Spatial transcriptomics enables the scoring of Cancer Hallmarks at a high resolution across a tissue biopsy, yet current analyses of these data remain largely descriptive, treating each spot as a static, independent observation. This thesis develops a coupled system of advection-diffusion-reaction partial differential equations to model the hallmark landscape as the steady-state solution of an underlying spatial process, and casts the recovery of its physical parameters as a PDE-constrained inverse problem. The rapid emergence of multidrug-resistant Klebsiella pneumoniae has significantly reduced the effectiveness of conventional antibiotics, highlighting the need for alternative therapeutic strategies. This study employed a comprehensive in silico pipeline to identify antimicrobial peptides (AMPs) targeting the essential DNA replication initiator protein DnaA. A total of 28,361 peptide sequences were collected from publicly available AMP databases and sequentially filtered based on peptide length, net charge, GRAVY score, instability index, antimicrobial activity, toxicity, hemolytic potential, aggregation propensity, sequence similarity and favorable amphipathic properties. The COVID-19 pandemic significantly disrupted children’s developmental environments by limiting access to childcare, healthcare, and opportunities for social interaction, raising concerns about speech and language outcomes in children born during this period. Grounded in Bronfenbrenner’s ecological systems theory, this qualitative study was an exploration of factors associated with speech and language delays and examined parents’ perceptions and experiences in Philadelphia. The qualitative study included a sample of nine parents who cared for children during the COVID-19 pandemic quarantine period. The effects of terahertz electromagnetic stimulation of the DNA molecule can be understood through modeling of the quantum logical mechanics of the DNA molecule.  Coherent conduction of electron spin states along the aromatic nucleotide base pairs and chirality-induced spin selectivity are two characteristics of the DNA molecule that are involved in the precise separation of the double-stranded DNA molecule at a specific location.  Such modeling can inform predictions of DNA strand separation and has significant implications for understanding DNA transcription bubble formation.

Background: OBI-999 is an antibody-drug conjugate consisting of the Globo H-targeting antibody OBI-888 linked to the cytotoxic payload monomethyl auristatin E. In a dose-escalation study, the OBI-999 recommended phase 2 dose (RP2D) was 1.2 mg/kg every 3 weeks. This dose-expansion study evaluated the efficacy and safety of OBI-999 in patients with pancreatic cancer, colorectal cancer (CRC), or other tumor types ("basket cohort") and tumoral Globo H H-scores ≥100 (NCT04084366).

Patients and methods: Patients were treated at RP2D of OBI-999. We assessed response, progression-free survival (PFS), treatment-emergent adverse events (TEAEs), and pharmacokinetics of OBI-999.

Chitin, a highly abundant environmental polysaccharide present in fungi, insects, and mites, is continuously encountered at mucosal barrier surfaces and functions as an immunostimulatory particulate substrate within the respiratory tract. Acidic mammalian chitinase (AMCase; CHIA) is an evolutionarily conserved mammalian enzyme that degrades chitin-containing particles and regulates inflammatory responses at barrier tissues. Breast cancer progression is associated with systemic immune dysregulation characterized by the expansion of immunosuppressive myeloid populations that promote tumor progression, metastasis, and resistance to therapy. However, the mechanisms that sustain pathological myeloid expansion remain incompletely understood. In particular, it remains unknown whether solid tumors functionally reprogram hematopoietic stem and progenitor cells (HSPCs) to drive enhanced myeloid output and if bone-derived signals can regulate tumor-associated hematopoietic remodeling. During early B cell development, the stepwise arrangement of the B cell receptor (BCR) by immunoglobulin (Ig) gene recombination defines developmental stages and coordinates checkpoints for differentiation. Early pre-B cells express an incomplete version of the BCR, referred to as the pre-BCR, which signals clonal expansion and additional Ig rearrangements. These cellular programs are essential for B cell development and are carried out in a highly regulated fashion during the pre-B cell checkpoint. This quantitative study examined the impact of stricter sexual harassment policies with protections against retaliation and stronger supervisory support on the likelihood that female law enforcement officers would report incidents of sexual harassment by male colleagues. The problem addressed was the underreporting of sexual harassment in policing, a critical issue given the negative impact on victims and workplace culture. Coercive control and domestic violence perpetrated by men against women are global social problems sustained by patriarchal norms that normalize gender-based violence. These norms often contribute to victims' maladaptive self-beliefs, leading women to believe they provoked the abuse. When survivors seek legal protection, they may experience revictimization through legal processes that minimize their claims and enable perpetrators to continue exerting power and control through the justice system. Purpose: Upper-limb edema commonly occurs among critically ill patients due to immobility, inflammation, and fluid overload, leading to discomfort, impaired mobility, and higher pressure injury risk. Based on lymphatic physiology and prior evidence, this quality improvement (QI) project aimed to reduce edema severity through a structured nurse-led lymphatic massage intervention in the medical intensive care unit (MICU) of a northern Taiwan medical center.

Objective: This study empirically investigated the drivers of patient-provider conflict and tested a conceptual model linking treatment expectations, communication, trust, and organizational constraints to conflict intensity in Nigerian tertiary care. Methods: A cross-sectional survey of 400 patients (response rate 82.3%) and 150 healthcare providers (response rate 71.2%) from three Nigerian tertiary hospitals was conducted using stratified random sampling. Validated, culturally adapted scales measured unrealistic treatment expectations, communication barriers, organizational constraints, trust, and conflict intensity. Data were analyzed using Structural Equation Modeling (SEM) with maximum likelihood estimation and bootstrapping for mediation testing. While free clinics provide essential healthcare to socioeconomically disadvantaged populations, patients' lived experiences in these settings remain underexplored. This qualitative study examined how uninsured patients at a U.S.-based free clinic perceive their illness experiences and interactions with healthcare providers. From October to December 2024, semi-structured interviews were conducted with 21 adult patients (ages 18–72; M = 49.14), most of whom were non–U.S.-born and primarily Spanish-speaking. Participants described illness as entangled with emotional distress, social isolation, stigma, and disruptions to identity and family roles—revealing a psychological burden that extends beyond physical symptoms. The reproductive anatomy of rare cervid species remains insufficiently documented, limiting comparative studies and preservation efforts. This study describes the anatomical details in the female reproductive organs of Thamin Eld’s deer (Rucervus eldii thamin) and Fea’s muntjac (Muntiacus feae), while the application of a modified glycerin impregnation technique was achieved for specimen preservation. Dissections of female reproductive tracts revealed morphological variations that correlate with the known female reproductive characteristics of domestic and closely related species. Guided by the Dynamic, Relational, Ecological Approach to Media Effects Research (DREAMER) model, this study examined longitudinal associations between MER and executive functioning (EF) in early childhood and explored whether respiratory sinus arrhythmia (RSA), a physiological index of regulation, predicted developmental changes. Small non-coding RNAs (sncRNAs) have garnered considerable attention in recent years, following accumulating evidence of their critical roles in many cellular processes. Among sncRNAs, microRNAs (miRNAs) and their isoforms (isomiRs), tRNA-derived fragments (tRFs), rRNA-derived fragments (rRFs), and Y RNA-derived fragments (yRFs) account for more than 95% of all sncRNAs found in cells. Background: Perinatal mood and anxiety disorders (PMADs) affect 20–25% of mothers worldwide and are a leading cause of maternal complications. Stigma, fragmented care, and limited provider training leave families vulnerable, while birth workers offer trusted, continuous support.

Problem: Birth workers' maternal mental health (MMH) training is minimal and often excludes matrescence, the developmental transition to motherhood, limiting their ability to distinguish normal adjustment from clinical PMADs and delaying intervention.
