Tumor progression and developmental morphogenesis require transient or stable Epithelial-Mesenchymal transitions (EMT) to modulate epithelial junctional complexes allowing programmed migration, invasion, and anoikis resistance of cells. Grainyhead-like-2 (GRHL2), an epithelial master-regulatory transcription factor, suppresses EMT and causes a mesenchymal-epithelial transition to the default epithelial phenotype. This body of work investigates the role of GRHL2 in kidney cancer, in kidney development using a kidney specific GRHL2 knockout mouse model, and in tubulogenesis using Madin-Darby Canine Kidney (MDCK) cells. In kidney cancer, GRHL2 was not expressed in the normal kidney proximal tubule epithelial cells which are thought to be the progenitors of …

The reactivity of transition metal complexes is greatly affected by the complex structure and the nature of the coordinated ligands. Ambiphilic ligands, a class of molecule termed a frustrated Lewis pair that possesses non-interacting functional groups of both Lewis acidic and basic character, are of particular interests few reports of their application to organometallic catalysis have emerged and their full potential has not been achieved. New, young drivers are over three times more susceptible to crashes in their first 1,000 miles of on road driving. In order to reduce the number incident rate for teenage drivers, Graduated Drivers Licensing (GDL) programs have been implemented across the country. West Virginia, along with all other states, aims to further improve the effectiveness of this program. Currently, GDL effectiveness is steeply limited by the willingness of teenager drivers and their parents to comply with the laws in place. This research centers on the conceptual development of a fastening system in a functional wearable - fishing footwear - by assimilating utilitarian design values in a product design model. Experiments were conducted to determine the effects of different ingredients and manufacture techniques on the quality of pellets being produced as well as to examine how broiler performance will be affected by the dose of phytase product included in the diet. Chapter 3 explores the effects of a manipulation of either feed ingredient or manufacture technique (referred to as concern areas) on manufacture and pellet quality variables. Automated white blood cell (WBC) counting systems process an extracted whole blood sample and provide a cell count. A step that would not be ideal for onsite screening of individuals in triage or at a security gate. Snapshot Hyper-Spectral imaging systems are capable of capturing several spectral bands simultaneously, offering co-registered images of a target. With appropriate optics, these systems are potentially able to image blood cells in vivo as they flow through a vessel, eliminating the need for a blood draw and sample staining. Reactivity controlled compression ignition (RCCI) is a form of dual-fuel combustion that exploits the reactivity difference between two fuels to control combustion phasing. This combustion approach limits the formation of oxides of nitrogen (NOX) and soot while retaining high thermal efficiencies associated with compression-ignition (CI) engines. Theoretical and applied research has been conducted by researchers at several other institutions detailing RCCI combustion characteristics, exhaust emissions, fuel efficiency, and operability. Little research has been done on the characteristics of surface disturbance between conventional (C) and unconventional (UC) wells in West Virginia. To understand the impacts of petroleum development in West Virginia, a total of 296 conventional wells and 206 unconventional wells were digitized for the purposes of this study. Using a combination of spatial and resource-related data, the total land area impacted, forest fragmentation characteristics, and forest resource impacts were investigated between conventional and unconventional wells. The first goal of this research was to explore the potential value of hardwood-derived carbon materials as fillers for reinforcing polymer materials, and investigate the effect of filler content on various properties of reinforced composites. Three loading levels of biochar particles, 2wt%, 6wt%, and 10wt% (by weight) were added to a 10wt% polyvinyl alcohol (PVA) solution (by weight) and composites were formed via the film-casting method. The morphological, tensile, thermal, and dynamic mechanical properties of PVA/biochar composite films were tested and analyzed. Alternative pre-mRNA splicing provides an important mechanism for generating the diverse array of proteins required to generate complex tissue and cell types from a limited genome. Therefore, the proper regulation of alternative splicing is vital to shape cellular identity and function. As consequence, defects in alternative splicing are associated with disease phenotypes that can range from systemic syndromes to the dysfunction of single cell types. The current study attempted to evaluated the effectiveness of a one-day training workshop used to train bachelor's level mental health staff in Staff-Child Interaction Therapy (SCIT), a newly developed intervention designed to reduce problem behaviors in children 2- to -9 years of age through the use of behavioral management techniques by home-basedtrained staff. A total of 39 therapists, known as Therapeutic Support Staff (TSS), were involved in this study, 20 who received training in SCIT (i.e., experimental group) and 19 who received received training in Compassion Fatigue (i.e., attention -control group). Radioactive Masculinity explores how the Cold War legacy, of nuclear weapons finding resonance in images of white maleness and masculinity, results in anxious hypermasculine performances. These discursive and physical masculine acts contingent on the symbolic and material power of nuclear weapons, I argue, represent radioactive masculinity, a form of hegemonic militarized masculinity, which is intrinsically linked to the concept of nationhood and sovereignty. This idealized masculinity is fluid and cannot be tangibly or materially realized, much like the constantly decaying radioactive bomb on which it is modeled. The Middle Ordovician Point Pleasant Formation has become a major focus in the United States for unconventional, horizontal drilling. It stretches throughout Ohio, Pennsylvania, New York, Kentucky, and West Virginia and is estimated to contain upwards of 38 trillion cubic feet of natural gas and 940 million barrels of oil. Metabolic Syndrome (Mets) is a clustering of cardiovascular risk factors that results in a three-fold increase in cardiovascular disease. MetS is known to be associated with adverse arterial remodeling, endothelial dysfunction, decreased nitric oxide (NO) bioavailability, increased inflammation, and conduit artery dysfunction. This thesis examines the distribution and habitat use of two sympatric sand darter species, the western sand darter (Ammocrypta clara) and the eastern sand darter (A. pellucida). This thesis includes three chapters: (1) an introduction and literature review on the western and eastern sand darters, habitat use, and species distribution models; (2) a substrate selection laboratory experiment; and, (3) an evaluation of sympatric sand darter habitat use at multiple scales in the Elk River. I aim to answer three main questions: After controlling for outside market forces, how did acid rain regulation impact Eastern coal production? LaModel uses a laminated overburden boundary-element model and can not only calculate seam-level stresses and displacements but also surface subsidence for thin tabular deposit such as coal seams. Up to this point, the material property wizards in LaModel were primarily designed for calculating accurate stress redistribution in single and multiple-seam situations and for investigating and optimizing pillar sizes and layouts in relation to overburden, abutment and multiple-seam stresses. Basel-like/ Triple Negative Breast Cancer (BTBC) are a subtype of Breast Cancer that are clinically characterized as an aggressive and highly metastatic disease phenotype and poor outcome for women who are diagnosed with this disease. Patients with BTBCs lack treatment options for target therapeutics that are available for other Breast Cancer subtypes (e.g. tamoxifen and herceptin) and have been shown to have a disproportional mortality rate in minority and in young woman of all races. The purpose of this study was to examine how an elementary literacy coach supported teachers learning to teach reading in ways that meet the needs of their students. Second, I determined how the coaching influenced the ways in which a classroom teacher plans for and carries out reading instruction. Finally, I understood how the educational context influences the nature of the coaching and how classroom teachers learned to teach reading.
