Fundamental cause theory encourages researchers to consider broad social conditions that put people at risk of individual-level health-related risks that can lead to health disparities between social groups. Stigma has recently been proposed as a fundamental cause of health disparities as it influences multiple disease outcomes, affects access to resources, and is consistently related to health inequities across historical and geographical contexts. Minority stress theory describes how sexual minorities endure excess stressors in the form of prejudice and discrimination due to their stigmatized status. Innovations in synthetic organic chemistry such as methodology improvements, new chemical transformations and technology development significantly impact medicinal chemistry and continue to drive the drug discovery process. Here, new cyclization strategies to the pharmaceutically underexplored 3,3-dimethylcyclopentane-fused aromatic rings are described. One approach highlighted oxidative cycloisomerization, intermolecular [2+2+2] cyclotrimerization and 5-exo-dig cyclization providing indanoylindole cannabinoids that possess high affinity for human CB2 receptors at nanomolar concentration and good selectivity index. Another featured a [4+2]-type benzannulation reaction which enabled the 5-step synthesis of illudalic acid — compared to the current available sequences (≥16 steps) — and its equipotent and LAR-selective analogs. …

Cerebral ischemia reperfusion injury, a common type of stroke, is a neurodegenerative disorder which is the leading cause of permanent disability in the United States. The underlying pathophysiology of ischemic stroke involves an occlusion in the vasculature supplying oxygen and glucose to the brain tissue, leading to infarction and eventual necrosis in the brain. MitoNEET is an outer mitochondrial protein that plays a role in the cellular bioenergetics and its redox physiology. The tissue-level response to pathogens involves an intricate series of signal transduction events, influenced by immune and healing mediators that alert the host to danger and eliminate the infection. Disruptions to normal signaling events can compromise the host’s ability to respond and lead to the development of chronic infections that cannot be resolved without clinical intervention. Previous research has demonstrated relationships among mindfulness, self-efficacy, and substance use relapse. Contrasting research has found mindfulness and self-efficacy are not consistently negatively associated with substance use relapse demonstrating a need for further research. An additional factor that has been found to associated with substance use is self-compassion; however, it has yet to be examined in relation to the process of relapse. The current study assessed self-efficacy, self-compassion, and mindfulness and their associations with substance use relapse, with the aim of predicting relapse risk. The USDA - Natural Resources Conservation Service (NRCS) supports private lands conservation across the United States to benefit imperiled focal wildlife species using conservation practices to restore habitat. The development of horizontal drilling for natural gas in the Marcellus Shale in 2004 resulted in rapid exploitation of the play in the Appalachian Basin. This “unconventional” gas well drilling accesses resources deeper beneath the surface and allows for multiple wells to be co-located on a single well pad. This results in fewer, but larger, well pads on the landscape than traditional vertical wells. Pipelines are required to transport the volumes of gas produced by unconventional wells to production facilities and market. In this study, analyses were conducted to compare fillet quality attributes of 4 rainbow trout families (40, 102, 159, and 187) fed fish meal (FM) and a fishmeal free diet (FMF), to two age endpoints, 9 and 11 months.

Previous studies with rainbow trout have evaluated the effect of age, diet, and family on growth performance and fillet quality. However, a comprehensive analysis of variables is lacking for rainbow trout, one of the main aquatic foods for domestic and international consumers.

The recent surge in the analysis of biomolecules has driven the need for robust, versatile, sensitive, and accurate techniques such as tandem mass spectrometry for compound structural elucidation. Many fragmentation techniques have been developed over the years, but each has their own limitations, such as the loss of post-translational modifications (PTMs), inability to fragment singly charged precursor ions or cost prohibitiveness. Corollary discharge (CD) circuits provide critical information about movement and behavior to provide context to sensory processing. However, to date, there has not been a comprehensive study of CD circuits at a single-cell level. In this thesis, I aimed to resolve the connectivity of ascending histaminergic neurons, consisting of two pairs, the mesothoracic pair (MsAHNs) and metathoracic pair (MtAHNs) at a single-cell level and characterize contexts of activation. Using transgenic techniques, connectomics and transcriptomics, we identify neural populations receiving input from the AHNs and neural populations with significant output to the AHNs. The goal of wetland mitigation in the United States is to achieve a no-net-loss of wetlands; however, mitigated wetlands must be monitored to ensure wetland function is comparable to natural wetlands. In this study, relationships between land use practices and freshwater turtle abundance, wetland connectivity, and heavy metal bioaccumulation were investigated. Child-caregiver attachment is an important factor in healthy child development and is often targeted by early intervention programs. To assess the efficacy of these interventions, attachment must be accurately measured across the toddler years in populations referred for treatment of externalizing behavior problems. The Strange Situation Procedure (SSP; Ainsworth et al., 1978) and Attachment Q-Set (AQS; Waters, 1987) are empirically-validated measures of child-caregiver attachment, each with unique strengths and weaknesses. The Ohio River is continuing to recover from a long history of anthropogenic impacts. Blue Catfish were considered locally extirpated from the West Virginia sections and have been reintroduced through fingerling stockings beginning in 2002. Flathead Catfish are naturally sustaining in the pool and combined with Blue Catfish provide two popular recreational fisheries. The Blue Catfish and Flathead Catfish studies shared similar objectives: (1) evaluating size structure, (2) estimating growth rates, and (3) estimating annual mortality. Little is known about how above-ground vegetation may differentially influence the below-ground microbial community structure, abundance, and function. Abundance of soil microbial N cycling genes responsible for nitrification (amoA), denitrification (nirK and nirS), and nitrous oxide reduction (nosZ) may vary with tree species and increasing N availability, and these variables may be used to predict production of NO3- and N2O from soil. Many forest ecosystems have been receiving elevated nitrogen (N) deposition due to human activity. Increased ecosystem N threatens soil fertility, water and air quality, and modifies the soil microbially-mediated N cycle in complex ways. The current study investigates how the abundance of key functional N cycling genes within soil bacteria is altered by N deposition and further mediated by different dominant tree species at the Fernow Experimental Forest, WV. With current pressures to seek out genetic solutions to combat antimicrobial resistance, genetic resistance to the gastrointestinal nematode Haemonchus contortus may reduce need for prophylactic antibiotic use. The Katahdin breed has effectively utilized post-weaning fecal egg count estimated breeding value (PFEC EBV) to reduce parasitism within their flocks, but there still remains significant opportunity for selection within breed. In a previous study, Katahdin sheep sired by low PFEC sires had greater survivability to weaning than lambs sired by high PFEC rams and preweaning was caused by other diseases than parasitism. As a post translational modification protein glycosylation plays a crucial role in protein signaling, binding, kinetics and folding. In disease diagnosis and prognosis, monitoring glycosylation has been identified as a biomarker. Sialylation and sialic acid linkage in N-glycans are markers of cancers including liver, pancreatic and kidney cancer. Quantification of sialic acid linkage is analytically challenging because of the diverse linkages and the presence of heterogenous branching. DuPont Washington Works chemical plant knowingly pumped hundreds of thousands of pounds of toxic C8 waste into local waterways, water tables, and landfills for over 40 years, contaminating the drinking water for more than 100,000 residents in and around Parkersburg, West Virginia. Traditional methods of collecting quantitative petrographic data from thin sections (modal mineralogy, size distribution, shapes, etc.) are time- and labor-intensive, and rarely have sample sizes adequate to statistically describe complex rocks (i.e. volcanic rocks). Although manual counting and measurements are now routinely supplemented by digital image analysis, the majority of quantitative petrographic studies still go through a manual digitization stage where object classes are traced before further analyses. Little is known about differences observed in early immunological signaling events that result in cellular infiltration and Th2 cytokine production that mediate parasite expulsion. Parasite resistant St. Croix (STC) sheep have been used as model for full host protective immunity as they present the ability to respond to larval stages of H. contortus. RNA sequencing analysis of peripheral blood mononuclear cells (PBMC) stimulated with H. contortus larval antigen (HcLA) revealed significant upregulation in NLRP3 and its pathway component TLR4. In order to maintain normal operations and prevent unnecessary morbidity and mortality during times of disease outbreak, institutions find a need to conduct frequent and widespread testing of their constituents, often under significantly limited testing resource constraints. The primary objective of my thesis was to develop and utilize a biocompatible multifunctional trityl spin probe for concurrent measurement of pO2, pHe, and [Pi] in vivo by electron paramagnetic resonance (EPR) spectroscopy (Chapter 2). My first goal was to synthesize the proposed probe we are terming HOPE71. Secondly, HOPE71 was characterized by X-band and L-band EPR spectroscopy. Next, the biocompatibility of HOPE71 was assessed through an albumin binding test, cytotoxicity assays, and in vivo intravenous tolerance. Huntington’s Disease (HD) is a genetic neurodegenerative disease caused by the expansion of polyglutamine (polyQ) domain within the first exon (exon1) of the huntingtin (htt) protein. Due to this mutation within the polyQ domain, htt aggregates into various toxic species such as oligomers, fibrils, and other amorphous aggregates. While the aggregation of htt strongly correlates with polyQ length, other factors, e.g. interaction with membranes or organelles and posttranslational modifications (PTMs), modulate aggregation. The first 17 N-terminal amino acids (Nt17) that precede the polyQ in htt-exon1 enhances aggregation and facilitated binding of htt to membranous organelles, promoting morphological changes and disfunction. …

The Effects Of Dementia Knowledge On Dementia Worry, Attitudes, Social Comfort, Empathic Concern, And Personal Distress, Alexandria Rose Ebert

The present study examined the effects of different types of dementia knowledge on attitudes and affective reactions towards persons with dementia. Work has demonstrated that individuals with higher levels of personhood-based knowledge have lower levels of personal dementia fear and higher levels of social comfort. However, to our knowledge, work has not examined dementia attitudes more broadly or differentiated causal relations among different forms of dementia knowledge and attitude outcomes. Participants (N = 334) aged 19-78 (M = 44.53, SD = 16.57) were randomized into one of five experimental conditions: (1) biomedical-knowledge (BK; read biological and medical facts about …
