Obesity is an established risk factor for colorectal cancer (CRC). Numerous studies have linked diets low in fruits and vegetables and decreased physical activity, risk factors for obesity, with increased cancer risk. The purpose of this study was to evaluate the effect of obesity on CRC mortality; the behavioral, socioeconomic and demographic predictors of obesity; and the knowledge attitudes, beliefs and behaviors of a Rhode Island population identified at high risk for both obesity and CRC. Two antimicrobial peptides of marine origin, tachyplesin from the horseshoe crab (Tachyplesus tridentatus) and pleurocidin from the winter flounder (Pseudopleuronectes americanus), were evaluated for potential use in feed-based delivery in oyster aquaculture. Tachyplesin, a beta sheet structure was more effective against a range of gram-positive and gram-negative pathogens, as well as against the protozoan oyster parasite Perkinsus marinus. In addition, tachyplesin's structure made it more resistant to the effects of proteases from the oyster and the parasite. The two peptides also had different effects on the host bacteria of the oyster digestive system, with tachyplesin selectively killing more gram-negatives and …

Deep basins in the Gulf of Maine act as refuge for a large population of resting Calanus finmarchicus during the summer and fall. Dynamics of this population are not well understood, however, and loss terms from advection and starvation may be large. A three part study was therefore undertaken to determine potential diapause duration in C. finmarchicus , monitor changes in the summer-fall population, and to define the relative importance of energetic limitation as a control on C. finmarchicus population dynamics. The present work presents the results of survey research conducted in 23 coastal communities of the Dominican Republic to evaluate the impacts of tourism and also the evaluation of a particular co-management system of a tourism-related activity (whale watching in Samaná Bay). Major findings include that tourism is having a positive impact on rural livelihoods as measured by increased household income, and higher levels of job satisfaction (and in the case of female-headed households, also improved material well-being). Corals and macroalgae often co-occur on temperate reefs, where macroalgae provide a majority of the bio-complexity and small scleractinian corals occur in low abundance. Macroalgae typically outcompete adjacent organisms for resources such as space, light, and nutrients, and one mechanism for this competitive success is abrasion. This study explores factors that may influence the interactions between corals and macroalgae, in particular macroalgal abrasion, on a temperate reef in Narragansett Bay, Rhode Island. This dissertation addresses the variability in settlement behavior, growth and development during metamorphosis of summer flounder. There were three objectives: (1) Expand basic knowledge of hormonal and environmental factors that regulate timing of metamorphosis, (2) develop hormonal and environmental techniques that promote synchronization of metamorphic growth and settling behavior in order to eventually reduce cannibalism in juveniles, and (3) identify stressful culture practices and evaluate their long-term effects on growth in juvenile flounder. The Cra protein is a global regulator of carbon and energy metabolism for glycolysis and gluconeogenesis. The live oral vaccine candidate, Salmonella typhimurium SR-11 Fad, is unable to utilize gluconeogenic substrates as sole carbon sources and is avirulent and protective in BALB/c mice. Furthemore, the era gene is interrupted in this strain of Salmonella. The cra gene was suspected to be regulated by another global regulator, acetyl phosphate. This research project targets the three leading causes of death in the U.S., cardiovascular disease, cancer, and stroke through the successful development and validation of six culturally sensitive Proactive Healthy Lifestyle measures (Stage of Change, Decisional Balance, Self-Efficacy, Skills, Relapse, and Processes of Change) targeting diet, exercise, and smoking. 574 college students were recruited and completed self-report surveys. Data from 418 non-Hispanics was used to develop (N = 209) and validate (N = 209) each of the measures. Data from 155 Hispanics was used to further successfully validate the measures, and explore differences in this group with the non-Hispanic group. …

Interventions to decrease unhealthy behaviors and to increase healthy behaviors are crucial for health promotion and disease prevention. Computerized tailored interventions provide a promising approach for creating positive health behavior change. The Transtheoretical Model (TTM) delineates a way to conceptualize behavior change, provides the foundation for developing assessments of an individual's readiness to change, and is utilized in tailoring interventions for actualizing behavior change. Animals metabolically route specific macronutrients from their diet to build and maintain tissue and researchers generally assume that the isotopic composition of an animal's tissues resembles the isotopic composition of their diet. However, animals regularly change diets and they may route dietary macronutrients differently to specific tissues. Thus, I studied metabolic routing of dietary macronutrients into animal tissue because it is fundamentally important for understanding the nutritional ecology of wild vertebrates and because such knowledge is essential for accurate interpretation of stable isotope patterns in consumers and consumed. This study examined the factors that influence middle level educators to incorporate community service-learning into the curriculum. Eleven participants, each a teacher of middle level students, were interviewed to determine the extent to which they were influenced by intended outcomes for students, the community, and themselves. Findings suggest that participants were influenced to include community service-learning in the curriculum by anticipated benefits to students and community members. Participants hoped that by engaging in community service-learning activities, their students would become better scholars, leaders, citizens, and/or people. E. coli K-12, strain MG1655, is a non-pathogenic laboratory strain of E. coli whose genome has been fully sequenced (1). The lily leaf beetle, Lilioceris lilii (Scopoli), first officially reported in the U.S. in 1992, has become a serious pest of native and cultivated lilies throughout its North American distribution. With no effective North American natural enemies, this exotic insect poses a potential economic threat to horticulturists and an environmental threat to native lilies. This study presents the results of an ongoing classical biological control program initiated against L. lilii in North America. This study explored the relationship between the age of development of 5 locomotor behaviors, the level of infant activity for the locomotor behaviors, and the performance of the sample of infants on standardized indices of infant development (Fagan Test of Infant Intelligence, FTII; Bayley Scales of Infant Development, BSID-II). A total of 157 infants were assessed at 6, 9, and 12 months for their activity levels on the 5 locomotor behaviors of interest, and the age at which each locomotor behavior occurred was obtained through records maintained by each infants' caregiver. The texture quality of canned fruits and vegetables can be severely degraded or softened due to thermal processing. Increased ferulic acid cross-linking and cell-cell adhesion has been associated with increased firmness of texture in fruits and vegetables. In order to test such a theory for maintaining firmness and cross-linkages of ferulic acid after thermal processing, vegetables were pre-treated/infused with peroxidase enzyme and ferulic acid. Carrots, beets and Chinese water chestnuts were chosen for their extreme differences in texture. Various methods for measuring texture were tested. A simple, efficient system for seed surface disinfestation and in vitro germination was developed for the carnivorous pitcher plants Darlingtonia californica and Sarracenia leucophylla. Of the disinfectants tested, hydrogen peroxide or 10% Clorox® were most effective for disinfesting seeds of D. californica, while concentrated sulfuric acid worked best for S. leucophylla. Differences in the effectiveness of sterilants were associated by differences in seed coat morphology. Seeds of D. californica imbibed at 4-7°C in sterile deionized water with surfactant and gibberellin germinated earlier than seeds without exposure to gibberellin. Many health maintenance organizations (HMOs) have implemented programs providing varying degrees of annual drug coverage for Medicare beneficiaries enrolled in their plans. Older adult plan members in an HMO operating in central Massachusetts were able to choose among 3 drug benefit options starting January 1, 1994: full coverage for prescription drugs, a maximum of $1000/year in coverage, or no drug coverage. As such, cost containment policies have been shown to affect prescription drug use and other types of health service utilization. The unintended effects of this policy are important to consider. Habitat enhancement and stock enhancement methods were examined as to their effect on abundance of the lobster, Homarus americanus, in Dutch Harbor in Narragansett Bay, Rhode Island. Six experimental artificial reefs for lobster were established in Dutch Harbor in February 1997. The reefs were constructed identically consisting of two portions made up of cobble and boulders. Adherence to self-management regimens among individuals with type 2 diabetes is a major issue. The purpose of this study was to examine the relationship of adjustment distress and depression, in relation to the Transtheoretical Model constructs of stage of change, processes of change, decisional balance, and self-efficacy for exercise behavior. IPC Synthase has been shown to be an essential membrane bound enzyme in yeast that catalyzes the transfer of PI to ceramide to yield the primary sphingolipid found in yeast, Inositolphosphorylceramide. A biochemical approach was taken to investigate the biological factors that regulate IC synthase activity in order to better understand the sphingolipid synthesis. A highly purified IPC synthase was purified over 1000-fold from the yeast Saccharomyces cerevisiae by a procedure that included Triton X-100 solubilization, as well as a series of ion exchange and hydrophobic column chromatography steps. The present research was designed to advance the theory and science of health behavior change by evaluating one of the most commonly used behavior change theories, the Transtheoretical Model (TTM). Two criteria were selected for evaluation, empirical adequacy (predictive power) and generalizability of the TTM to dietary fat intake. Predictive power was evaluated by testing evidence-based a priori predictions. Generalizability was evaluated by testing a priori predictions founded on smoking data and tested on diet data. Alzheimer's disease (AD) is characterized by the insidious onset of an array of cognitive deficits that progress over time and lead to changes in an individual's ability to even perform simple tasks, such as preparing meals. Recently, researchers have demonstrated that clients with mild AD have preserved procedural memory function despite significant cognitive deficits. Procedural memory is an automatic, unconscious process, requiring practice and repetition to activate the particular structures involved in learning tasks. Enzyme assays were developed for the measurement of β-D-glucuronide (GUS) as a rapid detection method for E. coli in food. A fluorescent assay utilizing 4-methylumbelliferyl-β-D-glucuronide as substrate was optimized and tested with growth cultures of E. coli. A relationship between E. coli and the level of GUS activity was established. A chemiluminescent assay using a 1,2-dioxetene derivative as substrate was compared to the fluorescent assay, and was found to be 10x more sensitive. Anti-E. coli GUS antibodies were covalently immobilized on magnetic beads and used in a 30 min immunocapture method. GUS was recovered from E. coli grown at 37°C …
