Multiple Sclerosis (MS) is a demyelinating disease of the central nervous system with autoimmune etiology. It affects approximately 2.3 million people worldwide, but prevalence is distributed unequally with countries closer to the equator manifesting a lower prevalence of MS. The Italian island of Sardinia is an exception, with prevalence rates that are among the highest in the world. Sardinia is inhabited by a unique, isolated population that was founded approximately 10,000 years ago. The reasons for this enrichment of MS cases in Sardinia are unknown. Fluorescence microscopy offers a non-invasive way to image biological species in real time. The phenomenon of diffraction, however, restricts the resolution of the conventional fluorescent microscopes to submicrometer dimensions in both the horizontal and vertical directions. This limitation can be overcome with the aid of photoactivatable fluorophores. Photoactivatable fluorophores switch from a nonemissive state to an emissive one under irradiation at an activation wavelength and then emit light in form of fluorescence upon illumination at an excitation wavelength. Studies of movement, fundamental to most life on earth, become complicated in the ocean. Many marine organisms have a complex lifecycle with a larval phase, giving them vast dispersal potential from powerful ocean currents while obscuring direct observation by virtue of their minute size. These research challenges are beginning to be circumvented through biophysical modeling of larval transport. Modeling simulates the potential pathways of larvae, but not fine-scale spatial movements. Estrogen receptor negative (ER-) breast cancer is more aggressive and associated with both shorter disease-free and overall survival than ER positive breast cancer. Anti-estrogen therapies are not effective in ER- breast cancers, thus identifying mechanisms underlying lack of ER expression in ER- breast cancers is imperative. We have previously demonstrated that hyperactivation of MAPK (hMAPK) downstream of overexpressed EGFR or overexpression/amplification of Her2 represses ER protein and mRNA expression. Dating violence has been identified as a serious public health problem among adolescents and emerging adults. Recently, there has been an increase in research focused on identifying risk and protective factors for dating violence across the individual-, relationship-, community-, and societal-levels, yet studies exploring associations between parent-child relationships and dating violence have been lacking. Gratitude may function to foster human sociality. Exactly how gratitude performs this putative function, however, is a question that has received little scientific attention. Many recent investigations—primarily concerned with the positive emotional outcomes associated with the experience of gratitude—have overlooked the fundamental mechanistic operations that might produce gratitude. My dissertation addresses this oversight by capitalizing on advancements in the evolutionary psychological study of function and internal regulatory variables. Graphene oxide (GO), a novel 2−dimensional carbon based nanomaterial, has shown potential applications in biomedical and biological field, including drug and gene delivery, sensing, and bioimaging. However, one critical question needs to be addressed before any actual application: how does GO interact with biological molecules, such as amino acids, peptides, proteins, and biomembranes? In this study, spectroscopy, microscopy, and surface chemistry were applied to fundamentally understand the nature of interactions between these molecules and GO. Tumor relapse is the primary cause of mortality in patients with hematologic cancers following autologous hematopoietic stem cell transplantation (HSCT). Vaccination early post-HSCT can exploit both the state of lymphopenia and minimal residual disease for the generation of anti-tumor immunity. Here, multiple vaccinations utilizing lymphoma cells engineered to secrete the heat shock protein fusion gp96-Ig within two weeks of T cell replete syngeneic HSCT led to antigen cross-presentation and increased survival of lymphoma bearing mice. Retrotransposons (RTs) comprise approximately half of the human genome and contribute to chromatin structure, regulatory motifs, and protein-coding sequences. Since RT insertions can disrupt functional genetic elements as well as introduce new sequence motifs to a region, they have the potential to effect the function of genes that harbour insertions as well as those nearby. Excitatory amino acid transporters (EAATs) remove glutamate from synapses in a reuptake process driven by the Na+ gradient to prevent glutamate concentrations from reaching neurotoxic levels. Malfunction of this reuptake mechanism is implicated in stroke. Peripheral substituents on cationic porphyrins play a significant role during binding with DNA hosts. Possible applications of these systems in photodynamic therapy as well as in anti-bacterial and anti-cancer therapies motivate the binding studies. For characterizing DNA binding motifs different methods are useful including absorption, emission, and circular dichroism spectroscopies, as well as viscometry and X-ray crystallography. With the classic H2 T4 porphyrin, or 5,10,15,20-tetra(N-methylpyridinium-4-yl)porphyrin, the mode of binding varies with the base composition of the DNA host. Electroosmotic flow with electrokinetic effects is the primary method of fluid handling in micro-total analysis systems. Under external applied electric fields, electrokinetic micro-devices allow for innovative functionality in a wide range of microfluidic applications including sample injection, separation, rapid micromixing, and miniaturized chemical and biochemical analysis and detection. This dissertation focuses on simulations of two electrokinetic phenomena, isotachophoresis (ITP) and electrokinetic instability (EKI). Emerging spin-devices like magnetic tunnel junctions (MTJ's), spin-valves and domain wall magnets (DWM) have opened new avenues for spin-based logic design. This work explored potential computing applications which can exploit such devices for higher energy-efficiency and performance. The proposed applications involve hybrid design schemes, where charge-based devices supplement the spin-devices, to gain large benefits at the system level. However, a lack of evidence-based practices makes matching difficult. Therefore, investigating the pathways that lead to mitochondrial protection following cerebral ischemic injury represents a major strategy in the development of therapeutic approaches. The PKCepsilon enzyme has been shown to translocate to the mitochondria where it is involved in providing global mitochondrial protection against ischemic insults. Research has demonstrated that taking care of a patient with schizophrenia has serious mental health costs to caregivers that include high levels of burden and poorer overall mental health. Thus, it is necessary to pinpoint predictors of caregiver distress and develop strategies to reduce it. We have developed a family focused, culturally-informed treatment for schizophrenia (CIT-S). We examined this intervention and its ability to decrease the maladaptive behaviors, beliefs, and values that we believe contribute to caregiver distress. Particularly, I investigate the effects of transnationality in works that represent queer subjectivities.
