1. Regiospecific expression of CYP1A1 and CYP1B1 in human uterus of nonsmokers. The goal of this study was to investigate the expression of these isoforms in different regions of the human uterus. Expression was determined in the endometrium, endocervix and squamous (exocervix) regions from six nonsmoking women by using RT-PCR. The transcripts encoding for CYP1A1 were significantly higher (p < 0.05) in the squamous (exocervix) region compared to other areas. However the expression of CYP1B1 was significantly higher (p < 0.05) in the endometrium. CYP1B1 expression appeared to be extremely low in a woman in the secretory stage of the menstrual phase, relative to the endometrium of the other patients who were all in the proliferative stage at hysterectomy. It appears that variability in the expression of these isoforms may be responsible for the differential susceptibility to cancer in women.;2. Influence of menstrual phase and smoking status on expression of CYP1A1 and CYP1B1 in different regions of the uterus. The goal of this study was to investigate the expression of these isoforms in different regions of the uterus from smokers and nonsmokers. These results indicate that CYP1A1 and CYP1B1 are expressed variably in different regions of the human uterus. The expression of CYP1B1 may be susceptible both to hormonal control and cigarette smoking. The variability in the expression of these isoforms may be responsible for the differential susceptibility to cancer in some women.;3. The effect of menstrual phase on CYP3A expression in human endometrium. The CYP3A family consists of four closely related proteins. Of these, CYP3A3/4 is the most abundant P450 isoform and is responsible for metabolizing estrogen as well as a variety of therapeutic agents, whereas CYP3A7, the fetal liver form, is involved in the biosynthesis of estriol. The goal of this study was to evaluate the expression of these two major CYP3A isoforms in human uterine tissue during the proliferative (PROL) and secretory (SEC) phases of the menstrual cycle. Expression of CYP3A7 in endometrium appeared to be several fold higher (∼10-fold) in the proliferative phase compared to the secretory phase. CYP3A4 expression was comparable between the two phases of the menstrual cycle. The RTPCR results indicated differential expression of CYP3A7 in various phases of the menstrual cycle. These results indicate that CYP3A isoforms may be subject to hormonal regulation during the menstrual cycle in endometrium of premenopausal women. (Abstract shortened by UMI.).

The objective of this study was the design, construction, and preliminary qualification of the Mobile Emissions Measurement System (MEMS) for in-use, on-road testing of heavy-duty diesel-powered vehicles. This included selection of emissions analyzers and sampling system components capable of producing emissions results of similar accuracy to those attained with laboratory-grade instruments. A thorough investigation of commercially available systems, and a literature review, identified no suitable designs. The dissertation explores several issues that arise from the restructuring of the American electricity industry. The first chapter discusses economic reasons for deregulation, describes possible deregulation scenarios, and outlines potential pitfalls of deregulation.;Chapter 2 develops a forecast of electricity prices after the transition to retail competition. The equilibrium prices are found from an n -firm Cournot model of oligopoly with non-uniform marginal costs. It is further shown how the translog cost function can be used in the traditional solution of the Cournot model. During the construction of a segment of Appalachian Corridor H between Buckhannon and Elkins, West Virginia, mine spoils and land use features related to historic mining were disturbed. An assessment of the water quality, benthic macro invertebrates, and fish populations was conducted to ascertain the condition of the water and to assess the biological productivity of Laurel Run, a tributary of the Tygart Valley River which runs parallel to Corridor H. On October 20, 1998 the West Virginia Division of Natural Resources stocked adult and intermediate size hatchery reared largemouth bass (Micropterus salmoides) into two Ohio River embayments in an attempt to supplement existing populations. Prior to stocking each fish was fitted with an anchor tag and a visible implant elastomer mark to help ensure recognition as a hatchery-origin fish. The response of bone to loads in the transverse orientation is not well understood and has implications to the stability of a press-fit hip implant. Total hip arthroplasty is a surgical procedure to replace the hip joint. It involves inserting an implant into the shaft of the femur. This experiment assessed whether FI performance is influenced by prior exposure to a temporal discrimination procedure. Pigeons first were exposed to a two-key free-operant psychophysical procedure for two sessions per day. Each trial lasted 60 s and reinforcement was available according to a variable-interval 60-s schedule for pecking one key (Key 1) during the first part of the interval and pecking another key (Key 2) during the second part of the interval. The purpose of this research was to identify if there was a significant difference between the OSHA recordable incidence rates at a selected steel manufacturer prior to a major work stoppage and subsequent to the work stoppage. Monthly and quarterly time periods were evaluated to determine the level of change that occurred between prior and post work stoppage periods. Raw steel production was also evaluated to determine the effects of the work stoppage on production totals. This study investigated the issues embedded in the transition of home schooled students entering or reentering public school in West Virginia. The research was primarily qualitative, however; quantitative research was also employed. Two rounds of interviews, for a total of 46 interviews, were conducted over an eight month period. The interviews included three families and their children who had been home schooled and were returning to public school. This study examines the influence of the cognitive controls of field independence and field dependence on the use of a multimedia-assisted reading program and reading recall. The participants were 11 university students of intermediate Spanish as a second language. The participants were classified as field dependent or field independent based on GEFT scores. The participants read a passage about Mayan architecture that was converted to a multimedia-assisted format. Participants could access as many or as few of the multimedia aids as they chose. Fifty-five randomly-selected West Virginia county boards of education members, or 20 percent of school board members in the state, participated in a year-long study of school board decision making, specifically identifying factors influencing their decision making in regard to the 441 decisions identified. During the data analysis, four broad decision-making categories emerged: Board Foundational Considerations (17.4 percent), School District Personnel Considerations (20.9 percent), School District Infrastructure Considerations (33.7 percent), and School District Student/Curricular Considerations (28.0 percent). In this dissertation, we used ab-initio full potential linear muffin-tin orbital (FP-LMTO) method to study native defects, chlorine and nitrogen doping, and chromium incorporation issues in ZnSe and ZnMgSe alloys. The method we used and the results we obtained are discussed in details. In chapter 1, an introduction to recent studies in ZnSe was given. The methodology was discussed at chapter 2. Although Cr(VI)-containing compounds have been widely recognized as carcinogens for over 100 years, the mechanisms of their actions are not fully understood. The overall objective of this study is to investigate the role of reactive oxygen species (ROS) in Cr(VI)-induced carcinogenesis, focusing on tumor suppressor protein p53 and regulatory signal transduction pathways. In the present study, the role of ROS in p53 activation was investigated and the following conclusions were obtained: (a) Cr(VI) is able to activate p53 protein; (b) ROS, especially ·OH radical, are responsible for the p53 activation induced by Cr(VI). The river continuum concept (RCC) asserts that the biological community in a stream system is largely regulated by geomorphological factors (Vannote, et al., 1980, 131). The concept states that community structure in natural stream systems is structured in order to capitalize on the most efficient energy utilization throughout. The goal of this thesis research was to show that spatial data do affect the results of the analysis of vegetation-environment relationships. Vegetation-environment relationships were modeled within the Allegheny Mountain section of the Appalachian Plateaus physiographic province in western Maryland. Specifically, this research focused on two state forests, namely Garrett and Potomac State Forests, in Garrett County, Maryland. Despite the reversion of Okinawa to Japan in 1972, thirty-eight U.S. military bases still remain in the prefecture. While the local residents’ entreaties to end their suffering from the consequences of the U.S. military presence remain strong, the U.S. and Japanese governments have adopted measures to maintain the U.S. military foothold. This study examined the viewpoints of six claimsmakers to illustrate an overlooked political process in the perpetuation of the U.S. military presence. In 1992 NASA launched its "faster, better, cheaper" initiative to increase the number of missions while decreasing mission cost and time. Accordingly, NASA began supporting research efforts to replace physically redundant systems on board spacecraft with analytically redundant systems. The sensor failure, detection, identification and accommodation (SFDIA) system eliminates physical redundancy by supplying neural network estimations for the pitch, roll, and yaw rate gyros. A Boolean logic scheme utilizes the estimations to detect and identify gyro sensor failures. Under high temperature and stress (as in the front of the crack tip) oxygen can diffuse to the grain boundaries and react with the constitutive elements of the alloy producing brittle oxide layers, which cause the embrittlement of the material, and a brittle elastic, intergranular failure. Crack growth tests were performed on INCONEL 783 three-point-bend specimens in air and controlled oxygen atmosphere at 650°C and 538°C on two types of specimens, with and without beta-phase. This study was to investigate the English-language needs of 182 second-year Thai undergraduate engineering students in a Thai public university in Thailand for their study in their second-year English program in engineering. A two-part self-assessment questionnaire was constructed based upon the previous research on the English-language needs of English as a Second Language (ESL)/English as a Foreign Language (EFL) students, particularly in Science and Technology in relation to English for Academic Purposes (EAP). School-business partnerships have proliferated in response to public outcry for education reform, yet little evidence exists to support that these collaborations are effective. The purpose of this study is to determine if a significant relationship exists between the factors influencing collaboration and school-business partnership effectiveness in West Virginia. The purpose of this study was to investigate the impact of gender dynamics and culture on interaction in the adult ESOL, from the teachers', students', and observer's points of view. Using an ethnographic methodology, the researcher drew from class observations, videotaped segments of classroom interaction, teachers' self-evaluations of gender equity in instruction, and their responses to gender dynamics scenarios, student responses to classroom photographs, as well as student, teacher, and administrator interviews.
