Computer Modeling Of Dapsone-Mediated Heteroactivation Of Flurbiprofen Metabolism By Cyp2c9, Robyn Renee Ayscue

Graduate Theses, Dissertations, and Problem Reports (ETD)

The occurrence of atypical kinetics in cytochrome P450 reactions can confound in vitro determinations of a drug's kinetic parameters. During drug development, inaccurate kinetic parameter estimates can lead to incorrect decisions about a lead compound's potential for success. It has become widely accepted that in certain CYP subfamilies more than one molecule can occupy the active site simultaneously, in some cases resulting in enhanced substrate turnover (heteroactivation). However, the specific mechanism(s) by which dual-compound binding results in heteroactivation remain unclear. It is known that orientation of the substrate in the active site, as dictated by interactions with active site residues, …

In Vitro Elucidation Of The Metabolic Fate Of The Anticancer Drug Busulfan, Islam Rasem Younis May 2008

In Vitro Elucidation Of The Metabolic Fate Of The Anticancer Drug Busulfan, Islam Rasem Younis

Graduate Theses, Dissertations, and Problem Reports (ETD)

Busulfan is a bifunctional alkylating agent that is used to treat mylogenous leukemia. The major elimination pathway of busulfan is through glutathione-S-transferase (GST) catalyzed conjugation to form glutathione sulfonium conjugate. The aim of this work is to elucidate the novel metabolic pathways of busulfan that may explain its toxicity. The observed data showed that busulfan is not a substrate for CYP450. The sulfonium ion conjugate of busulfan was found to be inactive as it did not inhibit GST in human liver cytosol, did not react with 4-(4-nitrobenzyl)pyridine, did not induce apoptosis in NCI-H460 cells, and was not stable in basic …

De Novo Asymmetric Syntheses Of Biologically Active Natural Product, Haibing Guo

Graduate Theses, Dissertations, and Problem Reports (ETD)

The enantioselective syntheses of daumone and four analogs have been achieved in 7 to 8 steps. This route relies upon a diasteroselective palladium catalyzed glycosylation reaction for the formation of the anomeric bond. The asymmetry of the sugar and aglycone portion of daumone were introduced by Noyori reduction of an acylfuran and a propargyl ketone. A highly diastereoselective epoxidation and reductive ring opening established the desired C-2 and C-4 stereochemistry of daumone.;A highly enantioselective and stereocontrolled approach to D-, L-swainsonine and 8, 8a-epi-D-swainsonine has been developed from achiral furan or furfural. A one-pot hydrogenolysis, followed by an intramolecular reductive amination …

Solutions For 2-Dimensional Stabilized Kuramoto -Sivashinsky System, Maomao Cai Jan 2008

Solutions For 2-Dimensional Stabilized Kuramoto -Sivashinsky System, Maomao Cai

Graduate Theses, Dissertations, and Problem Reports (ETD)

We study a stabilized Kuramoto-Sivanshinsky system in two-dimensional space. A model consists of a mixed Kuramoto-Sivanshinsky-Korteweg-de Vires equation, linearly coupled to an extra linear dissipative equation. The model is proposed to describe the surface waves on multi-layered liquid films. In this work, we investigate the stability of the solution to this system by establishing a priori energy estimate for the linearized problem of this non-linear system. We use linear iteration to prove the local existence of the solution to this system. Based on a weak global priori energy estimate, we further prove the global existence and uniqueness of classical solution …

Understanding The Motivations Of Ghanaian Bureaucrats, Christine Naa Norley Lokko Jan 2008

Understanding The Motivations Of Ghanaian Bureaucrats, Christine Naa Norley Lokko

Graduate Theses, Dissertations, and Problem Reports (ETD)

This study examines the factors that affect the work performance of Ghanaian bureaucrats. Three decades ago Robert Price (1975) examined the impact that kinship ties had on the motivation of Ghanaian bureaucrats. He found that Ghanaian bureaucrats behaved in particularistic fashions as opposed to acting in a universalistic manner, suggesting kinship was a major determinant of bureaucratic routine. This study followed the work of Price by replicating his mix of interviews and surveys, with the addition of questions that incorporate recent work on bureaucracy that suggest motivation involves a mix of solidary, functional, and pecuniary preferences (Brehm and Gates, 1997; …

Properties And Function Of Somatostatin-Containing Inhibitory Interneurons In The Somatosensory Cortex Of The Mouse, Yunyong Ma

Graduate Theses, Dissertations, and Problem Reports (ETD)

GABAergic inhibitory interneurons play a pivotal role in balancing neuronal activity in the neocortex. They can be classified into different classes according to their variable morphological, electrophysiological, and neurochemical properties, including two major groups: parvalbumin-containing (PV+), fast-spiking (FS) cells and somatostatin-containing (SOM+) cells. Using transgenic mice, we identified two subgroups, distinct by all criteria, of SOM+ cells in the somatosensory (barrel) cortex of the mouse, one (called X94) in layer 4 and 5B, and the other one (X98) in deep layers (Ma et al., 2006). We found that X98 cells were calbindin-expressing (CB+), infragranular, layer 1--targeting "Martinotti" cells, and had …

Study Of The Variations In Continuous Diesel Particulate Matter Size Measurements And Effect Of Fuel Properties On Dpm Size, Kevin Anthony Chappell Flaim

Graduate Theses, Dissertations, and Problem Reports (ETD)

Heavy-duty diesel engines (HDDE), because of their widespread use and reputation of expelling excessive soot, have frequently been held responsible for superfluous amounts of overall environmental particulate matter (PM). PM is a considerable contributor to air pollution, and a subject of primary concern to health and regulatory agencies worldwide. The U.S. Environmental Protection Agency (EPA) has provided PM emissions regulations and standards of measurement techniques since the 1980's. PM standards set forth by the EPA for HDDEs are based only on total mass, instead of size and/or concentration. The European Union is considering regulating particle size, and it may influence …

Cognitions Of The Community: The Worldview Of U.S. Intelligence, David Alex Mastro Ii

Graduate Theses, Dissertations, and Problem Reports (ETD)

This dissertation attempts to determine what the worldview of U.S. intelligence looks like so that it can be incorporated into America's intelligence identity. I argue that this is necessary to gain a better understanding of why the agencies that comprise the U.S. Intelligence Community (IC) share several behaviors despite having different interests and preferences. To answer my research question, which asks what the worldview of U.S. intelligence is, I conceptualize the IC's core belief system about world politics (its worldview) as the five philosophical beliefs in an operational code. I hypothesize about how the IC views each of these cognitions …

Graduate Theses, Dissertations, and Problem Reports (ETD)

The purpose of this study was to test this hypothesis examining the in vivo effects of ultrafine and fine particles on certain parameters of pulmonary damage and inflammation in the rat model. To do this each treatment group received a dosage based on mass particles. This dosage was then expressed as surface area of the particles (instead of mass) to determine if surface area rather than mass is the more appropriate metric of dose for pulmonary toxicity studies. To assess inflammation, BAL was conducted harvested and PMNs were counted as an indicator of pulmonary inflammation. LDH activity was also measured …

The Effect Of Follicular Aging On Gene Expression In Oocytes And Granulosal Cells., Brandon Michael Lingenfelter Jan 2008

The Effect Of Follicular Aging On Gene Expression In Oocytes And Granulosal Cells., Brandon Michael Lingenfelter

Graduate Theses, Dissertations, and Problem Reports (ETD)

Mammalian oogenesis usually occurs concomitantly with folliculogenesis, coordinated through paracrine effects and physical contacts. During this time, the oocyte acquires the ability to resume meiosis, undergo fertilization and sustain embryonic development. Synthesis of RNA occurs during oogenesis and is largely responsible for early embryonic development. Any perturbations that delay either activation or ovulation can result in early embryonic loss. Therefore, the interacting effects at the genomic level were investigated using a persistent follicle model. Experiment 1. The present study was conducted to analyze global gene expression profiles in granulosal cells from ovarian follicles in cows. Analysis of the microarray data …
