human–Elephant Conflict Mitigation, Robbie Ball, Sarah L. Jacobson, Matthew S. Rudolph, Miranda Trapani, Joshua M. Plotnik

Publications and Research

Elephants are well known for their socio-cognitive abilities and capacity for multi-modal sensory perception and communication. Their highly developed olfactory and acoustic senses provide them with a unique non-visual perspective of their physical and social worlds. The use of these complex sensory signals is important not only for communication between conspecifics, but also for decisions about foraging and navigation. These decisions have grown increasingly risky given the exponential increase in unpredictable anthropogenic change in elephants’ natural habitats. Risk taking often develops from the overlap of human and elephant habitat in Asian and African range countries, where elephants forage for food …

Sources And Types Of Social Supports And Their Association With Mental Health Symptoms And Life Satisfaction Among Young Adults With A History Of Out-Of-Home Care, Rhiannon Evans, Colleen C. Katz, Anthony Fulginiti, Heather Taussig

Publications and Research

Young adults with a history of out-of-home care report poorer mental health and life satisfaction compared to non-care-experienced peers. Social support is a known protective factor for mental health. There is limited evidence, however, on the relationship between sources (e.g., family members) and types (e.g., information) of social support and mental health symptoms and life satisfaction in this population. Reporting cross-sectional survey data from 215 young adults aged 18–22 years with a history of out-of-home care, the current study conducted descriptive, bivariate, and linear regression analysis to examine the different sources and types of support young adults receive and their …

For grazing herbivores, dung density in feeding areas is an important determinant of exposure risk to fecal- orally transmitted parasites. When host species share the same parasite species, a nonrandom distribution of their cumulative dung density and/or nonrandom ranging and feeding behavior may skew exposure risk and the relative selection pressure parasites impose on each host. The arid-adapted Grevy's zebra ( Equus grevyi ) can range more widely than the water-dependent plains zebra ( Equus quagga ), with which it shares the same species of gastrointestinal nematodes. We studied how the spatial distribution of zebra dung relates to ranging and …

Motivation Drug discovery has witnessed intensive exploration of predictive modeling of drug–target physical interactions over two decades. However, a critical knowledge gap needs to be filled for correlating drug–target interactions with clinical outcomes: predicting genome-wide receptor activities or function selectivity, especially agonist versus antagonist, induced by novel chemicals. Two major obstacles compound the difficulty on this task: known data of receptor activity is far too scarce to train a robust model in light of genome-scale applications, and real-world applications need to deploy a model on data from various shifted distributions.

Results To address these challenges, we have developed an end-to-end …

Pathological Human Tau Induces Alterations In The Brain Insulin Signaling Cascade, Abdeslem El Idrissi, Alejandra Del Carmen Alonso

Publications and Research

The process of neurodegeneration in Alzheimer’s disease has been associated with a disruption of insulin signaling cascade in neurons, and to insulin resistance. T2DM correlates with Alzheimer’s disease, but mechanisms of interaction are unknown. We have developed a mouse model of tau induced neurodegeneration expressing pseudo-phosphorylated tau [Pathological Human Tau (PH-Tau)] in neurons. This model (PH-Tau-Tg) recapitulated cognitive decline and neurodegeneration observed in AD. In this study we examined if expression of PH-Tau could affect neuronal excitability and insulin receptor signaling. Neuronal excitability was investigated using intracerebral recordings of extracellular field potentials from prefrontal cortex after insulin and kainic acid …

Integrating Natural Language Processing And Interpretive Thematic Analyses To Gain Human-Centered Design Insights On Hiv Mobile Health: Proof-Of-Concept Analysis, Simone J. Skeen, Stephen Scott Jones, Carolyn Marie Cruse, Keith J. Horvath

Publications and Research

Background: HIV mobile health (mHealth) interventions often incorporate interactive peer-to-peer features. The user-generated content (UGC) created by these features can offer valuable design insights by revealing what topics and life events are most salient for participants, which can serve as targets for subsequent interventions. However, unstructured, textual UGC can be difficult to analyze. Interpretive thematic analyses can preserve rich narratives and latent themes but are labor-intensive and therefore scale poorly. Natural language processing (NLP) methods scale more readily but often produce only coarse descriptive results. Recent calls to advance the field have emphasized the untapped potential of combined NLP …
